Induction of Autophagy by Rosa acicularis Leaves Extracts in RAW264.7 Cells (인가목(Rosa acicularis Lindl.) 잎 추출물의 대식세포에서 자가포식 유도활성)

  • Autophagy contributes to enhancing the immune system (innate and adaptive immune system) against foreign pathogens. Autophagy of macrophages is used as a major indicator for developing vaccine adjuvants to increase the adaptive immune response. In this study, water extracts from Rosa acicularis leaves (RAL) increased the production of immunostimulatory mediators and phagocytic activity in RAW264.7 cells. RAL increased p62/SQSTM1 expression. Inhibition of TLR4, JNK, and PI3K/AKT blocked RAL-mediated increase of p62/SQSTM1. RAL activated JNK and PI3K/AKT signaling. RAL-mediated activations of JNK and PI3K/AKT signaling were reversed by TLR4 inhibition. Taken together, it is believed that RAL-mediated autophagy may be dependent on activating via TLR4-dependent activation of JNK and PI3K/AKT signaling in macrophages.

Comparative Profiling of Volatiles in Flower Tea of Prunus mume, Rosa spp. and Rosa rugosa (매화, 장미 및 해당화 꽃차의 향기 성분 비교)

  • 꽃을 우려내어 차로 마시는 꽃차(flower tea)는 꽃이 가진 색과 향, 맛과 모양을 즐길 수 있어 최근 수요가 급증하고 있다. 본 연구는 매화(Prunus mume), 장미(Rosa spp.), 그리고 해당화(Rosa rugosa) 등 3종의 장미과 식물의 꽃차의 향기 성분 특성을 구명코자 꽃차를 headspace vial에 담은 후 solid phase microextraction(SPME)를 이용하여 추출한 후 gas chromatography-mass spectrometry를 이용하여 휘발성 성분을 분리하였다. 분리된 각 성분은 deconvolution 과정을 수행한 후 NIST masss spectral library를 이용하여 동정하였다. 매화꽃차에서는 총 58개의 휘발성 성분이 검출되었는데, benzaldehye와 nonane, 그리고 phenylmethanol이 주요 구성 성분으로 검출된 전체 휘발성 성분의 각각 64.7%, 16.1% 및 4.7%를 차지하고 있었다. 장미 꽃차의 경우 검출된 35종 중 주요 구성 성분은 nonane과 2-phenylethanol, 그리고 phenylmethanol로서 각각 전체 성분 중 54.0%, 18.0% 및 4.6%를 구성하고 있었다. 해당화 꽃차의 경우는 장미 꽃차와 동일하게 2-phenylethanol(62.2%)과 nonane(20.1%)이 주요 성분으로 동정되었으며 이들 2 성분의 합이 전체 43개 휘발성 성분의 82.3%를 차지하는 것으로 나타났다. 본 연구에 사용된 매화, 장미, 해당화 등 장미과 식물 3종의 꽃차 모두에서 공통적으로 검출된 휘발성 성분은 앞서 각 꽃차 종류별 주요 성분으로 언급된 4종의 성분을 포함하여 19종이었으며 매화꽃차에서만 검출된 성분은 29종으로 장미꽃차(6종)와 해당화 꽃차(8종)보다 향기성분의 다양성이 높은 것으로 나타났다.

Rosa acicularis Leaves Exert Anti-obesity Activity through AMPK-dependent Lipolysis and Thermogenesis in Mouse Adipocytes, 3T3-L1 Cells

  • It has been reported that Rosa acicularis has anti-obesity activity by inhibiting the digestive lipase activity. However, there is a lack of clear in vitro studies regarding the anti-obesity activity of Rosa acicularis. Therefore, in this study, we aimed to verify the anti-obesity activity of Rosa acicularis leaves (RAL) and elucidate its mechanism of action in 3T3-L1 preadipocytes. RAL dose-dependently inhibited the accumulation of lipid droplets and triacylglycerol. RAL had no effect on cell proliferation and survival in undifferentiated 3T3-L1 cells, but it inhibited cell proliferation in differentiating 3T3-L1 cells. RAL increased ATGL, p-HSL, and HSL, and decreased perilipin-1 in differentiating 3T3-L1 cells. In addition, RAL reduced lipid droplet accumulation and increased free glycerol content in differentiated 3T3-L1 cells. RAL increased ATGL and HSL in differentiated 3T3-L1 cells. Also, RAL increased p-AMPK, PPARγ, UCP-1, and PGC-1α in differentiating 3T3-L1 cells. AMPK inhibition by Compound C attenuated RAL-mediated increase of ATGL, HSL, PPARγ, and UCP-1 in 3T3-L1 cells. Taken together, it is thought that RAL may inhibit lipid accumulation through lipolysis and thermogenesis via the activation of AMPK in adipocytes.

Ecophysiological characteristics of Rosa rugosa under different environmental factors

  • Background: Ecophysiological characteristics of Rosa rugosa were analyzed under different environmental factors from May to October 2022. Photosynthesis, chlorophyll fluorescence, chlorophyll content, leaf water content (LWC), osmolality, carbohydrate content, and total ion content were measured to compare the physiological characteristics of R. rugosa at two study sites (i.e., in large pots and in the Goraebul coastal sand dune area). Results: When R. rugosa was exposed to high temperatures, photosynthetic parameters including net photosynthetic rate (PN) and stomatal conductance (gs) in both experiment areas declined. In addition, severe photoinhibition occurs when R. rugosa is continuously exposed to high photosynthetically active radiation (PAR), and because of this, relatively low Y(II) (i.e., the quantum yield of photochemical energy conversion in photosystem II [PSII]) and high Y(NO) (i.e., the quantum yield of non-regulated, non-photochemical energy loss in PSII) in the R. rugosa of the pot were observed. As the high Y(NPQ) (i.e., the quantum yield of regulated non-photochemical energy loss in PSII) of R. rugosa in the coastal sand dune, they dissipated the excessed photon energy through the non-photochemical quenching (NPQ) mechanism when they were exposed to relatively low PAR and low temperature. Rosa rugosa in the coastal sand dune has higher chlorophyll a and carotenoid content. The high chlorophyll a + b and low chlorophyll a/b ratios seemed to optimize light absorption in response to low PAR. High carotenoid content played an important role in NPQ. As a part of the osmotic regulation in response to low LWCs, R. rugosa exposed to high temperatures and continuously high PAR used soluble carbohydrates and ions to maintain high osmolality. Conclusions: We found that Fv/Fm was lower in the potted plants than in the coastal sand dune plants, indicating the vulnerability of R. rugosa to high temperatures and PAR levels. We expect that the suitable habitat range for R. rugosa will shrink and move to north under climate change conditions.

In vitro multiple shoot proliferation and plant regeneration in rose(Rosa hybrida L.)

  • This study was conducted to investigate an optimal condition for shoot proliferation and regenerate shoots from in vitro leaflet and embryogenic calli from in vitro roots in rose. The effect of BAP on shoot proliferation was somewhat different depending upon genotypes or gelling agents. Leaflets with petiole cut from donor shoots which had been cultured in MS medium supplemented with 0.1 $mg{\cdot}L^{-1}$ NAA for six weeks was effective for regeneration of adventitious buds(ABs) as well as shoot elongation of Rosa hybrida cv. Sweet Pink. Culturing seven leaflet explants per petri plate($100mm{\times}15mm$) was effective for regeneration of ABs. Embryogenesis was shown in the calli induced from roots of Rosa hybrida cv. Sweet Pink cultured in the SH medium supplemented with 11 $mg{\cdot}L^{-1}$ 2, 4-D for four weeks. Color of calli induced from roots was yellow although their color was a little different as type of basal medium.

Antioxidant and hepatoprotective action of the crude ethanolic extract of the flowering top of Rosa damascena

  • The hepatoprotective activity of the alcoholic extract of Rosa damascena was studied against paracetamol induced acute hepatotoxicity in rats. Liver damage was assessed by estimating serum enzyme activities of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and histopathology of liver tissue. Pre- and post-treatment with ethanolic extracts showed a dose-dependent reduction of paracetamol induced elevated serum levels of enzyme activity. The mechanism underlying the protective effects was assayed in vitro and the R. damascena extracts displayed dosedependent free radical activity using DPPH ($IC_{50}=162.525\;{\mu}g/ml$) and TBA method. The hepatoprotective action was confirmed by histopathological observation. The ethanolic extracts reversed paracetamol induced liver injury. These results suggest that the hepatoprotective effects of R. damascena extracts are related to its antioxidative activity.

Study on the Plants-preference Pattern According to Personality Type(Enneagram) for the Creating of Healing Garden - Focusing young generation - (치유정원 조성을 위한 성격 유형별(Enneagram) 식물 선호도에 관한 연구 - 청년층을 대상으로 -)

  • This study identifies the basic personality types of subjects who use the healing garden and investigates their plants-preference. The purpose of the study is to provide a basic information about planting plan, considered as a main parts of healing garden. Survey results are stated as follows: First, the order of preference of instinct type tree is Prunus yedoensis, zelkova serrata, Phyllostachys Bambusoides. On the other hands, the order of preference of feeling and thinking type is Prunus yedoensis, zelkova serrata, Metasequoia glyptostroboides. Second, the order of preference of instinct type of flowers is Rosa hybrida, Hibiscus syriacus, Forsythia koreana. However, the preference of emotional type is the order of Rosa hybrida, cosmos bipinnatus, Helianthus annuus and the preference of accident type is the order of Rosa hybrida, Hibiscus syriacus, Helianthus annuus. Third, for the color preferences, instinct type prefer yellow color, feeling type and thinking type prefer red color. Lastly, when people view plants, most personality types are interested in flower firstly, and secondly type of tree and scent. In other words, we can assume that people are interested in the flowers, most noticeable in plants. This study will enhance the effectiveness of the healing garden by providing relevant information regarding healing garden planting plan. Future research should consider to widen the range of target of investigation in order to perform additional comparison analysis.

Vegetation Structure of Peucedanum japonicum Thunb. Community in Southern Coast of Korea

  • This study was conducted to evaluate the vegetation structure of the Peucedanum japonicum community by the phytosociological method of floristic composition table and cluster analysis on the southern coast of Korea. The vegetation of the Peucedanum japonicum community was classified mainly into 2 communities such as the Miscanthus sinensis community and the Lysimachia mauritiana-Rosa wichuraiana community. The Carex boottiana and Sedum oryzifolium community were classified as the lower rank of Miscanthus sinensis community. On level 1 of the cluster analysis of plant species, they were classified into Lysimachia mauritiana and Rosa wichuraiana group, also Miscanthus sinensis, Carex boottiana and Sedum oryzifolium in Peucedanum japonicum community, which is similar to the community classification shown in the synoptic table. On level 1 of the cluster analysis of relev, inland coast with Jejudo was Lysimachia mauritiana and Rosa wichuraiana of group such as level 1 of the cluster analysis of plant species, and island coast with Geomundo was Miscanthus sinensis Carex boottiana and Sedum oryzifolium of group such as cluster analysis of plant species.

Aqueous Extract of Rosa rugosa Radix Dilates Vascular Smooth Muscle Via a NO-cGMP Pathway

  • While conducting an in vitro screening of various medicinal plant extracts, an aqueous extract of Rosa rugosa Radix (ARR) was found to exhibit a distinct vasorelaxant activity. ARR induced a concentration-dependent relaxation of the phenylephrine-precontracted aorta. This effect disappeared with the removal of functional endothelium. Pretreatment of the aortic tissues with NG-nitro-L-arginine methyl ester (L-NAME) or 1H-[1,2,4]-oxadiazole-[4,3-]-quinoxalin-1-one (ODQ) completely inhibited the relaxation induced by ARR. ARR-induced vascular relaxations were also markedly attenuated by addition of diltiazem or verapamil. However, the relaxant effect of ARR was not blocked by pretreatment with indomethacine, tetraethylammonium (TEA), glibenclamide, atropine, or propranolol. Taken together, the present study suggests that ARR dilates vascular smooth muscle via endothelium-dependent NO/cGMP signaling.

  • The aim of this study was to find optimal ratio of herbal extracts to ameliorate ethanol-induced psychomotor alterations. Four mixtures of Laurus nobilis (L. nobilis), Rosa roxburghii (R.R.) and Opuntia ficus indica (OFI) were pretreated 30 minutes before ethanol administration in rats. Behavioral activities were evaluated. Blood ethanol and acetaldehyde levels were also measured. These effects of mixture of R.R (50 mg/kg)+OFI (100 mg/kg)+L.nobilis (100 mg/kg) were better than those of other mixtures. This mixture could be used as an effective substance to develop a functional food.
