Preventing of Bone Loss in Ovariectomized Rats by Salvia miltiorrhiza Radix Root Extract

  • Salvia miltiorrhiza Radix root extracts (SMR) was evaluated for inhibition of the progress of bone loss induced by ovariectomy (OVX) in rats. We measured body weight and bone histomorphometry in sham, OVX and SMR-administered OVX rats. From light microscopic analyses, porous or erosive appearances were observed on the surface of trabecular bone of tibia in OVX rats, whereas those of the same bone in sham rats and in SMR-administered rats were composed of fine particles. The trabecular bone area and trabecular thickness in OVX rats were decreased by $50\%$ from those in sham rats, and these decreases were completely inhibited by administration of SMR for 7 weeks. In this study, the mechanical strength in femur neck was significantly enhanced by the treatment of SMR for 7 weeks. In OVX rats, free $T_3$ was normal in all cases, whereas free $T_4$ was significantly increased. Although there was no difference between OVX and SMR-administered rat in $T_3$ levels, we have found significant difference between them in $T_4$ level. These results strongly suggest that SMR may be beneficial for preventing bone loss in OVX rats.

Vitamin $B_2$ 결핍이 Streptozotocin 유발 당뇨 흰쥐의 에너지대사에 미치는 영향 (The Effect of Vitamin $B_2$ Deficiency on Fuel Metabolism in Streptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of vitamin B2 deficiency on fuel metabolism in streptozotocin-induced diabetic rats. Thirty rats were fed a vitamin B2 deticient diet(-B2) or a control diet (+B2) for 2 weeks and then subdivided into 3 groups respectively : base group, one day diabetic group and three day diabetic group. Diabetes of the rats were induced by streptozotocin injection into the tail vein. Glucose, glycogen, protein, alanine, triglyceride and free fatty acid were compared in plasma, liver, skeletal muscle of rats. Also, the total urinary nitrogen and glucose excertion were compared. Compared with +B2 rats, the increase of plasm glucose in -B2 rats due to the diabetes tended to be smaller. After diabetes were induced, the levels of plasma protein and alanine was significantly decreased and the urinary nitrogen excretion was significantly increased in -B2 rats. The level of plasma free fatty acid was increased continuously in B2 rats while increased at the first day and decreased at the third day diabetes was induced in +B2 rats. These results suggest that vitamin B2 deficiency increase protein catabolism due to the decrease of fatty acid oxidation. Thus, vitamin B2 deficiency in diabetes impair the adaptation of animals to the fuel metabolism and aggravate the body protein wasting which is one of the chronic complications of diabetes.

BHT-C의 허혈성 뇌졸중 동물에서의 뇌부종 억제효과 (Effect of BHT-C extract on the infarction in cerebral ischemic rats)

  • Objectives : BHT has been commonly used to treatment of brain disorders in Oriental clinic in Korea. The purpose of this study was to determine the inhibitory effect of modified BHT-C extract on the transient forcal cerebral ischemia in rats. Method : We prepared ischemic rats by the transient middle cerebral artery occlution(MCAO; 90 min occlusion and 144 h reperfusion) in rat brains. BHT-C extract (100 and 200 mg/kg, i.p.) was administered every day after the onset of MCAO until 6 day. Result : BHT-C extract increased survival rate of ischemic rats compared with vehicle-treated rats. BHT-C extract treated rats (100 and 200 mg/kg) were shown a significant reduction in infarct volume compared with vehicle-treated rats. Conclusions : These results suggest that BHT-C extract may contribute to its protective effects in brain ischemia through the reduction of brain infarction.

Fenofibrate reduces adiposity in pregnant and virgin rats but through different mechanisms

  • Fenofibrate has been proven to reduce adiposity. Since gestation produces an increase in white adipose tissue (WAT) mass, we comparatively studied this drug-effect in virgin and pregnant rats. Fenofibrate reduced lumbar WAT weight in both pregnant and virgin rats. Fenofibrate treatment did not modify plasma free fatty acid (FFA) concentration in virgin rats, it greatly increased it in pregnant animals. Remarkable differences between the two groups were obtained for two proteins related to fatty acid oxidation and esterification and storing. Respectively, the mRNA levels of carnitine palmitoyltransferase I (CPT-I) were increased by the fenofibrate only in the virgin rats and a similar finding was observed for the expression of phosphoenolpyruvate carboxykinase (PEPCK). These findings indicate that fenofibrate reduces adiposity in pregnant and virgin rats through different mechanisms: a) in virgin rats, by promoting fatty acid oxidation; and b) in pregnant rats, by enhancing fatty acid output.

Single Oral Toxicity Study of 3-Methoxy-6-Allylthiopyridazine in Rats

  • The single dose toxicity of 3-methoxy-6-allylthiopyridazine (K-6) was studied with Sprague-Dawley rats. K-6 was administered orally with dosages of 4.0, 3.0, 1.5, 1.0, 0.5, 0.25 and 0.1 g/kg to the rats. We observed daily the number of death, clinical signs, body weights and gross findings. All rats (6) were dead within a day after administration when doses of 4.0and 3.0 g/kg were administered. When dose of 2.0 g/kg was administered, 2 rats among 3 rats were dead. Any significant toxicity below 1.5g/kg of K6 was not observed when the different doses of 1.5, 1.0, 0.5, 0.25 and 0.1 g/kg were administered to 6 rats respectively.

갈근탕(葛根湯) 투여가 흰 쥐의 스트레스 저항 반응에 미치는 영향에 관한 연구 (Studies on the Effect of 'Gal Geun Tang' upon Stress Resistance of Rats)

  • Oxygen consumption rate has been determined to study the effect of 'Gal Geun Tang' upon stress resistance of rats. The oxygen consumption rate of liver tissue increased slightly when normal rats were given the Gal Geun Tang infusion only. When rats were given the infusion after an injection of typhoid vaccine, the elevated boby temperature and oxygen consumption rate due to typhoid vaccine fell to normal level. From this result, it is concluded that 'Gal Geun Tang' has a function that may normalize abnormally strained liver metabolism. Oxygen consumption rate of the rats significantly increased when rats were exposed to cold condition. However, under the same condition it rather maintained the normal consumption rate when the rats were given previously "Gal Geun Tang". It is suggested that 'Gal Geun Tang' has an effects that impairs the immune response from cold stress. The rats that were given Cinnamomi Ramulus('Ge Ji Tang') showed less effect than the group of 'Gal Geun Tang'.

흰쥐의 간 Xanthine Oxidase 활성에 미치는 Ethionine의 영향 (An Effect of Ethionine Treatment on the Activity of Hepatic Xanthine Oxidase in Rats)

  • Liver and serum xanthine oxidase(XOD) activity were determined in rats treated with DL-ethionine. Concomitantly, the enzyme activity was compared with that of rats treated with CCl$_{4}$, actinomycin D and those fed a protein depleted diet. The activity of XOD in liver was inhibited by injection of ethionine to the rats. But, there were no differences in activity of serum XOD between control group and ethionine-treated rats. And the pattern of changes in enzyme activity of serum and liver in ethionine-treated rats, was similar with actinomycin D treated rats or those fed a protein depleted diet. On the other hand, the activity of XOD was rather elevated both in serum and liver by injection of CCl$_{4}$ to rats.

흰쥐에 toluene 투여가 백혈구 xanthine oxidase 및 superoxide dismutase 활성에 미치는 영향 (Effect of Toluene Treatment on the Xanthine Oxidase and Superoxide Dismutase Activities in Leukocyte of Bacterial Infected Rats)

  • This study was designed to observe the effect of tohiene pretreatment on leukocyte variation in whole blood and the oxygen free radical generating, scavenging enzyme activities in neutrophil of bacteria infected rats. Toluene was administered 7 times intraperitoneally at levels of 9.45 mM/kg body weight to the rats and then infected with S. aureus $2\times10^7$ cfu/ml. The toluene treated-rats showed the significantly decreased numbers of lymphocyte and monocytes, but the similiar numbers of neutrophils with the control. Furthermore the increased neutrophils in blood of bacteria infected rats were reduced by the toluene pretreatment. Concomitantly the increased activities of xanthine oxidase and superoxide dismutase in neutrophil of bacteria infected rats were also decreased by the toluene pretreatment. On the other hand, injection of benzaldehyde to rats also led to similiar results in the count of leukocytes, xanthine oxidase and superoxide dismutase activities of neutrophil with those of toluene treated rats. These data suggest that toluene and its intermediate metabolite, benzaldehyde influence on the phagocytosis and defence mechanism of neutrophil.

Cholesterol Lowering Effect of Enzymatic Hydrolysates of Squid in Rats

  • This study evaluated effects of enzymatic hydrolysates of squid on cholesterol lowering in rats. Thirty male rats were blocked into 3 groups [high cholesterol diet (control), 5% normal squid, and 5% enzymatic hydrolysates of squid] and were raised for 10 weeks. Triglyceride level in enzymatic hydrolysates of squid-fed rats was lower than that in the control. Serum low density lipoprotein-cholesterol level followed in the order of control>normal squid>enzymatic hydrolysates. Serum high density lipoprotein-cholesterol level in enzymatic hydrolysates of squid-fed rats was higher than that in control rats. Liver cholesterol level in enzymatic hydrolysates of squid-fed rats was lower than that in control rats.

삶은 계란을 섭취한 흰쥐의 심전도 (Electrocardiograms in the Rats Fed Diets with Boiled Eggs)

  • Electrocardiograms in rats fed diets with boiled eggs for 30 days was investigated. Amplitudes of P,Q and R waves were not significant differences among treatment groups. Amplitude of S wave in rats fed the diet with 95% boiled eggs was significantly tended to be increased compared with other groups(P<0.05). Amplitude of T wave in the rats fed the diet with 0% boiled eggs showed the highest values, and there were significant difference among treatment groups fed diets with 0% boiled eggs, 25% and 95% boiled eggs (P<0.05). Durations of P and PQ(PR) waves were high in the rats fed diets with 25% and 50% boiled eggs (P<0.05). Duration of QRS complex showed low in the rats fed diet 0% boiled eggs but not significant difference among treatment groups. Duration of QT was high in the rats fed diet with 0% boiled eggs(P<0.05). This result is assumed that electrocardiograms in the rats is not changed to intake the boiled eggs.
