백서 간에서 방사선조사에 의한 Apoptosis와 Cytokine 발현과의 관계 (The Relationship between Radiation-Induced Apoptosis and the Expression of Cytokines in the Rat's Liver)

  • Purpose : 백서의 간에서 나타나는 방사선조사에 의한 조직손상에서 IL-1, IL-4, IL-6, TNF 등의 cyokine과 apoptosis와의 상관관계를 알아보고자 하였다. 대상 및 방법 : 백서의 전신에 8 Gy의 방사선조사 후 각 1일, 3일, 5일, 7일, 14일에 간 조직의 변화를 현미경으로 관찰하고, apoptosis의 발현빈도를 확인하였으며, IL-1, IL-4, IL-6군 및 TNF 등의 cytoklne에 대해서 면역조직화학염색 방법과, 간 조직과 혈액에서의 IL-6의 생물학적 활성도 측정 및 방사선 면역측정 등의 실험을 하였다. 결과 : 방사선조사에 의한 간 손상은 광학현미경 소견상 방사선조사 후 1일부터 관찰되어, 3일과 5일에 염증세포의 침윤과 함께 가장 심하게 나타났고, 7일 이후부터는 섬유화가 시작되었다. Apoptosis의 발현빈도는 광학현미경 소견과 in situ detection method 모두에서 방사선조사 후 1일에 가장 높게 나타났다. 면역조직화학 염색소견상, IL기과 TNF의 발현은 조사 후 1일과 3일에 보였고, IL-4는 모든 군에서 발현되지 않았다. IL순는 방사선조사 후 1일과 3일에 강양성으로 나타난 후 점차 감소하였고, 간 조직과 혈액에서의 IL푼의 생물학적 활성도와 방사선면역측정에서도 방사선조사 후 1일에 가장 높고 이후 점차 감소하였다. 결론 : 간에 방사선조사를 시행한 결과 IL-1, IL-6군 및 TNF와 같은 cytokine의 발현이 증가되었으며, 이들 cylokine의 분비가 apoptosis를 유도함으로써 apoptosls가 방사선조사에 의한 간 손상에 중요한 기전의 하나로 작용할 것으로 생각된다.

알긴산올리고당 처치 마우스의 방사선 유도 IL-6 (Radiation-Induced IL(interleukin)-6 in Mice with Algin-Oligosaccharide Treatment)

  • 본 연구에서는 미역이나 다시마에 많이 분포하면서 항산화작용이 탁월한 알긴산올리고당의 방사선 방어효과를 알아보기 위해 3 Gy 방사선이 전신 1회 조사된 마우스를 가지고 IL-6을 측정하였다. 측정 결과 방사선조사대조군과 비교하여 볼 때 소장과 간 조직 모두 방사선조사 전 7일간 알긴산올리고당의 처치를 시행한 그룹에서 IL-6 생성이 억제됨을 관찰하였다(p < 0.001). 이는 알긴산올리고당이 항산화작용을 통해 방사선이 피폭된 생체조직을 방어함으로써 IL-6의 생성을 억제한 것으로 사료된다. 결론적으로, 본 연구를 통해 알긴산올리고당의 일부 방사선 방어효과를 규명했고 아울러 화학적 독성이 없는 자연산생물이 방사선 방어제로 활용될 수 있을 가능성을 확인하였다.

Isoegomaketone Ameliorates Atopic Dermatitis via MAPK and STAT Pathway-based Pro-Inflammatory Cytokine Regulation

  • Isoegomaketone(IK), isolated from the radiation-induced mutant cultivar of Perilla frutescens var. crispa, is a major phytochemical compound that has attracted attention in pharmacological research. In this study, we demonstrated that IK exerts anti-inflammatory and protective effects on human mast cells and in an atopic dermatitis mouse model. IK inhibited tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6), and IL-8 expression in human mast cells (HMC-1) stimulated with phorbol myristate acetate(PMA) and calcium ionophore A23187 (PMACI). IK significantly reduced the PMACI-induced phosphorylation of ERK and JNK, but not p38. IK also inhibited the PMACI-induced phosphorylation of STAT1 and STAT3. Oral administration of IK in atopic dermatitis mouse model ameliorated skin inflammation severity, as measured by skin thickness and pro-inflammatory cytokine levels such as TNF-α, IL-8, IL-4, and IL-13. These results might suggest that IK is a potent therapeutic agent against skin inflammation and atopic dermatitis.

Effect of quercetin on impaired immune function in mice exposed to irradiation

  • Radiation used in cancer treatment may cause side effects such as inflammation. Quercetin is a polyphenol that reduces inflammation. This study evaluated the recovery efficacy of quercetin on impaired immune function in irradiation-induced inflammatory mice. Quercetin administered at two concentrations of 10 and 40 mg/kg body weight was initiated 2 weeks before irradiation and was continued 30 days after irradiation. The animals exposed/not exposed to radiation were sacrificed on radiation days 10 and 30. Splenocyte proliferation, which was diminished after irradiation, was enhanced significantly by quercetin supplementation after 30 days of irradiation. Cytokine secretion increased in the radiation group compared to that in the non-radiation control group. After 30 days of radiation, interleukin (IL)-$1{\beta}$ and IL-6 secretion decreased significantly in the radiation-quercetin groups. When quercetin was administered for 44 days, it showed a possible protective effect against irradiation-induced inflammation in mice. Quercetin could be beneficial in the recovery of irradiation-induced increases in cytokine secretion.

Anti-inflammatory Activity of an Ethanol Extract of Caesalpinia sappan L. in LPS-induced RAW 264.7 Cells

  • The anti-inflammatory activities of an ethanol extract of Caesalpinia sappan L. (CS) were investigated in LPS-induced RAW 264.7 cells. Result indicated that CS inhibited the LPS-induced NO production in a dose-dependent manner with an $IC_{50}$ of $10.9\;{\mu}g/mL$. In addition, CS attenuated the iNOS mRNA and protein expression by inhibiting NF-${\kappa}B$ activation. CS also suppressed the productions of IL-6 and MCP-1 in a dose-dependent manner, with $IC_{50}$ values of $15.9\;{\mu}g/mL$ and $5.47\;{\mu}g/mL$, respectively. In addition to the anti-inflammatory activities, CS decreased intracellular ROS formation in the same cells. In conclusion, CS inhibited the production of NO, IL-6 and MCP-1 via a suppression of the NF-${\kappa}B$ activation and intracellular ROS generation.

Preliminary Study of Protective Effects of Flavonoids against Radiation-induced Lung Injury in Mice

  • Background: Radiation therapy plays an important role in lung carcinoma treatment. However, the incidence of symptomatic radiation-induced lung injury is high. This study aimed to evaluate radioprotective effects of flavonoids extracted from Astragalus complanatus and mechanisms of action against radiation damage. Methods: Alteration in antioxidant status and levles of several cytokines were investigated in BABL/C mice treated with 4 mg/kg b.wt. flavonoids after exposure to 10Gy thoracic radiation. Results: Serum levels of SOD in the flavonoids+radiation group were significantly higher compared to the radiation control group, while TGF-${\beta}1$ and IL-6 were lower. Mice in the radiation control group displayed more severe lung damage compared with the flavonoids+radiation group. The expression of TGF-${\beta}1$ and TNF-${\alpha}$ in the radiation control group was markedly increased in alveolar epithelial cells and macrophages of the alveolar septum. Conclusions: From the results of the present study, flavonoids could be excellent candidates as protective agents against radiation-induced lung injury.

Radiation Induces Phosphorylation of STAT3 in a Dose- and Time-dependent Manner

  • Background: We have reported the radiation could activate STAT3, which subsequently promotes the invasion of A549 cells. We here explored the dose- and time-response of STAT3 to radiation and the effect of radiation on upstream signaling molecules. Materials and Methods: A549 cells were irradiated with different doses of ${\gamma}$-rays. The expression of and nucleus translocation of p-STAT3 in A549 cells were detected by immunoblotting and immunofluorescence, respectively. The level of phosphorylated EGFR was also assessed by immunoblotting, and IL-6 expression was detected by real time PCR and ELISA. Results: Radiation promoted the phosphorylation of STAT3 at Y705 in a dose- and time-dependent manner and nuclear translocation. The level of phosphorylated EGFR in A549 cells increased after radiation. In additional, the mRNA and protein levels of IL-6 in A549 cells were also up regulated by radiation. Conclusions: STAT3 is activated by radiation in a dose-and time-dependent manner, probably due to radiation-induced activation of EGFR or secretion of IL-6 in A549 cells.

  • Ginsan, a polysaccharide extracted from Panax ginseng. was earlier scrutinized for a biological-response modifier. We further studied the protective and restorative activity of Ginsan against sublethal dose irradiation owing to increase production of endogenous hematopoietic growth factors such as IL-1. Anti-inflammatory Activities of Ethylacetate Extract of Rehmannia glutinosa in LPS-induced RAW 264.7 Cells

  • This study is to investigate the anti-inflammatory effects of the ethylacetate extract of Rehmannia glutinosa (RGEAE). The anti-inflammatory activities using nitric oxide (NO), cytokine, and chemokine production in lipopolysaccharide (LPS)-induced RAW 264.7 cells were checked. Results indicated that RGEAE suppressed the NO, interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1) production in a dose-dependent manner. Inhibition of NO formation was due to a decrease in inducible NOS (iNOS) expression. It was also found that the anti-inflammatory activities of RGEAE resulted from its inhibitory role on the nuclear factor $(NF)-{\kappa}B$ activation and reactive oxygen species (ROS) production. Therefore, it is suggested that RGEAE has potential as a therapeutic material to attenuate the inflammatory disease such as rheumatoid arthritis.

생약복합조성물(HemoHIM)의 사람 비만세포주 활성 억제 효과 (Inhibitory Effects of a Herbal Composition (HemoHIM) on the Activation of Human Mast Cell Line (HMC-1))

  • 방사선에 대한 방호와 면역기능 조절을 목적으로 새로운 생약복합조성물인 HemoHIM을 개발하였다. 식품 원료로 사용 가능한 생약재 3종 당귀, 천궁, 백작약의 에탄올 분획을 열수추출물에 첨가하여 HemoHIM을 제조하였다. HemoHIM의 항알레르기 효과를 검증하기 위하여 사람 비만세포주 HMC-1을 사용해 compound 48/80으로 유도되는 히스타민 분비량과 PMA/A23187로 유도되는 염증성 사이토카인의 분비량을 측정하였다. 히스타민의 양은 형광분석법으로, 염증성 사이토카인 IL-6, IL-8, TNF-$\alpha$, GM-CSF의 양은 효소결합 면역측정법으로 측정하였다. 저농도의 HemoHIM에 의해 히스타민 분비량이 억제되었고, 모든 농도에서 IL-6, TNF-$\alpha$, GM-CSF의 분비량은 억제되었지만 IL-8은 고농도에서만 억제되었다. 사이토카인의 mRNA 발현량은 HemoHIM의 농도 의존적으로 억제되었다. 그리고 c-kit와 Fc$\varepsilon$RI의 mRNA 발현량도 모든 농도에서 억제되었지만, tryptase의 mRNA 발현량은 저 농도에서만 억제되었다. 이상의 결과로 HemoHIM이 비만세포의 활성을 억제하는 효과가 있다는 것을 알 수 있었으며, 상대적으로 독성이 적은 항알레르기 제재로 개발할 가능성을 제시한 것으로 생각된다.
