Anti-Inflammatory Effects of Extracts from Ligustrum ovalifolium H. Leaves on RAW264.7 Macrophages (RAW264.7 대식세포에서 왕쥐똥나무잎 추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effects of Ligustrum ovalifolium H. (LOH) leaf extracts on RAW264.7 macrophages. Cell toxicity was determined by MTT assay. We evaluated the anti-inflammatory effects of LOH extracts by measuring nitric oxide (NO), reactive oxygen species (ROS), inducible NOS (iNOS) production, and cyclooxygenase-2 (COX-2) expression by Western blotting. LOH ethanolic extracts (0.05, 0.1, and 0.2 mg/mL) significantly suppressed LPS-stimulated production of NO. The intracellular ROS level also significantly decreased. LOH ethanolic extracts reduced the expression of iNOS and COX-2 proteins. The present results show that LOH ethanol extract has potent anti-inflammatory effects on RAW264.7 macrophages. These results also suggest that the anti-inflammatory effects of LOH extracts may be related to the inhibition of LPS-stimulated ROS and NO production. Therefore, ethanolic extracts of LOH leaves may be utilized as a good source of functional foods for protection against inflammatory diseases.

Inhibitory Effects of Lyophilized Dropwort Vinegar Powder on Adipocyte Differentiation and Inflammation (미나리 발효 식초의 지방세포 분화억제 및 항염증 효과)

  • Obesity, which is characterized by a state of mild chronic inflammation, is known to cause metabolic diseases. This study was carried out to investigate the effect of lyophilized dropwort vinegar powder (DVP) on adipocyte differentiation and inflammation in T3-L1 preadipocyte and RAW 264.7 macrophage cell lines. DVP inhibited the differentiation of 3T3-L1 preadipocytes induced by a mixture of IBMX, dexamethasone, and insulin (MDI). Western blot analysis of cell lysates showed that DVP decreased the levels of two major transcription factors involved in adipogenesis, peroxisome proliferator- activated receptor-${\gamma}$ (PPAR-${\gamma}$) and CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$). DVP also significantly suppressed lipopolysaccharide (LPS)-induced production of nitric oxide (NO), and this was accompanied by a decrease in inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression. These results demonstrate that DVP inhibits MDI-induced adipocyte differentiation of 3T3-L1 cells and LPS-induced inflammation in RAW 264.7 macrophage cells. The findings indicate that this natural product may be a good candidate as to prevent metabolic diseases.

The Effects of Euonymi Ligunum Suberalatum Phamacopuncture on Nitric Oxide and Interleukin-6 Production in Macrophage (귀전우(鬼箭羽)약침액이 대식세포의 Nitric Oxide와 Interleukin-6의 생성에 미치는 영향)

  • Objectives : The objective of this study is to study the effects of Euonymi Lignum Suberatatum pharmacopuncture solution on NO and IL-6 production in macrophage. Methods : At first, the RAW 264.7 macrophage was subclutured. In order to evaluate cytotoxicity, MTT assay performed. Then, the cell was induced by LPS, INF-$\gamma$ and Experimental groups were divided into five(Normal, Control, Euonymi Lignum Suberatatum 100, 200, $300{\mu}g/m{\ell}$). Then Euonymi Lignum Suberatatum pharmacopuncture solution was put into cell. We measured IL-6, iNOS, NO. Results : The cytotoxic effect of Euonymi Lignum Suberatatum pharmacopuncture solution in RAW 264.7 macrophage was not appeared. $300{\mu}g/m\ell$ Euonymi Lignum Suberatatum pharmacopuncture solution inhibited IL-6 production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Euonymi Lignum Suberatatum pharmacopuncture solution inhibited iNOS revelation in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages. All group of Euonymi Lignum Suberatatum pharmacopuncture solution inhibited NO production in LPS, INF-$\gamma$-stimulated RAW 264.7 macrophages significantly. Conclusions : Our study demonstrated that Euonymi Lignum Suberatatum pharmacopuncture solution had an inhibition effect on NO production, iNOS revelation, IL-6 production. So Euonymi Lignum Suberatatum pharmaco puncture solution may have an Anti-inflammation effect.

Inhibitory Effects of Soyeum Pharmacopuncture (SPP) on LPS-induced Inflammation Related Cytokine Expressions of RAW 264.7 cells (소염약침의 LPS로 유도된 RAW 264.7 세포의 염증성 사이토카인 발현에 미치는 영향)

  • Aim This study was done to investigate whether SPP has inhibitory effects on the activation of RAW 264.7 cells. Method In tumor necrosis factor-a (TNF-a)/ interleukin-1b (IL-1b) and IL-6, the mRNA expression of molecular indicators related to inflammatory changes of the Reumatoid Arthritis (RA) were examined using quantitative real-time PCR. Results The treatment of SPP significantly suppressed the expression of proinflammatory cytokines and chemokines such as TNF-a, IL-1b, IL-6 compared with the control. The expression of NOS-II was considerably reduced, which was accompanied by a reduction in the production of nitric oxide (NO). It also reduced the expression of $TNF-{\alpha}$ in serum of Balb/c mice compared with control group. Conclusion SPP is an effective herbal material for suppressing the inflammation related cytokines of RAW 264.7 cells.

Anti-inflammatory effect of SD-01 in RAW 264.7 cells (마우스대식세포주인 RAW 264.7에서 SD-01의 항염증 활성 연구)

  • Objective : The aim of this study was to investigate anti-inflammatory activity of SD-01 methanol extract in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Methods : Cytotoxic activity of SD-01 methanol extract on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines and $PGE_2$ were measured by ELISA method. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), $I{\kappa}$-B-alpha and nuclear NF-${\kappa}$ B p65 expression were detected by western blot. Results : Our results indicated that methanol extract of SD-01 significantly inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-$\alpha$, IL-$1\beta$, IL-6 and MCP-1 production in RAW 264.7 cells. Moreover, methanol extract of SD-01 treatment also blocked LPS-induced NF-kB activation. Conclusion : These findings indicate that methanol extract of SD-01 inhibits the production of pro-inflammatory mediators and cytokines via suppression of NF-${\kappa}$ B activation. Take together, these results indicate that methanol extract of SD-01 has the potential for use as an agent of anti-chronic inflammatory diseases.

Effect of Scutellariae Radix Water Extract on Hydrogen Peroxide Production in LPS-induced RAW 264.7 Mouse Macrophages (황금(黃芩) 물추출물이 LPS로 유발된 마우스 대식세포의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate effects of Scutellariae Radix water extract on hydrogen peroxide production in LPS-induced RAW 264.7 mouse macrophages. Methods : Scutellariae Radix produced from South Korea (SK) and Scutellariae Radix produced from China (SC) were extracted by hot water. Effects of SK and SC on hydrogen peroxide production in LPS-induced RAW 264.7 were measured by dihydrorhodamine 123 assay after 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL. Results : SK significantly increased hydrogen peroxide production in LPS-induced RAW 264.7 cells for 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL (P < 0.05). But SC did not represent any significant effect on hydrogen peroxide production in LPS-induced RAW 264.7 cells. Conclusions : These results suggest that Scutellariae Radix, especially produced from South Korea, has the immune-enhancing property related with its increasement of bacteriocidal hydrogen peroxide production in LPS-induced macrophages.

Anti-Inflammatory Effects of Xanthoceras sorbifolia Seeds Oil on RAW264.7 Macrophages and TPA-Induced Ear Edema Mice (RAW264.7 대식세포와 급성염증유발 동물모델에서 문관나무 종자유의 염증억제 효과)

  • This study investigated the anti-inflammatory effects of Xanthoceras sorbifolia of seeds oil on RAW264.7 macrophages and TPA (12-O-tetra decanoylphorbol-acetate)-induced ear edema mice. MTT assay method to measure cytotoxicity was formed in RAW264.7 cell. The anti-inflammatory effect was measured by ability to inhibit production nitric oxide (NO) in RAW264.7 cell. Hexane and eight-percent methanol fractions from Xanthoceras sorbifolia seeds oil did not show cytotoxicity. Hexane and eight-percent methanol fractions were showed significantly inhibitory effect on NO production. TPA-induced acute edema was developed in the mouse ears, and Xanthoceras sorbifolia seeds oil dissolved in acetone was applied to inflamed ears. It was found out that Xanthoceras sorbifolia seeds oil could significantly reduce th ear swelling, compared to the control. Overall results indicate that the Xanthoceras sorbifolia seeds oil has anti-inflammatory activity and could be used as a resource of anti-inflammatory materials.

Chemical Constituents from the Aerial Parts of Artemisia absinthium and Its Inhibitory Effects of Nitric Oxide Production in RAW264.7 and BV2 Microglia (쓴쑥 지상부의 화학성분과 RAW264.7 및 BV2 미세아교세포에서 Nitric Oxide 생성 억제효과)

  • Six compounds, eupatilin (1), dammaradienyl acetate (2), glutinol acetate (3), $3{\beta}-acetoxyoleanan-12-one$ (4), taraxasterol (5) and quercetin-3,4'-dimethyl ether (6) were isolated from the aerial parts of Artemisia absinthium. The chemical structures of compounds 1-6 were determined by the basis of physico-chemical properties and spectroscopic methods such as 1D and 2D NMR. Among them, compounds 2-5 were isolated from this plant for the first time. The inhibitory effects of these isolated compounds against nitric oxide (NO) production in LPS-induced RAW264.7 cells or BV2 microglia were also examined. Among the tested compounds, compound 1, eupatilin, inhibited the production of NO in LPS-induced RAW264.7 cells and BV2 microglia, respectively.

In vitro Antiinflammatory Activity of the Essential Oil from Ligularia fischeri var. spiciformis in Murine Marcrophage Raw 264.7 Cells (넘취 정유의 Murine Macrophage Raw 264.7세포에서 In vitro 항염효과)

  • In the present study, effects of essential oils isolated from various plants have been evaluated on lipopolysaccharide (LPS)-induced release of nitric oxide (NO), prostaglandin E$_2$(PGE$_2$) and tumor necrosis factor-a (TNF-$\alpha$) by the macrophage RAW 264.7 cells. Among the tested essential oils, essential oil of Ligularia fischeri var. spiciformis (LF-oil) significantly inhibited the LPS-induced generation of NO, PGE$_2$ and TNF-$\alpha$ in Raw 264.7 cells. Consistent with these observations, the expression of inducible NO synthase (iNOS) and cyclooxygenase (COX)-2 enzyme was inhibited by LF-oil in a concentration-dependent manner. Thus, this study suggests that inhibition of release of iNOS, COX-2 expression, and TNF-$\alpha$ by the essential oil of Ligularia fischer may be one of the mechanisms responsible for the anti-inflammatory effects of this medicinal plant.

Anti-inflammatory Effect of Imyosan Extract is more potent than that of its Component Herb Extracts in Murine Macrophages (마우스 대식세포인 RAW 264.7에 대한 이묘산(二妙散)의 메탄올 추출물의 항염증 효과)

  • Objectives : Imyosan (IMS), a drug preparation comprised of Phellodendri Cortex (PC) and Atractylodis Rhizoma (AR), is commonly used as a traditional herbal medicine in Korea and China for the treatment of various inflammatory diseases. However, little is known about the effect of IMS and its component herbs on inflammatory mediators in RAW 264.7 cells. Therefore, in this study, methanol extracts of IMS and its component herbs were examined to determine if they inhibited inflammatory effects in RAW 264.7 cells. Methods : Cytotoxic activity of IJHT and its components on RAW 264.7 cells was using 5-(3-carboxymethoxyphenyl)-2H-tetrazolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were detected by western blot. Results : Methanol extract of IMS and its component herbs were significantly reduced iNOS and COX-2 expression as well as NO, PGE2, $IL-1{\beta}$ and IL-6 production in RAW 264.7 cells. Conclusions : The results of this study indicated that the anti-inflammatory effect of Imyosan extract is more potent than that of extracts of its component herbs in macrophages.
