Effect of pH on the elaboration of pullulan and the production of high molecular weight pullulan by Aureobasidium pullulans.

  • The effect of on the cell growth, the elaboration of pullulan, the morphology and were the effect of on the molecular weight of pullulan were investigated. A. pullulans showed maximum pullulan production when initial pH 6.5 was 11.98 g/l in shake-flask culture. In batch culture, the maximum pullulan production of 15.16 g/l was obtained at an aeration rate of 0.5 vvm. The mixture of yeast-like form and mycelial form of cells was found at the constant pH 4.5, at which condition, the elaboration of pullulan was high, about 13.31 g/l. However, pullulan with its higher molecular weight (>1,000,000) was produced at the constant pH 6.5.

  • For the maximum production of pullulan from glucose as a carbon source, the effects of glucose concentration, pH and dissolved oxygen concentration on the cell growth and mass production of high-molecular weight pullulan by A. pullulans ATCC 42023 were evaluated. A. pullulans showed optimum pullulan productivity when glucose concentration was 0.3M (54g/L). And inhibitory effects on the cell growth and the pullulan production were observed at the glucose concentration higher than 0.3M (54g/L). The influence of pH control and dissolved oxygen on the pullulan production and growth of A. pullulans was studied. In shake-flasks, maximum pullulan production was obtained with $11.98g/{\ell}$ when initial pH was 6.5. In the batch fermentation, the maximum pullulan production of $13.31g/{\ell}$ was obtained with constant pH 4.5. And it was found that pullulan yield and synthesis rate increased with oxygen availability. For the production of commercially useful pullulan with high-molecular weight, a mixed carbon source, which was a mixture of glucose and glucosamine, was used for the pullulan fermentation with A. pullulans. On the basis of 5% mixed carbon source, culture with 3% glucosamine with 2% glucose was optimum condition for the production of high (M.W.> 1,000,000) and medium (M.W.> 200,000) molecular weight pullulan with considerable yields of cell mass and product. And the influence of pH control on the molecular weight of pullulan was studied in batch fermentation. It was found that the productivity of high-molecular weight pullulan with pH control at 6.5 was higher than that with no pH control.

  • The effects of dissolved oxygen and pH on the cell growth and mass production of high-molecular weight pullulan by A. pullulans ATCC 42023 were evaluated. For the production of commercially useful pullulan with high-molecular weight, the influence of pH control on the pullulan production and growth of A. pullulans was studied in batch fermentation. It was found that the productivity of high-molecular weight pullulan with pH control at 6.5 was higher than that with no pH control. The influence of dissolved oxygen on the pullulan production and growth was studied. It was found that pullulan yield and synthesis rate increased with oxygen availability.

Mass Production of Pullulan with Optimized Concentrations of Carbon and Nitrogen Sources by Aureobasidium pullulans HP-2001 in a 100-L Bioreactor with the Inner Pressure

  • Cell growth and the production of pullulan by Aureobasidium pullulan HP-2001, the UV-induced mutant of A pullulans ATCC 42023, increased with increased concentration of glucose up to 15.0% (w/v). Maximal production of pullulan in the flask scale was 27.65 g/l, when concentrations of glucose and yeast extract were 15.0 and 0.25% (w/v), respectively. Maximal conversion rate of pullulan from glucose as the carbon source was 0.37, when those of glucose and yeast extract were 5.0 and 0.15% (w/v), respectively. On the basis of total amount of pullulan, the conversion rate of pullulan from glucose, and utilization rate of glucose to cell mass and pullulan by A. pullulans HP-2001, the optimal concentrations of glucose and yeast extract for the mass production of pullulan were determined to be 10.0 and 0.25% (w/v), respectively, at which concentrations the production of pullulan and its conversion rate were 27.14 g/l and 0.27, respectively. Maximal production of pullulan with optimized concentrations of carbon and nitrogen sources by A. pullulans HP-200l in a 7-1 bioreactor was 32.12 g/l for 72 h culture, and that in a 100-1 bioreactor with the inner pressure of $0.4 kgf/cm^2$ was 36.87 g/l. Increased inner pressure of a 100-1 bioreactor resulted in a higher concentration of dissolved oxygen in the medium, which might enhance the production of pullulan by A. pullulans HP-2001.

  • Electrospinning is a versatile process used to prepare micro or nano sized fibers from various materials dissolved in volatile solvents. This study reports electrospun pullulan fibrous webs fabricated through electrospinning using water as a solvent. The electrospinning conditions such as pullulan (PUL) concentration and applied voltage were optimized in order to obtain smooth electrospun fibers. The concentration of PUL greatly influenced the viscosity and surface tension of PUL solution. PUL beaded electrospun fibers were obtained from PUL solutions with concentrations lower than 5 wt%, while homogenous electrospun fibers were prepared from solutions with high concentration and high viscosity. The average diameters of PUL fibers were decreased to 200 nm when the polymer concentration was kept at 10 wt% and the applied voltage was fixed at 15 kV during electrospinning. PUL electrospun fiber exhibited higher solubility, flexibility, softness and adhesive strength.

A Study on the Pullulan Production Using Whey (Whey를 이용한 Pullulan의 발효생산에 관한 연구)

  • In this study, Pullulan Production from whey medium by Aureohsidium pullulans was investigated. When the Pullulan production from whey medium was carried outs final cell concentration was similar to sucrose basal medium but Pullulan concentration was less than 5g/$\ell$. For pullulan production from whey medium, adaptation culture technique was tried on lactose and galactose base medium When the adaptation culture technique was not applied, the maximum concentration of pullulan was 3.4g/$\ell$ for lactose, 2.5g/$\ell$ for galactose. Adapted strains produced 10~12g/$\ell$ from lactose, 10~11g/$\ell$ from galactose. The pullulan production from lactose basal medium was 13.5g/$\ell$ for lactose adapted strains and 18.6g/$\ell$ for galactose adapted strains. When the adapted strains were inoculated on whey medium, maximum pullulan production was obtained at initial pH 3.0.

  • pullulan은 maltotriose를 기본 단위로 하여 .alpha.(1.rarw.4), .alpha.(1.rarw.6)결합으로 구성된 일종의 .alpha. glucan으로, 'black yeast'로 알려진 Aureobasidium pullulans에 의해 세포외로 생산되는 다당류이다. pullulan은 다른 다당류에서 볼 수 없는 많은 우수한 성질이 있어, 식품에서 접착, 증점, 안정, 품질 개량 효과등의 이용을 비롯하여, 산소 불투과성 필름, 가식성 필름, 플라스틱 결착제, 코오팅제 등 여러가지 무공해 재료로써 광범위한 분야에 그 이용 가능성이 모색되고 있다. 본 고에서는 A. pullulans를 이용한 pullulan 생산의 연구 현황과 그 특성 및 응용 가능성에 대해 고찰해보고자 한다.

  • The pullulan production and morphological change of Aureobasidium pullulans ATCC 9348 were investigated both in batch fermentation and in continuous fermentation. The best carbon source for pullulan production was sucrose among seven different carbon sources. The pullulan production of A. pullulans was increased with increasing the carbon to nitrogen ratio of the medium using sucrose as a carbon source. In batch fermentation, production of pullullan occurred following exhaustion of the nitrogen source from the medium. The continuous fermentation showed that the pullulan production was closely parallelled with cell growth and was most effective at a dilution rate of 0.06~0.07 hr$^{-1}$-. The ratio of yeast-like cells(blastospores) of A. pullulans increased with the increase of growth rate, and reached 100% over the growth rate of 0.07 hr$^{-1}$. The growth rate, within a certain range, affected not only on the cell morphology, but on the specific pullulan productivity of A. pullulans.

  • For the production of pullulan from the high concentration of sugar, the utilization of sugars by a pullulan-producing fungus, Aureobasidium pullulans was examined. A. pullulans showed the different utilization patterns for sugars such as sucrose, maltose, and maltotriose. Especially for maltotriose, the hydrolysis of sugar was accompanied by a transferase activity. Glucose and maltose showed the inhibitory effect on the cell growth and the pullulan production at the sugar concentration higher than 0.28M, but sucrose showed the inhibitory effect at the sugar concentration higher than 0.14M. Among the sugars examined, sucrose gave the best result for the pullulan production. 27.5g/l of pullulan was obtained from 5% sucrose.

  • The effects of pH on the cell growth, the elaboration of pullulan, and the morphology of Aureobasidium pullulans IFO 4464 were examined. A. pulluans grew in yeast-like form at constant pH7.5 and in mycelial form at constant pH2.5. At the both pH conditions, the elaboration of pullulan was very low, about 6.0~6.5g/l. The mixture of yeast-like form and mycelial form of cells was found at the constant pH4.5, at which condition, the elaboration of pullulan was high, about 24.5g/l. The pH shift experimemts showed that the specific production rates of pullulan were 0.048($hr^{-1}$)for the mycelial form and 0.058($hr^{-1}$)for the yeast like form, which indicated that the yeast-like form has the similar, only slightly higher, biosynthetic activity of pullulan to the mycelial form at pH4.5 and the pH of culture broth is more important factor for the elaboration of pullulan than the morphology of A. pullulans.
