The giant mealworm beetle, Zophobas atratus (Coleoptera: Tenebrionidae) has been used as a protein source for small pets and mammals. Recently, it was temporarily registered in the list of the Food Code. We previously performed an in silico analysis of the Zophobas atratus transcriptome to identify putative antimicrobial peptides and identified several antimicrobial peptide candidates. Among them, we assessed the antimicrobial and anti-inflammatory activities of zophobacin 1 that was selected bio-informatically based on its physicochemical properties against microorganisms and mouse macrophage Raw264.7 cells. Zophobacin 1 showed antimicrobial activities against microorganisms without inducing hemolysis and decreased the nitric oxide production of the lipopolysaccharide-induced Raw264.7 cells. Moreover, ELISA and Western blot analysis revealed that zophobacin 1 reduced expression levels of pro-inflammatory enzymes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). We also investigated expression of pro-inflammatory cytokines (interleukin-6 and interleukin-1β) production through quantitative real time-PCR and ELISA. Zophobacin 1 markedly reduced the expression level of cytokines through the regulation of mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B (NF-κB) signaling. We confirmed that zophobacin 1 bound to bacterial cell membranes via a specific interaction with lipopolysaccharides. These data suggest that zophobacin 1 could be promising molecules for development as antimicrobial and anti-inflammatory therapeutic agents.
Study objectives: To determine the factors associated with mortality after an in-hospital diagnosis of tuberculosis in a region with low levels of HIV coinfection. Methods: From January 2003 to December 2004, all subjects who were > 15 years of age and had received a diagnosis of tuberculosis were registered. The clinical, radiological and laboratory aspects of the patients who died (n=27) were compared with those of an age and gender matched control population(n=54). Logistic regression analyses were carried out, which included age, gender, hospital admission source, initial site of admission, dyspnea, general weakness and initial laboratory data. Results: The mean age of the patients was $60{\pm}16$ years and male patients outnumbered female patients. Univariate analysis identified hemoglobin, blood urea nitrogen, albumin, cholesterol, aspartate aminotransferase (AST), C-reactive protein and the risk factors for tuberculosis to be significantly associated with mortality. Among the characteristics of disease presentation and treatment, emergency department admission, intensive care unit, disease severity, general weakness and dyspnea at the time of admission were associated with mortality. Multiple regression analysis revealed the initial management in the intensive care unit and lower serum albumin to be independently associated with mortality. Conclusion: The markers of disease chronicity and severity appear to be associated with in-hospital mortality. Identifying potentially reversible factors such as malnutrition and respiratory failure suggests specific intervention that might lead to an improvement in the patients' outcomes.
Because of our aging population, there is increasing concern about the impact of dementia and age-related cognitive decline. Intense research efforts on effective dietary interventions for the prevention or amelioration of dementia and age-related cognitive decline have indicated that dairy products affect physiological health and potentially healthy brain function during aging. Milk is a rich source of proteins and peptides with nutritional and immunotropic activities. The preparation of biologically active proteins and peptides generally requires enzymatic degradation, chemical modification, or the addition of specific co-factors. Milk-derived preparations are widely available in the food industry in the form of hygiene products and infant formulas. However, milk-derived products could also be applied as preventive or therapeutic measures for a wide-range of pathological conditions not only in neonates and infants but also in adults, including the elderly. Because they have no adverse side effects, milk-derived proteins and peptides could be used as a supplementary treatment for dementia and age-related cognitive decline.

Objective: Human embryonic stem cells (hESCs) can proliferate indefinitely and differentiate into all kinds of cell types in vitro. Therefore, hESCs can be used as a cell source for cell-based therapy. Transduction of foreign genes to hESCs could be useful for tracing differentiation processes of hESCs and elucidation of gene function. Thus, we tried to introduce enhanced green fluorescent protein (eGFP) gene to hESCs, XX and XY cell lines in this study. Methods: Lentivirus containing eGFP was packaged in 293T cells and applied to hESCs to transduce eGFP. Expression of transduced eGFP was evaluated under the fluorescence microscope and eGFP positive population was analyzed by FACS. Expression of undifferentiation state markers such as Oct4, Nanog, SSEA4 and Tra-1-81 was examined by RT-PCR and/or immunofluorescence in eGFP-hESCs after transduction. In addition, the ability of eGFP-hESCs to form embryoid bodies (EBs) was tested. Results: eGFP was successfully transduced to hESCs by lentivirus. eGFP expression was stably maintained up to more than 40 passages. eGFP-hESCs retained expression patterns of undifferentiation state markers after transduction. Interestingly, disappearance of transduced eGFP was notably observed during spontaneous differentiation of eGFP-hESCs. Conclusion: We established eGFP expressing hESC lines using lentivirus and showed the maintenance of undifferentiation characteristics of these eGFP-hESCs. This reporter-containing hESCs could be useful for tracing the processes of differentiation of hESCs and other studies.

This study was carried out to examine nutrients intake and food preference in 100 elderly people aged over 85 years residing in Namhae-gun Kyungnam. Mean daily energy intake and Korean recommended dietary allowances (RDA) percent were 1547.6$\pm$452.7 kcal and 91.5%, respectively. Mean daily intakes of protein (63.1$\pm$28.2 g), iron (16.7$\pm$11.2 mg), vitamin Bi (1.0410.4 mg) and vitamin C (97.0$\pm$45.3 mg) were higher than RDA. The intakes of calcium (418.1$\pm$189.7 mg) and vitamin B2 (0.94$\pm$0.5 mg) were shown to be 58.4% and 78.4% of the RDA. Females had higher intakes of energy and most other nutrients than males. Major food source of energy and protein intakes was rice and cereals, and that of lipid intake was meat, fish, egg and beans. Cooked rice, all kind of vegetables, fish, yogurt and sugars were preferred to the subjects. The most preferred taste was sweet.
Experiments were carried out to establish the effects of acids in neutralization after alkaline treatment of rice straw, with which cellulosic single cell protein can be produced by cellulose utilizing bacteria, Cellulomonas flavigena KIST 321, previously isolated by authors. Following results were obtained. 1. Rice straw as carbon source was pretreated with 10 volumes of 1 normality of NH$_4$OH or NaOH(NaOH/substrate:40%, and then washed with water or neutralized with H$_3$PO$_4$, H$_2$SO$_4$, HCl and CH$_3$COOH. Among the above mentioned methods, neutralization with H$_3$PO$_4$after alkaline treatment was proved to be the most effective on its digestibility and SCP production. Dry cell 12.28g/$\ell$ and 78% digestibility were obtained. 2. When rice straw was treated with NaOH solution, the result suggested that the productibity of cell-mass was attained on treatment of rice straw with 6% of NaOH (NaOH/substrate ratio) for 15~24hrs at room temperature. 3. When rice straw was treated with NaOH, a volume of water to substrate is adequate by two or three fold and the amount of NaOH can be economized up to 5% for the weight of rice straw. 4. The steaming of rice straw at 121$^{\circ}C$ for 30min. in alkaline treatment of rice straw gave the similiar effectiveness to that at room temperature for 15~24hrs. and accelerated the sterilization of the substrate. 5. Finally, the level of inorganic phosphate in a medium was investigated. 11.2g of dry cell was produced at the concentration of 0.2%, phosphate (phosphorous level 0.04%) in medium even though treated rice straw was neutralized with HCI instead of H$_3$PO$_4$, and 12.2g/$\ell$ at the concentration of 0.3% phosphate (phosphorous 0.04%) on neutralization with H$_2$SO$_4$.
Industrial wastes from pulp and food plants were treated with microorganisms to clarify organic waste-water and to produce cells as animal feed, and results were summarized as follows. (1) Waste-water from pulp, beer, bread yeast, and ethanol distillation plants contained $1.4{\sim}1.5%$ of total sugar, $0.25{\sim}0.35%$ nitrogen, and biological oxygen demand (BOD) was $400{\sim}25,000$, chemical oxygen demand (COD), $500{\sim}28,000$, and pH, $3.8{\sim}7.0$. The BOD and COD were highest in waste-water from ethanol distillation plants among others. (2) Bacterial and yeast counts were $4{\times}10^4-1{\times}10^9,\;2{\times}10^2-7{\times}10^4/ml$ in waste-water. (3) Bacteria grew better in pulp waste and yeasts in beer, bread yeast, and ethanol distillation waste. (4) Saccharomyces cerevisiae SAFM 1008 and Candida curvata SAFM 70 were the most suitable microorganisms for clarification of ethanol distillation waste. (5) When liquid and solid waste from ethanol distillation were treated with microbial cellulase, xylanase, and pectinase, solid waste was reduced by 36%, soluble waste was increased, and recuding sugar content was increased by 1.3 times which provided better medium than untreated waste for cultivation of yeasts. (6) Optimum growth conditions of the two species of yeast in ethanol distillation waste were pH 5.0, $30^{\circ}C$, and addition of 0.2% of urea, 0.1% of $KH_2PO_4$ and 0.02% of $MgSO_4$. (7) Minimum number of yeast for proper propagation was $1.8{\times}10^5/ml$. (8) C. curvata70 was better than cerevisae for the production of yeast cells from ethanol distillation waste treated with microbial enzymes. (9) S. cerevisiae produced 16 g of dried cell per 1,000ml of ethanol distillation waste and reduced BOD by 46%. C. curvata produced 17.6g of dried cell and reduced BOD by 52% at the same condition. (10) Yeast cells produced from the ethanol distillation waste contained 46-52% protein indicating suitability as a protein source for animal feed.

As a part of basic investigation for utilizing cultured eel and by-products as a food source, a mineral contents and fatty acid composition of bone and flesh were investigated. Flesh of cultured eel was higher in moisture and crude protein content, and lower in crude ash and lipid content than those of the bone. Mineral of bone were measured 220.72 mg/100 g of Ca, 169.87 mg/100 g of P, 117.05 mg/100 g of Na,92.75 mg/100g of K, 6.18 mg/100g of Cu,5.02 mg/100 g of Zn,2.56 mg/100 g of Fe, and flesh were measured 120.23 mg/100g of CL 150.36 mg/100 g of P, 136.36 mg/100 g of Na, 89.36 mg/100 g of K, 4.02 mg/100 g of Cu, 1.71 mg/100 g of Zn,2.03 mg/100 g of Fe. The major fatty acid in bone and flesh of cultured eel were generally oleic acid $(44.40\%, 43.76\%)$, palmitic acid $(24.19\%,\;21.30\%)$, palmitoleic acid $(8.18\%,\;7.72\%)$, eicosapentaenoic acid $(5.72\%,\;6.65\%)$, myristic acid $(3.96\%,4.34\%)$ in order. The fstty acid composition of total lipid was no significant difference among bone and flesh of cultured eel, However, bone of cultured eel revealed higher content in saturated and monounsaturated fatty acid, while lower in polyunsaturated fatty acid than those of the flesh. It is shown that cultured eel contains various nutrients such as protein, minerals, unsaturated fatty acid, so cultured eel can be regarded as a highly nutritious food.
The effects of different dietary lipids on growth, body chemical composition, and nutrient partition of the Korean rockfish (43 g of initial mean weight) were studied. Fish were fed three experimental diets containing 1) $4.5\%$ squid liver oil, 2) $1\%$ squid liver oil and $3.5\%$ beef tallow, and 3) $1\%$ squid liver oil and $3.5\%$ soybean oil. Growth and feed efficiency were detarmined during a 19-week feeding period at water temperature of $15.5^{\circ}C$, and at the end of which body chemical compositions were analyzed. Remained fish were fasted for 45 days, and four fish were taken at 6, 12 and 24 hours, and 3, 15 and 45 days after the fasting to determine tissue compositions and hematological changes. Growth, feed efficiency, protein and lipid retention efficiency, hepatosomatic index(HSI), viscerosomatic index (VSI), and chemical composition of whole body and dorsal muscle were not affected by the different dietary lipids. Liver moisture content of fish fed squid liver oil diet was higher than that of fish fed beef tallow or soybean oil diet. Liver lipid content of fish fed beef tallow diet was higher than that of fish fed the other diets. Fatty acid composition of dorsal muscle and liver were affected by the different dietary lipids ; high levels of 20 : 5n-3 and 22 : 6n-3 from fish fed the diet containing squid liver oil, 18 : 1 from fish fed the diet containing beef tallow, and high 18 : 2n-6 and 18 : 3n-3 from fish fed the diet containing soybean oil were observed. Both HSI and VSI of fish fed three diets decreased with time after the begining of starvation. Liver glycogen did not change during the first 15 days of starvation and decreased thereafter, and that was not affected by the different dietary lipids. Lipid and protein contents in the dorsal muscle of fish decreased up to 15 days of starvation and remained unchanged thereafter, these were not different from each other. Glucose, free fatty acid, triglyceride and phospholipid concentrations in Fish serum were varied for the first 15 days of starvation, after that the concentrations of fish serum remained relatively stable in all the treatment groups with prolonged starvation. The results indicate that Korean rockfish can utilize fish oil, animal fat or vegetable oil equally as energy source when n-3HUF A is adquate.
A series of rearing experiments were conducted to determine the growth rates and feed conversion efficiencies of tilapia in accordance with body size or age in nearly total closed system glass aquariums ($270\;\ell$ each in water volume) and concrete tanks ($4000\;\ell$) from April 10 to October 16, 1987. The fish used for the experiments was a Japanese strain of Oreochromis niloticus, and the size of the fish ranged from 7 g to more than 1,000 g in body weight. The starting stocking rates for each experimental lot were 10 to 20 kg in the glass aquarium ($3.7{\%}$ to $7.4{\%}$ of water volume) and 200 kg in the concrete tank ($5{\%}$ of water volume). A single experimental rearing term was 14 days with slight variations on occasions. Water temperature was designed to be kept at $26^{\circ}C$ but slight fluctuations were inevitable. Dissolved oxygen level was designed to be maintained at around $3\;mg/\ell$, but it also showed some variations. The ammonia level in the glass aquarium section once reached up to $18\;mg/\ell$, but generally remained at around $4\;mg/\ell$, and in the concrete tank section it was maintained at around $1\;mg/ell$. The feed was composed of mainly soybean meal with a small amount of fish meal as the protein source, and the crude protein content was about $32{\%}$. Mean daily growth rate was $3.5{\%}$ of body weight with 0.9 in food conversion ratio in the glass aquarium when the mean weight of fish was around 10 g with gradually reduced performances as the fish grew bigger. When the mean weight was 800 g, mean daily growth rate was $0.5{\%}$ with about 1.5 in food coversion for fish in the glass aquarium, and $0.8{\%}$ and 1.6 for fish in the concrete tank, respectively. According to the mean growth rate obtained from this experiment, it was calculated that the fish reared in the concrete tank require 223 days from 50 g to reach 1,000 g which is the ideal size for market in Korea, at the conditions provided as above, and 302 days from 10 g fingerlings to 800 g fish in the glass aquarium conditions of the closed recirculating water system.
