Background: Changes to work-life balance has increased the incidence of cervical cancer among younger people. A minor ginseng saponin known as ginsenoside Rk1 can inhibit the growth and survival of human cancer cells; however, whether ginsenoside Rk1 inhibits HeLa cell proliferation is unknown. Methods and results: Ginsenoside Rk1 blocked HeLa cells in the G0/G1 phase in a dose-dependent manner and inhibited cell division and proliferation. Ginsenoside Rk1 markedly also activated the apoptotic signaling pathway via caspase 3, PARP, and caspase 6. In addition, ginsenoside Rk1 increased LC3B protein expression, indicating the promotion of the autophagy signaling pathway. Protein processing in the endoplasmic reticulum signaling pathway was downregulated in Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses, consistent with teal-time quantitative PCR and western blotting that showed YOD1, HSPA4L, DNAJC3, and HSP90AA1 expression levels were dramatically decreased in HeLa cells treated with ginsenoside Rk1, with YOD1 was the most significantly inhibited by ginsenoside Rk1 treatment. Conclusion: These findings indicate that the toxicity of ginsenoside Rk1 in HeLa cells can be explained by the inhibition of protein synthesis in the endoplasmic reticulum and enhanced apoptosis, with YOD1 acting as a potential target for cervical cancer treatment.
Background: Ginsenoside Rk1, a saponin component isolated from heat-processed Panax ginseng Meyer, has been implicated in the regulation of antitumor and anti-inflammatory activities. Although our previous studies have demonstrated that ginsenoside Rg3 significantly attenuated the activation of NMDA receptors (NMDARs) in hippocampal neurons, the effects of ginsenosides Rg5 and Rk1, which are derived from heat-mediated dehydration of ginsenoside Rg3, on neuronal NMDARs have not yet been elucidated. Methods: We examined the regulation of NMDARs by ginsenosides Rg5 and Rk1 in cultured rat hippocampal neurons using fura-2-based calcium imaging and whole-cell patch-clamp recordings. Results: The results from our investigation showed that ginsenosides Rg3 and Rg5 inhibited NMDARs with similar potencies. However, ginsenoside Rk1 inhibited NMDARs most effectively among the five compounds (Rg3, Rg5, Rk1, Rg5/Rk1 mixture, and protopanaxadiol) tested in cultured hippocampal neurons. Its inhibition is independent of the NMDA- and glycine-binding sites, and its action seems to involve in an interaction with the polyamine-binding site of the NMDAR channel complex. Conclusion: Taken together, our results suggest that ginsenoside Rk1 might be a novel component contributable to the development of ginseng-based therapeutic treatments for neurodegenerative diseases.

Background and objective: Synthetic ginsenoside compounds G-Rp (1,3, and 4) and natural ginsenosides in Panax ginseng 20(S)-Rg3, Rg6, F4 and Ro have inhibitory actions on human platelets. However, the inhibitory mechanism of ginsenoside Rk1 (G-Rk1) is still unclear thus, we initiated investigation of the anti-platelet mechanism by G-Rk1 from Panax ginseng. Methodology: Our study focused to investigate the action of G-Rk1 on agonist-stimulated human platelet aggregation, inhibition of platelet signaling molecules such as fibrinogen binding with integrin αIIbβ3 using flow cytometry, intracellular calcium mobilization, fibronectin adhesion, dense granule secretion, and thromboxane B2 secretion. Thrombin-induced clot retraction was also observed in human platelets. Key Results: Collagen, thrombin, and U46619-stimulated human platelet aggregation were dose-dependently inhibited by G-Rk1, while it demonstrated a more effective suppression on collagen-stimulated platelet aggregation using human platelets. Moreover, G-Rk1 suppressed collagen-induced elevation of Ca2+ release from endoplasmic reticulum, granule release, and αIIbβ3 activity without any cytotoxicity. Conclusions and implications: These results indicate that G-Rk1 possess strong anti-platelet effect, proposing a new drug candidate for treatment and prevention of platelet-mediated thrombosis in cardiovascular disease.
Two isolates, Bacillus sp. BS87 and RK1, selected from soil in strawberry fields in Korea, showed high levels of antagonism towards Fusarium oxysporum f. sp. fragariae in vitro. The isolates were identified as B. velezensis based on the homology of their gyrA sequences to reference strains. BS87 and RK1 were evaluated for control of Fusarium wilt in strawberries in pot trials and field trials conducted in Nonsan, Korea. In the pot trials, the optimum applied concentration of BS87 and RK1 for pre-plant root-dip application to control Fusarium wilt was $10^5$ and $10^6$ colony-forming units (CFU)/ml, respectively. Meanwhile, in the 2003 and 2005 field trials, the biological control efficacies of formulations of RK1 were similar to that of a conventional fungicide (copper hydroxide) when compared with a non-treated control. The RK1 formulation was also more effective than BS87 in suppressing Fusarium wilt under field conditions. Therefore, the results indicated that formulations of B. velezensis BS87 and RK1 may have potential to control Fusarium wilt in strawberries.
목적: Cinnamic acid의 유도체들중에서 항돌연변이 효과가 보고된 바 있어, 본 연구에서는 E. coil PQ37균주를 사용한 SOS chromotest를 이용하여 약 170개 cinnamic acid의 유도체들의 항돌연변이 효과를 확인하고자 하였고, 1차로 확인된 수종의 유도체들에 대한 결과를 보고하고자 한다. 방법: SOS chromotest는 sul A:: lacz fusion strain 인 E. coli PQ37 에서의 chemical로 인한 DNA damage에 대한 SOS response를 $\beta$-galactosidase enzyme activity level로서 측정하는 실험이며, m-RNA 또는 protein synthesis에 대한 test chemicals의 cytotoxic저해효과를 알아보기 위하여 alkaline phosphatase를 병행측정하여 보완하였다. 결과 및 고찰: (-)S-9 경우는 4-nitroquinoline 1-oxide(4-NQO)를 유도물질로 하였으며 대략 Induction factor가 9.7 정도였고, (+)S-9의 경우는 aflatoxinB$_1$을 유도물질로 하였고 이때의 Induction factor는 13.8 정도였다. 이결과 1차로 test된 cinnamic acid유도체 6개중에서 RK001, RK002, RK003, RK004, RK005는 거의 항돌연변이 효과를 나타내지 않았으나, RK006은 항돌연변이 효과를 보여주어 이들의 Structure-Activity Relationship (SAR) 및 Dose-response와 RK006의 항돌연변이 효과의 mechanism에 관해 M13 mp2 viral DNA의 유전자 염기서열을 이용하여 항돌연변이 기전연구를 수행중이다.
In this paper, the finite element modeling of the RK4 rotor kit system (RK4) and then frequency analysis and transient analysis, and was compared with the actual experimental results. RK4 manufactured by General Electric for the purpose of education and research. It is composed of two shaft, Two shaft is connected using a flexible coupling, one disk is mounted. The analytical model is modeled by using the ANSYS finite element analysis program commercially available. Based on impact hammer test results, material properties and the stiffness of the bearing and coupling was tuned. Considering the operating conditions and the vibration response of the analytical model were compared with experimental results.
In this paper, an error-free Butcher algorithm is introduced to study the singular system of a linear electrical circuit for time invariant and time varying cases. The discrete solutions obtained using Runge-Kutta (RK)-Butcher algorithms are compared with the exact solutions of the electrical circuit problem and are found to be very accurate. Stability regions for the single term Walsh series (STWS) method and the RK-Butcher algorithm are presented. Error graphs for inductor currents and capacitor voltages are presented in a graphical form to show the efficiency of the RK-Butcher algorithm. This RK-Butcher algorithm can be easily implemented in a digital computer for any singular system of electrical circuits.

Meshfree methods are known to have the capability to overcome the strict regularization requirements and numerical instabilities that encumber the finite element method (FEM) in large deformation problems. They are also more naturally suited for problems involving material perforation and fragmentation. To take advantage of the high efficiency of FEM and high accuracy of meshfree methods, a coupled finite element (FE) and reproducing kernel (RK, one of the meshfree approximations) formulation is described in this paper. The coupling of FE and RK approximation is implemented in an evolutionary fashion, where the extent and location of the evolution is dependent on a triggering criteria provided by the material constitutive laws. To enhance computational efficiency, Gauss quadrature is applied to integrate both FE and RK domains so that no state variable transfer is required when mesh conversion is performed. To control the hourglassing that might occur with 1-point integrated hexahedral grids, viscous type hourglass control is implemented. Meanwhile, the FEM version of the K&C concrete (KCC) model was modified to make it applicable in both FE and RK formulations. Results using this code and the KCC model are shown for the modeling of concrete responses under quasi-static, blast and impact loadings. These analyses demonstrate that fragmentation phenomena of the sort commonly observed under blast and impact loadings of concrete structures was able to be realistically captured by the coupled formulation.
This study investigated changes in the quality of fermented red snow crab Chionoecetes japonicus sauce with or without Aspergillus kawachii koji and added salt. Samples were divided into four groups depending on whether koji was added and the amount of salt: RC15, 15% added salt, no koji; RC20, 20% added salt, no koji; RK15, 15% salt plus 10% koji; and RK20, 20% salt plus 10% koji. The samples were fermented at 20±2℃ for 4 months. During the fermentation period, the moisture contents of the four types of sauce decreased while the crude ash and protein contents increased. The pH of the RK groups decreased and was lower than in the RC groups. The acidity of the RK groups increased and was higher than in the RC groups. Both the total nitrogen (TN) and amino nitrogen (AN) levels increased continuously and were higher in the RK groups than in the RC groups. The volatile basic nitrogen (VBN) content increased rapidly and was higher in the RC groups than in the RK groups. The color did not differ significantly among the four groups. The viable cell counts in the four groups increased and no coliforms were detected. The total free amino acid and glutamic acid contents were highest in the RK15 group and the main amino acids in RK15 were aspartic acid, glutamic acid, alanine, leucine, phenylalanine, and lysine. Overall acceptance was significantly higher for the RK groups than the RC groups and RK15 ranked highest among the four sauces. These results suggest that Aspergillus kawachii koji is beneficial for processing fish sauce made using red snow crab.
