Changes on the Quercitrin Content in the Preparation for the Leaves of Cedrela sinensis (참죽나무잎의 조리시 quercitrin의 함량 변화)

  • When we prepared the cooked namul, bugak and kimchi by the leaves of Cedrela sinensis, the changes of quercitrin which was isolated from the leaves of this plant in each preparations were analysed by HPLC. Separation by reversed phase chromatography on u-Bondapak C/Sub 18/ column was achieved by isocratic elution with THF-dioxane-MeOH-HOAc-5% H/Sub 3/PO/Sub 4/-H/Sub 2/O (145: 125:50:20:2:658). When Kimchi was stored at 5$^{\circ}C$ for 20 days, the content of quercitrin in the methanol extract of Kimchi was 7.21% and Kimchi at 20$^{\circ}C$ was reduced by 5.78% (w/w). Contents of quercitrin in the leaves of Cedrela sinensis kimchi obsered to be gradually decreased during storage at 5$^{\circ}C$ and 20$^{\circ}C$. Contents of quercitrin of stored kimchi at 5$^{\circ}C$ was higher than that stored at 20$^{\circ}C$. The contents of quercitrin in Namul and Bugak were 13.06 and 5.03% (w/w), respectively, which were lower than control.

  • The Houttuynia cordata has been utilized for various beneficial purposes in humans mainly because of its potent antioxidant principle quercitrin present in this plant. This study examines the possibility of producing a functional sea food commodity containing active principle quercitrin by feeding H. cordata for a extended period. Spotted sea bass (Lateolabrax maculatus) were fed a diet containing H. cordata at 0.1-1.0% levels for 1 month and tissue concentrations of quercitrin were analyzed in serum, hepatopancreas and muscle. It was observed that quercitrin was found in the ranking order of hepatopancreas>muscle>serum. After a bolus administration of quercitrin (20 mg/kg, oral) to spotted sea bass and Nile tilapia (Oreochromis niloticus), idential rank order was observed after 48 hr. In contrast, the order was liver>serum>muscle in rat and mice, indicating that higher quercitrin distribution occurs to the muscle in fishes compared with in mammals tested. High residue concentration of qeurcitrin in the edible tissue can be an advantageous property in terms of functional food production. High level H. cordata extract inclusion of 1.0% seems to have detrimental effects in spotted sea bass leading to growth retardation and hepatic damage. It was concluded that incorporation of H. cordata extract into diet can be a way of producing healthy foods. However the level of active extract needs fine tuning to avoid toxicity to fishes.

Quercitrin Gallate Down-regulates Interleukin-6 Expression by Inhibiting Nuclear Factor-kB Activation in Lipopolysaccharide-stimulated Macrophages

  • Quercitrin gallate was previously isolated from Persicaria lapathifolia (Polygonaceae) as an inhibitor of superoxide production. In the present study, quercitrin gallate was found to inhibit interleukin (IL)-6 production in lipopolysaccharide (LPS)-stimulated macrophages RAW 264.7 with an $IC_{50}$ value of $63\;{\mu}M$. Furthermore, quercitrin gallate attenuated LPS-induced synthesis of IL-6 transcript but also inhibited LPS-induced IL-6 promoter activity, indicating that the compound could down-regulate IL-6 expression at the transcription level. Since nuclear factor (NF)-kB has been shown to play a key role in LPS-inducible IL-6 expression, an effect of quercitrin gallate on LPS-induced NF-kB activation was further analyzed. Quercitrin gallate exhibited a dosedependent inhibitory effect on LPS-induced nuclear translocation of NF-kB without affecting inhibitory kB (IkB) degradation, and subsequently inhibited LPS-induced NF-kB transcriptional activity in macrophages RAW 264.7. Taken together, quercitrin gallate down-regulated LPS-induced IL-6 expression by inhibiting NF-kB activation, which could provide a pharmacological potential of the compound in IL-6-related immune and inflammatory diseases.

Analytical Method Development of Avicularin and Quercitrin in Astilbe chinensis Extract Using HPLC (HPLC를 이용한 노루오줌 추출물의 Avicularin 및 Quercitrin 분석법 개발)

  • This study attempted to eatablish a high performance liquid chromatography (HPLC) analysis method for the determination of avicularin, quercitrin as a part of the quality control for the development of functional cosmetic materials from Astilbe chinensis extract. HPLC was performed on a Capcell Pak C18 MGII column with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at 30 ℃. The analyte was detected at 254 nm. The HPLC method was performed in accordance with the international conference on harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation of avicularin and quercitrin were 0.094 and 0.285 mg/mL, 0.031 and 0.095 mg/mL respectively. Calibration curves showed good linearity r2 > 0.99990 for avicularin and r2 > 0.99994 for quercitrin. Precision of analysis was satisfied with less than 0.59% for avicularin and 0.63% for quercitrin. Recoveries of quantified compounds ranged from 100.97 to 101.77% for avicularin and 100.18 to 100.32% for quercitrin. These result indicated that the established HPLC method is very useful for the determination of marker compounds in A. chinensis extracts.

Identification of Afzelin and Quercitrin from Pinus koraiensis and Their Contents in Genus Pinus Using HPLC/UV Analysis

  • Phytochemical constituents were isolated from Pinus koraiensis needles by open column chromatography. The structures isolated from the ethyl acetate fraction of P. koraiensis needles were elucidated as afzelin (1) and quercitrin (2) on the basis of spectroscopic analyses including $^1H$-NMR and MS. The amounts of afzelin (1) and quercitrin (2) in genus Pinus were determined by HPLC/UV analysis. Afzelin (1) and quercitrin (2) contents were highest in P. thunbergii needles. Because P. thunbergii needles contain high amounts of afzelin (1) and quercitrin (2), they have promising potential as new additives to natural medicinal products, health supplements, and beverages.

Antiherpetic Activities of Natural Quercitrin Alone and in Combinations with Nucleoside Antiherpetic Agents (천연 Quercitrin의 항허피스바이러스작용과 Nucleoside계 항허피스바이러스제와의 병용효과)

  • In order to find less toxic antiherpetic agents, antiviral activities of quercitrin against two strains of pathogenic viruses such as herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) were determined in Vero cells using plaque reduction assay in vitro. Quercitrin showed a concentration-dependent decrease in plaque formation of HSV-1 and HSV-2. It also exhibited more potent antiherpetic activity on HSV-1 with 50% effective concentration (EC$_{50}$) of 20.4 $\mu$g/ml than on HSV-2 with EC$_{50}$ of 30.4 $\mu$g/ml. The combined antiherpetic effects of quercitrin with nucleoside antiherpetic agents, acyclovir and vidarabine, were examined on the multiplication of these two strains of herpesviruses in Vero cells by the combination assay. The results of combination assay were evaluated by the combination index (CI) that was calculated by the multiple drug effect analysis. The combinations of quercitrin with acyclovir and vidarabine on HSV-1 showed more potent synergism with CI values of 0.27-0.81 for 50%, 70%, 90% effective levels than those on HSV-2 with CI values of 1.03~2.20..20.

Flavonoids and Antimicrobial Activity of the Ethanol Extract of Korean Cherry (Prunus tomentosa Thunberg) (앵두 Ethanol 추출물의 항균력과 Flavonoid)

  • Quantitaties of flavonoids were determined by HPLC in three different ethanol extract (50, 70 and 95%, v/v) from Korean cherry. Antimicrobial activity of the ethanol extract and its solvent fraction (ethyl acetate and n-butanol fractions) against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidermidis (ATCC 12228) was evaluated using MIC and disk test. Additionally, UV protective effect was also determined. The highest flavonoid quantity was found in 70% ethanol extract of Korean cherry. The quantities of quercitrin were 12.7 mg% and 2.1 mg% in ethyl acetate fraction and in buthanol fraction, respectively. In addition, the flavonoid in 70% (v/v) ethanol extract of Korean cherry included 14.9 mg% of quercitrin, 6.0 mg% of tannic acid and 5.2 mg% of catechin. Therefore, it was confirmed that main flavonoid of Korean cherry was quercitrin. For antimicrobial activity of ethanol extract of Korean cherry against Staphylococcus aureus (ATCC 6538) and Staphylococcus epidemidis (ATCC 12228), MIC and disk test using ethyl acetate fraction and butanol fraction were carried out. As a result, ethyl acetate fraction with higher quantity of flavonoid showed higher antimicrobial activity. Therefore, it was confirmed that the higher antimicrobial activity was brought about by flavonoid in Korean cherry. For UV protective effect of Korean cherry, ethanol extract showed the absorption zone of ultraviolet rays in 210∼380 nm including 290∼320 nm that is the wave length of UV-B harmful to the skin of human body. The pattern is the same as the absorption zone of 220∼300 nm in catechin and 210∼400 nm in quercitrin. Then, the functionalities of catechin and quercitrin were anticipated. It is expected that quercitrin in Korean cherry has the antimicrobial effect and UV protective effect related to the skin.

  • To obtain the basic information for commercial process and high quality production of Saururus chinensis, useful components were determined at different growth stages and organs. The contents of quercetin, quercitrin and tannin at different growth stages were decreased before flowering time but slightly increased after flowering. The contents of quercetin, quercitrin and tannin collected on July 26 were 5.72, 5.45g/kg and 1.5%, respectively. The contents of quercetin, quercitrin and tannin were the highest in white leaf, leaf and flower, respectively.

  • The seasonal variation of two flavonoid, quercitrin and afzelin, in the leaves of Cedrela sinensis A. Juss. from May until November was investigated by high performance liquid chromatography. Separation by reversed phase chromatography on ${\mu}-Bondapak\;C_{18}$ column was achieved by isocratic elution. The concentration of quercitrin was 5.81% (w/w) in May and high in July(8.89%, w/w), August(7.94%, w/w), and decreased from September to November. The content of another flavonoid, afzelin was 0.06% (w/w) in May and highest in June (0.12%, w/w), and was reduced from July.

Purification and Characterization of Quercitrin-Hydrolyzing ${\alpha}$-L-Rhamnosidase from Fusobacterium K-60, a Human Intestinal Bacterium

  • An ${\alpha}$-L-rhamnosidase (EC 3.2.1.40.), which transforms quercitrin to quercetin, was purified from Fusobacterium K-60, a human intestinal anaerobic bacterium. The specific activity of the purified ${\alpha}$-L-rhamnosidase was 2.89 mol/min/mg protein. ${\alpha}$-L-Rhamnosidase, whose molecular size was 170 kDa by gel filtration, was composed of four subunits ($M_r$ 41,000 Da) with pI and optimal pH values of 5.2 and 5.5-7.0, respectively. The apparent $K_m$ and $V_{max}$ values for p-nitrophenyl-${\alpha}$-L-rhamnopyranoside and quercitrin were determined to be 0.057 mM and 3.4 mol/min/mg, and 0.077 mM and 5.0 mol/min/mg, respectively. This enzyme was strongly inhibited by $Cu^{2+},\;Mn^{2+}$, L-rhamnose, and p-chlormercuriphenylsulfonic acid. These findings suggest that the biochemical properties and substrate specificity of the purified enzyme are different from those of the previously purified ${\alpha}$-L-rhamnosidase. This is the first reported purification of quercitrin-hydrolyzing ${\alpha}$-L-rhamnosidase from intestinal bacteria.
