  • A metal complex, copper (II) L-proline was chemically bound to ethylene glycol dimethacrylate and divinylbenzene crosslinked chloromethylated polystyrene and they were used as chiral chelate resin matrix for column chromatography to resolve enantiomers of DL-amino acids. The L-enantiomers eluted first and the degree of resolution on the polymer crosslinked with ethylene glycol dimethacrylate was superior to the polymer crosslinked with divinylbenzene.

  • Enantiomeric separation of underivatized amino acids using N-alkyl-L-proline (octyl, dodecyl or hexadecyl) coated HPLC has been accomplished. The anchoring N-alkyl groups of L-proline provides a permanent adsorption of there solving chiral agent on the hydrophobic interface layer of a reversed phase. The factors controlling retention and enantioselectivity such as the Cu(II) concentration, pH of the eluent, the type and concentration of organic modifier in the hydroorganic eluent, and extent of coating were examined. The elution orders between D- and L-amino acids were consistent, L-forms eluting first, except histidine and asparagine. The extremely high enantioselectivity $(\alpha$ upto 13 for proline) is observed. The retention mechanism for the chiral separation can be illustrated by a complexation and hydrophobic interaction.

Comparison of Structural Types of Proline Pentamer by Quantum Chemical Calculation (QCC)

  • In this study, Proline pentamer model was used to investigate change in the dihedral angle, intramolecular hydrogen bonding and formation energies during structural optimization. L-Proline (LP, as an imino acid residue) pentamers having four conformation types [β: φ/ψ=t−/t+, α: φ/ψ=g−/g−, PPII: φ/ψ=g−/t+ and Plike: φ/ψ= g−/g+] were carried out by QCC [B3LYP/6-31G(d,p)]. The optimized structure and formation energy were examined for designated structure. In LP, P-like and PPII types did not change by optimization, and β types were transformed into PPII having no H-bond independently of the designated ψ values. PPII was more stable than P-like by about 2.2 kcal/mol/mu. The hydrogen bond distances of d2(4-6) type H-bonds were 1.94 - 2.00Å. In order to understand the processes of the transformations, the changes of φ/ψ, distances of NH-OC (dNH/CO) and formation energies (ΔE, kcal/mol/mu) were examined.

Effects of Kinetin on Alleviating Manganese Chloride Toxicity during Rice(Oryza sativa L.) Germination (벼 발아중 염화망간 독성경감에 미치는 Kinetin의 효과)

  • The experiment was conducted to find changes of growth response, free proline content and organic acids on manganese chloride toxicity($4,000mg\;l^{-1}$) for germination and early growth in rice. Root growth was increased in kinetin $10^{-3}M$ compared with control and germination rate was also increased in kinetin $10^{-3}M$ as 89%. Chlorophyll contents was slightly increased in kinetin $10^{-3}M$. Free proline content at 3days in Mn $4,000mg\;l^{-1}$ was higher than at 7 and 10 days, and all kinetin concentrations promoted free proline content. In organic acids, particularly, malic acid was remarkably increased in kinetin $10^{-3}M$ compared with control and Mn $4,000mg\;l^{-1}$.

  • Takju (Korean native Sake) was mashed with two different materials. One of the material was polished rice and Nuruk(mold wheat), the other one was corn and Nuruk. The amino acids in those fermenting mashes were identified by paper partition chromatography at regular intervals. The results were summarized as follows; a) Following 14 kinds of amino acids were identified in the mash of rice material; lysine, valine, proline, leucine, serine, glycine, aspartic acid, alanine, cystine, tyrosine, histidine, glutamic acid, tryptophan and phenylalanine b) Following 12 kinds of amino acids were identified in the mash of corn material; lysine, valine, proline, leucine, serine, glycine, aspartic acid, alanine, cystine, histidine, argrinine and tryptophan. c) The main amino acids in the Takju mash according to the color density of the each amino acid spot on the paper chromatograms were checked as lysine, valine, leucine, serine, proline and glycine.

Contents of Free Amino Acids, and Nucleotides and Their Related Compounds of Dried Cod (대구 건제품의 핵산관련물질 및 유리아미노산 함량)

  • In this experiment, the contents of free amino acids, and nucleotides and their related compounds of dried cod products were analysed by amino acid autoanalyzer and HPLC. Proline, histidine, lysine and alanine were dominant amino acids in raw extracts, having $65.4\%$ of total free amino acid contents, but the contents of aspartic acid, serine, isoleucine and phenylalanine were low, and methionine, tyrosine and arginine were detected in trace amount. In free amino acid composition of dried products, abundant amino acids were glycine, histidine, lysine, proline and alanine. Such amino acid as aspartic acid, methionine, tyrosine and phenylalanine were poor. In extracts of storage sample, most free amino acids were increased in both sun dried products and hot air dried products, and glycine, histidine, lysine, proline and alanine were abundant amino acid, such amino acid as methionine, tyrosine and phenylalanine were poor. In raw cod, inosine and IMP were dominant contents which 18.5, $10.0{\mu}mole/g$ on dry base, respectively, but the contents of ATP, ADP, hypoxanthine and AMP were detected less than $3.2{\mu}mole/g$. ATP and ADP were decreased while IMP, inosine and hypoxanthine were increased during drying and storage of cod.

Increased Production of Amino Acids in an Escherichia coli rpoS Mutant (RpoS 대장균 돌연변이 균주에서 아미노산의 생산 증가)

  • An RpoS factor is a transcriptional regulator which participates in numerous biological processes. In this work, we investigated the transcriptional regulation of proBA and proC composing proline biosynthetic pathway in Escherichia coli. While the proBA and proC genes were greatly induced in an exponential growth phase, they were dramatically repressed in a stationary growth phase in the wild type E. coli. Unlike the wild type E. coli, the proBA and proC genes were not repressed even in the stationary growth phase in its isogenic rpoS mutant. These results suggest that the RpoS factor acts as a transcriptional repressor of proBA and proC genes. The production of threonine, methionine, lysine, and arginine in the rpoS mutant were also increased by more than two times compared to its parental wild type, suggesting that the mutant is able to be used as an useful host strain for the amino acid overproduction.

Modeling and Measurements of the Activity Coefficients and Solubilities of Amino Acids in the L-valine/electrolyte and L-proline/electrolyte Aqueous Solutions (L-Valine/전해질 및 L-Proline/전해질 수용액에서 아미노산의 활동도계수와 용해도의 측정 및 모델링)

  • Activity coefficients and solubilities of L-Valine and L-Proline in aqueous solutions containing each of four electrolytes such as NaCl, KCl, $NaNO_3$ and $KNO_3$ were measured at 298.15 K. The measurements of activity coefficients were carried out in the electrochemical cell coupled with two ion-selective electrodes (cation and anion), and the solubilities were measured by the gravimetric analysis of saturated solutions in equilibrium with the solid phase of amino acid. The measured activity coefficients of electrolytes and amino acids were correlated with the theoretical thermodynamic model presented in the previous work [Korean Chem. Eng. Res. 48(4), 519(2010)]. It was found that the activity coefficients of amino acids and electrolytes described based on the our previous model were well agreeable with experimental data. Also the experimental solubility data of L-Valine and L-Proline were successfully correlated with the thermodynamic relation mentioned in the previous work.
