  • Objectives : Jakyakgamcho decoction is a traditional prescription known to be an effective pain control medication and muscle relaxant. For more localized treatment outcomes achieved in a shorter period of time, Jakyakgamcho decoction was reprocessed into a form of pharmacopuncture. An analysis of Jakyakgamcho decoction pharmacopuncture showed that there was a significant loss of paeoniflorin(Jakyak's index component). This study was designed to investigate ways to minimize this loss. Methods : After making changes to the processing methods of Jakyakgamcho decoction pharmacopuncture, we measured the quantity of paeoniflorin using high performance liquid chromatography(HPLC) for a before-and-after analysis Results : Paeoniflorin loss was observed 15 minutes after sterilization with $Na_2HPO_4$ at $121^{\circ}C$ Conclusions : It was found that paeoniflorin loss did not occur when pH was not controlled for during processing.

  • Triacylglycerols of the seeds of Ginkgo biloba have been resolved by high-performace liquid chromatography(HPLC} in the silver-ion and reverse-phase modes. The fatty acids were identified by a combination of capillary gas chromatography and gas-chromatography /mass spectrometry as the methyl and /or picolinyl ester. The main components are $C_{18:2{\omega}6}$(39.0mol%), $C_{18:1{\omega}7}$(asclepic acid 21.5mol%), and $C_{18:1{\omega}9}$(oleic acid, 13.8mol%). Considerable amounts of unusual acid such as $C_{20:3{\Delta}^{5,11,14}$ (5.7mol%), $C_{18:2{\Delta}^{5,9}$(2.8mol%), and $C_{18:3}{\Delta}^{5,9,12}$(1.6mol%), were checked. In addition, an anteiso-branched fatty acid, 14-methylhexadecanoic acid, was also present as a minor component(0.9 mol%). The triacylglycerols were separated into 17 fractions by reverse-phase HPLC, and the fractionation was achieved according to the partition numnber(PN) in which a ${\Delta}^5$-non methylene interrupted double bond($^5$-NMDB) showed different behaviour from a methylene interrupted double bond in a molecule with a given cahinlength. Silver-ion HPLC exhibited excellent resolution in which fractions(23 fractions) were resolved on the basis of the number and configuration of double bonds. In this instance, the strength of interaction of a ${\Delta}^5$-NMDB system with silver ions seemed to be weaker than a methylene interrupted double bond system. The principal triacylglycerol species are as follows ; $(C_{18:2{\omega}6)2}/C_{18:1{\omega}7}$, $C_{18:1{\omega}9}/C_{18:1{\omega}7}/C_{18:2{\omega}6}$, $(C_{18:1{\omega}7)2}/C_{18:2{\omega}6}$, $C_{16:1{\omega}7}/C_{18:1{\omega}9}/C_{20:3}{\Delta}^{5,11,14}$, $C_{16:1{\omega}7}/C_{18:1{\omega}7}/C_{20:3}{\Delta}^{5,11,14}$, $C_{18:1{\omega}9}/C_{18:1{\omega}7}/C_{18:2{\omega}6}$, $C_{18:1{\omega}9}/C_{18:2}{\Delta}^{5,5}/C_{20:3}{\Delta}^{5,11,14}$, $(C_{18:1{\omega}7)2}/C_{18:2{\omega}6}$ and $(C_{18:1{\omega}9)2}/C_{18:2{\omega}6}$, while simple triacylglycerols without $C_{18:2{\omega}6})_3$ were not present. Stereospecific analysis showed that fatty acids with ${\Delta}^5$-NMDB system and saturated chains were predominantly located at the site of sn-3 carbon of glycerol backbones. It is evident that there is asymmetry in the distribution of fatty acids in the TG molecules of Ginkgo nut oils.

  • The principal objective of this study was to estimate the measurement uncertainty associated with determination of deoxynivalenol (DON), a mycotoxin generated by Fusarium strain, in food. In service of this goal, wheat as a food matrix was analyzed via high performance liquid chromatography-ultraviolet (HPLC-UV) detection using an immunoaffinity column for clean-up. The uncertainty sources in the measurement process were identified by sample weight, final volume, and sample concentration in extraction volume with components including standard stock solution, working standard solution, 5 standard solutions, calibration curve, matrix, and instrument. The expanded uncertainty for DON at a concentration of 300 ${\mu}g/kg$ was estimated as 71.62 ${\mu}g/kg$ using a coverage factor of two, which provides a confidence level of approximately 95%. The most influential component in the uncertainty sources was the recovery of the wheat matrix, followed by the calibration curve. These results indicate that all efforts may be directed toward reducing the uncertainties of the recovery of the wheat matrix and the calibration curve to obtain a reliable HPLC-UV method for DON analysis in wheat.

  • Effects of pH (1.8-10.2) and time (56 sec-15 min 4 sec) at high hydrostatic pressure (500 MPa) on the reduction of volatile compounds in garlic were studied. Volatile components of garlic were obtained from the headspace, analyzed, and identified by gas chromatography (GC)-mass spectrometry and an electronic nose. Nineteen sulfur compounds were identified as major compounds in garlic, and furan, aldehydes, alcohols, and ketones were also detected. Off-flavor compounds were more effectively reduced under strong acidic conditions. As the residence time at 500 MPa increased from 56 sec to 15 min 4 sec, the total amount of volatile compounds decreased significantly. The total amount of sulfur compounds decreased about 70% compared to those of raw garlic when the garlic was soaked in buffer (pH 6.0) and treated at 500 MPa for 15 min 4 sec. A principal component analysis showed that the off-flavors of garlic were reduced by the operating time of high hydrostatic pressure as well as pH treatment. The correlation coefficient of the results between GC and the electronic nose analysis was 0.9620. Therefore, pH and high hydrostatic pressure treatment could be used as an efficient method for reducing of garlic off-flavor.

  • Background: Although flowers of Panax ginseng Meyer (FPG), Panax quinquefolius L. (FPQ), and Panax notoginseng Burk. (FPN) have been historically used as both medicine and food, each is used differently in practice. Methods: To investigate the connection between components and enhancing immunity activity of FPG, FPQ, and FPN, a method based on a rapid LC coupled with quadrupole time-of-flight MS and immunomodulatory activity study evaluated by a carbon clearance test were combined. Results: According to quantitative results, the ratio of the total content of protopanaxatiol-type ginsenosides to protopanaxadiol-type ginsenosides in FPN was 0, but ranged from 1.10 to 1.32 and from 0.23 to 0.35 in FPG and FPQ, respectively. The ratio of the total content of neutral ginsenosides to the corresponding malonyl-ginsenosides in FPN ($5.52{\pm}1.33%$) was higher than FPG ($3.2{\pm}0.64%$) and FPQ ($2.39{\pm}0.57%$). The colorimetric analysis showed the content of total ginsenosides in FPQ, FPG, and FPN to be $13.75{\pm}0.60%$, $17.45{\pm}0.42%$, and $12.45{\pm}1.77%$, respectively. The carbon clearance assay indicated that the phagocytic activity of FPG and FPQ was higher than that of FPN. A clear discrimination among FPG, FPQ, and FPN was observed in the principal component analysis score plots. Seven compounds were confirmed to contribute strongly by loading plots, which may be the cause of differences in efficacy. Conclusion: This study provides basic information about the chemical and bioactive comparison of FPG, FPQ, and FPN, indicating that protopanaxtriol-type ginsenosides and malonyl-ginsenosides may play a key role in their enhancing immunity properties.

  • Isoflavone contents in soybean seeds are affected by both genetic and environmental factors. Correlation analysis between these factors and isoflavone contents are considered as valuable inputs when breeding improved soybean cultivars. In this study, the seeds of 49 soybean accessions grown in Korea were grouped as black, yellow, green, yellowish-green, pale yellow, and green with a black spot based on their seed coat colors. The contents of 12 isoflavones were analyzed and the association between isoflavone content and seed coat color was determined. The accessions were also grouped as early, intermediate, and late-maturing based on their days to maturity. Out of the 12 isoflavones, 11 were found in 2 accessions, 9 in 18 accessions, 8 in 11 accessions, 7 in another 11 accessions, and 6 in 7 accessions. The total isoflavone content (TIC) in black, yellow, green, yellowish-green, pale yellow, and green with black spot soybeans was in the ranges 2.110 ~ 5.777, 2.487 ~ 4.733, 2.185 ~ 4.413, 2.681 ~ 4.065, 1.827 ~ 4.085, and 3.376 ~ 4.133 mg/g, respectively. The average TIC was highest in green with black spot soybeans (3.616 mg/g), and lowest in pale yellow soybeans (2.875 mg/g). Besides, the average TIC was lowest in early maturing accessions compared to late- and intermediate-maturing accessions. TIC was strongly correlated to malonylgenistin (r = 0.91) and malonyldaidzin (r = 0.78) contents, and poorly correlated to glycitein (r = 0.04) and malonylglycitin (r = 0.18) contents. Also, days to maturity showed strong correlation with malonylgenistin (r = 0.47) content and TIC (r = 0.38). The principal component analysis outlined accessions with high TIC and diverse isoflavones along the first and second components, respectively. The results of the present study depicted that green soybeans with a black spot could be sources of high TIC. Furthermore, late-maturing accessions with diverse isoflavones in their seeds could be useful in future agricultural systems in Korea.

  • Sea ice is generally accepted as an important factor to understand the process of earth climate changes and is the basis of earth system models for analysis and prediction of the climate changes. To continuously monitor sea ice changes at kilometer scale, it is demanded to create more accurate grid data from the current, limited sea ice data. In this paper we described a downscaling method for Advanced Microwave Scanning Radiometer 2 (AMSR2) Sea Ice Concentration (SIC) from 10 km to 1 km resolution using a weighting scheme of sea ice days ratio derived from Moderate Resolution Imaging Spectroradiometer (MODIS) sea ice cover product that has a high correlation with the SIC. In a case study for Okhotsk Sea, the sea ice areas of both data (before and after downscaling) were identical, and the monthly means and standard deviations of SIC exhibited almost the same values. Also, Empirical Orthogonal Function (EOF) analyses showed that three kinds of SIC data (ERA-Interim, original AMSR2, and downscaled AMSR2) had very similar principal components for spatial and temporal variations. Our method can apply to downscaling of other continuous variables in the form of ratio such as percentage and can contribute to monitoring small-scale changes of sea ice by providing finer SIC data.

  • Solid phase microextraction and gas chromatography-mass spectrometry (SPME/GC-MS), electronic nose, and electronic tongue were used to characterize the sensory profiles of cider vinegars from Korea (K1-2), China (C1-2), Japan (J1-2), and US (U1-2). SPME-GC/MS detected acetic acid as the common volatile compound in all vinegars, in addition to isovaleric acid, octanoic acid, and phenethyl acetate. Acids and acetic esters were the major components of Korean and US vinegar samples, respectively. Chinese vinegars had high ethyl acetate content, while Japanese samples were characterized by a low content of acetic acid. Principal component analysis (PCA) pattern provided a clear categorical discrimination of Chinese vinegars by E-nose and E-tongue analyses. The instrumental sensory scores and the taste attributes for flavor ($r^2=0.9431$), sourness ($r^2=0.9515$), and sweetness ($r^2=0.8325$) were highly correlated. Therefore, SPME/GC-MS, E-nose, and E-tongue analyses may be useful tools to discriminate the sensory profiles of cider vinegars of different origins.
