Modification of cell wall structural carbohydrate in the hybrid poplar expressing Medicago R2R3-MYB transcription factor MtMYB70

  • The isolation, cloning, and characterization of an R2R3-MYB transcription factor gene (MtMYB70) from the model legume Medicago truncatula is reported. MtMYB70 consists of a 768-bp coding sequence corresponding to 255 amino acids. Sequence alignment revealed that MtMYB70 cDNA contains conserved R2R3-type MYB domains with highly divergent C terminal regions. MtMYB70 was found to have relatively low sequence homology with known R2R3-MYB genes. Phylogenetic analysis placed the R2R3-MYB domain of MtMYB70 closest to PtMYB1, a known activator of lignin biosynthesis. Overexpression of MtMYB70 under the control of the 35S promoter in transgenic poplar did not cause a significant difference in total lignin content relative to the control, but glucan content was significantly increased in transgenic poplar. Therefore, MtMYB70 might have regulatory role in the biosynthesis of cell wall structural carbohydrates.

Experimental study for the pressure drop of R-22 and R-4O7C during the condensation in the micro-fin tubes (마이크로핀관내에서 R-22와 R-4O7C의 응축압력강하 특성에 관한 실험적 연구)

  • Experiments were conducted for the investigation of pressure drop inside horizontal micro-fin tubes during the condensation of R-22 and ternary refrigerant. R-407C(HFC-32/125/134a 23/25/62 wt%) as a substitute of R-22. The condenser is a double-tube and counterflow type heat exchanger which is consisted with micro-fin tubes having 60 fins with a length of 4000mm, outer diameter of 9.53mm and fin height of 0.2mm. The mass velocity varied from 102.1 to $301.0kg/(m^2{\cdot}s)$ and inlet quality was fixed as 1.0. From the experimental results. the pressure drop for R-407C was considerably higher than that for R-22. The value of PF(penalty factor) for both of refrigerants was not bigger than the ratio of micro-fin tube area to smooth tube area. Based on the experimental data. correlation was Proposed for the prediction of frictional pressure drop during the condensation of R-22 and R-407C inside horizontal micro-fin tubes.

  • Interactions between factor Va (HFVa) and membrane phosphatidylserine (PS) regulate the activity of the prothrombinase complex. I have previously shown that two solvent exposed hydrophobic residues located in the C2-domain, Trp2063 and Trp2064, are required for binding to immobilized PS and for expression of procoagulant activity on membranes containing 5% PS. In order to fully define the functional importance of these two residues I have expressed and isolated recombinant factor Va (rHFVa) W2063A/W2064A double mutant. In contrast to the native protein the two glycoforms resulting from alternative glycosylation of Asn2181 eluted as a single peak with rHFVa1 W2063A/W2064A eluting on the leading edge and rHFVa2 W2063A/W2064A eluting on the trailing edge. The double mutant rHFVa2 W2063A/W2064A expressed little or no procoagulant activity on membranes containing 1-10% mol % PS. In contrast, the procoagulant activity of this mutant was slightly greater than the native protein on membranes containing>18 mol % PS. The binding of rHFVa2 W2063A/W2064A to immobilized phospholipid vesicles was markedly reduced compared to the native protein in a surface plasmon resonance binding assay. I conclude that Trp2063 and Trp2064 are required for high affinity binding of factor Va to PS membranes and that this interaction is necessary for assembly of the prothrombinase complex on membranes containing physiological concentrations of PS.

  • Upon Pd(0)-catalyzed coupling reaction of (2R)-2-(N,N-ditosylimido)-3-butenyl methyl malonate (4) which was selected for the total synthesis of A-factor, (3R)-2-(6-methylheptanoyl)-3-hydroxy methyl-4-buttinolide (1), an unexpected 14-membered cyclic compound, bis(2-methoxycabonyl-(4E)-hexenolide) (15) was obtained. The structure of this compound was conformed by X-ray crystallography. This result implies that this method can be applied the synthesis of various size of symmetrical macrocyclic compounds.

  • In this pacer, it is examined and confirmed that the R factor which is compensated after analyzing the characteristics of superconductor resulted from this experimented theory with the computer simulation, is refined to R$\sub$WP/=8.83[%], R$\sub$P/=6.47[%], R$\sub$I/=10.08[%], RF=7.19[%], R/eub E/=3.76(%), and on the basis of these experimental data, the significant parameter such as the scale factor(s) and the zero point shift(Z) and FWHM value(U,V,W) are optimized as follows; S =72.0827E-3, Z=0.2146, U=4.2761E-2, V=1.7983E-2, and W=2.6768E-2

  • In this study, we estimated a CES production function for the Korean economy. We have found in the empirical results that the elasticity of the factor substitution is less than one and that the Korean economy exhibits labor-saving technological progress. In addition, we obtained the regression coefficient of R&D investment on technological change, i.e., the elasticity of R&D investment with respect to the technological change was 0.26% point. It implies that if R&D stock increases by 1%, labor efficiency increases 0.26% point through technological progress which is Hicksian non-neutral. It confirms that innovation-based growth strategy by increasing R&D investment would be effective on the one hand. Some policy consideration on the other might be needed for an increase in employment which is offset by technological progress.

  • MatR in Rhizobium trifolii is a malonate-responsive transcription factor that regulates the expression of genes, matABC, enabling decarboxylation of malonyl-CoA into acetyl-CoA, synthesis of malonyl-CoA from malonate and CoA, and malonate transport. According to an analysis of the amino acid sequence homology, MatR belongs to the GntR family The proteins of this family have two-domain folds, the N-terminal helix-turn-helix DNA-binding domain and the C-terminal ligand-binding domain. In order to End the malonate binding site and amino acid residues that interact with RNA polymerase, a site-directed mutagenesis was performed. Analysis of the mutant MatR suggests that Arg-160 might be involved in malonate binding, whereas Arg-102 and Arg-174 are critical for the repression activity by interacting with RNA polymerase.

  • A development in method of evaluating the measurement precisions using approximate F tests and variance components from expected mean square (EMS) is investigated. The research proposes three-factor mixed measurement models with the fixed and random factors. Unrestricted and unconstrained design work was rarely studied, while restricted and constrained designs have been significantly discussed. The unrestricted and unconstrained designs assume to be an independence of interaction. The proposed evaluation method about the measurement precisions can be extended to four-factor random measurement model or mixed measurement model. The study also presents the three evaluation indexes of precisions such as R&RTR (Reproducibility & Repeatability-To-Total Precision Ratio), PTR (Precision-To-Tolerance Ratio), and SNR (Signal-To-Noise Ratio). Numerical examples are proposed to evaluate the approximate F tests with Satterthwaite degrees of freedom and three indexes using the measurement precisions from EMS.

Investigation on the Selection of Capillary Tube for the Alternative Refrigerant R-407C

  • The capillary tube performance for R-407C is experimentally investigated. The experimental setup is a real vapor-compression refrigerating system. Mass flow rate is measured for various diameter and length while inlet pressure and degree of subcooling are changed. These data are compared with the results of a numerical model. The mass flow rate of the numerical model is about 14% less than the measured mass flow rate. It is found that mass flow rate and length for R-407C are less than those for R-22 under the same condition. Based on this experimental study and the numerical model, a set of design charts for capillary tube of R-407C is proposed.

The Reliability and Validity of a Drinking Refusal Self-Efficacy Questionnaire-Revised (DRSEQ-R) in Korean College Students (한국 대학생을 대상으로 한 음주거절효능감 도구 [Drinking Refusal Self-Efficacy Questionnaire-Revised: DRSEQ-R]의 신뢰도 및 타당도 검증)

  • Purpose: This study was to test the reliability and validity of a Drinking Refusal Self-Efficacy Questionnaire-Revised (DRSEQ-R) in Korean college students, Methods: The DRSEQ-R was designed to assess an individual's belief in their ability to refuse drinking alcohol by Oei et al. (2005) and consisted of three factors (social pressure, opportunity, and emotional relief). A methodological study design with an exploratory factor analysis for validity and correlation coefficients for reliability was used. DRSEQ-R was translated into Korean and a translation equivalency was obtained. DRSEQ-R was tested with a sample of 201 college students in Korea. The subjects consisted of 106 males and 95 females. Principal component factor analysis was used for construct validity and Cronbach's alpha was used to evaluate for internal consistency of the instrument. Results: The factor analysis showed three factors explaining 66.3% of total variance and the corresponding factors were emotional relief (48.01%), drinking contextual opportunity (11.18%), and social pressure (7.14%). The internal consistency was acceptable (Cronbach's alpha=.937). All three factors of DRSEQ-R negatively correlated with alcohol dependency and a heavy drinker showed a lower DRSEQ-R than a moderate drinker. Conclusion: Based on the findings, DRSEQ-R is a reliable and valid instrument to measure for drinking refusal self-efficacy in Korean college students.
