화약물질 현장검출시약 EXPRAY$^{(R)}$를 이용한 토양내 화약물질 스크리닝 및 준정량화 가능성 (Screening and Possibility of Semi-quantitative Analysis of Explosive Compounds in Soil Using EXPRAY$^{(R)}$ Explosives Field Detection Kit)

  • 환경 시료 (토양 및 수질)내 화약물질을 신속하고 간편하게 검출할 수 있다면, 현장조사에서 오염도 조사를 위한 스키리닝 단계로 이용하여 불필요한 시료의 갯수와 고가의 실험실 정량분석비용을 줄일 수 있다. 상용 EXPRAY$^{(R)}$ Explosives Field Detection Kit(EXPRAY)를 이용하여 14종의 화약물질을 대상으로 화약물질 표준용액에 대한 최저검출농도를 결정 하고 및 준정량화 가능성을 확인하기 위한 실험을 실시하였다. 실험결과 EXPRAY는 14종의 화약물질 중에서, TNT, RDX, HMX, Tetryl및 TNB등 5종의 화약물질을 검출할 수 있었다. Nitramine계열인 HMX와 RDX에 대한 최저검출농도는 모두 14ng/mm$^2$이었다 EXPRAY는 nitroaromatic계열에는 더욱 민감하게 반응하여, Tetryl, TNB및 TNT에 대한 단위면적당(mm$^2$)최저 검출량이 각각 3ng, 3ng및 0.3ng이었다. 실험이 통제되는 조건에서 화약물질 표준용액에 EXPRAY를 적용하였을 경우에는 발색 강도의 비교를 통해 10배 단위의 준정량화가 가능하였다. 그러나 오염토양으로 실험한 경우에는 화약물질의 존재여부 및 종류만을 확인할 수 있었다. EXPRAY Explosives Detection Kit$^{(R)}$가 화약물질을 검출할 수 있는 경제적이고 민감한 방법이지만, 현장시료의 스크리닝 단계에만 적용할 수 있는 방법이다.

Breast Imaging Using Electrical Impedance Tomography: Correlation of Quantitative Assessment with Visual Interpretation

  • Background: Electrical impedance tomography (EIT) is a new non-invasive, mobile screening method which does not use ionizing radiation to the human breast; allows conducting quantitative assessment of the images besides the visual interpretation. The aim of this study was to correlate the quantitative assessment and visual interpretation of breast electrical impedance tomographs and associated factors. Materials and Methods: One hundred and fifty mammography patients above 40 years and undergoing EIT were chosen using convenient sampling. Visual interpretation of the images was carried out by a radiologist with minimum of three years experience using the breast imaging - electrical impedance (BI-EIM) classification for detection of abnormalities. A set of thirty blinded EIT images were reinterpreted to determine the intra-rater reliability using kappa. Quantitative assessment was by comparison of the breast average electric conductivity with the norm and correlations with visual interpretation of the images were determined using Chi-square. One-way ANOVA was used to compare the mean electrical conductivity between groups and t-test was used for comparisons with pre-existing Caucasians statistics. Independent t-tests were applied to compare the mean electrical conductivity of women with factors like exogenous hormone use and family history of breast cancer. Results: The mean electrical conductivity of Malaysian women was significantly lower than that of Caucasians (p<0.05). Quantitative assessment of electrical impedance tomography was significantly related with visual interpretation of images of the breast (p<0.05). Conclusions: Quantitative assessment of electrical impedance tomography images was significantly related with visual interpretation.

Quantitative Analyses for the Quality Evaluation of Salviae Miltiorrhizae Radix by HPLC

  • In this study, quantitative analysis for the quality evaluation of Salviae Miltiorrhizae Radix using HPLC/UV was developed. For quantitative analysis, six major bioactive compounds were determined. The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6\;mm$, $5\;{\mu}m$) with gradient condition of A (1% formic acid in $H_2O$) and B (acetonitrile : methanol : formic acid = 100 : 75 : 1) as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 280 nm. These methods were fully validated with respect to the linearity, accuracy, precision and recovery. The HPLC/UV method was applied successfully to the quantification of six major compounds in the Salviae Miltiorrhizae Radix. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis.

  • 유비쿼터스 컴퓨팅 시스템은 하나 이상의 컴퓨터가 네트워크로 상호 연결된 프로세서 시스템이다. 하지만, 기존의 보안 유지 방법은 정성적 탐지 및 대응책으로 공격이 발생한 이후 대응에만 치중하여 능동적 차원의 보안 유지 방법에 대한 연구가 부족하다. 따라서, 본 논문은 정량적인 분석을 통해 범용적 인프라의 개선뿐만 아니라 특정 인프라 공격에 대해서 탐지 및 데응할 수 있는 방법에 대해 제안한다. 이를 위해 시스템의 고정적 요소 정보, 임의의 요소 정보, 공격 유형 모델링을 근간으로 시스템의 보안성을 정량적으로 분석할 수 있도록 생존성에 대한 정의 및 모델링 기법을 사용하였다. 그리고 제안한 기법의 검증을 위해 TCP-SYN 공격과 Code-Red 웜 공격에 대한 생존성 분석을 수행하였다.

Direct and Quantitative Analysis of Salmonella enterica Serovar Typhimurium Using Real-Time PCR from Artificially Contaminated Chicken Meat

  • For quantitative PCR assay of Salmonella enterica serovar Typhimurium in food samples, a real-time PCR method was developed, based on DNA genome equivalent. Specific primers and probe designed based on the STM4497 gene of S. Typhimurium LT2 showed the specificity to S. Typhimurium. Threshold cycle (Ct) values of real-time PCR were obtained from a quantitative standard curve with genomic DNA of Salmonella Typhimurium. In addition, the recovery of S. Typhimurium inoculated artificially to chicken samples with $4.5{\times}10^5$ to 4.5 CFU/ml was evaluated by using real-time PCR and plate-count methods. Result showed that the number of cells calculated from the real-time PCR method had good correlation with that of the plate-count method. This real-time PCR method could be applicable to the detection and quantification of S. Typhimurium in food samples.

Quantitative Analysis of Bioactive Marker Compounds from Cinnamomi Ramulus and Cinnamomi Cortex by HPLC-UV

  • In this study, quantitative and pattern recognition analysis for the quality evaluation of Cinnamomi Ramulus and Cinnamomi Cortex using HPLC/UV was developed. For quantitative analysis, three major bioactive compounds were determined. The separation conditions employed for HPLC/UV were optimized using an ODS $C_{18}$ column ($250{\times}4.6$ mm, 5 ${\mu}m$) with gradient conditions of acetonitrile and water as the mobile phase, at a flow rate of 1.0 mL/min and a detection wavelength of 265 nm. This method was fully validated with respect to linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of three major compounds in the extract of Cinnamomi Ramulus and Cinnamomi Cortex. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of thirty eight Cinnamomi Ramulus and thirty five Cinnamomi Cortex samples. The results indicate that the established HPLC/UV method is suitable for quantitative analysis.

Quantitative and Pattern Recognition Analyses for the Quality Evaluation of Magnoliae Flos by HPLC

  • In this study, quantitative and pattern recognition analysis for the quality evaluation of Magnoliae Flos using HPLC/UV was developed. For quantitative analysis, eleven major bioactive lignan compounds were determined. The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6\;mm$, $5\;{\mu}m$) with isocratic elution of acetonitrile and water with 1% acetic acid as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 278 nm. These methods were fully validated with respect to the linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of eleven major compounds in the extract of Magnoliae Flos. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of twenty one reference samples corresponding to seven different species of Magnoliae Flos and nine samples purchased from market. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis and quality control of multi-components in Magnoliae Flos.
