니겔라 사티바 오일의 미백 효능에 관한 연구 (Effect of Nigella sativa Oil on Melanogenesis)

  • 니겔라 사티바(Nigella sativa Linn.) 오일의 미백 효능을 확인하기 위하여 니겔라 사티바 오일 및 오일에서 분리된 유효성분들을 버섯 타이로시네이즈 효소, B16 멜라노마 세포를 이용하여 멜라닌 생성에 관련된 다양한 실험을 실시하였다. B16-F10 멜라노마 세포를 이용한 멜라닌 저해 활성시험 결과에서 니겔라 사티바 오일은 10 mg/mL의 농도에서 약 86 %의 멜라닌 생성을 억제하였으며, RT-PCR과 Western blot을 통한 멜라닌 생성 기작에 대한 영향을 조사한결과, 멜라닌 합성의 주요 단백질인 타이로시네이즈 발현 저해효과가 우수하게 나타났다. 또한, tyrosinase related protein-1 (TRP-1) 및 tyrosinase related protein-2 (TRP-2)의 발현이 억제되는 것을 확인하였다. 따라서, 니겔라 사티바오일은 멜라닌 생합성 저해 효과뿐만 아니라, 멜라닌 합성에 필수적인 효소(타이로시네이즈, TRP-1, TRP-2)의 발현저해를 통해 미백 효과를 나타내는 것으로 확인되었으며, 이에 따라 니겔라 사티바 오일은 멜라닌 생합성을 저해하는미백 소재로 활용할 수 있을 것으로 사료된다.

  • The present study was conducted to evaluate the immunomodulatory effects of Dangguijakyak-san(당귀작약산). We investigated the effects of cell proliferation in mouse spleen cell and RAW 264.7 macrophages cells. Dangguijakyak-san enhanced mitogenic activity in the dose-response manner in mouse spleen cells and RAW 264.7 macrophages cells. In nitric oxide (NO) synthesis and iNOS mRNA expression by Dangguijakyak-san, Dangguijakyaksan alone had an effect on NO synthesis and iNOS mRNA expression in RAW 264.7 cells. NO production and iNOS mRNA expression which is excessively induced by LPS decreased after treatment of Dangguijakyak-san. The expressions of cytokine gene by Dangguijakyak-san investigated using reverse transcription polymerase chain reaction (RT-PCR). In RT-PCR, IL-1α, IL-1β and IL-6 mRNA expressions induced in Dangguijakyak-san-treated RAW 264.7 cells. These data indicate that 1) Dangguijakyak-san can modulate various immune response and 2) the immunomodulatory effects of Dangguijakyak-san may be, in part, associated with the regulation of NO synthesis, the expressions of these cytokine as well as the mitogenic effect on spleen cells and macrophages cells.

Effects of Liriope Platyphylla on LPS-stimulated Expression of COX-2 and iNOS in Mouse BV2 Microglial Cells

  • Objective: In this study, the effects of Liriope Platyphylla against LPS-induced inflammation was investigated. Methods: Cell viability was determined using the MTT assay. To identify expressions of COX-2 and iNOS mRNA, RT-PCR was performed. Assessment of PGE2 synthesis was performed using the PGE2 immunoassay. Measurement of NO synthesis was performed using the NO detection. Result : The MTT assay revealed that Liriope Platyphylla exerted no significant cytotoxicity in the microglial BV2 cells. RT-PCR analysis revealed that the mRNA levels of COX-2 and iNOS were significantly decreased in the LPS- and 5 mg/ml Liriope Platyphylla treated group. From the PGE2 immunoassay and NO detection, PGE2 and NO synthesis was significantly suppressed in the LPS- and 5 mg/ml Liriope Platyphylla treated group. Conclusion : In these study, Liriope Platyphylla was shown to suppress PGE2 and NO production by inhibiting LPS-stimulated enhancement of COX-2 enzyme activity and iNOS expression. It is very possible that Liriope Platyphylla can offer a valuable mode of therapy for the treatment of brain inflammatory diseases.

  • 피부에서 melanin은 자외선 차단의 주요한 역할을 한다. Tyrosinase는 멜라닌 생합성과정에서 초기 단계에 관여하는 중요한 효소로서 이것의 조절을 통한 피부 멜라닌화 억제에 관해 많은 연구가 되어져왔다. 본 연구에서는 해녀콩 추출물에서 mushroom tyrosinase 활성억제, B16F10 melanoma 세포를 이용한 dopa oxidase 활성억제 및 멜라닌 합성 억제 효과를 확인하였다. Tyrosinase mRNA 발현에서의 억제 효과를 확인하기 위하여 RT-PCR을 이용하였으며, $CHCI_3$ 층에서 분리해낸 A 분획에서 tyrosinase mRNA 발현을 억제시킴을 확인하였다.

  • The reverse transriptase (RT) of HIV-1 is a proven target for inhibition of HIV-1 replication. Many nonnucleoside RT inhibitors (NNRTls) are in development stage for the clinical use: Among them. trovirdine (PETT). (thiophene) ethylpyridylthioureas (TET). and phenylethylpyridylureas (urea-PETT) are simple and flexible arylalkylarylureas. DNA·RNA Heteroduplex Containing 8-Oxo-7,8-dihydroguanosine: Base Pairing, Structures, and Thermodynamic Stability

  • Oligoribonucleotides containing 8-oxo-7,8-dihydroguanosine (8-oxoG) and 8-oxo-7,8-dihydro-2'-O-methylguanosine (8-oxoG-Me) were synthesized. The base pairing properties of 8-oxoG and 8-oxoG-Me in oligoribonucleotide in cDNA synthesis by reverse transcriptases were studied. dCMP was preferentially incorporated into the site opposite 8-oxoG or 8-oxoG-Me than into other dNMPs. TMP and dCMP were inserted preferentially into sites opposite 8-oxoG or 8-oxoG by reverse transcriptases. HIV-RT did not incorporate TMP, but RAV2-RT incorporated 50% more TMP than dCMP into the site opposite 8-oxoG. In the site opposite 8-oxoG-Me TMP was substantially incorporated by HIV-RT or RAV2-RT. Thermodynamic analysis of the DNA. RNA heteroduplex containing 8-oxoG revealed that 8-oxoG and 8-oxoG-Me formed base pairs with cytidine and thymidine with similar stability. The thermodynamic parameter (${\Delta}G^{\circ}$) demonstrated that the formation of duplexes between 8-oxoG or 8-oxoG-Me and cytidine or thymidine is more thermodynamically favorable than with adenosine and guanosine. However, differences in the melting temperature and ${\Delta}G^{\circ}$'s of 8-oxoG/dC and 8-oxoG/T were much smaller than between G/dC and G/T. CD spectra showed that DNA . RNA containing 8-oxoG or 8-oxoG-Me duplexes showed similarities between the A-type RNA and B-type DNA conformations.

최적 모듈 선택 아키텍쳐 합성을 위한 전력 감소 Force-Directed 스케쥴링 (Low Power Force-Directed scheduling for Optimal module selection Architecture Synthesis)

  • 본 논문은 최적 모듈 선택 아키텍쳐 합성을 위한 천력 감소 Force-directed 스케줄링을 제안한다. 제안한 전력 강소 스케줄링은 행위 수준 언어를 업력으로 스위칭 활동-(switching activity) 을 고려하여 기존의 FDS 스케쥴링을 저 전력으로 고려한 FDS_LP 앙고리듬을 수행한다. 제안한 FDSL LP 알고리듬은 스위칭 활동을 최소로 하는 동적 파워를 포스 개념에 적용하여 전력 감소를 수행한다. 모듈 선택에서는 전력, 면적, 지연의 매개 변수를 고려하여 최척 모율 성택 RT 라이브러리를 구축한다. 구축한 RT 라이브러리에서 최적 파라메터를 구하기 위해서 프렌치 앤드 바운드 방법을 사용한 최걱 요율 선택 방법을 제안한다. 비교 실험에서는 최적 모율 선택을 고려한 제안한 FDS LP 앙고리듬과 기존의 FDS 알고리듬간의 전력 차이를 비교하여 최대 23.9 % 까지 전력 감소를 얻을 수 있다.

1-Benzyl-3-butylimidazolium Hydroxide 이온성액체 합성 및 전해질 특성 조사 (Synthesis and Electrolyte Characterization of 1-Benzyl-3-butylimidazolium Hydroxide Ionic Liquid)

  • 본 논문에서는 수산화기를 음이온으로 이미다졸리움, 즉 1-benzyl-3-butylimidazolium [BzBIM]을 양이온으로 구성한 친수성의 알칼라인 이온성 액체 전해질을 합성하였다. 합성한 이온성 액체의 전기화학적, 물리적 및 구조적 특성을 순환전압전류법, 이온전도도, 점도, 열중량분석기, 시차 주사 열량 측정법, FT-IR과 1H-NMR을 이용하여 측정하였다. 합성된 이온성 액체는 0.1 M KCl 전해질과 유사한 높은 이온전도도와 낮은 점도를 나타내었으며, 또한 약 4.4 V 이상의 전위창을 나타내었다. 따라서 상기 이온성 액체는 대체 전해질로 다양한 에너지 및 환경 응용분야에 활용될 수 있을 것으로 전망된다.

  • Alpha-viniferin was isolated from Carex humilis (Cyperaceae), and showed anti-inflammatory effects on carrageenin or histamine-induced paw edema in mice. To understand mode of the anti-inflammatory action. effects of alpha-viniferin on cyclooxygenase (COX)-2, iNOS, oxygen radicals and proinflammatory cytokines have been analyzed. Alpha-viniferin showed selective inhibitory effect with an IC50 value of 5 $\mu\textrm{m}$ on COX-2 activity but showed weak inhibitory effect on the synthesis of COX-2 transcript which was identified by RT-PCR.   • In this paper, we present a high-level synthesis system that generates area-efficient RT-level datapaths with multiport memories. The proposed scheduling algorithm assigns an operation to a specific control step such that maximal sharing of functional units can be achieved with minimal number of memory ports, while satisfying given constraints. We propose a measure of multiport memory cost, MAV (Multiple Access Variable) which is defined as a variable accessed at several control steps , and overall memory cost is reduced by equally distributing the MAVs throughout all the control steps. Experimental results show the effectiveness of the proposed algorithm. When compared with previous approaches for several benchmarks available from literature, the proposed algorithm generates the datapaths with less memory modules and interconnection structures by reflecting the memory cost in the scheduling process.
