Rosmarinic Acid Attenuates Cell Damage against UVB Radiation-Induced Oxidative Stress via Enhancing Antioxidant Effects in Human HaCaT Cells

  • This study was designed to investigate the cytoprotective effect of rosmarinic acid (RA) on ultraviolet B (UVB)-induced oxidative stress in HaCaT keratinocytes. RA exerted a significant cytoprotective effect by scavenging intracellular ROS induced by UVB. RA also attenuated UVB-induced oxidative macromolecular damage, including protein carbonyl content, DNA strand breaks, and the level of 8-isoprostane. Furthermore, RA increased the expression and activity of superoxide dismutase, catalase, heme oxygenase-1, and their transcription factor Nrf2, which are decreased by UVB radiation. Collectively, these data indicate that RA can provide substantial cytoprotection against the adverse effects of UVB radiation by modulating cellular antioxidant systems, and has potential to be developed as a medical agent for ROS-induced skin diseases.

  • Astronauts are constantly exposed to space radiation at a low-dose rate during long-tenn stays in space. Therefore, it is important to determine correctly the biological effects of space radiation on human health. Space radiations contain various kinds of different energy particles, especially high linear energy transfer (LET) particles. Therefore, we have to study the relative biological effectiveness (RBE) of space radiation under microgravity environment which may change RBE from a stress for cells. Furthermore, the research about space radiation might give us useful information about birth and evolution of life on the earth. We also can realize the importance of preventing the ozone layer from depletion by use of exposure equipment to sunlight at International Space Station (ISS).

Nondestructive Evaluation of the Characteristics of Degraded Materials Using Backward Radiated Ultrasound

  • The frequency dependency of Rayleigh surface wave is investigated indirectly by measuring the angular dependency of the backward radiation of the incident ultrasonic wave in two kinds of degraded specimens by scuffing or corrosion. Then, the frequency dependency is compared with the residual stress distribution or the corrosion-fatigue characteristics for the scuffed or corroded specimens, respectively. The width of the backward radiation profile increases with the increase of the variation in residual stress distribution for the scuffed specimens. In the corroded specimens, the profile width decreases with the increase of the effective aging layer thickness and is inversely proportional to the exponent, m, in the Paris' law that can predict the crack size increase due to fatigue. The result observed in this study demonstrates high potential of backward radiated ultrasound as a tool for nondestructive evaluation of subsurface gradient of material degradation generated by scuffing or corrosion.

Neurobehavioural Changes and Brain Oxidative Stress Induced by Acute Exposure to GSM900 Mobile Phone Radiations in Zebrafish (Danio rerio)

  • The impact of mobile phone (MP) radiation on the brain is of specific interest to the scientific community and warrants investigations, as MP is held close to the head. Studies on humans and rodents revealed hazards MP radiation associated such as brain tumors, impairment in cognition, hearing etc. Melatonin (MT) is an important modulator of CNS functioning and is a neural antioxidant hormone. Zebrafish has emerged as a popular model organism for CNS studies. Herein, we evaluated the impact of GSM900MP (GSM900MP) radiation exposure daily for 1 hr for 14 days with the SAR of 1.34W/Kg on neurobehavioral and oxidative stress parameters in zebrafish. Our study revealed that, GSM900MP radiation exposure, significantly decreased time spent near social stimulus zone and increased total distance travelled, in social interaction test. In the novel tank dive test, the GSM900MP radiation exposure elicited anxiety as revealed by significantly increased time spent in bottom half; freezing bouts and duration and decreased distance travelled, average velocity, and number of entries to upper half of the tank. Exposed zebrafish spent less time in the novel arm of the Y-Maze, corroborating significant impairment in learning as compared to the control group. Exposure decreased superoxide dismutase (SOD), catalase (CAT) activities whereas, increased levels of reduced glutathione (GSH) and lipid peroxidation (LPO) was encountered showing compromised antioxidant defense. Treatment with MT significantly reversed the above neurobehavioral and oxidative derangements induced by GSM900MP radiation exposure. This study traced GSM900MP radiation exposure induced neurobehavioral aberrations and alterations in brain oxidative status. Furthermore, MT proved to be a promising therapeutic candidate in ameliorating such outcomes in zebrafish.

Radioprotective Effect of Extracts from Plants Indigenous to Korea

  • We have screened the cytoprotective effect on $\gamma$-ray radiation induced oxidative stress from eighteen Korean plant extracts. Querus salicina, Clerodendron trichotomum, Lamium amplexicaule, Lozoste lancifolia and Malus baccata were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of these plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by $\gamma$-ray radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Quercus salicina, Clerodendron trichotomum, Lamium amplexicaule, Lozoste lancifolia and Malus baccata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

살리실산이 오이 잎의 산화적 스트레스와 UV-B 내성에 미치는 영향 (Effects of Salicylic Acid on Oxidative Stress and UV-B Tolerance in Cucumber Leaves)

  • The effect of salicylic acid(SA) on antioxidant system and protective mechanisms against UV-B induced oxidative stress was investigated in cucumber(Cucumis sativus L.) leaves. UV-B radiation and SA were applied separately or in combination to first leaves of cucumber seedlings, and dry matter accumulation, lipid peroxidation and activities of antioxidant enzymes were measured in both dose and time-dependant manner. UV-B exposure showed reduced levels of fresh weight and dry matter production, whereas SA treatment significantly increased them. SA noticeably recovered the UV-B induced inhibition of biomass production. UV-B stress also affected lipid peroxidation and antioxidant enzyme defense system. Malondialdehyde(MDA), a product of lipid peroxidation, was greatly increased under UV-B stress, showing a significant enhancement of a secondary metabolites, which may have antioxidative properties in cucumber leaves exposed to UV-B radiation. Combined application of UV-B and SA caused a moderate increase in lipid peroxidation. These results suggest that SA may mediate protection against oxidative stress. UV-B exposure significantly increased SOD, APX, and GR activity compared with untreated control plants. Those plants treated with 1.0 mM SA showed a similar pattern of changes in activities of antioxidant enzymes. SA-mediated induction of antioxidant enzyme activity may involve a protective accumulation of $H_2O_2$ against UV-B stress. Moreover, their activities were stimulated with a greater increase by UV-B+SA treatment. The UV-B+SA plants always presented higher values than UV-B and SA plants, considering the adverse effects of UV-B on the antioxidant cell system. ABA and JA, second messengers in signaling in response to stresses, showed similar mode of action in UV-B stress, supporting that they may be important in acquired stress tolerance. Based on these results, it can be suggested that SA may participates in the induction of protective mechanisms involved in tolerance to UV-B induced oxidative stress.

  • Floating breakwaters have been recently studied to reduce the transmission ratio of wave energy. The numerical study shows how wave pressure and stress act on the rectangular floating breakwater under various regular wave conditions. In order to evaluate hydrodynamic pressure on the floating breakwater, the infinite element is applied to the linear wave diffraction and radiation problems. SAP2000, a structural analysis program, is used to evaluate stress on the floating breakwater.
