Present experiment was carried out to determine maintenance energy requirements for growing Hanwoo steers. Six Hanwoo steers (BW = $180.6{\pm}3.1$ kg) were used in two 3 ${\times}$ 3 latin square design with three different energy intake levels; TDN 1.70 kg (Low), 2.05 kg (Medium), 2.80 kg (High), respectively, based on the Korean Feeding Standards. Each period lasted 18 days including a 14-day adaptation and a 4-day measuring period. The steers were in the head hood chamber system (one cattle per chamber) during each measuring time to measure heat and methane production for 1 day. Dry matter intake was 2,058, 3,256 and 3,881 g/day for Low, Medium and High TDN, respectively. Increase in energy intake did not affect digestibilities of dry matter, crude protein, crude fiber, crude fat, NDF, ADF and nitrogen-free extract. Gross energy intake averaged 180.21, 292.74 and 337.15 kcal/$BW^{0.75}$ for Low, Medium and High TDN, respectively. Energy loss was 28.7% in feces and 2.1% in urine of gross energy intake. Further, energy loss from methane produced during rumen fermentation was 6~8.3%, while body heat loss averaged 34~60%. Intercept of regression equation between ME intake and retained energy indicated that the energy requirement was 109.84 kcal ME/$BW^{0.75}$.

This experiment was conducted to evaluate forage production and quality, and soil improvement of three legumes at Cheonan Yonam College. The three legumes used in the experiment were crimson clover (Trifolium incarnatum L.) red clover(Trifolium pratense L.) and chinese milk vetch(Astragalus sinicus L.). Flowering stage was observed on the 20th of April for chinese milk vetch and on the 30th of April for crimson clover, but the red clover did not produce flower until harvest time. The dry matter(DM) content of crimson clover was the highest among the three legumes. In terms of DM yield, crimson clover has the highest yield, the highest yield in crude protein(CP) and total digestible nutrients(TDN) was red clover. In terms of forage quality, the highest CP content was red clover, while the chinese milk vetch was the lowest for both neutral detergent fiber(NDF) and acid detergent fiberr(ADF). Red clove. and chinese milk vetch were highest fur TDN content, and the chinese milk vetch has the highest for relative feed value(RFV). Assessment of the chemical properties of soil after harvest showed that the nitrogen content of soil increased in all legumes. The organic matter(OM) content of soil in both crimson and red clover increased, while the chinese milk vetch decreased. The available $P_2O_5$ decreased in all three legumes, but the biggest decrease was in the chinese milk vetch. The exchangeable potassium in the soil for both crimson and red clover increased, but in chinese milk vetch it decreased. Based on the results of this study, the chinese milk vetch was superior in terms of forage quality, but crimson and red clover are excellent in forage yield and in maintaining soil quality in upland.

Four ruminally cannulated Hanwoo steers (BW 600 ${\pm}$ 48.4 kg) fed whole crop rice silage (WRS) as forage were used to investigate the effects of its dietary on rumen fermentation and blood metabolites at different harvesting time such as milk, dough, yellow ripe and mature stages of WRS, respectively. Mean ruminal pH sampled at 0, 1, 2, 4, 6 and 8 h after the morning feeding was not significantly (p>0.05) different between the WRS by the harvesting time. In diurnal pattern in ruminal pH, the lowest pH for the mature stage treatment (Mature) appeared at 6 h after the feeding while it appeared at 2-4 h for the other treatments. This may indicate that retention time in the rumen of Hanwoo fed WRS harvested at the Mature increased. Although feeding WRS harvested at different stages did not significantly (p>0.05) affect ruminal ammonia N concentration, the ammonia N for WRS harvested at the milk stage (Milk) tended (p=0.11) to decrease compared with that for the yellow stage treatment (Yellow). In the blood metabolites analysis, BUN concentration for Yellow numerically decreased compared with that for Milk. This, taking lower concentration of ruminal ammonia N for Milk than the other treatments into account, clearly indicates that the BUN concentrations may not increase in proportion to ruminal ammonia N concentration according to ammonia N concentration entering into the lower gastric tracts. Lower (p=0.03) concentration of blood albumin for the dough stage treatment than that for Mature may indicate high protein synthesis in Hanwoo fed WRS at the dough stage, but further studies in terms of mechanism of nutrients distribution should be required. Present results indicate that different harvesting time did not affect rumen fermentation and blood metabolites in Hanwoo. Thus, based on the results of the present and the previous studies, utilizing WRS harvested at yellow stage could be recommendable for Hanwoo steers.
Heme oxygenase-1 (HO-1) is a stress-responsive protein that is known to regulate cellular functions such as cell proliferation, inflammation, and apoptosis. In this study, we investigated the role of NADPH oxidase on the expression of HO-1 in human liver hepatoma cell line HepG2. Diphenyleneiodonium (DPI), an NADPH oxidase inhibitor, markedly inhibited HO-1 expression and the nuclear translocation of transcription factor Nrf2 in cobalt protoporphyrin (CoPP) or hemin-treated HepG2 cells. Similarly, the knockdown of $p47^{phox}$, a cytosolic factor for NADPH oxidase activity, by siRNA inhibited the CoPP-induced expression of HO-1. In addition, GSHmee, an intracellular antioxidant, blocked the expression of HO-1 in CoPP-treated cells. Based on these results, we conclude that the blockage of NADPH oxidase with DPI or $p47^{phox}$ siRNA inhibits CoPP-induced HO-1 expression in HepG2 cells, and also suggest that the expression of HO-1 in CoPP-induced HepG2 cells is associated with increase of intracellular ROS by NADPH oxidase activity.
In this study, fibrinolytic activities of fermented yellow agabean (FYA) and black agabean (FBA), and the antioxidation efficiencies of 70% ethanol extract of fermented yellow agabean (FYAE) and black agabean (FBAE) were investigated by selecting Bacillus sp. sm26 strain. Fibrinolytic activities of FYA and FBA were $6.38{\pm}0.5$ and $6.83{\pm}0.5\;U/ml$, which were 1.3 and 1.4 times higher than that of FSB, respectively. With regard to total phenolic contents, FYAE and FBAE were $3.40{\pm}0.44\;mg/g$ and $2.45{\pm}0.20\;mg/g$ respectively, suggesting that their contents were about twice as high as that of fermented soybean extract (FSBE) used as a control. In comparison with FSBE, total protein and sugar contents of FYAE were $0.56{\pm}0.11$ and $2.41{\pm}0.48\;mg/g$, respectively, and those of FBAE were $0.39{\pm}0.12$ and $2.72{\pm}0.63\;mg/g$, respectively. This result suggests that FYAE was 4.7 and 1.7 times higher than FSBE, respectively. The DPPH radical scavenging activity of FBAE was 79% at 1 mg/ml, which was highest among the fermented bean extracts, and was twice as high as FSBE in regards to activity. In addition, FBAE exhibited the highest reducing power at 1 mg/ml, which was higher than FSBE by two-fold. With regard to lipid peroxidation, FBAE and FYAE were 93% and 80% at 1 mg/ml, which were 3 and 2.5 times higher than FSBE, respectively. Of note, the hydrogen peroxide scavenging activities of FBAE and FYAE were 82% and 54% at 1 mg/ml, offering activity that was 4 and 2.5 times higher than FSBE, respectively. Based on these results, the fibrinolytic activity and antioxidation efficiency of the fermented agabeans were significantly higher than other soybeans. Therefore, these studies may suggest that the functional agabeans can be a potential candidate for a natural functional food.
In the previous paper, we isolated a bacterium that can hydrolyze various organic materials from soybean paste, including cellulose, lipids, starch, and protein. The activity and chemical properties of the crude enzymes produced by the isolate Bacillus subtilis CK-2 were further investigated. Cellulase showed the highest activity at pH 5.0 and $55^{\circ}C$. The stability of cellulase was maintained within the ranges of pH 5.0~10.0 and $20{\sim}50^{\circ}C$. Cellulolytic enzymes were activated by a $Co^{2+}$ ion, demonstrating the highest activity at a 0.45%(w/v) concentration of $Co^{2+}$. The optimal conditions for amylase were pH 5.0 and $50^{\circ}C$. The activity of amylase was stable within the ranges of pH 4.0~5.0 and $20{\sim}50^{\circ}C$. The $Co^{2+}$ ion was also necessary for amylase activity, which was the highest at a 0.2%(w/v) concentration of $Co^{2+}$. The optimal pH and temperature conditions of protease were pH 8.0 and $50^{\circ}C$. The activity of protease was stable within the ranges of pH 7.0~8.5 and $20{\sim}50^{\circ}C$. Protease activity was catalyzed by $Mn^{2+}$, which was the highest at a 0.125%(w/v) concentration of $Mn^{2+}$. The isolate B. subtilis CK-2 demonstrated a high activity of autolysin. Based on these results, we identified and suggested the optimal pH, temperature, and metal ion concentration in the use of the hydrolytic enzymes of B. subtilis CK-2 for industrial purposes.

A unicellular green alga was axenically isolated from a tidal pool on Ulleung-do, Korea. Morphological, molecular, and biochemical analyses revealed that the isolate belonged to Auxenochlorella protothecoides. The current study is the first record of this species in Korea. The microalgal strain was named as A. protothecoides MM0011 and its growth, lipid and pigment compositions, and biomass properties were investigated. The strain is able to thrive in a wide range of temperatures ($5{\sim}35^{\circ}C$) and to withstand up to 1.5 M NaCl. The results of GC/MS analysis showed that the isolate was rich in nutritionally important polyunsaturated fatty acids (PUFAs). Its major fatty acids were linoleic acid (27.6%) and ${\alpha}-linolenic$ acid (39.6%). Thus, this indigenous microalga has potential as an alternative source of ${\omega}3$ and ${\omega}6$ PUFAs, which currently come from fish and plant oils. Also, the HPLC analysis revealed that the value-added antioxidant, lutein, was biosynthesized as the accessory pigments by the microalga. A proximate analysis showed that the volatile matter content was 85.6% and an ultimate analysis indicated that the gross calorific value was $20.3MJ\;kg^{-1}$. Since 40.5% of total nitrogen and 27.9% of total phosphorus were removed from the medium, respectively, it also has potential as a feedstock for biofuel applications which could be coupled to wastewater treatment. In addition, the biomass may also serve as an excellent animal feed because of its high protein content (51.4%). Therefore, A. protothecoides MM0011 shows promise for application in production of microalgae-based biochemicals and as a biomass feedstock.
Background: Early diagnosis of tuberculosis is critical, especially in Korea, an area where tuberculosis is endemic. Because antibody responses to some membrane proteins of Mycobacterium tuberculosis are not comparable, and the policy of BCG vaccination and the prevalence of tuberculosis are different from country to country, the usefulness of the serological diagnostic tests is questionable in Korea, even though they have been confirmed to be useful in other countries. In the specific context of Korea, we tried to evaluate the validity of the ICT Tuberculosis Test (ICT), a membrane-based antibody kit that purports to detect the 5 M. tuberculosis complex-specific antigens including 38-kDa protein. Method: 68 patients with tuberculosis were tested : 37 had no history of previous tuberculosis, and 31 were reactivated cases. The control group comprised 77 subjects : 25 healthy adults, 35 hospital workers with frequent contact with tuberculosis patients, and 17 in-patients with non-tuberculous respiratory diseases. Results: The diagnostic sensitivities of the ICT were 87% and 73% in patients with versus without previous history of tuberculosis, respectively. The sensitivities of smear-positive and smear-negative patient groups were 81% and 73%, respectively. Both of the two patients with extrapulmonary tuberculosis tested positive through the ICT. The specificities of the ICT were 88%, 94%, and 94% in healthy adults, hospital workers, and non-tuberculous patients, respectively, with an overall specificity of 92%. Conclusion: It is suggested that when combined with traditional techniques, the ICT is an useful tool for the diagnosis of pulmonary tuberculosis. The procedure is simple, easy to perform, rapid, and needs no equipment. It shows 73% sensitivity and 92% specificity for the diagnosis of tuberculosis.

Purpose : The objective of this study was to determine whether the expressions of the two components of DNA-dependent protein kinase, Ku70 and DNA-PKcs, influence the response to radiotherapy (RT) and outcome of treatment of non-disseminated nasopharyngeal carcinoma (NPC) in patients who received definitive RT. Materials and Methods : Sixty-six patients with NPC who were treated with radiotherapy alone or with concurrent chemotherapy between June 1995 and December 2001 were divided into groups based on the levels of immunoreactivity for Ku70 and DNA-PKcs in pretreatment biopsy specimens. The over-expression of Ku70 or DNA-PKcs groups Included patients whose biopsy specimens showed at least 50% immunopositive tumor cells; patients in which less than 50% of the tumor cells in the biopsy tissues were immunopositive were placed in the low Ku70 and DNA-PKcs groups. The immunoreactivities for Ku70 and DNA-PKcs were retrospectively compared with the sensitivity of the tumor to radiation and the patterns of therapy failure. Univariate analyses were peformed to determine the prognostic factors that influenced locoregional control of NPC. Results : The five-year locoregional control rate was significantly higher in the low Ku70 group (Ku(-)) (85%) than in the high Ku70 group (Ku(+)) (42%) (p=0.0042). However, there were no differences in the metastases-free survival rates between the two groups (Ku70 (+), 82%; Ku70 (-), 78%; p=0.8672). Univariate analysis indicated that the over-expression of Ku70 surpassed other well-known predictive clinocopathologic parameters as an Independent prognostic factor for locoregionai control. Eighteen of 22 patients who had locoregional recurrences of the tumor displayed an over-expression of Ku70. No significant association was found between the level of DNA-PKcs expression and the clinical outcome. Conclusion : Our data suggest that the level of Ku70 expression can be used as a molecular marker to predict the response to RT and the locoregional control after RT and concurrent chemotherapy in patients with non-disseminated NPC.
In the current study, the mesenchymal stem cells (MSCs) isolated and propagated from the human umbilical cord blood (UCB) were tested for their capabilities of differentiation into chondrocytes in vitro. The mesenchymal progenitor cells (MPCs) collected from UCB were cultured in a low glucose DMEM medium with 10% FBS, L-glutamine and antibiotics. The human MSC colonies were positively stained by PAS reaction. When the immunophenotypes of surface antigens on the MSCs were analyzed by fluorescence-activated cell sorter (FACS) analysis, these cells expressed positively MSC-related antigens of CD 29, CD44, CD 90 and CD105, whereas they did not express antigens of CD14, CD31, CD34, CD45, CD133 and HLA-DR. Following induction these MSCs into chondrocytes in the chondrogenic differentiation medium for 3 weeks or more, the cells were stained positively with safranin O. We clearly confirmed that human MSCs were successfully differentiated into chondrocytes by RT-PCR and immunofluorescent stain of type-II collagen protein. These data also indicate that the isolation, proliferation and differentiation of the hUCB-derived MSCs in vitro can be used for elucidating the mechanisms involved in chondrogenesis. Moreover this differentiation technique can be applied to developing cell-based tissue regeneration or repair damaged tissues.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
