Antioxidant effects of the Root of Vitis labrusca Water and Aqua-acupuncture Solution (포도근 열수 추출액 및 약침액이 항산화에 미치는 효과)

  • This study was purposed to investigated the antioxidant effects of water (VLWS) and aqua-acupuncture solution (VLAS) from root of Vitis labrusca. VLWS and VLAS was were assessed to determine the mechanisms of its antioxidant activities. The VLWS and VLAS exhibitde a concentration-dependent inhibition of DPPH free radicals. This VLWS-mediated antioxidant activity was similar to that L-ascorbic acid (vitamin C). In addition, the VLWS and VLAS showed dose-dependent free radical scavenging activity, including superoxide anions and hydroxyl radicals, using different assay systems. The VLWS and VLAS were also found to be strongly inhibited linoleic acid oxidation in a ferric thiocyanate assay (FTC) system. Finally, the VLWS and VLAS significantly prevented Fenton's reagent-induced DNA nicking.

Removal of 2,4-D by an Fe(II)/persulfate/Electrochemical Oxidation Process (Fe(II)/과황산/전기화학적 산화 공정에 의한 2,4-D의 제거)

  • The removal of 2,4-dichlorophenoxyacetic acid (2,4-D) in aqueous solution by coupled electro-oxidation and Fe(II) activated persulfate oxidation process was investigated. The electrochemical oxidation was performed using carbon sheet electrode and persulfate using Fe(II) ion as an activator. The oxidation efficiency was investigated by varying current density (2 - 10 mA/㎠), electrolyte (Na2SO4) concentration (10 - 100 mM), persulfate concentration (5 - 20 mM), and Fe(II) concentration (10 - 20 mM). The 2,4-D removal efficiency was in the order of Fe(II) activated persulfate-assisted electrochemical oxidation (Fe(II)/PS/ECO, 91%) > persulfate-electrochemical oxidation (PS/ECO, 51%) > electro-oxidation (EO, 36%). The persulfate can be activated by electron transfer in PS/ECO system, however, the addition of Fe(II) as an activator enhanced 2,4-D degradation in the Fe(II)/PS/ECO system. The 2,4-D removal efficiency was not affected by the initial pHs (3 - 9). The presence of anions (Cl- and HCO3-) inhibited the 2,4-D removal in Fe(II)/PS/ECO system due to scavenging of sulfate radical. Scavenger experiment using tert-butyl alcohol (TBA) and methanol (MeOH) confirmed that although both sulfate (SO4•-) and hydroxyl (•OH) radicals existed in Fe(II)/PS/ECO system, hydroxyl radical (SO4•-) was the predominant radical.

  • In the photocatalytic degradation of benzene using $TiO_2$ as photocatalyst, anionic effects were investigated. When near UV and visible light was irradiated, the photodegradation of benzene was slightly increased in which $S_2O{_8}^{2-}$ or $NO{_3}^-$ coexisted with $TiO_2$. But $NO{_2}^-$ or $Cl^-$ diminished it remarkably, because these anions scavenged hydroxyl radical. While in the case of UV light irradiation, $S_2O{_8}^{2-}$ and $NO{_3}^-$ enhanced photodegradation of benzene due to photosensitization of these anions, but $NO{_2}^-$ or $Cl^-$ diminished it little.

In-vitro Antithrombosis Activity of Different Parts of Sorbus commixta from Ulleung Island (울릉도 마가목의 다양한 부위의 항혈전 활성)

  • Sorbus commixta, a flowering plant in the Rosaceae family, is native to Japan and Ulleung Island, Korea. This plant is also called maga-mok or mai-mok in Korea because the bud of the stem has a similar shape to the teeth of a horse. In this study, hot water extracts from different parts of S. commixta, such as leaf, stem, and immature and mature fruits, were prepared, and their antithrombosis and antioxidant activities were evaluated. The extraction yield and pH of stem extracts were 3.99% and 5.5, respectively. The stem extracts contained 89.2 mg/g of total polyphenols and 28.3 mg/g of total flavonoids. The hot water extracts prepared from the leaf, stem, immature, and mature fruit of S. commixta exhibited no hemolytic activity against human red blood cells, up to a concentration of 0.5 mg/ml. In an anticoagulation assay, the stem extracts showed strong extension in thrombin, prothrombin, and activated partial thromboplastin times, whereas the other extracts had no anticoagulation activity. In a platelet aggregation inhibitory activity assay, all the extracts tested had no inhibitory activity against human platelets. With regard to antioxidation activity, the stem extracts showed stronger radical scavenging activity and reducing power activity than the other extracts. The calculated RC50s, the concentration required for 50% radical scavenging activity, for DPPH anions, ABTS cations, and nitrite of the crude stem extracts were 119.7, 53.3, and 117.5 μg/ml, respectively, whereas they were 13.7, 5.2, and 14.9 μg/ml for DPPH anions, ABTS cations, and nitrite, respectively, for vitamin C. The results suggest that the stem extracts of S. commixta have strong potential for use as a novel resource for antithrombosis agents.

Effect of Curcuma longa Hot Water Extract on Activity of Neuronal Cells Related to Oxidative Stress (산화적 스트레스와 관련하여 신경세포의 활성에 미치는 강황 열수추출물의 영향에 대한 연구)

  • The aim of this study is to screen a therapeutic agent with a cognitive function. The inhibitory effect of $Curcuma$ $longa$ hot water extract (CLWE) on the angiotension-converting enzyme and acetylcholinesterase derived from rabbit lungs and neural cells (PC12), as well as its antioxidant effect, was investigated in this study. Thus, for the first time, the direct scavenging effect of CLWE on DPPH radicals, superoxide anions, hydroxyl radicals, lipid peroxidation, reducing power, and the protective effect of DNA oxidation related to oxidative stress was evaluated in vitro. In addition, it was observed that CLWE especially exhibited a scavenging effect on reducing power and superoxide anions in this study. CLWE showed a protective effect on DNA oxidation produced by hydroxyl radicals. Furthermore, CLWE inhibited the activity of angiotensin-converting enzymes above 0.25%. Additionally, the extract inhibited oxidative stress and inducible nitric oxide in neuronal cells. Therefore, these results demonstrated that CLWE has antioxidant activity and neuronal cell protective effects, suggesting that it may have great potential as a natural source for human health.

Resistance to Reactive Oxygen Species and Antioxidant Activities of Some Strains of Lactic Acid Bacteria from the Mustard Leaf Kimchi (갓김치에서 분리된 유산균의 활성산소종에 대한 저항성과 항산화 활성)

  • In present study, five strains of Lactobacillus acidophilus GK20, Lactobacillus brevis GK55, Lactobacillus paracasei GK74, Lactobacillus plantarum GK81, and Leuconostoc mesenteroides GK104 isolated from the mustard leaf kimchi were investigated for resistance to reactive oxygen species (ROS) and antioxidant activity. L. acidophilus GK20, L. brevis GK55, L. paracasei GK74, and L. plantarum GK81 were resistant to hydrogen peroxide (0.5 mM), showing a survival rate of 50% or more. In particular, L. acidophilus GK20 and L. paracasei GK74 were the most superoxide anions-resistant and L. paracasei GK74 and L. plantarum GK81 were most likely survive hydroxyl radicals. Meanwhile, the intracellular cell-free extract (ICFE) from L. plantarum GK81 exhibited significantly higher DPPH radical scavenging values ($96.4{\pm}2.8%$) than the intact cells (IC). The ICFE of L. plantarum GK81 showed the highest superoxide radical scavenging ability and chelating activity for $Fe^{2+}$ ions among the 5 lactic acid bacteria (LAB) tested, and IC and ICFE from L. plantarum GK81 demonstrated excellent reducing activity, which was higher than those of BHA and vitamin C as a positive control.

Electrochemical Behavior of Oxovanadium (IV) Complex of Benzohydroxamic Acid (옥소바나듐 (IV) 과 벤조히드로옥사믹산 간에 형성되는 착물의 전기화학적 성질에 관한 연구)

  • The redox properties of benzophydroxamic acid (Hben) and its oxovanadium complex, $VO(Ben)_2$ has been studied by the use of polarograpy and cyclic voltammetry. The radical anions of Hben seem to be generated in acetone. The wave at -0.05V vs. Ag/AgCl electrode might be attributed to the formation of radical anion and the wave at -1.78V vs. Ag/AgCl electrode might be attributed the formation of radical dianion. The $VO(Ben)_2$ exhibits one oxidation wave at + 0.55V and two reduction waves at -0.15V and -1.30V vs. Ag/AgCl electrode; the oxidation is reversible one electron process $(VO(ben)_2 {\rightleftharpoons} VO(ben)^+ + e)$. The reduction wave at -0.15V is quasireversible and is arised from the formation of radical anion,$VO(Ben)_2^-$. The second reduction wave at -1.30V is irreversible and this reduction process produces vanadium(III). This oxygen containing ligand of Hben seems to reduce the stability of + 4 oxidation state of vanadium while the sulfur or nitrogen donor of the ligands stabilize the + 4 oxidation state of vanadium when comparisons are made among several oxovanadium complexes.

  • CNDO/2 calculations on $XC_6H_4CH_2Cl,\;where\;X = H,\;p-CH_3,\;p-NH_2\;and\;p-NO_2$, and on benzyl radicals, cations and anions have been carried out in order to investigate nonlinear Hammett behavior. Main conclusions reached are: 1. Benzyl chloride exhibits borderline behavior due to ${\sigma}-{\pi}$ conjugation between C-Cl bond and the ring-system. 2. The extent of mutual conjugation can be judged by $\pi$-charge and bond alternation and interfrontier level separation narrowing effects. 3.The electron donating para substituent reduces the HOMO AO coefficient of the benzylic carbon, while the electron withdrawing para substituent reduces the LUMO AO coefficient of the benzylic carbon.

Inhibitory Effects of Eel (Anguilla japonica) Extracted Carnosine on Protein Glycation (뱀장어(Anguilla japonica)로부터 추출된 Carnosine의 단백질당화 억제효과)

  • Glycation and oxidation induce formation of carbonyl (CO) groups in proteins, which can be used to develop an index of cellular aging. Methyl glyoxal (MG) and hypochlorite anions are deleterious products of oxygen free-radical reaction. The effects of eel carnosine on protein modification mediated by MG and hypochlorite were studied. MG and hypochlorite induced formation of carbonyl groups with high molecular weight and cross-linked forms of ovalbumin. The presence of eel carnosine effectively inhibited these modifications in a concentration-dependent manner. Imidazole ring in eel carnosine might have a primary role in inhibition of protein glycation. Our data suggests that the eel carnosine may be useful as a "natural" anti-glycating agents.

Combination of Light Emitting Diode at 375 nm and Photo-reactive TiO2 Layer Prepared by Electrostatic Spraying for Sterilization

  • The objective of this work was to increase the efficiency of ultraviolet-light emitting diodes at 375 nm for sterilization. Since $TiO_2$ had antibacterial properties, which were attributed to the appearance of hydroxyl radicals and superoxide radical anions on the surface species under ultra violet radiation at about 387 nm, photo-reactive layers such as Ag-doped $TiO_2$ were coated on aluminum substrates by electrostatic spraying. Crystallinity and surface morphology of the coating layer were examined by X-ray diffraction ${\theta}-2{\theta}$ scan and field emission-scanning electron microscope, respectively. In an antibacterial test, we observed above 99% reduction of Escherichia coli populations on 3 or 5 mol% Ag-doped $TiO_2$ layers after irradiation for 2 hrs at 375 nm, while very low inactivation on bare aluminum substrates occurred after irradiation as the same condition.
