Promoting Effect of Hydrogen Peroxide on 1-Methyl-4-phenylpyridinium-induced Mitochondrial Dysfunction and Cell Death in PC12 Cells

  • The promoting effect of hydrogen peroxide ($H_2O_2$) against the cytotoxicity of 1-methyl-4-phenylpyridinium ($MPP^+$) in differentiated PC12 cells was assessed by measuring the effect on the mitochondrial membrane permeability. Treatment of PC12 cells with $MPP^+$ resulted in the nuclear damage, decrease in the mitochondrial transmembrane potential, cytosolic accumulation of cytochrome c, activation of caspase-3, increase in the formation of reactive oxygen species (ROS) and depletion of GSH. Addition of $H_2O_2$ enhanced the $MPP^+-induced$ nuclear damage and cell death. Catalase, Carboxy-PTIO, Mn-TBAP, N-acetylcysteine, cyclosporin A and trifluoperazine inhibited the cytotoxic effect of $MPP^+$ in the presence of $H_2O_2$. Addition of $H_2O_2$ promoted the change in the mitochondrial membrane permeability, ROS formation and decrease in GSH contents due to $MPP^+$ in PC12 cells. The results show that the $H_2O_2$ treatment promotes the cytotoxicity of $MPP^+$ against PC12 cells. $H_2O_2$ may enhance the $MPP^+$-induced viability loss in PC12 cells by promoting the mitochondrial membrane permeability change, release of cytochrome c and subsequent activation of caspase-3, which is associated with the increased formation of ROS and depletion of GSH. The findings suggest that $H_2O_2$ as a promoting agent for the formation of mitochondrial permeability transition may enhance the neuronal cell injury caused by neurotoxins.

Factors Affecting Job Satisfaction of In-Home Child Care Helpers: Focusing on Child-Care Efficacy, Job Efficacy, and Health Promoting Lifestyle (아이돌보미의 직무만족에 영향을 미치는 요인: 보육효능감, 직무효능감, 건강증진 생활양식을 중심으로)

  • Purpose: The purpose of this study was to investigate the influence of child-care efficacy, job efficacy, and health promoting lifestyle on the job satisfaction of in-home child care helpers who are called Idolbomi. Methods: Participants for this survey were 153 in-home child care helpers who worked in Seoul. Data were collected from May 23 to October 17, 2014 using self-report structured questionnaires. Data were analyzed using IBM/SPSS 18.0 program. Results: There were significantly positive correlations between child-care efficacy, job efficacy, health promoting lifestyle, and job satisfaction. Stepwise multiple regression analysis revealed that the predictors of job satisfaction were spiritual growth, education level, job efficacy, effect of income. Conclusion: Therefore, we need to develop strategies to enhance the job efficacy and health promoting lifestyle of in-home child care helpers to improve their job satisfaction.

The Determinants of Health Promoting Lifestyle in High School Students (여고생의 건강증진 생활양식과 관련요인 연구)

  • This descriptive correlational study was conducted to identify the variables related to health promoting lifestyle of girls in High school students. 156 high school students and 88 vocational high school students who lives in Taegu city were selected by a convenience sampling method. The data were collected by self-reported questionnaires from August 24 to September 4. 1998. Pender and others' Health Promoting Lifestyle Profile. Shere et al's. Self-Efficacy scale. Wallston et al.' s Internal Health Locus of Control scale. Wares' Health self rating scale and Cohen & Hoberman's Social support scale were used. The data were analyzed by using descriptive statistics, Pearson correlation coefficient. t-test. MANOVA. ANOVA. Tukey verification and Stepwise multiple regression with SAS progrom. The results of this study were as follows : 1. The average score of health promoting lifestyle performance was 2.34(SD=.36)points by the 4 point scale. 2. The combination of self-efficacy. internal health locus of control. mather's educational level and type of school explained $41.31\%$ of the variance of health promoting lifestyle. 3. Health promoting lifestyle showed significant positive correlations with self efficacy(r=.5173. p=.001), perceived health status(r=.254, p=.001) and internal health locus of control(r=.321. p=.001). On the basis of the above findings, self-efficacy in cognitive factors. and social support in modifying factors were identified as the variables which explained the most part of the Pender's health-promotion model. Nursing strategies enhancing self-efficacy which has the most significant effect on health promoting lifestyle should be developed. Further research is required to find out the factors influencing health pormoting lifestyle of high school students.

Study on Health Promoting Behavior Determinant of Nursing Students (간호대학생의 건강증진 행위 결정요인에 관한 연구)

  • This descriptive correlational study was conducted to identify the variables related to a health promoting behavior in nursing students. 238 nursing students were selected by a convenience sampling method. The data were collected by self - reported questionnaires from 12 to November 19, 2001. The data were analyzed by using descriptive statistics, pearson correlation coefficient, stepwise multiple regression with an SPSS program. The result of this study were as follows: 1. The average score of health promoting behavior was 2.47(SD=0.36)of a 4 point scale. Self actualization (mean=2.94, SD=0.50), interpersonal support (mean=2.91, SD=0.53), stress management(mean=2.58, SD=0.48), health responsibility(mean=2.07, SD=0.48)) nutrition & exercise(mean=2.07, SD=0.48)) were also analyzed. 2. Health promoting behavior showed significant positive correlations with self­esteem(r=0.446, p<0.001) perceived health status(r=0.180, p<0.01), perceived benefits (r=0.183. p<0.01). self-efficacy(r=0.311, p<0.001), social support(r=0.447, p<0.001), control(r=0.169, p<0.01). 3. The combination of self-esteem, social support, affect related to action explained $38.0\%$ of the variance in health promoting behavior. On the basis of the above findings. self-esteem and social support were identified as the variables which explained most of Pender's health promoting model. Nursing strategies enhancing self-esteem and social support which have a more significant effect on health promoting lifestyles should be developed.

Detection of Phagocytosis-Promoting Factor of Culture Supernatant from Feline Peripheral Blood Mononuclear Cells Cultured with Egg White Derivatives (계난백유래물질로 배양한 고양이 말초혈액 단핵구세포 배양상층액중의 탐식촉진인자 검출)

  • The aim of this study is to determine the phagocytosis-promoting factor(s) for feline peripheral blood polymorphonuclear cells (PMN) by culture supernatant from mono-nuclear cells (MNC) treated with egg white derivatives (EWD). The phagocytic activity of PMN was analyzed by a flow cytometry system. The EWD did not show direct effect on the phagocytic response of PMN. The phagocytic activity of PMN was enhanced by culture supernatant from MNC but not PMN treated with EWD. Therefore, it was suggested that the enhanced phagocytic activity of feline PMN could be mediated by humoral factor(s) released from MNC treated with EWD. Thus, the phagocytosis-promoting factor(s) in supernatant fraction from MNC culture treated with EWD were isolated by reverse phase high pressure liquid chromatography. The resulting supernatant fraction on 29.02 minutes of retention time showed high phagocytic activity of PMN. The molecular weight of this supernatant fraction was 16 to 18 kDa when analyzed by capillary electrophoresis. The isoelectric point was pH 5.76 when assessed by ion-exchange chromatography. These results suggest that EWD stimulates feline MNC to elaborate a phagocytosis-promoting factor, 16 to 18 kDa of molecular weight, which could be an important mediator for the enhancement of phagocytic activity of feline peripheral blood phagocytes. Further study will be needed to elucidate this phagocytic factor.

Health-Promoting Life-Style and Related Factors Among Teachers (교사의 건강증진생활양식 실천도와 관련요인)

  • This study was aimed at examining the self-reported health-promoting life- style (HPL) and related factors among teachers assumed to be role models for students. The subjects were 300 teachers who were conviently drawn from nine elementary schools and three high schools. The conceptual framework for this study was based on the Pender's revised Health Promotion Model(1996). Data was collected by a mailed survey (response rate, 62.5%) with structured questionnaire. The score of health-promoting life-style was 2.8(full mark: 5.0), harmonized relationship was the highest(3.2), and professional helath management was the lowest(2.0). The related factors to health-promoting life-styles were age, marital stauts, career, perceived health status, self esteem, intermal locus of control, perceived benefit, perceived barrier, self efficacy, and social support in univariate analysis. In the final regression model, predictors of HPL were social support, self esteem, perceived benefit. self efficacy, and perceived health status after control the effects of demographic characteristics (p<.0001, R2=0.494). The results generally supported the Pender Model. It is recommended to develop the health promotion program for teachers based on these results, and to evaluate the effect of that program for teacher.

The carcinogenicity study of Folpet in rats (랫드에서 Folpet의 발암성에 관한 연구)

  • This study was performed for assessing carcinogenicity of Folpet using medium-term carcinogenicity bioassay. Sprague-Dawley rats aged six weeks divided into four grout's and were initially given an intraperitoneal injection of diethylnirosamine at 200mg/kg body weight. Two weeks later, group 1(negative control) was treated with basal diet. A Folpet was given per oral administration to group 2(100 ppm) and goup 3(1,000 ppm). Group 4 was fed on water containing 0.05% phenobarbital sodium as a promtor for six weeks. At three weeks after beginning of the experiment, partial hepatectomy was performed in all rats. The tumor-promoting effects were examined by the numbers and areas per $cm^2$ of induced glutathion S-tranferase placetal form(GST-P) positive foci in liver, and silver stained nucleolar organizer regions(AgNORs) which have recently introduced as one of the indicators for the cell proliferative activity. As the results, Folpet didn't have tumor-promoting effects on GST-P positive foci developement and AgNORs during promoting stage after initiation, whereas phenobarbital sodium treatment group showed promoting effect. It was concluded that Folpet didn't have promoting effect at 500, 1,000 ppm using this midium-term carcinogenicity bioassay model.

Effect of Inoculation of Azospirillum brasilense and Methylobacterium oryzae on the Growth of Red Pepper Plant (고추의 생장에 미치는 Azospirillum brasilense 및 Methylobacterium oryzae 접종 효과)

  • BACKGROUND: Rhizosphere bacteria may improve plant growth and productivity both by supply nutrients and hormonal stimulation. Although many experiments have shown improvements in plant growth with inoculation of bacterial cultures to the rhizosphere, the main obstacle in the applications of plant growth promoting rhizobacteria in a large scale is the inconsistency of the results. We tested the growth promoting effects of Azospirillum and Methylobacterium strains on red pepper plant. METHODS AND RESULTS: Red pepper seedlings were grown for 25 days in a growth media inoculated with A. brasilense CW903 or M. oryzae CBMB20. The seedlings were transplanted and grown for 45 days in pots with soil in a greenhouse, at half the recommended level of fertilizer. Bacterial culture, $4.0{\times}10^9$ for A. brasilense CW903 and $5.8{\times}10^8$ CFU for M. oryzae CBMB20, was applied in root zone soil periodically every 10 days during the experiment. Inoculation of M. oryzae CBMB20 significantly increased the red pepper plant growth in terms of leaf number, height and mass of shoot, or root mass compared to uninoculated control plants. Although beneficial effects of A. brasilense on plant growth of many crops were observed, the growthpromoting effect of A. brasilense CW903 on red pepper plant was not found in this study. CONCLUSION(s): The factors responsible for the irregularities in plant growth promoting of rhizobacteria are difficult to elucidate. Extensive inoculation experiments in the greenhouse and in the field should enable us to define the factors critical to obtain successful application of plant growth promoting rhizobacteria.

Effect of Fertilization Promoting Peptide on Kinematic Parameters, Capacitation and Acrosome Reaction in Human Spermatozoa (Fertilization Promoting Peptide가 사람 정자의 운동양태, 수정능력획득 및 첨체반응에 미치는 영향)

  • Objective: This study has been carried out to evaluation the effect of fertilization promoting peptide (FPP) on the kinematic parameters, capacitation and acrosome reaction of the frozen-thawed human spermatozoa. Methods: After FPP treatment, we examined kinematic parameters, capacitation and acrosome reaction, using the methods of computer-aided sperm analysis (CASA) and chlortetracycline (CTC) fluorescence analysis. Results: We have obtained the evidence that FPP can promote the capacitation and inhibit the spontaneous acrosome reaction of frozen-thawed human spermatozoa in vitro. FPP ($25{\sim}100$ nM) induced a significant increase in the proportion of B-pattem capacitated spermatozoa, and a significant decrease in the proportion of F-pattem uncapacitated ones without significant stimulation of acrosomal exocytosis. In the kinematic parameters treatment, FPP treated groups maint3ined higher LIN, BCF and STR than those of control. The VAP, VSL, VCL and ALH were not different. Therefore it is suggested that FPP in human seminal plasma may play a positive role in promoting human sperm function.

Sleep-promoting and Anti-anxiety Effects of Shihogayonggolmoryo-tang in Mice (동물모델에서 시호가용골모려탕(柴胡加龍骨牡蠣湯)의 수면유도 및 항불안 효과)

  • Shihogayonggolmoryo-tang (ST) is a Korean medical herb cocktail which has been used to treat anxiety induced insomnia. In this study, we will examine sleep-promoting and anti-anxiety effects of ST, and investigate its mechanism. ICR mice were divided into three groups for the first examination : control group (n=11), ST50 group (50 mg/kg, po, n=11), ST200 group (200 mg/kg, po, n=11). Sleep-promoting effect was confirmed by measuring the sleeping duration time and sleeping onset time after thiopental sodium treatment (50 mg/kg, ip). ICR mice were divided into five groups for the second examination : control group (n=11), ST200 group (200 mg/kg, po, n=11), ST200+Flumazenil group (ST 200 mg/kg, po, flumazenil 0.3 mg/kg, ip, n=11), diazepam group (1 mg/kg, ip, n=11), diazepam+flumazenil group (diazepam 1 mg/kg, ip, Flumazenil 0.3 mg/kg, ip, n=11). Anxiety behavior and sleep-promoting effect was confirmed by open field test and measuring the sleeping duration time and sleeping onset time. Expression levels of c-fos in tuberomammillary nucleus (TMN) and ventrolateral preoptic nucleus (VLPO) were analyzed by immunohistochemistry. ST treated group showed significantly decreased anxiety behavior and enhanced sleeping duration time and sleeping onset time concentration dependently. The expression of c-fos was significantly upregulated in VLPO as sleep-inducing center and TMN as downregulated in arousal center by ST treatment. In addition, all effects of ST were reversed by flumazenil. Our results suggest that ST has sleep-promoting and anti-anxiety effects through regulating arousal center (TMN) and sleep-inducing center (VLPO).
