Preparation and Characterization of New Immunoprotecting Membrane Coated with Amphiphilic Multiblock Copolymer

  • New immunoprotecting membranes were prepared by spin coating the amphiphilic random multiblock copolymers of poly(ethylene glycol) (PEG) and poly(tetramethylene ether glycol) (PTMEG) or poly(dimethyl siloxane) (PDMS) on porous Durapore(R) membrane. The copolymer coating was intended to make a biocompatible, immunoprotecting diffusional barrier and the supporting porous substrate was for mechanical stability and processability. By filling Durapore(R) membrane pores with water, the penetration of coating solution into the pores was minimized during the spin coating process. A single coating process produced a completely covered thin surface layer (~1 ${\mu}{\textrm}{m}$ in thickness) on the porous substrate membrane. The permselectivity of the coated layer was influenced by PEG block length, polymer composition, and thickness of the coating layer. A composite membrane with the coating layer prepared with PEG 2 K/PTMEG 2 K block copolymer showed that its molecular weight cut-of fat any 40 based on dextran was close to the molecular size of IgG (Mw = 150 kDa). However, IgG permeation was detected from protein permeation test, while glucose oxidase (Mw = 186 kDa) was not permeable through the coated membrane.

Chitosan/whey Protein (CWP) Edible Films Efficiency for Controlling Mould Growth and on Microbiological, Chemical and Sensory Properties During Storage of Göbek Kashar Cheese

  • The objective of present study was to evaluate the effects of the application of chitosan and chitosan/whey protein on the chemical, microbial and organoleptic properties of Göbek Kashar cheese during ripening time (on 3rd, 30th, 60th and 90th d). Difference in microbiological and chemical changes between samples was found to be significant (p<0.05) during ripening period. Cheese samples with edible coating had statistically lower mould counts compared to the uncoated samples. Furthermore the highest and lowest mould counts were determined in control (4.20 Log CFU/g) and other samples (<1 Log CFU/g) at 60th and 90th d of storage. All samples exhibited higher levels of water soluble nitrogen and ripening index at the end of storage process. At the end of 90 day storage period, no signicant dierences in salt and fat values were observed among the cheeses studied. The edible coatings had a beneficial effect on the sensory quality of cheese samples. In the result of sensory analysis, while cheese C and the chitosan coated cheese samples were more preferred by the panellists, the chitosan/whey protein film-coated cheese samples received the lowest scores. This study shows coating suggests could be used to improve the quality of cheese during ripening time.

수분거동 패턴에 따른 차폐막 설정을 통한 모발단백질 소실방지 (Prevention of Protein Loss Using A Shield Coating According to Moisture Behavior in Human Hair)

  • 세정 과정에서 물에 의한 단백질 소실을 차폐막을 통해서 방지 하고자 먼저 모발 내부의 물 거동 경로를 파악하였다. 이를 위해서 소수성을 가지는 건강한 모발과, 모발 내부는 정상이지만 표면의 최외곽 지질층에서 18-methyleicosanoic acid (18-MEA)만 소실시켜 친수성이 된 모발과, 다시 표면에 18-MEA를 결합시킨 회복모를 이용해서 모발 수분의 거동을 파악하였다. 모발에 열을 가할 때 일어나는 수분 증발에 따른 질량 변화를 관찰한 결과 표면이 소수성인 모발들은 39 s와 151 s의 두개의 시간 상수로 감소하였다. 이에 반해 표면이 친수성으로 변한 손상 모발에서는 83 s의 하나의 빠른 시간 상수로 감소함을 확인하였는데, 이는 모발 내부에 있는 비결합과 결합 수분들이 외곽으로 빠져나올때 모발 표면에 소수성 막이 없음으로써 바로 용출됨을 반증한다. 따라서 세정과정에서 모발 내부의 단백질 용출을 효율적으로 방지하기 위해서는 모발 표면을 소수성 코팅하여 물분자의 거동을 저지시킬 필요가 있을 것이라는 가정한 결과 친수성 polyethylene glycol (PEG)를 코팅한 경우 179 ㎍/mL의 단백질이 용출한데 반해서 소수성 polydimethylsiloxane (PDMS)를 코팅한 모발은 보다 적은 148 ㎍/mL를 용출하였으며, 표면을 소수성 및 친수성으로 코팅하여 모발 내부 단백질을 검량하여 비교하였다. 마침내 소수성 차폐막이 세정 과정에서 모발 단백질 소실을 방어하는 방법이 됨을 확인하였고, 이 소수성 지표를 lateral force microscopy (LFM) 값으로 환산하여 정리하였다. 마침내 이 연구 결과물은 세정 과정에서 발생하는 단백질 소실을 소수성 코팅막을 부여함으로써 막을 수 있는 모발 세정제품 개발에 기여할 수 있다.

대두단백 코팅 종이의 수분저항성 및 물리적 성질 (Effect of Water Resistance and Physical Properties of Soy Protein Isolate coated Liner Board)

  • 대두단백 필름을 코팅한 라이너지의 효과를 알아보기 위하여 대두단백 $2\sim8%$의 농도 범위에서 필름을 제조하였다. 필름 제조시 친수성 소재인 대두단백의 불용화를 위하여 모든 필름은 formaldehyde로 포화된 데시케이터에서 2시간 동안 흡착하여 사용하였다. 필름의 연신강도, 연신율, 투습도 및 수분용해도를 측정한 뒤 최적 농도를 5%로 판단하였다. 필름의 제조적성을 위해 첨가된 가소제 glycerol은 대두단백 대비 40% 농도에서 필름제조에 가장 적합하였다. 앞의 조건을 이용하여 제조된 용액을 라이너지에 코팅하여 그 물성과 수분저항성을 측정하였다. Formaldehyde 처리된 대두단백 코팅 라이너지는 미처리 라이너지에 비하여 연신강도는 15 에서 21 MPa로 증가하였고, 수분용해도와 투습계수는 1.17%와 $2.06ng{\cdot}m/m^2{\cdot}s{\cdot}Pa$로 감소하여 물리적 성질과 수분저항성 모두 증진된 것을 알 수 있었다.

PDMS 블레이드 코팅법을 이용한 종이-기반 바이오센서칩 제작 (Fabrication of Paper-based Biosensor Chip Using Polydimethylsiloxane Blade Coating Method)

  • 본 연구는 적은 비용으로 분석 장치 없이 질병 진단 및 경과를 모니터링할 수 있는 종이-기반 분석 장치(paper-based analytical device, PAD)를 제작하기 위해 polydimethylsiloxane (PDMS) 블레이드 코팅 방법을 제안하였다. PAD 디자인은 레이저 커팅 기술로 쉽게 몰드에 적용할 수 있으며, 제작된 몰드로 블레이드 코팅을 수행하여 완전한 소수성 장벽 형성에 필요한 조건을 확립하였다. 코팅 조건인 잉크의 두께와 종이와의 접촉시간에 따라 PDMS 소수성 장벽의 구조와 친수성 채널의 크기 변화를 분석하여 안정적으로 소수성 장벽을 형성할 수 있는 조건을 최적화하였다. 최적화된 방법을 바탕으로 PAD를 제작하여 특별한 분석기기 없이 단백질, 당, 메탈이온을 검출하여 바이오센서에 응용가능함을 증명하였다.

Effect of chitosan coating combined with hypotaurine on the quality of shrimp (Litopenaeus vannamei) during storage

  • This study aimed to investigate the effect of different coating materials on the quality of shrimp (Litopenaeus vannamei) during chilled storage for 10 days. Fresh shrimp were randomly divided into five groups: the control group, the hypotaurine treatment group (2%), the chitosan group (1%), the hypotaurine + chitosan group (2% hypotaurine solution with 1% of chitosan), and the sodium metabisulfite treatment group (1.25%). Compared with other treatments, the lower accumulation of total visible counts (TVC, 5.25 Log10 CFU/g), total volatile basic nitrogen (TVB-N, 22.5 mg/100 g) and thiobarbituric acid values (TBA, 0.58 mg MDA/kg) suggested that coating of chitosan-hypotaurine could retard the microbial activity, protein degradation and lipid oxidation of shrimp. Meanwhile, results demonstrated that the chitosan coating combined with hypotaurine showed an excellent performance in inhibiting quality deterioration (pH 7.5, ∆E 7.0, hardness 393 g, and elasticity 0.69). Furthermore, the melanosis degree of shrimp was alleviated, and the sensory parameters, including appearance, odor and texture, were maintained to the acceptable level by chitosan based hypotaurine treatment during the chilled storage.

단백질 칩 기판의 표면에 미치는 용매 효과 (Effect of Solvent on the Surface of Protein Chip Plate)

  • Nickel chloride coated protein chip plate was developed by using a spin coating method. The ability of histidine tagged protein adsorption was investigated at various solvents. The surface of plate has a large aggregated nickel complex with high density in water. However, the surface of plate has a very small size of aggregated nickel complex with low density in isopropanol. The ability of protein adsorption decreased as increasing the size of alkyl chain in various alcohol solvents. The mechanism on the ability of protein adsorption at the plate surface is discussed.

친화성 막모듈에 의한 단백질 크로마토그래픽 특성 (Characteristics of Protein Chromatography by Affinity Membrane Mudule)

  • Protein affinity membrane was prepared via the coating of chitosan gel on the porous flat polysulfone membrane surface, followed by the immobilization f the reactive dye (Cibacron Blue 3GA) to the chitonsan gel. The maximum protein binding capacity of affinity membrane was about 70${\mu}g/cm^2$ determined by the batch adsorption experiments of human serum albumin (HSA). Using module of this membrane, the characteristics of protein chromatography were investigated through the experiments of elution and frontal chromatography of HSA. This membrane module promises as a chromatography column, since it represented a lower pressure drop and a greater reproducibility. The protein separation ratio was significantly influenced by the flow rate of mobile phase and the injection quantity of HSA. The dynamic protein binding capacity of module decreased from the equilibrium binding capacity with increasing flow rate and approached the value of 15 - 20 ${\mu}g/cm^2$ for flow rates above 6 mL/min.

The Effect of Gelatin Coating and Sonication on the Quality Properties of Wet-Aging Pork Loins

  • In this study, we evaluate the effect of gelatin coating and sonication of wet-aged pork loin on quality. The moisture content of wet-aged pork loin with sonication and gelatin coating was the highest in the G5S sample (5% gelatin coating and sonication), while the moisture content of wet-aged pork loin with sonication was higher than that without sonication. The pH of wet-aged pork loin with sonication was lower than that without sonication. The aging loss of 5% gelatin coating with sonication was significantly lower than that of G0 (control), while the cooking loss was the lowest in G0 wet-aged pork loin. The water holding capacity of the wet-aged pork loin was the highest in G1. The thiobarbituric acid reactive substances value of wet-aged pork loin was significantly decreased with coating and not affected by sonication. The gelatin coating and sonication treatment significantly increased the myofibrillar fragmentation index of the samples. Shear force of wet-aged pork loin significantly decreased as the samples were gelatin-coated and sonicated. The myofibrillar and total protein solubilities were not significantly different between samples. In conclusion, the 1% gelatin coating with sonication can enhance the quality of wet-aged pork loin.

단백질 칩 기판의 플라즈마 효과 (Effects of Plasma on the Surface of Protein Chip Plates)

  • 수소 플라즈마 처리된 유리 기판에 스핀 코팅 시스템을 이용하여 nickel chloride를 코팅하여 단백질칩 플레이트를 제조하였다. 다양한 플라즈마 처리 시간대에서 histidine tagged 단백질의 부착 능력 특성을 연구하였다. 유리 기판 표면에서 nickel chloride와 단백질 특성을 particle size analysis를 이용하여 관찰하였고, 단백질의 부착 능력 정도를 bio imaging analyzer system으로 측정하였다. 실험 결과에 따르면, 플라즈마 처리 시간이 증가할수록 단백질 부착 능력은 감소하는 것으로 나타났다. 기판 표면에서의 단백질 부착능력 특성에 관한 mechanism은 본문의 결과 및 토의에서 논의되었다. 플라즈마 처리된 단백질칩 기판에 대한 표면 안정화는 바이오센서 시장에서 큰 관심을 끌 것으로 기대된다.
