Effect of Different Kimchi Diets on Oxidation and Photooxidation in Liver and Skin of Hairless Mice (김치 종류를 달리한 식이가 Hairless Mice의 간과 피부에서 산화억제 및 광산화에 미치는 영향)

  • Kimchi contains various antioxidative components, therefore it is expected that kimchi have the inhibitory activity on aging. Among the many factor related to aging, UV radiation is the easiest way to induce skin aging. This experiment was designed to demonstrate the antioxidative activity of dietary kimchi with and without UV radiation in tissues. Lyophilized Korean cabbage kimchi, mustard leaf kimchi and buchu kimchi were used for measurement of the contents of antioxidative components. Chlorophyll contents in buchu and mustard leaf kimchi were higher than cabbage kimchi, and the contents of carotene and ascorbic acid were higher in the mustard leaf kimchi. To clarify the antioxidative activity of various kimchi diets in vivo system, hairless mice were fed the diet containing 10% lyophilized kimchi for 20 weeks. Contents of superoxide anion and hydroxyl radical in liver were not shown significant differences, however hydrogen peroxide were significantly decreased in mustard leaf and buchu kimchi diet groups. Supplementation of all kinds of kimchi diets attenuated the hepatic NDA accumulation, especially in mustard leaf and leek kimchi group. In the skin, the contents of MDA were decreased in kimchi groups compared to control group, especially the buchu kimchi diet significantly inhibited the lipid peroxidation. To elucidate the effect of kimchi diets on photooxidation, the extent of oxidative damage in liver and skin was measured after UVB radiation treatment. The contents of lipid peroxides was decreased in mustard leaf kimchi group and cabbage kimchi group compared to control, while lipid peroxidation was increased in buchu kimchi diet group. It suggests that chlorophylls and other antioxidative nutrients in the various kimchi diets are related to antioxidation or photooxidation and it might be needed further study to certify the exact mechanisms.

Effects of Water Extract from Orostachys japonicus on Lipid and Antioxidant Metabolism in the Liver of Obese Mice (와송이 고지방 식이로 유도된 비만 쥐의 간내 지질 및 항산화 대사에 미치는 영향)

  • The purpose of this study was to investigate the changes in the liver lipid and antioxidant metabolism in obese rats following ingestion of Orostachys japonicus (OJ) hot water extract. The rats were fed a high fat diet for 8 weeks to induce obesity. In the experimental group, 2.5% or 5% OJ hot water extract was added to the diets for 6 weeks. To determine the changes in the liver lipid and antioxidant metabolism in obese rats, the livers were collected and weighed, lipid content was measured, and antioxidant enzyme activity was determined. A significant decrease in liver weight was observed in the group fed with OJ hot water extracts and decreased lipid accumulation was observed in the morphological observation of liver tissue. Total cholesterol and triglyceride contents were also decreased. Both OJ extracts significantly decreased 2,2-diphenyl-1-picrylhydrazyl radical and intracellular reactive oxygen species levels. Further, the levels of the antioxidant enzymes superoxide dismutase and catalase were significantly increased in rats treated with OJ extracts. These results suggest that the intake of OJ hot water extracts may improve liver lipid metabolism by activating antioxidant enzymes.

Effects of fermentation by the commercial starter ABT-5 on the flavor and antioxidant activities of dark chocolate (복합 유산균 스타터 ABT-5를 이용한 발효 다크 초콜릿의 항산화 활성 및 향기 성분)

  • Chocolate, one of the most popular confectioneries in the world, is known for its aromatic flavor and high antioxidant activities. In this study, we investigated the effects of fermentation with commercially available lactic acid bacteria, ABT-5, on the flavor and antioxidant activities of dark chocolate. During 24 h fermentation, pH decreased from 5.52 to 3.97 and total acidity increased from 0.51 to 1.85%, whereas total polyphenol and flavonoid contents as well as DPPH and ABTS radical scavenging activities remained unchanged. Furthermore, compared with control HepG2 cells treated with unfermented dark chocolate, those treated with the fermented dark chocolate showed significantly lower levels of reactive oxygen species and higher viability under $H_2O_2-induced$ oxidative stress. Finally, GC-MS and headspace GC-MS analysis detected 4-hydroxy-2,5-dimethyl-3(2H)-furanone and 2-furanmethanol, known to enhance flavor, in the fermented dark chocolate. Collectively, these results suggest that ABT-5-fermented dark chocolate could be utilized for developing value-added dark chocolate products.

Anti-Oxidative, Anti-Inflammatory, and Anti-Melanogenic Activities of Endlicheria Anomala Extract (Endlicheria anomala (Nees) Mez 추출물의 항산화, 항염증 및 미백 활성)

  • In this study, the anti-oxidative, anti-inflammatory, anti-melanogenic activities of Endlicheria anomala (Nees) Mez methanol extract (EAME) were evaluated by use of in vitro assays and cell culture model systems. The results revealed that EAME scavenges various radicals such as 1,1-diphenyl-2-picryl hydrazyl hydrogen peroxide induced reactive oxygen species, and lipopolysaccharide induced nitric oxide. Furthermore, EAME induced the expression of anti-oxidative enzymes such as heme oxygenase 1, thioredoxin reductase 1, NAD(P)H dehydrogenase 1, and their upstream transcription factor, nuclear factor-E2-related factor 2. Moreover, EAME inhibited in vitro DOPA oxidation and 3-isobutyl-1-methylxanthine induced melanogenesis in B16F10 cells. Its anti-melanogenic activity will have originated from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Taken together, these results provide the important new insight that E. anomala possesses various biological activities such as anti-oxidative, anti-inflammatory, and anti-melanogenic. Therefore, it might be utilized as a promising material in the fields of nutraceuticals and cosmetics.

Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity (제주도 자생식물들에 대한 미백 및 항산화 효능 탐색)

  • In this study, we investigated the whitening and anti-oxidant activity of 37 Jeju island native plants. The active ingredients of the plants were prepared by methanol extraction. Whitening activity of plant extracts was examined from the inhibitory effect of tyrosinase and the inhibition of melanin synthesis of the B16-F1 cell line. Their anti-oxidant activity was measured by electron donating ability of DPPH (1,1-diphenyl-2-picrylhydrazyl) and ROS (reactive oxygen species) scavenging activity in V79-4 lung fibroblast cells using DCF-DA (dichlorofluorescin diacetate). Cytotoxicity of the extracts on cell s based experiments was investigated by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay. Also, the toxicity test using a rabbit and the human skin patch test were carried out for examining the safety of the extracts which showed the high whitening activity. It is interesting that the extracts of Lespedeza cuneata, Ligustrum lucidum (stem), Morus bombycis (stem) and Prunella vulgaris var. lilacina showed both potent whitening and anti-oxidant activities.

Antioxidative Activities of Whole Plant Extracts of Solanum nigrum L. (까마중(Solanum nigrum L.) 전초 추출물의 항산화 활성)

  • In this study, the antioxidative effects of 50% ethanol extract, ethylacetate fraction and aglycone fraction obtained from dried whole plant of Solanum nigrum L. were investigated. The free radical scavenging activities ($FSC_{50}$) were $215.46{\mu}g/mL$, $42.43{\mu}g/mL$ and $52.28{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system were $25.25{\mu}g/mL$, $7.05{\mu}g/mL$ and $6.25{\mu}g/mL$, respectively. 50% ethanol extract and aglycone fraction showed the cellular protective effect against $^1O_2$ induced cellular damage of rabbit erythrocytes at $5{\sim}25{\mu}g/mL$, but not at high concentrations. These results indicated that S. nigrum extract/fractions could be used as an antioxidative agent. However, it could induce cellular damage at high concentrations. In conclusion, a special caution is required to use S. nigrum extracts as a cosmetic ingredient.

Modulation of Chemical Stability and Cytotoxic Effects of Epigallocatechin-3-gallate by Different Types of Antioxidants (Epigallocatechin-3-gallate의 화학안정성 및 세포독성에 미치는 각종 항산화제의 영향)

  • Epigallocatechin-3-gallate (EGCG) is a polyphenolic compound frequently found in green tea, and its physiological actions have been extensively investigated. In the present study, changes in chemical stability and cytotoxic properties of EGCG in the presence of different types of antioxidants were investigated. The antioxidants used modulated the chemical stability of EGCG. Superoxide dismutase (SOD) significantly increased EGCG stability; EGCG was less stable in the presence of catalase. Ascorbic acid, N-acetylcysteine (NAC), and glutathione (GSH) stabilized EGCG concentration dependently. The $H_2O_2$ level generated from EGCG was decreased by catalase, SOD, and NAC but not by GSH. The cytotoxic effects of EGCG also decreased in the presence of NAC, catalase, and SOD. GSH, however, showed a complicated modulatory pattern according to the EGCG and GSH concentrations, and ascorbic acid rather enhanced EGCG toxicity. The results suggest that certain antioxidants could modulate the cytotoxic properties of EGCG in a cell culture system not only by removing reactive oxygen species but by modulating chemical stability and other factors, which should be considered carefully when studying reactive oxygen species-dependent mechanisms of EGCG.

Effect of Ursolic Acid on the Development of Mouse Embryonic Stem Cells under Hypoxia (저산소 상태에서 우르솔산이 배아줄기세포 성장에 미치는 효과)

  • Ursolic acid (UA) a bio-active ingredient found in a variety of fruits and vegetables, and it has potent antioxidant activity. However, the role of UA in mouse embryonic stem (ES) cells is poorly understood. This study investigated the functional role of UA in regulating the development of mouse ES cells under hypoxia. Hypoxia did not exert a significant effect on the undifferentiated state of mouse ES cells. However, it induced reactive oxygen species (ROS) generation and increased the level of lactate dehydrogenase (LDH) production at 48 h of hypoxic exposure. Conversely, oxidative stress induced by hypoxia was significantly inhibited by UA ($30{\mu}M$) pretreatment. Hypoxia significantly decreased cell survival and the level of [$^3H$] thymidine incorporation, both of which recovered following pretreatment of UA. In addition, UA decreased the apoptotic effect of hypoxia by attenuating caspase-3 cleavage or by recovering cellular inhibition of the apoptotic protein (cIAP)-2 and Bcl-2 expression. We further found that UA decreased senescence-associated beta-galactosidase activity. We suggest that UA is a natural antioxidant and one of the functional modulators of hypoxia-induced survival, apoptosis, proliferation, and aging in mouse ES cells.

Protective effects of red ginseng according to steaming time on HCl/ethanol-induced acute gastritis (염산/에탄올로 유도된 급성 위염 동물모델에서 증숙시간에 따른 홍삼의 보호 효과)

  • The objective of the present study was to evaluate the protective effect of red ginseng (RG) according to steaming time on 150 mM HCl/60 % ethanol induced gastric ulcer models in mice. The sample was divided into 3 groups-G (dried ginseng), RG 4 (steamed 4 h and dried ginseng), RG 6 (steamed 6 h and dried ginseng), and determined through in vitro experiments, such as 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid radical scavenging activity, HPLC analysis, total polyphenol, and flavonoid contents. In vitro experiment results were depended on steaming hours. Based on the results, we chose two samples (G, RG 6) and conducted in vivo experiments. Mice were divided into 5 groups: Nor (normal group), Con (acute gastritis mice treated with distilled water), G (gastris induced by HCl/Ethanol treated with 100 mg/kg G), RG 6 (gastris induced by HCl/ethanol treated with 100 mg/kg RG 6), and SC (gastris induced by HCl/Ethanol treated with 10 mg/kg sucralfate). In our results revealed that RG 6 suppressed elevated reactive oxygen species, and inflammatory related makers, such as cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor alpha, and interleukin-1 beta. In addition, gastric lesion area was improved. These results suggest that RG 6 protects the stomach by attenuating oxidative stress and inflammatory response under gastric ulcer conditions. Therefore, RG 6 should be a potential therapeutic agent for the treatment of acute gastric ulcer.

Anti-oxidant and Anti-inflammation Activity of Fractions from Aster glehni Fr. Schm. (섬쑥부쟁이(Aster glehni Fr. Schm.) 분획물의 항산화 및 항염증 활성)

  • The Plants and their extracts containing polyphenol have been shown to be associated with decreased the cause of aging and variety of disease such as reaction oxygen species (ROS) and reactive nitrogen species (RNS) in several recent studies. We conducted to investigate whether the extracts and fractionation isolated from Aster glehni Fr. Schm. has an inhibitory effect association with oxidation or inflammation. The Aster glehni Fr. Schm. 70% aq. MeOH was fractioned according to polarity with n-hexane layer, EtOAc layer, n-BuOH layer and water layer. The electron donating ability of EtOAc, n-BuOH solvent fraction from Aster glehni Fr. Schm. was about 58.0%, 46.4% at $50\;{\mu}g/mL$, respectively. The superoxide anion radical inhibitory effect of EtOAc extracts was about 64.65% at $50\;{\mu}g/mL$, and n-BuOH extracts was 35.66% at $50\;{\mu}g/mL$. EtOAc layer to the inhibition activity of hyaluronidase and lipoxygenase were inhibited about 24.37%, 29.5% at $5\;{\mu}g/mL$. In the anti-inflammation effect of EtOAc layer inhibited the generation of nitric oxide. also, these results showed that EtOAc extract inhibited 81.5% at $50\;{\mu}g/mL$ on the expressions of iNOS protein in Raw264.7 cell line.
