Free radical scavenging activity and protective effect of three glycyrrhiza varieties against hydrogen peroxide-induced oxidative stress in C6 glial cells (종류별 감초의 라디칼 소거능 및 H2O2에 의한 C6 glial 세포의 산화적 스트레스 개선 효과)

  • Oxidative stress is common cause of neurodegenerative diseases. The purpose of this study is to investigate the in vitro free radical scavenging activity and protective effect of three Glycyrrhiza species including Glycyrrhiza uralensis, G. glabra, and a new variety of Glycyrrihza (Shinwongam, SW) against hydrogen peroxide-induced oxidative stress in C6 glial cells. In vitro assays, radical scavenging activities of G. uralensis, G. glabra, and SW against 2,2-diphenyl-1-picrylhydrazyl, ·OH, and O2- increased as concentration-dependent manner. In addition, the SW was found to contain the highest polyphenol and flavonoid contents. The treatment of H2O2 to C6 glial cell induced oxidative stress, whereas G. uralensis, G. glabra, and SW significantly increased the cell viability as dose-dependent manner. In particular, SW exerted stronger protective effect on H2O2-induced cytotoxicity, than G. uralensis and G. glabra. Furthermore, reactive oxygen species (ROS) formation was significantly elevated by H2O2 in C6 glial cells. However, treatments of G. uralensis, G. glabra, and SW decreased ROS formation. In addition, SW decreased pro-inflammatory related protein expression levels such as inducible nitric oxide synthase and cyclooxygenase-2, compared to H2O2-treated control group. These results indicated that G. uralensis and G. glavra, especially SW, may be useful for preventing from oxidative stress-induced neuronal damage by regulating inflammatory reaction.

Effect of Gami-Chunggisan on Antioxidant and Pro-Inflammatory Cytokine (가미청기산(加味淸肌散)이 항산화와 염증성 사이토카인에 미치는 영향)

  • Objectives : Gami-Chunggisan extract (GCE) is one of the oriental traditional medicine. We investigated the antioxidant effect and reduction of pro-inflammatory cytokine as a functional ingredient for cosmetic products from the GCE. Methods : GCE was prepared by extracting with 80% ethanol. We analyzed total polyphenol and antioxidant activities. To evaluate antioxidant activity, we measured 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging assay. Also we measured the production of reactive oxygen species (ROS) and nitric oxide (NO) on Raw264.7 cells. We researched reduction of anti-inflammatory cytokines from concentration of GCE on Raw264.7 cells. Results : Total polyphenol quantity of GCE was included 46.6 mg/g. The GCE showed ABTS free radical scavenging ability with more than 89% at $1000{\mu}g/m{\ell}$. In addition the DPPH free radical scavenging ability from the GCE was activated over 93% at $1000{\mu}g/m{\ell}$. Production of the ROS was decreased by approximately 26%, upon the GCE treatment at concentration of $100{\mu}g/m{\ell}$. The GCE at $100{\mu}g/m{\ell}$ concentration showed inhibitory effect on NO production by 38%. Production of IL-$1{\beta}$ and IL-6 were decreased by approximately 56% and 36%, respectively upon GCE treatment at $100{\mu}g/m{\ell}$. Also, production of TNF-${\alpha}$ was decreased by approximately 79% at $100{\mu}g/m{\ell}$. Moreover, the GCE showed inhibitory effects on the expression of the IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ genes in LPS-induced Raw 264.7 cells. Conclusions : From the results above, we conclude that the GCE indicated significant antioxidant effects and induced reduction of pro-inflammatory cytokine.

The Antioxidative and Antimicrobial Effects of Celastrus orbiculatus (노박덩굴 추출물의 항산화 및 항균 효과)

  • This study was carried out to investigate the antioxidative and antimicrobial activities of Celastrus orbiculatus (CO). CO was extracted with methanol and then further fractionated with n-hexane (COMH), butanol (COMB), methanol (COMM) or aqueous (COMA) to get active fractions. The antioxidative activity of fractions from CO was investigated by measuring the scavenging activities of CO against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen speicies (ROS). Among the various solvent fractions, the COMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species(ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, the COMM and COMB fractions of CO showed strong antimicrobial activities. The results suggest that The CO may be suitable for development as a food preservative and alternative antioxidant.

The Antioxidative and Antimicrobial Effects of Internal Organs of Aplysia kurodai Fractions (군소내장 분획물의 항산화 및 항균효과)

  • This study was carried out to investigate the antioxidative and antimicrobial activities of internal organs of Aplysia kurodai (AK). The internal organs of AK were extracted with methanol (AKM), which was then further fractionated into four subfractions by using solvent partition method, affording hexane (AKMH), methanol (AKMM), butanol (AKMB), and aqueous (AKMA) soluble fractions. The antioxidative activity of fractions from AK was investigated by measuring the scavenging activities of AK against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). Among the various solvent fractions, AKMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, AKMM fractions of AK showed the strongest antimicrobial activities. The methanol extracts exhibited antimicrobial activity against all organisms tested, while the hexane extracts showed antimicrobial activity only against Proteus vulgaris. The results suggest that the AK may be suitable for development as a food preservative and alternative antioxidant.

The Effect of Lonicera japonica Extract in Wound-induced Rats (창상을 유발한 흰쥐에서 금은화(金銀花) 추출물의 치료 효과)

  • Objectives This study is carried out to investigate the effects of Lonicera japonica in wound-induced rats. Methods Rats were divided into 5 groups; normal (Nor), control (Veh), positive comparison (PC), Lonicera japonica 100 mg/kg (LL), Lonicera japonica 200 mg/kg (LH), each n=8. Total polyphenol and flavonoid were quantified. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3 ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging activation were measured. Reactive oxygen species (ROS) was measured in serum. Antioxidant factors and inflammatory factors were measured in skin tissue, and also hydroxyproline content. Skin tissue was analyzed by Hematoxylin & Eosin and Masson's trichrome staining method. Results Total polyphenol and flavonoid were 32.86±0.14 mg/g and 67.17±0.57 mg/g. The IC50 values of DPPH and ABTS free radical scavenging activation were 26.69±1.50 ㎍/mL and 49.33±4.52 ㎍/mL. ROS was significantly lower in LL and LH groups. Nuclear factor-erythroid 2-related factor 2 (Nrf2) was significantly higher in LH group and higher in LL group but not significant. Superoxide dismutase 1 (SOD-1), catalase, and heme oxygenase 1 (HO-1) were significantly higher in LL and LH groups. Nuclear factor kappa-B p65 (NF-κBp65), phosphorylated iκBα (p-iκBα), cyclooxygenase 2 (COX-2), and tumor necrosis factor alpha (TNF-α) were significantly lower in LL and LH groups. Hydroxyproline was significantly higher in LL and LH groups. The histopathologic analysis showed that skin tissue had recovered further more in LL and LH groups than in Veh group. Conclusions These results suggest that Lonicera japonica has the anti-oxidant, anti-inflammatory and healing effects in wound-induced rats.

Effect of the Processed Selaginella tamariscina on Antioxidation and Inhibition of Matrix Metalloproteinase (수치에 의한 권백의 항산화 효과와 MMP 발현 저해 효과)

  • Selaginella tamariscina with the popular Korean name Keoun Back, is a traditional medicinal plant for therapy of advanced cancer patients in the Orient. In this study, we evaluated anti-aging activity of S. tamariscina using processed technology and investigated diverse biological activities of processed S. tamariscina (PST) as an anti-aging ingredient of cosmetics. PST, heated with sand, used to different purpose compared with origin in medicine. PST raises total phenol concentration and enhances the DPPH radical scavenging activity. For testing intracellular ROS scavenging activity, the cultured human dermal fibroblasts were analyzed by increase in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20 mJ/cm^2$ after treatment of PST. UVA-induced MMP-1 expression in human dermal fibroblasts was reduced in a dose-dependent manner by PST. Taken together, 4hese results suggest that PST may act as an anti-aging agent by preventing the skin cell from damage induced by UV irradiation, and imply that PST may be useful as a new ingredient for anti-aging cosmetics.

Effect of Heme-rich Nutrient on Anaerobic Bacterial Growth and Survival: A Model Study on Lactobacillus gasseri (헴철이 풍부한 영양원이 혐기성 세균의 생장과 생존에 미치는 영향: 락토바실러스 가세리 모델연구)

  • Lactic acid bacteria (LAB), belonging to the Firmicutes phylum, lack heme biosynthesis and, thus, are characterized as fermentative and catalase-negative organisms. To verify the hypothesis that heme-rich-nutrients might compensate the heme-biosynthesis incapability of non-respiratory LAB in animal gut, a heme-rich-nutrient was fed to a dog and its fecal microbiome was analyzed. Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog was 99%, compared to 92% in the control dog. To clarify the reason of increased Firmicutes abundance in the feces from the heme-rich-nutrient-fed dog, Lacobacillus gasseri were used as model anerobic LAB to study a purified heme (hemin). The anaerobic growth of L. gasseri in the medium with 25 µM hemin supplementation was faster than that in the medium without hemin, while the growth in the 50 µM hemin-supplemented medium did not vary. Cellular activities of the cytochrome bd complex were 1.55 ± 0.19, 2.11 ± 0.14, and 2.20 ± 0.08 U/gcell in the cells from 0, 25, and 50 µM hemin-supplemented medium, while intracellular ATP concentrations were 7.90 ± 1.12, 11.95 ± 0.68, and 12.56 ± 0.58 µmolATP/gcell, respectively. The ROS-scavenging activities of the L. gasseri cytosol from 25 µM and 50 µM hemin-supplemented medium were 68% and 82% greater than those of the cytosol from no hemin supplemented-medium, respectively. These findings indicate that external hemin could compensate the heme-biosynthesis incapability of L. gasseri by increasing the cytosolic ROS-scavenging and extra ATP generation, possibly through increasing the electron transfer. Increase in the number of anaerobic bacteria in heme-rich-nutrient-fed animal gut is discussed based on the results.

Antioxidative and Cellular Protective Effects of Jeju Native Plant Extracts against Reactive Oxygen Species (I) (제주 자생 식물 추출물의 항산화 및 세포 보호작용에 관한 연구(I))

  • In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of 36 plant extracts collected from self-growing plants in Jeju island. Their anti-oxidant activities were measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical), reactive oxygen species (ROS) scavenging activities on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay, and cell protecting activities using the rose-bengal sensitized photohemolysis of human erythrocytes. In addition, the inhibitory activities of tyrosinase for whitening effect and elastase for anti-wrinkle were investigated. The results showed that the Rumex crispus (all grass) extract has the most significant free radical scavenging activity ($FSC_{50};\;10{\mu}g/mL$), Plantago asiatica and Rumex crispus extracts for the prominent ROS scavenging activity ($OSC_{50};\;0.006{\mu}g/mL$, $0.04{\mu}g/mL$ respectively), Rumex crispus ($\tau_{50};\;1,140 min $at $50{\mu}g/mL$), Machilus thunbergii leaf (216 min), and Celastrus orbiculatus (200 min) for cell protecting effects, Morus alba stem for the inhibitory activity on tyrosinse (94.8% at $200{\mu}g/mL$), Rumex crispus (81.8% at $200{\mu}g/mL$), Morus alba (74.6%), and Celastrus orbiculatus leaf/stem/flower (63.1%) for the activity on elastase. These results indicated that the extracts of Rumex crispus, Plantago asiatica, Machilus thunbergii leaf, Morus alba stem, Celastrus orbiculatus leaf/stem/flower could have the functional effects when they are added as ingredients in cosmetics. Thus, it is concluded that further experiments are needed to apply for cosmetic products.

Comparison of Flavonoid Content and Antioxidant Effect of Extracts from Stachys sieboldii Miq. and Lycopus lucidus Turcz (초석잠 및 택란 추출물의 플라보노이드 함량 및 항산화 활성 비교)

  • The flavonoid content and antioxidant effects of extracts from Stachys sieboldii Miq. and Lycopus lucidus Turcz were compared. The flavonoid content of the acetone + methylene chloride (A+M) extract of L. lucidus Turcz was 233.2 mg/g, suggesting that the extract was greater than that of S. sieboldii Miq. In the DPPH assay and the A+M and methanol (MeOH) extracts from L. lucidus Turcz had greater scavenging effects than those of S. sieboldii Miq. (p<0.05). The A+M extract from L. lucidus Turcz (0.5 mg/ml concentration) had an 82% scavenging effect in the DPPH assay. In the ABTS assay, A+M extracts from both S. sieboldii Miq. and L. lucidus Turcz (0.5 mg/ml concentration) had scavenging effects of 90% and 88%, respectively (p<0.05), suggesting that both A+M extracts had greater scavenging effects than those of both MeOH extracts. In a 120 min ROS production assay, all tested extracts dose-dependently decreased the cellular ROS production that was induced by $H_2O_2$, as compared to those produced by exposure to the extract-free control. The A+M extracts from both S. sieboldii Miq. and L. lucidus Turcz had greater inhibitory effects on cellular ROS production than those of both MeOH extracts at all concentrations tested. Treatment with the A+M extracts from S. sieboldii Miq. and L. lucidus Turcz (0.25 mg/ml concentration) inhibited the cellular ROS production by 60% and 86%, respectively. These results suggest that the A+M extracts of Stachys sieboldii Miq. and L. lucidus Turcz inhibit cellular oxidation and may contain valuable bioactive compounds, such as flavonoids.

Anti-oxidant and Anti-aging Activity on Saxifraga stolonifera MEERBURGH Ethanol Extract (바위취 에탄올 추출물의 항산화 및 항노화 작용)

  • To investigate the effect of ethanol extract of Saxifraga stolonifera MEERBURGH on skin care, we measured anti-oxidant and anti-aging activity. S. stolonifera ethanol extract itself had anti-oxidant activity in a dose-dependent manner in DPPH radical scavenging. Silica dose-dependently increased the intracellular ROS generation in RAW 264.7 cells. S. stolonifera ethanol extract inhibited silica-induced intracellular superoxide anion generation, $H_2O_2$ and hydroperoxide generation in RAW 264.7 cells. S. stolonifera ethanol extract significantly inhibited both hyaluronidase and elastase activity, also significantly inhibited MMP-1(collagenase) activity as well. In NIH 3T3 fibroblast cells, S. stolonifera ethanol extract significantly increased collagen-like polymer synthesis, which suggesting the S. stolonifera ethanol extract might be used as hydration and anti-wrinkle agents. From the above results, it is suggested that the main ingredients of S. stolonifera ethanol extract play an important role in anti-oxidant and anti-aging activity.
