Effects of Samyoo-tang Extract on Pulmonary Artery and Cerebral Blood Flow in Rabbits and Rats (삼요탕이 폐혈관 및 뇌혈류량에 미치는 영향)

  • Objective : This study aimed to investigate the effects Samyoo-tang Extract (SE) on the vascular systems, including changes in blood pressure and regional cerebral blood flow (rCBF), of male Sprague-Dawley rats. Methods : The changes in rCBF were determined by Laser-Doppler flowmetry through the opened cranial method and norepinephrine (NE)-induced blood vessel contractions were determined by physiograph in the pulmonary artery of isolated rabbits. Results and Conclusion : 1. Contractions evoked by NE ($ED_{50}$) were inhibited significantly by SE in the pulmonary artery. 2. SE inhibited the relaxation of NE induced contractions pretreated with propranolol. 3. SE did not inhibit the relaxation of NE induced contractions pretreated with ODQ and L-NNA. 4. Blood pressure was not affected by SE in rats. 5. rCBF was increased by SE in a dose-dependent manner. 6. Pretreatment with propranolol was increased by SE in a dose-dependent manner in blond pressure. 7. Pretreatment with methylene blue, ODQ and L-NNA did not inhibit SE induced increased in rCBF. These results indicate that SE can relax NE-induced contraction of rabbit blood vessels and increased the changes of rCBF in rats, that relate to the sympathetic nerve system.

Effects of Flos Sophora japonica Extract on the Changes of Cerebral Hemodynamics

  • The purpose of this study was to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Flos Sophora japonica L.(FSJ) water extract. The measurement was continually monitored by Laser-Doppler Flowmeter and pressure transducer in anaesthetized adult Sprague-Dawley rats for 2 hours to 2 hours and a half through the data acquisition system. FSJ increased the changes of rCBF in rat significantly. The rCBF of FSJ did not change by pretreated propranolol, atropine, L-NNA($N^G$-nitro-L-argininine) and indomethacin. But the rCBF of Flos Sophora japonica L. was increased by pretreated methylene blue. FSJ decreased the changes of BP, significantly. The BP of FSJ did not change by pretreated propranolol, atropine, L-NNA and indomethacin. But the BP FSJ was decreased by pretreated methylene blue. There results indicated that FSJ can increase the rCBF and decrease the BP, that is related to guanylyl cyclase activity.

  • These studies were carried out to investigate the effect of Jungchuntang water extract on the inhibitory contractile action of acetylcholine in rat. The results of these studies were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in rat was significantly inhibited by Jungchuntang. 2. Dose-response of acetylcholine pretreated Jungchuntang in rat was not significantly changed. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in rat was decreased. 4. Inhibitory contractile action of acetylcholine pretreated indomethacin was not significantly changed by Jungchuntang. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue was significantly changed by Jungchuntang. 6. Contractile force of acetylcholine from trachea smooth muscle in rat was significantly inhibited by Jungchuntang. 7. Dose-response of acetylcholine pretreated Jungchuntang in rat was not significantly changed. 8. Inhibitory contractile action of acetylcholine pretreated propranolol in rat was decreased. 9. Inhibitory contractile action of acetylcholine pretreated indomethacin was not significantly changed by Jungchuntang. 10.   • This study was carried out to investigate the effects of Gooboeum extract on the inhibitory contractile action of acetylcholine in the control and sensitized rat. The results were obtained as follows: 1. The acetylcholine contractile force of the trachea smooth muscle with epithelium was significantly relaxed by Gooboeum. 2. Dose-response of acetylcholine from the trachea smooth muscle pretreated Gooboeum was not changed. 3. Effect of Gooboeum on the inhibitory contractile action of trachea smooth muscle pretreated propranolol was not significantly inhibited. 4. The inhibitory contractile action of acetylcholine of trachea smooth muscle pretreated indomethacin was not significantly changed by Gooboeum. 5. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of trachea smooth muscle pretreated methylene blue was not significant. 6. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated propranolol was significantly inhibited. 9. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated indomethacin decreased. 10. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated methylene blue was not significant.

Influence of Sopung-Tang on the Blood Pressure Response of the Rat (소풍탕이 흰쥐의 혈압에 미치는 영향)

  • This study was attempted to examine the effect of Sopung-Tang(SPT) on the arterial blood pressure in rats and to elucidate its mechanism of action. SPT given into a femoral vein produced a dose-related vasopressor responses followed by vasodepressor responses. SPT-induced hypotension was significantly inhibited by pretreatment with atropine or propranolol while was not affected by chlorisondamine, Prazosin and cyproheptadine. SPT-evoked hypertensive activity was markedly blocked by pretreatment with prazosin but was not influenced by atropine, chlorisondamine, propranolol and cyproheptadine. Infusion of SPT(15.0 mg/kg/30min) did not affect norepinephrine-induced pressor responses. These experimental results suggest that SPT causes biphasically initial hypertensive activity followed by hypotensive activity, and that this hypertension may be due to the stimulation of peripheral adrenergic alpha-receptors and hypotension may be elicited through stimulation of peripheral cholinergic muscarinic receptors and adrenergic beta-receptors.

Relaxative Effect of Transmural Nerve Stimulation via ${\beta}$-adrenergic Nerve on the Isolated Uterine Smooth Muscle Motility of Pigs (돼지 적출 자궁 평활근의 운동성에 있어서 transmural nerve stimulation에 대한 ${\beta}$-adrenergic 신경의 이완작용)

  • The effects of transmural nerve stimulation induced releasing neurotransmitters on the changes of swine uterine smooth muscle motility were examined by polygraph through isometric force transducer. The frequency dependent relaxation and rebound contraction were revealed on precontraction with histamine by transmural nerve stimulation. The rebound contraction by transmural nerve stimulation was inhibited by nonselective ${\alpha}$-adrenergic receptor antagonist, phentolamine, and the relaxation by transmural nerve stimulation was blocked by nonselective ${\beta}$-adrenergic receptor antagonist, propranolol. The relaxation induced by nonselective ${\beta}$-adrenergic receptor agonist, isoproterenol on precontraction with histamine were the dose dependent manner and this relaxation was blocked by nonselective ${\beta}$-adrenergic receptor antagonist, propranolol in isolated uterine smooth muscle of pig. These results suggest that endogenous neurotransmitters on smooth muscle relaxation was influenced by ${\beta}$-adrenergic receptor in swine.

Involvement of Phospholipase D in Norepinephrine Uptake in PC12 Cells

  • Phospholipase D (PLD) is an enzyme hydrolyzing phosphatidylcholine to phosphatidic acid (PA) and choline. We investigated the involvement of PLD1 in the uptake of norepinephrine (NE) in PC12 cells, pheochromocytoma cells. NE uptake was specific in PC12 cells because nomifensine, a specific blocker of NE transporter, blocked NE uptake. Inhibition of PLD function in PC12 cells by the treatment of butanol suppressed the NE uptake. In contrast, overexpression of PLD1 in PC12 cells increased NE uptake efficiently. These results suggest that PLD activity is involved in NE uptake. We explored the action mechanism of PLD in NE uptake. PA phosphatase inhibitor, propranolol, blocks the formation of PKC activator diacylglycerol from PA. Propranolol treatment to PC12 cells blocked dramatically the uptake of NE. Specific PKC inhibitors, GF109203X and Ro31-8220, blocked NE uptake. Taken together, we suggest for the first time that PLD1 activity is involved in NE uptake via the activation of PKC.

  • The authors studied the adrenotropic receptors of isolated urinary bladder from Sebastes inermis, using adrenergic activators such as epinephrine, nor-epinephrine, isoproterenol and phenylephrine and adrenergic blocking agents such as phenoxybenzamine, pronethalol and propranolol. The studies have revealed the following results. 1) The spontaneous motility of isolated bladder from Sebastes inermis was inhibited by epinephrine nor-epinephrine, isoproterenol and phenylephrine. 2) The inhibitory effect of phenylephrine on the Sebastes inermis bladder was blocked by phenoxybensamine. 3) The inhibitory effect of isoproterenol was blocked by pronethalol and propranolol. 4) The effect of epinephrine and nor-epinephrine on the Sebastes inermis bladder was usually not blocked by either kind of blocking agent alone, but was blocked by a combination of ${\alpha}\;and\;{\beta}$ blockades. 5) It is, therefore, concluded that the Sebastes inermis bladder has alpha and that both receptors, and that both receptors subserve retaxation or inhibition.

  • Multifocal atrial tachycardia (MAT) is a rare arrhythmia in the newborn. MAT can be difficult to diagnose; it is frequently confused with atrial fibrillation. MAT is difficult to treat but often resolves spontaneously within the first year of life. A newborn with a rapid and irregular pulse rate was diagnosed with multifocal atrial tachycardia by eletrocardiography (ECG) using a hand-made transesophageal electrode. Treatment with propranolol was attempted but ineffective. Treatment with digoxin and sotalol was attempted. The heart rhythm gradually reverted to a sinus rhythm with this treatment. We report our experience managing a neonate with MAT diagnosed by ECG using a hand-made transesophageal electrode.

Study on the Effects of Chukdamtanggamibang on blood pressure, regional cerebral blood flow(rCBF) and smooth muscle (척담탕가미방이 혈압, 뇌혈류량 및 평활근에 미치는 효능에 관한 연구)

  • This study was aimed to investigate the effect Chukdamtanggamibang on the vascular systems including changes in blood pressure and regional cerebral blood flow(rCBF) of male Sprague-Dawely rats, contractile force of guinea pig`s tracheal smooth muscle and abdominal aorta and femoral artery in rabbits. Blood pressure was not affected by Chukdamtanggamibang in rats. rCBF was significantly increased by Chukdamtanggamibang in a dose-dependent manner. Contractile force of isolated guinea pig`s tracheal smooth muscle evoked by His ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. Propranolol, indomethacin and methylene blue did not significantly alter the inhibitory effect of Chukdamtanggamibang. Contractile force of isolated rabbit`s abdominal aorta and femoral artery evoked by NE ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. ODQ and L-NNA significantly attenuated the inhibitory effects of Chukdamtanggamibang in abdominal aorta, whereas propranolol did not significantly alter the inhibitory effect of Chukdamtanggamibang. These results indicate that Chukdamtanggamibang can relax hitamine-induced contraction of guinea pig`s tracheal smooth muscle and that this inhibition involves, in part, the relation to the soluble guanylyl cyclase synthesis and nitric oxide (NO) synthesis.
