해당화(Rosa rugosa) 잎의 항산화물질 (Antioxidants in Leaves of Rosa rugosa)

  • To search for useful antioxidants from plant materials, we investigated the antioxidative activity in the methanol extracts of 30 Chinese medicinal plants using DPPH method. The highest activity $(RC_{50}:\;12\;{\mu}g)$ was showed in the methanol extract of Rosa rugosa, followed by Potentilla fruticosa $(14\;{\mu}g)$, P. fragarioides $(16\;{\mu}g)$. And Geum aleppicum $(18\;{\mu}g)$. From the leaves of R. rugosa, two antioxidative compounds were isolated by a bioassay guided purification and identified as isoquercitrin and ${\beta}-glucogallin$ on the basis of $^1H-,\;^{13}C-\;NMR$, and FAB-MS data. ${\beta}-Glucogallin$ is the first report in this plant. The DPPH radical scavenging activity of ${\beta}-glucogallin\;(RC_{50}:\;8.5\;{\mu}g)$ was more effective than those of ${\alpha}-tocopherol\;(12\;{\mu}g)$ and BHA $(14\;{\mu}g)$.

Sanguisorba officinalis Inhibits Immediate-type Allergic Reactions

  • The effect of aqueous extract of Sanguisorba officinalis L. (Rosaceae) root (SOAE) on the immediate-type allergic reactions by anal therapy was investigated. SOAE (0.01 to 1 g/kg) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80 in mice. When SOAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. SOAE (0.1 and 1 g/kg) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. SOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. The level of cyclic AMP (cAMP) in RPMC, when SOAE (1 mg/ml) was added, transiently and significantly increased compared with that of basal cells. These results provide evidence that anal therapy of SOAE may be beneficial in the treatment of allergic diseases.

Genetic Variation and Conservation of the Endangered Species Cotoneaster wilsonii (Rosaceae) from Ulleung Island

  • The genetic diversity plays a significant role in determining a species' survival and perseverance. Endangered species often lack genetic variation, which makes them vulnerable to numerous dangers of extinction including selection, genetic drifts and human interference. Knowing an endangered species' genetic background greatly enhances conservation efforts since it reveals why, what and how to conserve that species. Cotoneaster wilsonii is an endangered plant species endemic to Ulleung island, but not enough genetic research has been done on this taxon for its effective conservation plans. In this study, three populations of C. wilsonii in Ulleung island underwent allozyme analysis through starch gel electrophoresis. 10 loci were analyzed and F-statistics was calculated. Overall data indicated that C. wilsonii possessed low genetic diversity with intense inbreeding, heterozygote deficiency and low differentiation among populations. These results implied that C. wilsonii was recently introduced to the Ulleung island from ancestor species, and did not have much time to differentiate. Current status of C. wilsonii habitats is very fragile and vulnerable, with increasing tourism constantly threatening the species' survival. It is very likely that C. wilsonii will become extinct in near future unless organized conservation protects its populations and genetic diversity.

Sexuality of Aruncus aethusifolius (Rosaceae)

  • Aruncus L. is a small genus which is distributed in the circumpolar regions of the Northern Hemisphere, including Europe, Asia, and North America. In Korea, there are two taxa, A. dioicus var. kamtschaticus and A. aethusifolius. Aruncus aethusifolius is a species endemic to Korea, occurring only on Jeju Island. An important characteristic of Aruncus is dioecy. However, there is some controversy regarding the sexuality of this genus, and little is known about A. aethusifolius. To characterize the sexuality of A. aethusifolius and to provide insight into the evolution of its sexual system, we investigated natural populations and herbarium specimens of A. aethusifolius. The results indicate that the species has carpellate, staminate, and bisexual flowers. Carpellate flowers are always borne on separate individuals, and staminate flowers are borne either on separate individuals or mixed with bisexual flowers on the same individuals. The sexuality of A. aethusifolius is defined as polygamo-dioecious. The polygamo-dioecious type is also found in A. dioicus var. dioicus and A. dioicus var. kamtschaticus and thus may be a general condition in Aruncus.

Identification of Self-incompatibility Genotypes of Apricot (Prunus armeniaca L.) by PCR and Test Crosses

  • Apricot (Prunus armeniaca L.) cultivars show a gametophytic self-incompatibility (GSI) system, like other fruit species of Rosaceae family. Thus, it is necessary to determine their S-genotypes in order for stable fruit set in commercial cultivation. S-genotypes of apricots were determined by PCR and test crosses. Three sets of consensus primers designed from Prunus S-RNases were used to amplify fragments containing the first and second S-RNase intron, respectively. Through the results obtained from the 3 PCRs, we could identify SI genotypes of 33apricot cultivars. Several cultivars such as 'Heiwa', 'Yamagata No.3' and 'Shinsuoomi' had the self-compatible (Sc) allele. Self-pollination tests revealed that cultivars with Sc allele were self-compatible. Cross-pollination tests confirmed that there was cross-incompatibility between the cultivars with the same S-genotypes. These results might be very useful for growers for effective pollination and for breeders using these in cross breeding programs.

Hepatoprotective Effect of Solvent Fractions from Raphiolepis indica against Oxidative Stress

  • Raphiolepis indica (R. indica) is one of evergreen shrubs belonging to the Rosaceae and is grown wildly in Jeju. This study was performed to evaluate the hepatoprotective effect of different fractions (n-hexane, dichloromethane, ethyl acetate, butanol, water) from R. indica. Anti-oxidative effects were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and total phenol contents. Hepatoprotective effect was identified by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay in Huh7 cells. Among various fractions, ethyl acetate and butanol fractions showed the lowest DPPH remained rate reaching approximately 78.7 and 65.5% at $400{\mu}g/mL$. Ethyl acetate and butanol fractions showed the total phenolic content at 164.5 and 137.3 mg GAE/g extract. The ethyl acetate and butanol fractions were resistant against oxidative stress in MTT assay and showed higher hepatoprotective effect than other fractions. Therefore, these results suggest that the ethyl acetate and butanol fractions of R. indica might have therapeutic value in liver damage.

왕벚나무 껍질로부터 Naringenin, Sakuranetin, Prunetin의 분리 및 함량 분석 (Isolation and Quantitative Analysis of Naringenin, Sakuranetin and Prunetin from the Barks of Prunus yedoensis)

  • Prunus yedoensis Matsum. (Rosaceae) has been used for cough, urticaria, pruritus, dermatitis, asthma and relaxation in traditional Korean medicine. In this study, naringenin, sakuranetin and prunetin were isolated from the barks of P. yedoensis and quantification were performed by using high performance liquid chromatography (HPLC) method with diode array detector. The structures of naringenin and sakuranetin, prunetin were identified NMR spectroscopic data analysis. The content of each compound was analyzed by HPLC and the analytical method was validated through linearity, precision, accuracy and specificity test. The result showed that calibration curves of three compounds naringenin, sakuranetin and prunetin indicated great linearity with a correlation coefficient ($R^2$) of 1.00, 1.00 and 0.99. Intra and inter day measurement accuracy of the three compounds ranged from 92.70 ~ 112.70%, and showed precision was less than 3%. Therefore, the content analysis showed that naringenin ($0.132{\pm}0.016%$), sakuranetin ($0.108{\pm}0.023%$), and prunetin ($0.059{\pm}0.014%$).

Evaluation of Anti-inflammatory activity of Fissistigma poilanei and Rubus laxus Focke

  • The aim of the study was to determine the anti-inflammatory activities of the plants with origin of China. The Fissistigma poilanei, which is a species of plant in the family Annonaceae and Rubus laxus Focke, which is a species of plant in the family Rosaceae from China were tested for anti-inflammatory activities. Samples were prepared using 95% ethanol using nitric oxide (NO) assay for assessing the anti-inflammatory activity. NO assay experiment showed that extracts of the Fissistigma poilanei give 28.3% increases in anti-inflammatory activity and extracts of Rubus laxus Focke give 57.1% decreases in anti-inflammatory activity, compared to control. To determine the cell toxicity, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was used. MTT assay experiment showed that Fissistigma poilanei and Rubus laxus Focke might have less toxicity of 60.5% and 62.3%, respectively, compared to control. Taken together, these experiments showed that Fissistigma poilanei extracts might have significantly higher anti-inflammatory activities and relatively lower toxicity, compared to control. This implies that this study might give a tool to develop a new potential anti-inflammatory therapeutic candidate.

An Isocoumarin with Hepatoprotective Activity in Hep G2 and Primary Hepatocytes from Agrimonia pilosa

  • In connection with our studies on the isolation of hepatoprotective constituents from natural products. we have recently reported hepatoprotective compounds including phenolic bakuchiol. diarylheptanoids. furocoumarins. In the course of continuing efforts. the aqueous extract of the roots of Agrimonia pilosa Ledeb. (Rosaceae) was found to exhibit promising hepatoprotective activity. A. pilosa is a perennial herb distributed throughout South Korea. and its roots have been used as the hemostatic. antimalarial. and antidysenteric agent in oriental medicine. Chemical investigation of the aqueous extract of the roots of this plant. as guided by hepatoprotective active catechin (2). Compound 1 showed hepatoprotective effects on both tacrine-induced cytotoxicity in human level derived Hep G2 cells and tert-hydroperoxide-induced cytotoxicity in rat primary hepatocyles with $EC_{50}$ values of 66.2 $\pm$ 2.8 and 22.9 $\pm$ 2.6 $\mu\textrm{M}$ respectively.

Enhancing the Effect of Aronia Extract on Hyaluronic Acid Synthesis through Liposome Formation

  • Background and objective: Aronia melanocarpa, called black chokeberry, is a natural product belonging to the family rosaceae, and is known to contain polyphenolic antioxidants including cyanidin-3-galactoside, cyanidin-3-arabinoside, cyanidin-3-xyloside, and cyanidin-3-glucoside Because of the abundance of anthocyanins, Aronia has been studied to be used in various industries. Methods: Aronia melanocarpa extract was treated 24 hours a day to RAW 264.7 cells with inflammations induced by LPS. After extracting total RNA, the amount of inflammatory cytokine expression was measured using RT-PCR. After processing the Aronia liposome using Aronia extract and the layer-by-layer electrostatic deposition method in keratinocyte cells at the same time, we checked the synthesis of Hyaluronic acid enhanced through the formation of Aronia liposome using ELISA. Results: The treatment of Aronia extract in inflammation-induced RAW 264.7 cells conducted to check the anti-inflammatory efficacy of Aronia extract inhibited inflammatory cytokines including TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased the mRNA expression of HAS2 genes related to moisturizing. Based on the anti-inflammatory and moisturizing effect of Aronia extract, the Aronia liposome technology was introduced to Aronia extract to produce Aronia liposome. Conclusion: The liposome formation of Aronia extract is expected to be used as a functional material in treating various inflammatory skin diseases by controlling the moisture content of the corneocytes by increasing the expression rate of genes associated with the synthesis of hyaluronic acid, while retaining the efficacy of its components.
