Studies on Application of Sterilization Method Using Ultra Violet Radiation for Liquid Preparations (자외선살균법(紫外線殺菌法)의 액제(液劑)에 대(對)한 응용(應用)의 검토(檢討))

  • The stabillities of taurine, niacinamide, and pyridoxine hydrochloride under exposure to ultra violet radiation in liquid preparations were studied. And sterilization effects for E. coli in both water and liquid preparations were also comparatively evaluated. The above mentioned organic compounds were stable under this experimental conditions and viable count of E. coli reveals that organic compounds dissolved in solution display protective action for microorganisms under UV-irradiation.

Effect of Additives on Disintegration of Enzyme Tablets (효소정제의 붕해에 미치는 첨가제의 영향)

  • In case of the slowly disintegrating tablets such as enzyme preparations, disintegration time (DT) may be the important factor in formulating those preparations. The effects of tablet hardness, lubricants and disintegrants on DT were investigated in this approach. Disintegration time was significantly affected by disintegrants, moderately by lubricants, but not by tablet hardness. The effect was in the order of magnesium stearate >talc, PEG, sodium benzoate in case of lubricants, and of Ac-Di-Sol>LHPC>Primogel >Kollidon in case of disintegrants. Because lubricants and disintegrants influenced the tablet hardness and DT profile showed complicated pattern, it should be remembered that all factors mentioned above should be simultaneously considered in the formulation of enzyme tablets.

  • A simple, accurate GC analytical technique for the determination of phthalates. commonly used as plasticizers during the manufacturing process of PVC bags, in blood component preparations was developed and validated. The blood component preparations were extracted with n-hexane. The n-hexane layer was evaporated to dryness and the residue was dissolved in 1 $m\ell$ of n-hexane and analyzed by GC and GC/MS.   • Each ferritin molecule consists of light subunit 19,000 dalton and heavy subunit 22,000 dalton. Twenty-four protein subunit about $440,000{\sim}500,000$ dalton apoferritin which contained $20{\sim}30%$ Fe as ferric hydroxyphosphate polymer form. Horse spleen-derived ferritin consists of 90% light subunit. These genetic characteristics of ferritin preparations were able to determine by cellulose acetate electrophoresis, but these ferritin preparations contained other components to be disturbed during refining, extraction and making finish products and have difficulties in deciding to be just. So, this study was performed to establish the scientific method for determine the quality of ferritin preparations with immunodiffusion methods which has high specificity between heterogeneous proteins.

  • In order to investigate the efficacy of combined preparations of crude drug, first, 'Kamisamhwang-Tang' were examined for pharmacological effect. Crude drug preparations were extracted successively with $H_2O$ and MeOH, and the extracts were developed on silicagel G plate were examined using the Dual-Wavelength TLC Scanner CS-910(Schimadzu) in ultra violet region. To estimate the clinical usefulness of 'Kamisamhwang-Tang' in heart disease (Symbyung syndrome), the above extracts were made an experiment on effects of the anticonvulsion, antipyretic, analgesic and sedative in mice, rats, and rabbits.

An Active Monomeric Form of Bovine Milk Xanthine Oxidase

  • Upon gel filtration, the commercial bovine milk xanthine oxidase preparation was fractionated into two preparations showing enzyme activity. Native polyacrylamide gel electrophoresis showed that one was in a dimeric form and the other was a monomer having molecular weight of 150 kDa. It was also found that this commercial enzyme existed mostly in an active monomeric form without loss of enzyme activity. The rabbit antisera produced against two enzyme preparations cross-reacted well each other. In SDS-polyacrylamide gtel electro-phoresis, however, both enzyme preparations yielded two smaller protein bands below 150 kDa, which appeared to bind with both antisera with high affinity but not to retain enzyme activity. It implies that bovine milk xanthine oxidase can lose its activity when monomeric subunit is further degraded.

Identification of Cinnamon Components and Quantitative Determination of Cinnamlc Acid from Crude Drug Drink Preparations (생약복방 드링크제 중 계피성분의 확인 및 계피산의 분리정량)

  • For the quality control of crude drug drink preparations, methods for identification of cinnamon components and quantitative determination of cinnamic acid were established. Cinnamon components were identified by TLC with benzene/ethyl acetate (1:1, v/v) on silica gel plate by spraying p-anisaldehyde-sulfuric acid. Cinnamic acid contents were determined at UV 280 nm by HPLC on $\mu$-Bondapak Cls column with acetonitrile/water/acetic acid (40:60:2, v/v). Recoveries of cinnamic acid in three crude drug drink preparations were between 84.1-90.2% compared to the content of the cinnamon extract.

  • The effects of various drugs on the alleviation of the symptoms of chemical bums were evaluated in mice to formulate topical antiblister preparations. After a chemical bum was induced on the mouse dorsal skin with 2-chloroethylethyl sulfide, the drug was applied on the disease site. The effectiveness of the drug was evaluated by determining blister size, necrosis score of skin and appearance of the chemical burns induced. It showed that steroids and aminoglycoside antibiotics had a tendency to protect skin cell, and antihistamines decreased the size of chemical bums. While oleaginous base resulted in deleterious effect, hydrophilic base didn't show a significant difference on the alleviation of the chemical burn symptoms compared to the control.

Development of Recipe for the Korean Typical Wild-Vegetable Preparations and Their Storage (전통채소 밑반찬의 Recipe 개발 및 저장성에 관한 연구)

  • Recipe for Korean typical wild.vegetable preparations were developed and changes in pH, total acidity and ascorbic acid were measured during the storage period at 4~$5^{\circ}C$. Crude fiber and minerals were also analyzed and sensory evaluation was conducted. Changes in pH and total acidity were not significant in most of the samples and the results reveal that the vegetable preparations were in good conditions throughtout the storage period at 4~$5^{\circ}C$. The contents of ascorbic acid have been gradually reduced during the first 10-day storage showing 30~70% retention and continuous loss to 30~40% left on 20th-day. Crude fiber and mineral (Ca, p, Fe, K, Mg) contents were relatively higher in the wild-vegetables than in the everyday use vegetables. The results of the sensory evaluations revealed that the tested samples were in good scores (overall scores>6) except sancho-seed pickles.

  • A new application of high pressure liquid chromatography for the determination of fenitrothion and cis- and trans-neopynamin in insecticidal preparations was investigated. Optimum conditions for a good separation and determination were determined; solvent system: dichloromethane + n-hexane = 17 + 83; Bow rate: 0.5ml/min; column: u-porasil ($4mm{\times}3Ocm$); absorbance wavelength: 254nm; 0.05 AUFS and sample size: 30 ul. Recovery of fenicrothion, cis- and trans-neopynamin from mixed artificial preparations was 99.6%, 99.7% and 99.8% respectively. Also reproducibility tests showed that the coefficient of variation was 0.89% for fenitrothion, 0.74% cis-neopynamin and 1.1% for trans-neopynamin. There was no interference with insecticidal preparation containing DDVP, allethrin, S-421 and kerosene. HPLC method was rapid, accurate and it gave better reproducibility and higher sensitivity than any other analytical method. It was considered that HPLC could be greatly applied to the analysis of fenitrothion and neopynamin in insecticidal preparations.
