Development of Landslide Hazard Map Using Environmental Information System: Case on the Gyeongsangbuk-do Province (환경정보시스템을 이용한 산사태 발생위험 예측도 작성: 경상북도를 중심으로)

  • The purpose of this research was develop tailored landslide hazard assessment table (LHAT) in Gyeongsangbuk-do Province and propose building strategies on environmental information system to estimate landslide hazard area according to LHAT. To accomplish this purpose, this research investigated factors occurring landslide at 172 landslide occurred sites in 23 city and county of Gyeongsangbuk-do Province and analyzed what factors effected landslide occurrence quantity using the multiple statistics of quantification method(I). The results of analysis, factors affecting landslide occurrence quantity were shown in order of slope position, slope length, bedrock, aspect, forest age, slope form and slope. And results of the development of LHAT for predict mapping of landslide-susceptible area in Gyeongsangbuk-do Province, total score range was divided that 107 under is stable area(IV class), 107~176 is area with little susceptibility to landslide(III class), 177~246 is area with moderate susceptibility to landslide(II class), above 247 area with severe susceptibility to landslide(I class). According to LHAT, this research built landslide attribute database and made 7 digital theme maps at mountainous area located in Goryeong Gun, Seongju-Gun, and Kimcheon-City. The results of prediction on degree of landslide hazard using environmental information system, area with little susceptibility to landslide(III class) occupied 65.56% and severe susceptibility to landslide(I class) occupied 0.51%.

A Usability Assessment Metric for Ubiquitous Services: Quantification of the Interactivity Attribute in Inter-personal Services

  • Objective: The main objective of this study is to propose a user-centered assessment metric for ubiquitous services. Background: As the ubiquitous era took off, the interactions between ubiquitous services and users have come to take an important position. It is essential to conceptualize a new assessment model that considers human-system interaction capability with a user-centered design perspective. Method: The evaluation model for the interactivity of ubiquitous service was approached from the concept of usability and inter-personality of services. As a validation study, suggested assessment metric was utilized to evaluate the u-Home service. Priority weighting of each assessment metric was derived using the quantification type-I analysis. Results: To evaluate interactivity, this study suggested a quantitative metric for user testing performed after classifying the interactivity characteristics to contextualization; ubiquity; user experience; and service capability. Conclusion: This study suggest the metric for the ubiquitous service that are experienced in real life, and introduced the concept of ubiquitous service interactivity. Application: The suggested evaluation metric can be used to evaluate interactivity level of ubiquitous service and identify the potential problem and usability requirements at the early stage of service development.

Analyzing nuclear reactor simulation data and uncertainty with the group method of data handling

  • Group method of data handling (GMDH) is considered one of the earliest deep learning methods. Deep learning gained additional interest in today's applications due to its capability to handle complex and high dimensional problems. In this study, multi-layer GMDH networks are used to perform uncertainty quantification (UQ) and sensitivity analysis (SA) of nuclear reactor simulations. GMDH is utilized as a surrogate/metamodel to replace high fidelity computer models with cheap-to-evaluate surrogate models, which facilitate UQ and SA tasks (e.g. variance decomposition, uncertainty propagation, etc.). GMDH performance is validated through two UQ applications in reactor simulations: (1) low dimensional input space (two-phase flow in a reactor channel), and (2) high dimensional space (8-group homogenized cross-sections). In both applications, GMDH networks show very good performance with small mean absolute and squared errors as well as high accuracy in capturing the target variance. GMDH is utilized afterward to perform UQ tasks such as variance decomposition through Sobol indices, and GMDH-based uncertainty propagation with large number of samples. GMDH performance is also compared to other surrogates including Gaussian processes and polynomial chaos expansions. The comparison shows that GMDH has competitive performance with the other methods for the low dimensional problem, and reliable performance for the high dimensional problem.

Suitable Site Prediction of Erosion Control Dam by Sediment (산지사면에 있어서 퇴사량에 의한 사방댐의 시공적지 예측)

  • This study was carried out to analyze the characteristics of forest environmental and stream morphological factors by using the quantification theory (I) for prediction of the suitable dam site. The results obtained from this study were summarized as follows; The selection of suitable site for erosion control dam was estimated by normalized score of each category. And the prediction method of suitable site for erosion control dam divided into class I (Very suitable site), II (Suitable site), and III (Poor suitable site) for the convenience of use. In conclusion, if we select the suitable site for construction of erosion control dam for disaster prevention, we could save the loss of tremendous budget, avoid the poor suitable site due to subjective judgment, and it would promote the functions of erosion control dam.

Discovery of a new primer set for detection and quantification of Ilyonectria mors-panacis in soils for ginseng cultivation

  • Background: Korean ginseng is an important cash crop in Asian countries. However, plant yield is reduced by pathogens. Among the Ilyonectria radicicola-species complex, I. mors-panacis is responsible for root-rot and replant failure of ginseng in Asia. The development of new methods to reveal the existence of the pathogen before cultivation is started is essential. Therefore, a quantitative real-time polymerase chain reaction method was developed to detect and quantify the pathogen in ginseng soils. Methods: In this study, a species-specific histone H3 primer set was developed for the quantification of I. mors-panacis. The primer set was used on DNA from other microbes to evaluate its sensitivity and selectivity for I. mors-panacis DNA. Sterilized soil samples artificially infected with the pathogen at different concentrations were used to evaluate the ability of the primer set to detect the pathogen population in the soil DNA. Finally, the pathogen was quantified in many natural soil samples. Results: The designed primer set was found to be sensitive and selective for I. mors-panacis DNA. In artificially infected sterilized soil samples, using quantitative real-time polymerase chain reaction the estimated amount of template was positively correlated with the pathogen concentration in soil samples ($R^2=0.95$), disease severity index ($R^2=0.99$), and colony-forming units ($R^2=0.87$). In natural soils, the pathogen was recorded in most fields producing bad yields at a range of $5.82{\pm}2.35pg/g$ to $892.34{\pm}103.70pg/g$ of soil. Conclusion: According to these results, the proposed primer set is applicable for estimating soil quality before ginseng cultivation. This will contribute to disease management and crop protection in the future.

Evaluation of DNA Extraction Methods from Low Copy Number (LCN) DNA Samples for Forensic DNA Typing

  • DNA isolation for PCR-based short tandem repeat (STR) analysis is essential to recover high yields of amplifiable DNA from low copy number (LCN) DNA samples. There are different methods developed for DNA extraction from the small bloodstain and gloves, commonly found at crime scenes. In order to obtain STR profiles from LCN DNA samples, DNA extraction protocols, namely the automated $iPrep^{TM}$ $ChargeSwitch^{(R)}$ method, the automated $QIAcube^{TM}$ method, the automated $Maxwell^{(R)}$ 16 DNA $IQ^{TM}$ Resin method, and the manual $QIAamp^{(R)}$ DNA Micro Kit method, were evaluated. Extracted DNA was quantified by the $Quantifiler^{TM}$ Human DNA Quantification Kit and DNA profiled by $AmpFISTR^{(R)}$ $Identifiler^{(R)}$ Kit. Results were compared based on the amount of DNA obtained and the completeness of the STR profiles produced. The automated $iPrep^{TM}$ $ChargeSwitch^{(R)}$ and $QIAcube^{TM}$ methoas produced reproducible DNA of sufficient quantity and quality trom the dried blood spot. This two automated methods showed a quantity and quality comparable to those of the forensic manual standard protocols normally used in our laboratory. In our hands, the automated DNA extraction method is another obvious choice when the forensic case sample available is bloodstain. The findings of this study indicate that the manual simple modified $QIAamp^{(R)}$ DNA Micro Kit method is best method to recover high yields of amplifiable DNA from the numerous potential sources of LCN DNA samples.

Development of Questionnaires for Pathogenesis Analysis of Bojungikgitang Symptom( II ) (보중익기탕증(補中益氣湯證)의 병인론적(病因論的) 분석을 위한 설문문항(說問問項) 개발(開發)( II ))

  • Background and purpose: We previously developed questionnaire of Bojungikgitang systom on the Delphi method through the pathogenesis analysis. But developed a questionnaire was not verified in the clinical. So, to ensure objectivity, quantification and validity, verification is needed for questionnaire items before applying a clinical. On this study, we looked at whether questionnaire items had been validity in the clinical. Methods: Surveys conducted about 191 patients at 12 oriental medicine hospitals. Among them, patients with Bojungikgitang systom(group I) were 95, and patients with no Bojungikgitang systom(group II) were 96. We calculated that the sum of each item in the survey and then the sum was reviewed statistically significant difference through Independent samples T test between group I and II. Results: Between group I and II, the total sum survey of the percent difference is meaningful (P<0.05). Conclusions: Reliability analysis of the Bojungikgitang systom survey research is needed in the future. Also I think that research should proceed about a lot of people.

Illite Polytypes: The Characteristics and the Application to the Fault Age Determination (일라이트 폴리타입: 그 특성과 단층 활동연대 결정에의 활용)

  • The 1M and $2M_1$ stacking sequences are the most frequently encountered in the illite species among the possible 6 polytypes. The $1M_d$, derived from the 1M polytype which exhibits a variable degree of disorder in the stacking sequence, is also observed in illite samples. In this paper, the author introduces and reviews the theoretical background of the quantitative analysis method of illite polytypes, and considers the possibility to determine the fault age and its reactivation age using K/Ar age-dating based on the quantification of illite polytypes in the fault system. For the increase of the accuracy and precision of the illite age analysis method, the occurrence, identification, and mineralogical characterization of illite polytypes should be defined in detail. The broadening effect of (hkl) reflections, due to disordering of 1M polytype and the presence of I/S minerals with expandability, are also considered as the main parameters controlling the quantification of illite polytypes using the WILDFIRE(C)simulation.

Characterization and Tissues Distribution of Vinculin, Agouti-relating Protein and Melanocortin 4 Receptor Genes in Rainbow Trout, Oncorhynchus mykiss

  • As in the O. mykiss electrophoretic profiles of RNA, the signals of each RNA sample from 9 individual tissues such as liver, muscle, brain, heart, pituitary gland, kidney, intestine, spleen and gill similar to positive control were obtained. The tissue distributions of the complimentary DNA (cDNA) of O. mykiss four genes were analyzed using quantitative real-time PCR with primer sets for tissue expression analysis. In this rainbow trout species, author obtained bands of various sizes, ranged from 700 bp to 1,400 bp. A dissociation curve was made at the end of each run to make sure that there was no non-specific amplification. Supplementarily, the Ct of each DNA was compared. The Ct values of vinculin with rainbow trout tissues were determined in a manner similar to those for agouti-related protein (AgRP) and melanocortin receptors (MC4R I and MC4R II). Further, obtained Cts for standard curve of each DNA were affected by specific product (vinculin, AgRP and MC4R II genes). After several experiments with four individual genes of rainbow trout, author estimated a variation ratio of the mean Ct value of the DNA extracted using the comparative CTt method was 37.27, and the standard deviation was 5.33. The correlation coefficient between the Ct values and the concentration of cDNA was -0.98, -0.99, -0.91 and -0.86, respectively (vinculin, AgRP, MC4R I and MC4R II genes). Since this correlation showed high linearity, the straight line obtained was used as a standard for the O. mykiss tissues reared in aquarium. A PCR efficiency of 100% is ideally achieved when the slopes are close to the theoretical value of -3.31. According to quantification method, the results of quantification are strongly affected by the DNA fragmentation. The size of most DNA fragments obtained from various tissues of rainbow trout used in the experiment was approximately 100 bp. According to the four slopes, an efficiency of nearly 100% was estimated for four genes detection methods. Additionally, further analysis with more individuals and primers will be required to fully establish optimization in rainbow trout.

Development of Analytical Method for Polychlorinated Biphenyls and Organochlorine Pesticides in Air (대기 중 PCBs와 유기염소계 살충제의 정량을 위한 분석 방법 개발)

  • An analytical method was investigated for the meaiiurement of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) concentrations in air samples. Procedures required for column chromatographic clean up. silicagel (stage I) and gel permeation chromatography (stage II), were discussed. Identification and quantification of PCBs and OCPs were performed using a combination of gas chromatography/mass spectrometry/ selected ion monitoring. Recovery tests calculated from six samples are 68∼137% for PCBs and 58∼130% for OCPs except for endrin aldehyde. Instrumental detection limits determined for the PCBs and OCPs varied from 0.05 to 0.18 pg/m3 and from 0.71 to 16.82 pg/㎥, respectively. The method has been applied to the analysis of air samples collected at Ansung city, Kyonggi province, Korea. This method may serve as a screening protocol for the simultaneous determination of PCBs and OCPs in air.
