Effects of Saengkanggamchotang (SKT) on MIA-Induced Osteoarthritis in Rats (생강감초탕(生薑甘草湯)이 골관절염 유발 Rat에 미치는 영향)

  • Objectives This study was performed to investigate the effects of Saengkanggamchotang (SKT) on the monosodium iodoacetate (MIA) induced osteoarthritis in rats. Methods Osteoarthritis was induced by injection of MIA (50 ul, 60 mg/ml) into knee joints of rats. Rats are divided into a total of 4 groups (normal, control, positive comparison group, SKT treated group, each n=6). Normal group are not treated at all without inducing osteoarthritis whereas control group were induced for osteoarthritis by MIA and oral medicated with 20 ml of distilled water per day. Positive comparison group was injected with MIA and after 7 days, that was taken indomethacin (30 mg/kg/mouse). SKT treated group was injected with MIA and after 7 days that was taken SKT (30 mg/kg/mouse). Positive comparison group and SKT treated group were oral medicated for each substance a total of 4 weeks with one time per day. After experiments (from 1 week after injection of papain to 4 weeks elapsed), the functions of liver and kidney, Prostaglandin E2, inflammatory cytokine (IL-$1{\beta}$, IL-6, TNF-${\alpha}$), osteocalcin, TIMP-1, MMP-9 within serum. Knee joint structures were observed by H&E, safranin-O staining method, and amount of cartilage were measured by ${\mu}CT$-arthrography. Results 1) Hind paw weight bearing ability was significantly improved. 2) Functions of liver and kidney were not affected. 3) Prostaglandin E2, osteocalcin, TIMP-1, MMP-9 in serum were significantly decreased. 4) Inflammatory cytokine IL-$1{\beta}$ was significantly decreased, and IL-6, TNF-${\alpha}$ were decreased but had not significant. 5) In terms of histopathology, significantly reduced subsidence of cartilage and bone in H&E staining. And in Safranin O staining, proteoglycan content in synovial membrane was significantly increased compared with control group. 6) Destruction of cartilage on ${\mu}CT$-arthrography was significantly reduced. Conclusions Based on all results mentioned above, Saengkanggamchotang (SKT) is believed to be meaningful for suppressing the progress of osteoarthritis and its treatments.

Effects of Leejung-tang (理中湯) on MIA-Induced Osteoarthritis Rat (이중탕(理中湯)이 골관절염 유발 생쥐에 미치는 영향)

  • Objectives This study intends to clarify how Leejung-tang (here in after reffered to LJT) affect Wistar Rat whose osteoarthritis was induced by MIA. Methods Osteoarthritis was induced into rat by injecting MIA in its knee joint. Rats are divided into a total of 4 groups (n=6). Normal group are not treated at all without inducing osteoarthritis whereas control group were induced for osteoarthritis by MIA and oral medicated with 2 ml of physiological saline per day. Positive comparison group (Indomethacin) was injected with MIA and after 7 days, 2 mg/kg of Indomethacin was medicated. Experimental group (LJT) was injected with MIA and after 7 days that was medicated with 23 mg/kg of LJT. Indomethacin and LJT were oral medicated for each substance a total of 4 weeks with one time per day. After experiments (from 1 week after injection of MIA to 4 weeks elapsed), Hind paw weight bearing ability, Functions of liver and kidney, Serum prostaglandin $E_2$, TNF-${\alpha}$, IL-1${\beta}$, IL-6, Osteocalcin, TIMP-1, MMP-9, LTB4 and amount of cartilage were measured and histopathological variations for knee joint structures were observed. Results 1) Hind paw weight bearing ability of LJT administration group was increased but there was no statistical significance. 2) Functions of liver and kidney were not affected. 3) Serum prostaglandin $E_2$, IL-1${\beta}$, Osteocalcin, MMP-9 were significantly decreased and TNF-$\alpha$, IL-6, TIMP-1, LTB4 were also decreased but there were no statistical significance. 4) In H&E staining and Safranin-O staining, there were small histopathological changes in LJT administration group than control group. 5) In micro CT (computed tomography)-arthrography, cartilage destruction was more suppressed in LJT administration group than control group. Conclusions Based on all results mentioned above, Leejung-tang (LJT) is believed to be meaningful for suppressing the progress of osteoarthritis and its treatments because of its anti-inflammatory effects and alleviation of pain with histopathological effective efficacy.

Morphologic Study of Effects of Prostaglandin E2 and Evening Primrose Oil on Early Orthodontic Tooth Movement in Rats (백서의 구치 이동 초기에 Prostaglandin E2와 Evening Primrose Oil의 영향에 관한 형태학적 연구)

  • To study the effect of prostglandin $E_2$ and evening primrose oil on orthodontic tooth movement in rats, one hundred and sixty rats were divided into four groups of 40 rats each. One group, injected with saline on the palate subperiosteally, served as a control group. A second and third group were injected subperiosteally on the palate with $PGE_2$ $10{\mu}g$ and evening primrose oil 10mg respectively. The fourth group was given indomethacin $20{\mu}g/m{\ell}$ orally by water bottle. The maxillary first molar was moved mesially from the incisors using a 50gm force rubber band. In each group at the 1, 2, 3, 5, and 7th day, 4 rats were examined by light microscope, and 4 by electron microscope. The obtained results were as follows: 1. Osteoclastic activity was maximum at the 3rd day in the $PGE_2$ group on the interradicular alveolar bone of the first molar, followed by the evening primrose oil group, control group, and indomethacin group. 2. Root resorption and vacuolar changes were maximum in the $PGE_2$ group. 3. At the 3rd day of the $PGE_2$ group, the osteoclasts showed well developed ruffled borders and clear zones. At the same day, the evening primrose oil group also showed well developed ruffled borders and clear zones, but less than the $PGE_2$ group. 4. At the 3rd and 5th day of the $PGE_2$ group, fibroblasts showed phagocytized fragmented collagen fibers in the cytoplasm. At the 7th day of the $PGE_2$ group, fibroblasts showed collagen fibers forming at the cell membrane surface.

Effect of Paeoniae Radix Rubra Extract on the Production of NO and Prostaglandin $E_2$ in LPS-stimulated RAW264.7 Marcrophages (적작약(赤芍藥) 추출물이 대식세포(大食細胞)에서 NO 및 $PGE_2$ 생성에 미치는 영향)

  • Objectives : The objective of this study is to study the effects of hot aqueous extract and ethanol extract from Paeoniae Radix Rubra on nitric oxide(NO) and prostaglandin $E_2(PGE_2)$ production in macrophage. Methods : Paeoniae Radix Rubra were extracted in 2 ways. one was extracted with hot aqueous for 4 hr in $100^{\circ}C$ and the other one was extracted with 70% ethanol for 4 hr in $70^{\circ}C$. RAW264.7 cells, a mouse macrophage lines, were incubated with different concentrations of the extract for 30 min and then stimulated with LPS at indicated times. Cell toxicity was determined by MTT assay. The concentrations of NO and $PGE_2$ were measured by griess assay and enzyme immunoassay(EIA). Results : The hot aqueous and ethanol extracts of Paeoniae Radix Rubra significantly inhibited the NO productions in LPS-stimulated RAW264.7 cells. The hot water extract of Paeoniae Radix Rubra significantly inhibited the $PGE_2$ productions in LPS-stimulated RAW264.7 cells. Conclusions : Our results demonstrated that Paeoniae Radix Rubra extract is able to significantly inhibit the production of NO, $PGE_2$ expression. Hot aqueous extract of Paeoniae Radix Rubra has more effective anti-inflammation than ethanol extract.

Antiproliferative effect of Houttuynia cordata Thunb was Associated with the Inhibiton of Cyclooxygenase-2 Expression in Human Breast Carcinoma Cells (인체유방암세포에서 Cyclooxygenase-2 활성 및 Prostaglandin E2 생성에 미치는 어성초 추출물의 영향)

  • Houttuynia cordata Thunb, well known as 'E-Sung-Cho' in Korea, is a traditional medicinal plant generally used in Oriental medicine therapy. In the present study, we investigated the effect of water extract of H. cordafa (WEHC) on the growth of human breast carcinoma MCF-7 cells. Exposure of MCF-7 cells to WEHC resulted in growth inhibition and apoptosis in a dose-dependent manner as measured by MTT assay and fluorescent microscope. The anti-proliferative effect of WEHC was associated with a dose-dependent up-regulation of cyclin-dependent kinase inhibitor p21 in a p53-independent fashion. We found WEHC decreased the levels of cyclooxygenase (COX)-2 and inducible nitric oxide synthease (iNOS) expression without significant changes in the expression of COX-1 , which was correlated with a decrease in prostaglandin $E_2{\;}(PGE_2)$ synthesis. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of H. cordata.

Imperatorin Suppresses Degranulation and Eicosanoid Generation in Activated Bone Marrow-Derived Mast Cells

  • Imperatorin has been known to exert many biological functions including anti-inflammatory activity. In this study, we investigated the inhibitory effects of imperatorin on the production of inflammatory mediators in mouse bone marrow-derived mast cells (BMMC). Imperatorin inhibited degranulation and the generation of eicosanoids (leukotriene $C_4$ ($LTC_4$) and prostaglandin $D_2$ ($PGD_2$) in IgE/antigen (Ag)-stimulated BMMC. To elucidate the molecular mechanism involved in this process, we investigated the effect of imperatorin on intracellular signaling in BMMC. Biochemical analyses of the IgE/Ag-mediated signaling pathway demonstrated that imperatorin dramatically attenuated degranulation and the production of 5-lipoxygenase-dependent $LTC_4$ and cyclooxygenase-2-dependent $PGD_2$ through the inhibition of intracellular calcium influx/phospholipase $C{\gamma}1$, cytosolic phospholipase $A_2$/mitogen-activated protein kinases and/or nuclear factor-${\kappa}B$ pathways in BMMC. These results suggest that the effects of imperatorin on inhibition of degranulation and eicosanoid generation through the suppression of multiple steps of IgE/Ag-mediated signaling pathways would be beneficial for the prevention of allergic inflammation.

Effects of Gagambojungikgi-tang on the Immune Mediators Regulation and Wound Healing in the rSj26 or Chemical Antigen induced Atopic Dermatitis Model Mice (rSj26 또는 화학 항원 유도 아토피 피부염 마우스 모델에서 가감보중익기탕의 면역 매개물 조절 및 상처치료 효과)

  • Objectives : The aim of this study was to investigative the effects of Gagambojungikgi-tang (GBT), a Korean herbal medicine, on the immune mediators, T cell proliferation and wound healing in the recombinant Sj26 (rSj26) antigen induced atopic dermatitis(AD) model mice. Methods : GBT is the water extracts prepared from mixture of Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Atractylodes Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisia iwayomogi Herba, Scutellaria Radix, Lonicera japonica Flos. This is a modified prescription of Bojungikgi-tang, which has been used for the treatment of indigestion, and immunological disease in east-asian countries. GBT was orally administered or externally applied at difference doses. The levels of immune mediators [(IgE, IgG1, prostaglandin E2 (PGE2), Th1/Th2 cytokines], T cell proliferation, and wound healing in the rSj26 or chemical antigen induced AD model BALB/c were investigated. Results : GBT dose-dependently suppressed the release of TNF-${\alpha}$, IL-$1{\beta}$ (Th1 cytokines), IL-4, IL-10 (Th2 cytokines), PGE2 (inflammatory mediators) and T cell proliferation. But GBT increased the production of IFN-${\gamma}$ (Th1 cytokine). Furthermore, A wound healing effect of GBT was similar to external application of dexamethasone. Conclusions : These results suggest that GBT suppresses the inflammatory mediators and regulates the Thl/Th2 cytokines, and promotes the wound healing. Therefore, these properties may contribute to the strong anti-AD effect of GBT.

A Comparison of the Responses of Lower Vertebrate Intestines to Prostaglandin $E_1\;and\;E_2$

  • 1. The isolated strips of guinea-pig, fowl and reptiles (snake and tortoise) showed consistenly excitatory responses to $PGE_1\;and\;E_2$, which were dose-dependent. 2. Frog intestine revealed inhibitory responses to both $PGE_1\;and\;PGE_2$ except a small of $PGE_2$ (1-10 ng/ml) caused slight contraction. 3. The intestines of pieces showed inconsistent responses to $PGE_1\;and\;E_2$. In fresh-water fish(carp), $PGE_1$ produced relaxation under the dose of 50 ng/ml, and contraction by the large doses, but $PGE_2$ consistently caused contraction in dose-dependent manner. However, the strips of sea-water fish revealed the different responses to PGE compound: $PGE_1$ caused relaxation and $PGE_2$ conversly contraction even though in small degree. 4. These results that there are genera differences in the responses of the longitudinal strips of intestine to $PGE_1\;and\;PGE_2$ was assumed to be possibly correlated with evolutionally primitive function of gut.

Change of Antioxidative Activity at Different Harvest Time and Improvement of Atopic Dermatitis Effects for Persimmon Leaf Extract (감잎(柿葉) 추출물의 채취시기에 따른 항산화 활성 변화 및 아토피성 피부염 개선효과)

  • Objectives : The purpose of this study was to investigate the anti-oxidative and anti-atopic dermatitis effects of persimmon leaf extract obtained from Cheongdo-gun, where more than 60% of Korean persimmon is produced. Methods : Anti-oxidative effects of the crude persimmon leaf extract harvested monthly between May and November were determined by in vitro assay using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide dismutase (SOD)-like reaction. Anti-atopic dermatitis effects of the crude persimmon leaf extract were determined by using collagenase type I inhibition assay and by quantitative assays including serum histamine, prostaglandin E metabolite and leukotriene $B_4$ levels in animal model of atopic dermatitis using Balb/c mice. Results : Persimmon leaf extract harvested in May had higher levels of total phenolic compounds (182.24 mg/g) and flavonoids (23.05 mg/g) than the ones of different month extract. Also, persimmon leaf extract harvested in May showed the most effective extract scavenging activities of DPPH free radical ($13.39{\pm}0.21\;{\mu}g/ml$) and superoxide anion radical ($40.52{\pm}2.32\;{\mu}g/ml$), leading to use the persimmon leaf extract harvested in May for the experiments hereafter. Persimmon leaf extract showed $326.71{\pm}4.6\;{\mu}g/ml$ of 50% inhibitory concentration ($IC_{50}$) for collagenase type I which is responsible for the degradation of extracellular matrix. In addition, persimmon leaf extract application group could decrease serum levels of histamine, prostaglandin E metabolite and leukotriene $B_4$ compared to the negative control in animal model of atopic dermatitis. Especially, persimmon leaf extract showed a significantly decreased serum leukotriene $B_4$ level relative to the levels of histamine and prostaglandin E metabolite. Conclusions : Persimmon leaf extract showed anti-oxidative and anti-atopic dermatitis effects in vitro and in vivo. These results suggest that persimmon leaf extract may have immunoregulatory function for alleviating atopic dermatitis by decreasing collagenase activity and mast cell activation.

Anti-inflammatory Activity of Wild Yeasts, Meyerozyma guilliermondii YJ34-2 and Rhodotorula graminis YJ36-1, on Lipopolysaccharide-induced Nitric Oxide in RAW 264.7 Cells Through the Inhibition of Nitric Oxide and Cytotoxic Effects (Lipopolysaccharide로 유도한 RAW 264.7 세포에 대한 Meyerozyma guilliermondii YJ34-2와 Rhodotorula graminis YJ36-1의 항염활성과 Nitric Oxide 생성 저해물질의 생산)

  • The anti-inflammatory effects of cell-free extracts from wild yeasts, Meyerozyma guilliermondii YJ34-2 and Rhodotorula graminis YJ36-1, caused by the inhibition of nitric oxide (NO) activity and cytotoxic effects were determined. Cell-free extracts from these two yeast strains had dose-dependent inhibitory effects on the production of lipopolysaccharide-induced NO and there were no cytotoxic effects on the treated cells or negative effects on their proliferation. Their cell-free extracts were also shown to have inhibitory effects on pro-inflammatory cytokines, such as tumor necrosis factor $(TNF)-{\alpha}$ and $prostaglandin-E_2$, in a dose-dependent manner. Maximal inhibitory activity on NO production occurred in cell-free extracts of Meyerozyma guilliermondii YJ34-2 cultivated at $30^{\circ}C$ for 24 hr and Rhodotorula graminis YJ36-1 cultivated at $25^{\circ}C$ for 24 hr in the yeast extract-peptone-dextrose (YPD) media.
