Hepatic Coccidiosis in a Native Korean Goat (한국 재래 산양에서의 간 콕시디움 감염증)

  • A case of chronic cholangiohepatitis associated with Eimeria spp. is reported in a 6-month-old, male, native Korean goat. The goat died after having a 1-week history of diarrhea, anorexia and weight loss. At necropsy, numerous multifocal to coalescing, pale and mottled red foci were present throughout the liver. Histologically, numerous coccidian parasites in both sexual and asexual stages were found in the intrahepatic biliary epithelia and bile duct lumens. Based on the light microscopic and ultrastrastructural features, the parasites present in the liver were compatible with the genus Eimeria ; however, the species was not speciated.

  • The number of companian animals has recently been increasing. Also the tumor incidence rates of those has been increasing due to aging. But the survey of canine neoplasm was sparse. From 1995 to 2002 we collected canine biopsy cases which were suspected as neoplasms and submitted by practising veterinarians.(omitted)

Immunological relationships of FMD vaccine strain and Asia1 field isolate from East Asia (동아시아 유래 구제역바이러스 Asia1혈청형과 백신항원의 면역학적 상관성)

  • Foot-and-mouth disease (FMD) is the most contagious disease of mammals. The use of inactivated vaccine can be chosen to prevent or control FMD. However, vaccination against one serotype of FMDV doses not cross-protect against other serotypes and may not protect fully against some strains of the same serotype. Appropriate selection of vaccine strain is an important element in the control of FMD. The immunity of vaccine antigens should be matched against newly circulating viruses. The phylogenetic analysis of serotype Asia1 reported from China, Mongolia, North Korea and Russia since 2005 shows that they are all classified into genetic group V, but the strain, Asia1/Shamir (ISR/89) which have been used as a vaccine strain in Korea, is clustered into different genetic group. So, in this study the serological relationship between the isolate (Asia1/MOG/05; MOG) and the Shamir strain was determined by ELISA and virus neutralization test. Even though the matching value of the virus (MOG) against the vaccinated sera in target animals was not so high, the vaccinated animals elicited antibodies enough for protection after vaccinated once or twice. Conclusively, we suggest that the vaccine containing Asia1/Shamir antigen could protect the genetic group V strains circulating in East Asia currently if vaccinated twice or the more.

  • Foot-and-mouth disease (FMD) is a severe vesicular disease of cloven-hoofed animals including domesticated ruminants and pigs. Acute clinical signs may be mild in sheep and goats but are associated with lameness in pigs and mouth lesions with vesicles in cattle. The required condition for a successful pathogen appears to be the ability to counteract both the host innate and adaptive immune response. FMD virus (FMDV) inhibits the induction of antiviral molecules and interferes with the secretory pathway in the infected cell. The surface expression of Major Histocompatibility Complex (MHC) class I molecules is reduced in infected cells. Thus, the ability of the host to recognize and eliminate virus infected cells is decreased. Furthermore, FMDV infection results in a rapid, but transient lymphopenia, reducing the number of T and B cells, and affecting T cell function. The virus appears to premature apoptosis-mediated cell death because it has a very short replication cycle and is able to rapidly produce large amounts of virus. FMDV engages the host protective response at multiple steps to ensure its effective replication and pathogenesis. This review describes the recent pathological and immunological studies to overcome the powerful abilities of FMDV to counteract defense mechanism of host.

Prevalence of canine scabies in the Korean stray dogs (국내 유기견의 개 옴 감염 상황)

  • Recently, the number of stray dogs is proportionate to the increase of the number of the companion dogs. Sarcoptic scabiei var canis, causing scabies, is one of the most important canine zoonotic arthropods in Korea and around the world. Thus, we have tried to know the prevalence of canine scabies in the stray dogs in Korea. A total of 565 stray dogs were collected from the rescue centers all over the country from Jan. 2006 to Dec. 2006. They were grouped with euthanasia or natural death and examined for the gender and age estimated by dental formula. To identify the lesions, the whole body was grossly examined and tested pathologically. Thirty two (5.66%) of 565 dogs were diagnosed as canine scabies. Dogs from urban areas had fewer scabies (0.62%) than those of rural areas (12.5%). Prevalence of scabies in male and female dogs was no difference as 5.96% and 5.25%. Euthanasia group showed higher prevalence (6.48%) than natural death group (2.44%) in scabies. Old dogs over five years showed lower infestration (1.82%) in scabies. In histopathological findings, there were mites in the burrows formed in the subcorneal space. Acanthosis, hyperkeratosis with crust, and vascular dilatation were main findings. One hundred thirteen (20%) of 565 stray dogs were diagnosed to have skin disease. Among them, canine scabies is the most prominent ectoparasite as 5.66 %. With previous reports on human infection in Korea, canine scabies must be regarded as the important zoonotic canine skin disease. Accordingly, for the human and canine hygiene it is imperative that stray dogs with skin problems are segregated and tested for the parasites to treat properly as soon as arriving at rescue shelter.

Characterization of Yersinia ruckeri isolated from the farm-cultured eel Anguilla japonica in Korea

  • Yersinia (Y.) ruckeri has been recognized as a serious bacterial pathogen to several kinds of fish, including rainbow trout. However, there are no reports about the characteristics and pathogenicity of Y. ruckeri isolated from farm-cultured eels. In this study, we isolated and characterized Y. ruckeri from the farm-cultured eel Anguilla japonica in Korea. We investigated the phenotypic and genotypic characteristics of Y. ruckeri and tested the virulence of Y. ruckeri isolates on experimentally infected eels. Examination of the flagellar morphology of Y. ruckeri by electron microscopy showed peritrichous flagella in its cell body. Biochemical reaction studies showed overall identical profiles between the isolates and the reference strain of Y. ruckeri in API 20E and API ZYM tests. We sequenced the 16S rRNA of the Y. ruckeri (1,505 bp) for the genotypic characterization (National Center for Biotechnology Information accession number EU401667). Comparison of the 16S rRNA sequences with previously reported Y. ruckeri strains revealed similar phylogenetic relationships. In the virulence assay of the Y. ruckeri on eels, the eels exhibited listlessness, but Y. ruckeri was reisolated from those of the gills and kidneys.

Establishment of a diagnostic method for porcine proliferative enteropathy using polymerase chain reaction (중합효소연쇄반응을 이용한 돼지 증식성 장염 진단기법 확립)

  • Porcine Proliferative Enteropathy(PPE) is an infectious enteric disease and a major cause of economic loss in swine industry due to weight loss, poor growth and sudden death in growing and finishing pigs at 6 to 20 weeks of age. PPE has been diagnosed by clinical signs, syndrom and lesions in the intestine in Korea. However, the diagnostic method had several problems in the detection of infected or carrier pigs. Therefore, in this study, we established the polymerase chain reaction(PCR) which was a fast, specific and sensitive method for identification of Lawsonia intracellularis (L intracellularis). We designed and synthesized primer on the 16S rDNA and p78 gene encoding L intracellularis. Specificity of the method was confirmed by comparison of the PCR results using other enteric bacteria and the study has shown that PCR method was sensitive to detect 1ng of genomic DNA as a template. Identity of the PCR products was confirmed by comparison of pattern of restriction endonuclease analysis with restriction enzyme Hae III and Pst I. Also, the PCR method was applicable to the naturally affected pigs with PPE. Based on the results from this study, the PCR method could be used as a fast and specific diagnostic tool for PPE.

Development of competitive enzyme linked immunosorbent assay for detection of Coxiella burnetii antibody in animal (동물에서 Coxiella burnetii 항체를 진단하기 위한 경쟁효소면역법 개발)

  • Coxiella burnetii (C burnetii) is the causative agent of Q fever in animal and human. The distribution of the disease has been documented around world. In this study we developed the competitive enzyme linked immunosorbent assay(cELISA) and compared it with indirect immunofluorescent assay(IFA). A monoclonal antibody(Mab) against C burnetii and a peroxidase-conjugated anti-mouse IgM were used as an indicator system competing against antibody in animal serum or as an indicater of the absence of antibody. Sera were considered antibody positive when the percentage inhibition index(PI index) is upper than 30. PI index is calculated as 100-[sample OD/Mab OD)${\times}100$]. Among 162 bovine serum samples, 23 samples were antibody positive both in cELISA and IFA. And 156 samples showed same results. From goat with experimentally induced infection with C burnetii the antibody was detected 20 days early in cELISA compared to IFA. On the basis of present findings, it was demonstrated that cELISA is a reliable diagnostic method for The detection of specific antibodies against C burnetii infection.

  • Plasmid-mediated quinolone resistance (PMQR) determinants have been contributed to quinolone resistance of gram-negative bacteria worldwide. However, little data on the prevalence of these determinants in bacteria from animals are available in Korea. In this study, the prevalence of PMQR genes was investigated with E. coli originating from diseased animals. Among 55 E. coli tested, 11 showed PMQR genes by PCR. The most prevalent genotype was qepA (14.5%), followed by aac(6')-Ib-cr (7.3%) and qnrS (1.8%). Interestingly, two isolates with PMQR genes did not show quinolone resistance in this study. The isolates exhibited higher fluoroquinolone resistance in aac(6')-Ib-cr in combination with qnrS or qepA compared with aac(6')-Ib-cr only. In a conjugal transfer test, PMQR genes were transferred from donor to recipient.

Histopathological classification of the bovine lymphoma caused by bovine leukemia virus in Korea (소 백혈병 바이러스 감염에 의한 소 림프종의 병리조직학적 분류)

  • Several studies have been performed on the bovine leukemia since bovine leukemia virus (BLV) had been detected in 1982 in Korea. We have conducted histopathological study on the bovine lymphoma caused by BLV because only limited results were reported on the pathological characterization of lymphoma. Lymphoma tissues were obtained from cattle necropsied and slaughtered during a designated period. Lymphoma was classified histopathologically according to the National Cancer Institute Working Formulation. Leukotic tissues consisted of fairly uniform sheets of closely packed medium to large lymphocytes without any architectural arrangement in all 30 cases. Twenty five cases belong to diffuse large cell type, while three cases were diffuse mixed cell type, and two cases were immunoblastic large cell type among 30 cases. Follicular type lymphoma was not detected in this study. The mitotic index of tumor cells showed average 2.5 in the field of 400X. Nuclear cleavage was detected in 53% of cases. Multi-nucleated cells were detected among tumor cells in 30% of lymphoma cases. In conclusion, the most common morphologic cell type of bovine lymphoma in Korea was a diffuse large cell type with multinucleated cells and nuclear cleavages.
