제주 중산간지에서 감자 수확후 휴경기 동안 연맥 및 사료용유채의 파종시기와 혼파비율이 생육특성 및 생산성에 미치는 영향 (Effect of Seeding Times and Mixture Rates of Oat and Forage Rape on the Growth Characteristics and Forage Yields After the Harvest of Potatoes in Jeju.)

  • 본 연구는 제주 중산간지에서 목초지를 전용하여 감자 재배지로 활용하고 있는 경작지에서 가을감자 수확 후 3월부터 6월까지 휴경기 동안 연맥 및 사료용 유채의 파종시기 및 혼파비율을 달리 했을 때 생육과 건물수량에 미치는 영향을 구면하기 위하여 수행하였다. 연맥(Swan)과 사료용 유채(Akella)의 파종시기를 주구로 3월 초순, 3월 중순, 3월 하순 파종구 등 3처리로 하고, 혼파비율을 세구로 연맥 단파구. 사료용유채 단파구, 연맥 75%+사료용유채 25%, 연맥 50%+사료용유채 50%, 연맥25%+사료용유채 75% 혼파구 등 5처리를 두어 분할구 배치 3반복으로 수행하였다. 연맥 및 사료용 유채의 발아 및 출현상태는 파종시기와 혼파비율에 따른 차이는 크지 않았으나, 수확시 연맥의 생육단계는 3월 초순 파종구가 호숙기로 가장 빨랐다. 파종시기에 따른 ha당 평균 건물수량은 3월 중순 파종구가 7,171kg으로 다른 파종시기에 비해 높았으며, 혼파비율에 따른 건물수량은 연맥 단파구가 7,676kg으로 가장 높았고, 연맥/사료용 유채 혼파구 및 사료용 유채 단파구 순으로 나타났다(p<0.05). 조단백질 함량은 사료용 유채 단파구가 15.9%, 혼파구가 12.4∼14.5%, 연맥 단파구는 8.5%로 나타났다.

국내 개발 이탈리안 라이그라스와 청보리 주요 품종의 생산성과 사료가치 비교 (Forage Productivity and Quality of Domestic Italian Ryegrass and Barley Varieties)

  • 본 연구는 국내 개발 이탈리안 라이그라스(IRG)와 청보리 품종의 생산성과 사료가치를 비교분석하여 영농현장에 필요한 기술정보를 제공하고자 국립축산과학원 수원 시험포장에서 2009년 9월부터 '10년 6월까지 수행하였다. 처리내용은 IRG 2품종(조생종 Kowinearly, 중생종 Kowinmaster), 청보리 5품종(영양, 우호, 유연, 다미, 유호) 및 혼파구(Kowinearly + 유연, Kowinmaster + 유연) 등 9처리였다. IRG의 출수기는 Kowin early 5월 14일, Kowinmaster 5월 18일이었다. 5월 22일 조사한 청보리의 숙기는 영양과 우호는 유숙후기, 다미는 호숙초기, 유연과 유호는 호숙초 중기로 품종간 약간의 차이가 있었다. IRG에서 도복이 다소 있었으나 혼파구에서는 관찰되지 않았고, 혼파구에서 IRG의 초장은 117~118 cm로 IRG나 청보리 단파구에 비해 길었다. 수확 시 건물률은 20.7~25.4%였으며, 혼파구에서 시기별로 조사한 IRG의 식생비율은 평균 43.1%였다. 청보리의 이삭비율은 성숙이 진행될수록 높아지는 경향으로 유숙후기, 호숙기, 황숙기, 완숙기에서 각각 35.7%, 44.1%, 54.8%, 57.2%였으며, 품종별로는 영양과 우호에서 다소 높은 경향이었다. 조단백질 함량은 IRG에서 9.0~10.0%로 높은 경향이었으며 청보리는 7.0~8.5%, 혼파구는 7.3~9.1%였고, NDF와 ADF 함량은 청보리구에서 낮은 경향이었다. 건물 소화율은 Kowinearly 64.4%, Kowinmaster 64.1%, Kowinmaster + 유연 혼파구는 64.5%였으며, 청보리는 60.2 (유호)~66.4% (우호) 범위로 우호에서 높은 경향이었다. 건물수량, 조단백질 수량, 가소화 건물수량은 Kowinmaster + 유연 혼파구에서 ha당 각각 11,508 kg, 1,046 kg, 7,422 kg으로 가장 많았으며(p<0.05), IRG와 청보리 품종별 생산성 차이는 크지 않아 지역 특성과 재배 목적에 맞는 초종 및 품종선택과 함께 IRG (Kowinmaster) + 청보리 혼파이용이 권장되었다.

Haematological and Immunological Response in Lambs Fed on Raw and Variously Processed Cottonseed Meal

  • An experiment was conducted with twenty crossbred male lambs to assess the effect of cotton (Gossypium) seed meal (CSM) on blood constituents and immunity. Lambs were randomly assigned to a reference diet (30% deoiled peanut meal, DPNM) and four test diets containing 40% of either raw, 45 minutes cooked, 1% $Ca(OH)_2$ and iron (1 free gossy-pol, FG : 0.3 Fe) treated CSM (replacing approximately 50%, reference concentrate mixture). These isonitrogenous and isocaloric concentrate mixtures were fed to meet 80% of protein requirements (NRC, 1985) along with ad lib maize hay for 180 days. Blood was collected at 60, 120 and 180 days post feeding. The lambs were sensitized with Brucella abortus S99 antigen after 140 days and were subjected to ELISA and delayed type hypersensitivity. Blood haemoglobin, erythrocyte count, leucocyte count, total protein, total albumin, total globulin, urea, creatinine concentration and aspartate aminotransferase activity in lambs fed on raw or processed CSM were comparable to the values of reference lambs. The higher (p<0.01) blood glucose levels observed in CSM fed lambs at 60 days of feeding was latter reduced to the levels comparable with those on reference diet at 120 and 180 days of feeding. The alanine amino transferase activity was lower in lambs fed raw and cooked CSM containing diets at 120 and 180 days of feeding. A marginal increase in serum iron and alkaline posphatase activity was observed in iron treated group and raw CSM fed lambs, respectively. The humoral immune response and DTH reactivity was lower (p<0.05) in lambs fed raw CSM (consuming 302.83 mg FG/day). Cooking, $Ca(OH)_2$ and iron treatment of raw CSM showed a positive response in alleviating the suppression of immune response owing to the reduced consumption of FG by 40.19, 17.40% and 26.73%, respectively in these diets. The present study thus indicated that consumption of 40% raw CSM (302.83 mg FG/day) though did not affect majority of the haematological and blood biochemical parameters, but markedly suppressed the immune mechanism of lambs.

Two groups of S-layer proteins, SLP1s and SLP2s, in Bacillus thuringiensis co-exist in the S-layer and in parasporal inclusions

  • We screened four B. thuringiensis strains whose parasporal inclusions contained the S-layer protein (SLP), and cloned two slp genes from each strain. Phylogenetic analysis indicated these SLPs could be divided into two groups, SLP1s and SLP2s. To confirm whether SLPs were present in the S-layer or as a parasporal inclusion, strains CTC and BMB1152 were chosen for further study. Western blots with isolated S-layer proteins from strains CTC and BMB1152 in the vegetative phase showed that SLP1s and SLP2s were constituents of the S-layer. Immunofluorescence utilizing spore-inclusion mixtures of strains CTC and BMB1152 in the sporulation phase showed that SLP1s and SLP2s were also constituents of parasporal inclusions. When heterogeneously expressed in the crystal negative strain BMB171, four SLPs from strains CTC and BMB1152 could also form parasporal inclusions. This temporal and spatial expression is not an occasional phenomenon but ubiquitous in B. thuringiensis strains.

Performance of Growing Lambs Fed Urea Ammoniated and Urea Supplemented Wheat Straw Based Diets

  • Twelve growing male lambs ($10.05{\pm}0.41kg$, 5 months age) were assigned to three equal groups in a completely randomized design and fed respectively wheat straw (WS), ammoniated wheat straw (AWS) or urea supplemented wheat straw (USWS) along with concentrate mixtures of varying protein and energy contents to meet their requirements as per NRC (1985). Despite comparable nutrient intake and digestibility, the lambs fed A WS based diet digested lower (p<0.05) crude protein. The digestibility of NDF and hemicellulose were lower while the digestibility of cellulose was higher in lambs on AWS as compared to lambs on USWS based diet. The lambs of all the three groups were in positive and comparable N, Ca and P balance except higher Ca balance in lambs fed WS based diet. The body weight change, average daily gain and feed conversion efficiency were similar among the dietary groups. All the parameters of rumen fermentation pattern were comparable between lambs of AWS and USWS based diets except ammonia-N concentration that was higher in the latter however it was lower in WS fed group. Though, the feeding cost per unit gain was comparable in lambs fed all the three diets, the cost incurred towards roughage during 120 days of experimental feeding was significantly less in lambs fed AWS than UTWS. Thus, feeding of USWS can be practiced to minimize labour cost and environmental pollution involved in the process of urea treatment without affecting the performance of growing lambs.

Study of Novel Markers for Early Diagnosis of Cardiovascular Diseases

  • In our country, cardiovascular disease (CVD) and Coronary heart diseases (CHD) are the leading causes of death. It is well known that CHD is multifactorial, involving environmental factors such as diet, level of exercise and cigarette smoking, and inherited factors. According to the statistical data in 2003, the cause of death with the highest mortality was including hypertension, ischemic heart disease and atherosclerosis, which accounted for $24.7\%$ of total mortality. In spite of, there have been few study reports on the change of biochemical markers and mechanisms concerned. The development of biochemical markers is required for an early diagnosis and treatment of cardiovascular diseases that are related with dietary habits of Korean people enjoying mixtures of traditional dietary style and westernized food-styles. Therefore, the most efficient cost-saving biochemical marker was established in this study, through analysis of biochemical markers related with dietary habits which are susceptibly being changed in association to cardiovascular diseases from the pre-disease phase, and through reanalysis and assessment of early diagnosis of and preventive effects of diagnosis of cardiovascular diseases by demographical character including sex, age, and socioeconomic level with use of biochemical markers that are identified and selected among the parameters in consideration of the effectiveness and appropriateness of early diagnosis of diseases. The appropriateness of biochemical markers was reviewed by professionals (medical, pharmaceutical area and food/ nutrition area) and CRP(C-Reactive Protein) and was identified to be possible in Korea. It is thought that these biochemical markers may be used as the basic data for early diagnosis and prevention of cardiovascular diseases (CVD) which may be used for Korean people.

이차원 전기영동과 펩타이드 지문 검색법을 이용한 초파리의 프로테옴 분석 (Proteome Analysis of Drosophila melanogaster Used 2-DE and MALDI- TOF-MS)

  • 초파리는 유전학, 발달 생물학, 행동 유전학, 노화 연구에 이르기까지 수많은 연구에 이용 되어져 왔다. 최근 초파리 전체 유전자의 염기서열이 보고되었으며, 유전자의 기능 분석과 발현 단백질에 대한 연구가 진행되고 있다. 본 연구에서는 야생형 초파리에서 추출한 단백질을 이차원 전기영동을 통해 400여개 이상의 단백질 스폿으로 분리하였으며, 각 스폿을 적출하여 트립신으로 처리하여 얻어진 펩타이드 단편을 MALDI-TOF-MS를 이용한 펩타이드 지문 검색으로 질량을 측정하였다. 측정된 질량을 초파리 데이터 베이스를 이용하여 분리한 단백질을 동정함으로써 59개의 유전자에서 발현되는 65개의 단백질 스폿을 동정하였다. 이러한 결과는 향후의 발생단계, 외부 자극, 노화 등에 관련되는 특이적 단백질 연구의 기초 자료로 활용될 수 있을 것이다.

Effects of Soy Protein Hydrolysates Prepared by Varying Subcritical Media on the Physicochemical Properties of Pork Patties

  • This study investigated the effect of soy protein hydrolysates (SPH) prepared by varying subcritical media on the physicochemical properties of pork patties. For resource of SPH, two different soybean species (Glycine max Merr.) of Daewonkong (DWK) and Saedanbaek (SDB) were selected. SPH was prepared by subcritical processing at 190℃ and 25 MPa under three different of media (water, 20% ethanol and 50% ethanol). Solubility and free amino group content revealed that water was better to yield larger amount of SPH than ethanol/water mixtures, regardless of species. Molecular weight (Mw) distribution of SPH was also similar between two species, while slightly different Mw distribution was obtained by subcritical media. For pork patty application, 50% ethanol treatment showed clear red color comparing to control after 14 d of storage. In addition, ethanol treatment had better oxidative stability than control and water treatment based on thiobarbituric acid-reactive substances (TBARS) analysis. For eating quality, although 20% ethanol treatment in SDB showed slightly higher cooking loss than control, generally addition of SPH did not affect the water-binding properties and hardness of pork patties. Consequently, the present study indicated that 50% ethanol was the best subcritical media to produce SPH possessing antioxidant activity, and the SPH produced from DWK exhibited better antioxidant activity than that produced SDB.

A Simple Carbamidomethylation-Based Isotope Labeling Method for Quantitative Shotgun Proteomics

  • In this study, we present a new isotope-coded carbamidomethylation (iCCM)-based quantitative proteomics, as a complementary strategy for conventional isotope labeling strategies, with providing the simplicity, ease of use, and robustness. In iCCM-based quantification, two proteome samples can be separately isotope-labeled by means of covalently reaction of all cysteinyl residues in proteins with iodoacetamide (IAA) and its isotope (IAA-$^{13}C_2$, $D_2$), denoted as CM and iCCM, respectively, leading to a mass shift of all cysteinyl residues to be + 4 Da. To evaluate iCCM-based isotope labeling in proteomic quantification, 6 protein standards (i.e., bovine serum albumin, serotransferrin, lysozyme, beta-lactoglobulin, beta-galactosidase, and alpha-lactalbumin) isotopically labeled with IAA and its isotope, mixed equally, and followed by proteolytic digestion. The resulting CM-/iCCM-labeled peptide mixtures were analyzed using a nLC-ESI-FT orbitrap-MS/MS. From our experimental results, we found that the efficiency of iCCM-based quantification is more superior to that of mTRAQ, as a conventional nonisobaric labeling method, in which both of a number of identified peptides from 6 protein standards and the less quantitative variations in the relative abundance ratios of heavy-/light-labeled corresponding peptide pairs. Finally, we applied the developed iCCM-based quantitative method to lung cancer serum proteome in order to evaluate the potential in biomarker discovery study.

Condensed Molasses Solubles(CMS) / 당밀 혼합물이 반추위 미생물 단백질 합성량에 미치는 영향 (The Effects of Condensed Molasses Solubles(CMS) / Molasses Mixtures on Ruminal Microbial Protein Synthesis)

  • 본 연구는 monosodium glutamate의 생산 후 발생되는 부산물인 condensed molasses solubles(CMS)가 반추가축의 질소 공급원으로서 반추위 미생물 단백질 합성에 미치는 효과를 조사하기 위하여 수행하였다. 반추위 canulae가 부착된 4마리의 비착유소를 4 ${\times}$ 4 라틴 방각법에 적용하여 실험을 수행하였으며, 4개의 처리구는 다음과 같다. (1) 기초사료(앞착된 보리 3kg/일과 보리짚의 자유채식), (2) 기초사료에 당밀 200g/일 그리고 물 300g/일 첨가, (3) 기초사료에 당밀 200g/일, CMS 100g/일 그리고 물 200g/일 첨가 (4) 기초 사료에 당밀 200g/일, CMS 200g/일, 그리고 물 100g/일 첨가. CMS의 첨가수준에 따른 반추위내 발효양상은 처리구간에 유의적인 차이가 없었으나, CMS를 200g/일을 첨가한 처리구가 다른 처리구들에 비해 반추위내 미생물 단백질 합성량의 표시로서 이용된 allantoin/creatinine의 비율을 증가시키는 경향이 나타났다(P < 0.10).
