식물 호르몬에 의한 옥수수 rbcL mRNA의 양적 변화 (Quantitative Change in rbcL mRNA of Maize by Phytohormones)

  • In order to investigate the effects of plant hormones on the quantitative changes in mRNA of maize (Zea mays L.) rbcL, we used GA3, IAA, ABA and BAP. GA3 at the concentration of 10-4M resulted in decrease in rbcL gene transcript to 62%. IAA decreased the amount of rbcL transcript to about 70% at all the hormone concentrations tested. ABA did not cause a noticeable change in the amount of rbcL transcript, but BAP increased the amount of rbcL transcript to 153% at 10-8M and 123% at 10-5M, respectively. Thus, it appears that BAP is related to the increase in the amount of rbcL transcript by light.

The Application of RNA Transcript Conformation Polymorphism in Resolving Mixed Infection of PVY Isolates

  • A method based on RNA-transcript conformation polymorphism (TCP) was tested for detection of two PVY isolates in a mixed infection. Differences in electrophoretic mobility of RNA transcripts copied from PCR products of each virus isolate enabled the distinction between the two virus isolates in a mixed infection. The identities of the RNA transcripts and hence of the infecting virus isolates were determined by annealing to reference oligonucleotides containing unique strain-specific sequences visualized by retardation of transcript mobility in gel. The ratio at which both virus isolates could be detected was as low as 1:10. The suitability of this procedure for the study of mixed virus infections is discussed.

In silico approach to calculate the transcript capacity

  • We sought the novel concept, transcript capacity (TC) and analyzed TC. Our approach to estimate TC was through an in silico method. TC refers to the capacity that a transcript exerts in a cell as enzyme or protein function after translation. We used the genome-wide association study (GWAS) beta effect and transcription level in RNA-sequencing to estimate TC. The trait was body fat percent and the transcript reads were obtained from the human protein atlas. The assumption was that the GWAS beta effect is the gene's effect and TC was related to the corresponding gene effect and transcript reads. Further, we surveyed gene ontology (GO) in the highest TC and the lowest TC genes. The most frequent GOs with the highest TC were neuronal-related and cell projection organization related. The most frequent GOs with the lowest TC were wound-healing related and embryo development related. We expect that our analysis contributes to estimating TC in the diverse species and playing a benevolent role to the new bioinformatic analysis.

Expression of Cyclin D3 Transcripts in the Postmeiotic Male Germ Cells of the Mouse

  • D-type G1 cyclins are known to be crucial for the progression of mitotic cell cycle in mammals. Although many studies have been performed to elucidate the roles of D-type cyclins, it is largely unknown whether D-type cyclins are directly involved in the regulation of meiotic germ cell development. In the present study, we examined the expression patterns of D-type cyclins (cyclin D1 and D3) during male germ cell development by northern blot and in situ Hybridization analyses. In the adult testes, we detected a 4.2 kb cyclin D1 mRNA and two different sizes (2.3 kb and 1.8 kb) of cyclinD3 mRNAs. The short form of the cyclin D3 transcript was testis-specific. Along with the testicular development, expression of cyclin D3 mRNA was increased whereas cyclin D1 mRNA was gradually decreased. in situ hybridization study also revealed that the expression of cyclin D3 was restricted to the postmeiotic germ cells. Furthermore, the 2.3 kb transcript was highly expressed in the round spermatids and decreased in the elongated spermatids/residual bodies, while the 1.8 kb transcript was expressed in elongated spermatids/residual bodies more abundantly. Sucrose-gradient separation of polysomal RNA fractions demonstrated that some portions of the 2.3 kb transcript are translationally active, while the 1.8 kb transcript is likely to be inactive. Taken together, the present data suggest a functional importance of cyclin D3 expression in the differentiated postmeiotic male germ cells.

Identification of a novel circularized transcript of the AML1 gene

  • The AML1 gene is an essential transcription factor regulating the differentiation of hematopoietic stem cells into mature blood cells. Though at least 12 different alternatively spliced AML1 mRNAs are generated, three splice variants (AML1a, AML1b and AML1c) have been characterized. Here, using the reverse transcription-polymerase chain reaction with outward-facing primers, we identified a novel non-polyadenylated transcript from the AML1 gene, with exons 5 and 6 scrambled. The novel transcript resisted RNase R digestion, indicating it is a circular RNA structure that may originate from products of mRNA alternative splicing. The expression of the novel transcript in different cells or cell lines of human and a number of other species matched those of the canonical transcripts. The discovery provides additional evidence that circular RNA could stably exist in vivo in human, and may also help to understand the mechanism of the regulation of the AML1 gene transcription.

  • In human U-937 cell exposed to ${\gamma}$-irradiation and $H_2O$$_2$, the level of mRNA efrpression in ceruloplasmin gene was measured by using comparative RT.PCR (reverse transcriptase-polymerase chain reaction). At the normal growth condition, the level of ceruloplasmin transcript was estimated as 8.2% and 0.0068% of hprt (hypoxantine phosphoribosyl transferase) transcript and of $\beta$-actin transcript, respectively. In U-937 cells exposed to a dose of 100 rad ${\gamma}$-irradiation, the level of ceruloplasmin transcript was increased about 2.7 and 1.6 fold compared to un-treated cell by using compensation with the levels of hprt and $\beta$-actin transcript. By contrast, the expression of ceruloplasmin gene in U-937 cells exposed to $H_2O$$_2$(50 $\mu$M, 24 h), was shown no significant difference compared to un-treated cell. These results indicated that the expression system of ceruloplasmin gene may react only some specific oxygen species, such as reactive oxygen species induced by ${\gamma}$-irradiation.

  • 연구배경 : 유전자 재결합 반응에 있어서 다른 종류의 RNA의 첨가에도 불구하고 유전자 반응에 영향이 없어야 여타 실험의 정량적 분석에 이용이 가능하다. 이에 저자들은 쥐를 대상으로 filter hybridization방법과 SP-A mRNA을 이용하여 비특이성 RNA 즉, 쥐의 비장 RNA의 첨가가 surfactant protein A (SP-A)의 유전자 재결합반응의 linearity, 상관계수 및 특이성에 미치는 영향을 알아보기 위하여 이 연구를 시행하였다. 방 법 : SP-A transcript mRNA의 정량, 즉 0, 0.1, 0.5, 1 및 2.5 ng에 비특성 RNA 즉 비장 RNA를 각각 0,1, 5 및 $10{\mu}g$을 첨가하여 filter hybridization 방법을 이용하여 SP-A mRNA양과 cpm과의 연관성을 비교정량측정하여 각각의 linearity, 상관계수 및 특이성의 분자생물학적 정도관리에 대한 비교 관찰을 하기 위하여 이 연구를 시행하였다. 결 과 : 1. 쥐의 spleen RNA 0, 1, 5, 10 및 $20{\mu}g$에 대한 cpm과의 표준곡선 및 상관계수는 Y=0.13X-19.35(X=cpm, Y=spleen RNA input)이고, 상관계수는 0.98이었다. 2. SP-A sense 전사체 0, 0.1, 0.5, 1.0, 2.5 및 5 ng에 대한 cpm과의 표준곡선 및 상관계수는 Y=0.00066X-0.046 (X=cpm, Y=SP-A mRNA 전사체)이고, 상관계수는 0.99이었다. 3. 쥐의 비장 RNA $1{\mu}g$을 첨가 후 SP-A sense 전사체 0, 0.1, 0.5, 1.0, 2.5 및 5 ng에 대한 cpm과의 표준곡선 및 상관계수는 Y=0.00056X-0.051(X=cpm, Y=SP-A mRNA 전사체)이고, 상관계수는 0.99이였다. 쥐의 비장 RNA $5{\mu}g$을 첨가 후 표준곡선은 Y=0.00065X-0.088 (X=cpm, Y=SP-AmRNA 전사체)이고, 상관계수는 0.99이였다. 쥐의비장 RNA $10{\mu}g$을 첨가 후 표준곡선은 Y=0.00051X-0.10 (X=cpm, Y=SP-A mRNA 전사체)이고, 상관계수는 0.99이었다. 결 론 : 이상의 결과는 비특이성 RNA인 비장 RNA의 첨가 후 SP-A sense mRNA양과 cpm과의 상관관계는 sense 유전자와 anti-sense 유전자의 유전자 재결합 반응에 있어서 다양한 양의 비특이성 RNA의 첨가나 오염에도 불구하고 linearity, 상관계수 및 그 특이성이 잘 유지됨을 입증해 준 결과라 생각된다.

Story of Johnsongrass Mosaic (Poty)virus in Australia

  • One of the major aims in studying plant viruses is to minimise the development of symptoms in infected plants. With the advent of in vitro transcript mediated research on plant viruses, substantial progress has been made. This article describes the biology of a plant specific RNA virus, Johnsongrass mosaic virus (JGMV), important to Australian sorghum and corn agriculture and, in particular, at a molecular level which of the RNA sequences in its genome that make it possible for the virus to move from cell to cell, and eventually spread systemically throughout the entire plant. The JGMV has caused considerable yield losses in maize and sorghum over a number of years in Australia. Incidents where 100% of the crop has been infected are on record. The use of this virus is convenient under laboratory conditions because it can be readily transmitted by mechanical inoculation with infected leaf sap, which obviates the need for maintaining aphid colonies. The JGMV is a single stranded positive sense RNA virus.

Upregulation of long non-coding RNA XIST has anticancer effects on epithelial ovarian cancer cells through inverse downregulation of hsa-miR-214-3p

  • Objective: The present study is to evaluate the biological functions of long non-coding RNA (lncRNA), X-inactive specific transcript, X-inactive specific transcript (XIST) in human epithelial ovarian cancer (EOC). Methods: XIST was upregulated in EOC cell lines, CAOV3 and OVCAR3 cells by lentiviral transduction. The effects of XIST overexpression on cancer cell proliferation, invasion, chemosensitivity and in vivo tumor growth were investigated, respectively. Possible sponging interaction between XIST and human microRNA hsa-miR-214-3p was further evaluated. Furthermore, hsa-miR-214-3p was overexpressed in XIST-upregulated CAOV3 and OVCAR3 cells to evaluate its effect on XIST-mediated EOC regulation. Results: Lentivirus-mediated XIST upregulation had significant anticancer effects in CAOV3 and OVCAR3 cells by suppressing cancer cell proliferation, invasion, increasing cisplatin chemosensitivity and inhibiting in vivo tumor growth. Hsa-miR-214-3p was confirmed to directly bind XIST, and inversely downregulated in XIST-upregulated EOC cells. In EOC cells with XIST upregulation, secondary lentiviral transduction successfully upregulated hsa-miR-214-3p expression. Subsequently, hsa-miR-214-3p upregulation functionally reversed the anticancer effects of XIST-upregulation in EOC. Conclusion: Upregulation of lncRNA XIST may suppress EOC development, possibly through sponging effect to induce hsa-miR-214-3p downregulation
