Separation and Identification of Antimicrobial Substances from Prunus mume extract (매실추출물로부터 항균물질의 분리 및 구조동정)

  • Prunus mume was extracted using various solvents including ethyl ether, ethyl acetate and n-butanol. The extracts were fractionated by column chromatography. Antimicrobial compounds (A, B and C) in fractions showing antimicrobial activity were purified by Sephadex LH 20 column chromatography, and identified by $^{1}H$- and $^{13}C$-NMR analysis as isoeugenol, nomilin and $\beta$-sitosterol, respectively.

  • Prolyl endopeptidase (PEP) is proline-specific serine protease, cleaving peptide bonds on the biologically active neuropeptides such as substance P, vassopressin, and thyrotropin-releasing hormone and is, therefore, suggested to play important roles in learning and memory process. In this work, the inhibitory effect of plant extracts on PEP was investigated. Out of 200 plant extracts, Prunus mume, Pyrola. japonica, Hypericum ascyron, Astilbe chinensis var. typica, and Elaeagnus umbellata inhibited more than 90% of PEP activity at the concentration of 5 ppm.

Antitumor and Free Radical-Scavenging Activities of Various Extract Fractions of Fruits and Leaves from Prunus mume (금매와 매화 잎 추출물의 프리라디칼 억제 활성 및 항암 효과)

  • This study investigated the free radical-scavenging and antitumor activities of hot water, water, acetone, ethanol, ethyl acetate, chloroform, and hexane extracts of fruits and leaves from Prunus mume. The various extracts were evaluated for their total polyphenol, flavonoid, and tannin contents, scavenging activities by DPPH and ABTS analyses, reducing power, protective effects against oxidative stress in L-132 cells, and antitumor activities against A549, HeLa, and U87 cancer cells. Ethanol extracts of fruits and leaves showed the highest total polyphenol content (336.41 and 523 mg GAE/100 g, respectively). DPPH and ABTS radical-scavenging activities increased according to concentration of fruit. DPPH radical-scavenging activity of ethanol extracts from leaves was 65.48% at $200{\mu}g/mL$. All extract fractions of leaves showed high ABTS radical-scavenging activities. The reducing power activities increased according to increasing concentration of fruits and leaves. All extracts of leaves performed better than extracts of fruits in terms of protective effects against oxidative stress in L-132 cells. Ethyl acetate, chloroform, hexane, ethanol extracts of fruits and leaves showed anticancer activities against A549, HeLa, and U87 cancer cells. However, ethanol extracts of fruits and leaves showed no toxicity in normal cells (BNLCL2). This study suggests that antioxidant activities of fruits and leaves from P. mume depend on polyphenol contents. Thus, fruits and leaves from P. mume can be useful as natural antioxidant compounds.

Antioxidant and α-Glucosidase Inhibition Activity of Solvent Fractions from Prunus mume Ethanol Extract (매실 순차분획물의 용매별 항산화 활성 및 α-glucosidase 억제 효과)

  • Prunus mume, known as maesil in Korea, has been widely cultivated in East Asia and used as medication and food. However, because most of the previous studies concerning P. mume had been investigated its under extract state, detailed studies are still required for its extensive utilization. In this study, we evaluated the antioxidant and ${\alpha}-glucosidase$ inhibitory activities of solvent fractions of P. mume ethanol extracts. The ethyl acetate fraction showed higher DPPH radical scavenging activity, ABTS radical scavenging activity, reducing power, and hydrogen peroxide scavenging activity than other fractions. The DPPH radical scavenging activities of ethyl acetate fraction was 67.79%; ABTS radical scavenging activity was 60.03%; reducing power ($OD_{670}$) was 1.26; and hydrogen peroxide scavenging activity was 93.18% at $500{\mu}g/ml$. Also, the ethyl acetate and methanol fraction showed effective levels of ${\alpha}-glucosidase$ inhibition activity (69.25% and 72.29% at $500{\mu}g/ml$). Total polyphenol contents and total flavonoid contents of the ethyl acetate fraction were 88.28 mg/g (gallic acid equivalent) and 70.38 mg/g (quercetin equivalent), respectively. These results suggest that the physiological activities of the ethyl acetate fraction are associated with its polyphenol and flavonoid contents. Therefore, this study can be used as basic data for developing natural antioxidants and potential functional material using P. mume.

Antioxidant Activities of Plunus mume flower buds Extract by Various Solvents (매화꽃봉오리의 추출용매별 항산화 효과)

  • This study investigated the antioxidant activities of distilled water, ethanol and methanol extracts of Plunus mume flower buds (PFB). The various solvent extracts of PFB were evaluated for their total polyphenol, flavonoid, reducing power and free radical scavenging activities by FRAP and DPPH analysis. The ethanol extract of PFB contained significantly higher amounts of total polyphenols (145 mg GAE/g) and flavonoids (25.43 mg QE/g) than methanol (132 and 25.42) and distilled water (113.6 and 18.04). Among solvent extracts of PFB, the ethanol extract showed the highest antioxidant activities. The 100% ethanol extract of PFB contained significantly higher amounts of total polyphenols and flavonoids than 70% and 50% ethanol extracts. Moreover, the 100% ethanol extract of PFB showed high efficacy in DPPH radical scavenging activity and in collagenase inhibition activity. This results suggest that 100% ethanol extract of PFB has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from PFB for usage of cosmeceutical and/or food industries.

Changes of Characteristics in Kochujang Fermented with Maesil (Prunus mume) Powder or Concentrate (매실분말 및 농축액을 첨가한 고추장의 숙성중 품질 변화)

  • Powder, or a concentrate, of maesil (Prunus mume) were used in the making of kochujang, and were added to kochujang at 0.5 or 1.0% (w/w). Kochujang fermented with maesil powder lower moisture content than did samples fermented with maesil concentrate. Moisture content slowly increased during fermentation, and was $3{\sim}4%$ higher than the initial value after 8 weeks of fermentation. Kochujang fermented with maesil powder or maesil concentrate showed lower pH values and higher total acidities than did the control. The total microbial count was $3.6{\times}10^7\;5.4{\times}10^7\;cfu/g$, and yeast and mold levels were $2.0{\times}10^7{\sim}3.6{\times}10^7\;cfu/g$, as kochujang fermentation commenced. Microbial load reached a maximum by 4 weeks of fermentation, and decreased thereafter. L (lightness), a (redness) and b (yellowness) values of kochujang gradually decreased during fermentation. Compared to methanol extracts of kochujang, water extracts and ethanol extracts showed higher free radical scavenging abilities at the initial stage of fermentation. Control, and samples with 0.5% (w/w) maesil powder or maesil concentrate, attained higher scores in sensory evaluation tests than did samples with higher maesil levels, and were not significantly different at P>0.05 as determined by Duncan's multiple range test.

Antioxidant Activation of Plum (Prunus mume Sieb. et Zucc.) Flesh, Pericarp and Stone Extracts (매실 과피, 과육과 핵의 품종별 추출액의 항산화 활성)

  • 매실에는 구연산, 사과산, 주석산, 피루브산 등의 유기산 등이 함유되어 있어서 체내의 대사작용을 돕고 세포와 근육에 필요물질인 젖산을 분해하여 피로를 감소시켜주며 신진대사를 촉진하고 있다. 또한 피루브산은 간의 피로, 해독작용을 활발하게 도와주며 카테킨산은 장내의 유해균의 번식을 억제하여 식중독을 예방하고 있다. 또한 이러한 작용은 설사와 변비를 치료하는데도 효과가 있으며 다량의 비타민C의 함유로 인하여 체내의 피로회복을 돕고 있다. 이러한 효능을 가진 매실은 전라남도 해안가에서 주로 재배하고 있다. 특히 순천과 광양지역에서 다양한 품종들을 재배하고 있다. 따라서 본 실험에서는 순천과 광양지역에 재배되고 있는 남고, 천매, 고성, 앵숙, 풍후, 옥영, 임주, 청축, 백가하 등의 잎과 과육, 핵의 추출물로부터 폴리페놀 성분은 핵, 과육보다 잎에서 양호한 반응을 나타내었다. 이러한 결과들을 중심으로 매실의 각 부위별 항산화 활성을 조사하였던 바, 매실의 대부분의 품종에서 항산화 활성이 양호하게 나타났으며 부위별에서는 핵보다는 잎과 과육에서 양호한 항산화 활성을 보였다. 품종별로는 앵숙, 남고, 청축에서 다소 양호한 항산화 활성을 나타내었다. 이러한 결과들로 미루어 볼 때 매실의 항산화 활성은 품종별, 부위별로 상이한 반응을 나타내었다.

The Effect of Prunus Mume Extracts on the Growth of HepG2 and HeLa Cell Lines (간암 및 자궁암 세포주 증식에 미치는 오매 추출물의 영향)

  • This study was undertaken to investigate the inhibitory effect of prunus fume extracts on the growth of Hep G2 and HeLa cells. Prunus mums was extracted using the following solvents hexane, chloroform, ethylacetate, methanol, and hot water. The effect on the growth of each cancer cell line was examined by MTT (3-[4, 5-dimethylthiaeol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, cytotoxicity testing, and microscopic observation. The ethylacetate extracts of Prunus muse at the concentration of 250 $\mu\textrm{g}$/ml exhibited the greatest inhibitory effect on the growth of Hep G2 in the MW assay. In cytotoxicity testing, the treatment of the Hep G2 cells with ethylacetate extracts (1000 $\mu\textrm{g}$/ml for 72 hrs) destroyed 75% of the cells, and morphological changes were also observed. futhermore, the hexane extracts of Prunes muse at the concentration of 250 $\mu\textrm{g}$/ml exhibited the greatest inhibitory effect on the growth of HeLa cells in the MTT assay. The treatment of the HeLa cells with the hexane extracts (1000 $\mu\textrm{g}$/ml for 72 hrs) resulted in the destruction of 68% of the cells. Fibroblasts were not affected by either ethylacetate or hexane extracts of prunus muse.

Quality Characteristics of Kochujang Prepared with Maesil (Prunus mume) Extract during Aging (매실추출액 첨가 고추장의 숙성 중 품질특성 연구)

  • Kochujang was prepared with various concentrations of Maesil extracts (0, 1, 2, and 5%) and the physicochemical characteristics of Maesil extracts added Kochujang ('Maesil Kochujang') were investigated during aging of 100 days. pH decreased slightly during aging. Titratable acidity, on the other hand, increased with aging, reaching at the highest level at the 40 days of aging and then decreased slowly. The changes of moisture content were complicated in the beginning, but after 60 days it increased slowly. The water activity decreased but salt concentration increased consistently in all samples during aging. L* and a*-values decreased during aging and L*, a*, and b*-values decreased significantly with high amount of Maesil extract in the sample. Soluble solid contents increased slowly while reducing sugar contents increased for up to 40 days and then decreased. Amino nitrogen contents increased steadily after 40 days of aging and they reached at $230.00{\sim}246.00mg%$ upon 100 days of aging.

Apoptosis Induction by Methanol Extract of Prunus mume Fruits in Human Leukemia U937 Cells (인체 백혈병세포에서 매실 추출물에 의한 apoptosis 유도)

  • In the present study, the pro-apoptotic effects of methanol extract of Prunus mume fruits (MEPM) in human leukemia U937 cells were investigated. It was found that exposure to MEPM resulted in growth inhibition in a concentration-dependent manner by inducing apoptosis. The induction of apoptotic cell death in U937 cells by MEPM was correlated with a down-regulation of inhibitor of apoptosis protein (IAP) family, such as X-linked inhibitor of apoptosis protein (XIAP) and survivin, anti-apoptotic Bcl-2, up-regulation of FasL and cleavage of Bid. MEPM treatment also induced the proteolytic activation of caspase-3, caspase-8 and caspase-9, and degradation of caspase-3 substrate proteins, such as poly (ADP-ribose) polymerase (PARP) and ${\beta}$-catenin. In addition, apoptotic cell death induced by MEPM was significantly inhibited by z-DEVD-fmk, a caspase-3 specific inhibitor, which demonstrates the important role of caspase-3 in the apoptotic process by MEPM in U937 cells. Taken together, these findings suggest that P. mume extracts may be a potential chemotherapeutic agent for the control of human leukemia cells and further studies will be needed to identify the active compounds.
