A Study on the Quantification of PrP 106-126 Peptide by Fluorescamine and Alpha-imager (플로래스카민과 알파이미저를 이용한 PrP 106-126 펩타이드 정량에 관한 연구)

  • Recent social interests on mad cow disease and Creutzfelt-Jacob disease lead researcher' focus onto pathogenic prion proteins causing those diseases. The purpose of this study is to introduce a novel method for micromolecular level quantification of PrP 106-126 peptide, which is a part of prion protein. Fluorescamine has been chosen due to its fluorescence emission characteristics upon reaction with primary amines and Alpha-imager is used to detect the intensity of fluorescence. We succeeded in setting optimal conditions for quantification of PrP 106-206, amyloidogenic prion peptide, at micromolecular level. This study will contribute to identify prion protein aggregation inhibitor and develop new prion protein drug.

Satzmodelle im Koreanischen und Deutschen im Hinblick auf ihre didaktische Verwertbarkeit ($\cdot$독 문형의 비료설정 시론)

  • In der vorliegenden Arbeit werden die deutschen Satzmodelle $f\"{u}r$ Zwecke des DaF-Unterrichts in Korea untersucht. Dabei gehen wir davon aus, dass man die deutschen Satzmodelle vergleichend mit den koreanischen neu aufbauen sollte, um sie im Deutschunterricht fur Koraner recht benutzen zu konnen. Dafur haben wir hier zunachst untersucht, wie man die koreanischen Satzmodelle im Hinblick auf den didaktischen Zweck modifizieren muss. Unter $Ber\"{u}cksichtigung$ des dabei erzielten Ergebnisses wurden $anschlie{\ss}end$end die deutschen Satzmodelle rekonstruiert. Im Koreanischen wurden die Satzmodelle ab Ende der 1960er Jahre verschiedenartig untersucht. Zwar gibt es bisher noch keine Arbeit, die Ergebnisse dieser Forschungen zusammenfasst, dennoch versuchen wir hier aus einigen bisherigen Untersuchungen einen begrenzten Rahmen von koreanischen Satzmodellen herauszufinden. Als Resultat haben wir funf grundlegende Satzmolle hergestellt: (1) Subjekt + $Pr\"{a}dikat$, (2) Subjekt + $Pr\"{a}dikativ$ + $Pr\"{a}dikat$, (3) Subjekt + Objektiv + $Pr\"{a}dikat$, (4) Subjekt + Adverbiale + $Pr\"{a}dikat$, (5) Subjekt + Obj. + Obj./$Pr\"{a}dikativ$/Advb. + $Pr\"{a}dikat$. Das Modell (1) $umfa{\ss}t$ drei $Pr\"{a}dikate$, die sich nach den Wortarten unterscheiden: (intransitives) Verb, Adjektiv und Substantiv. Auch beim Fall (5) $k\"{o}nnen$ drei verschiedene Untermodelle nach der $Kombinationsm\"{o}glichkeit$ auftreten: Sub + Obj. + $Pr\"{a}d$., Sub. + Obj. + $Pr\"{a}dikativ$ + $Pr\"{a}d$., Sub. + Obj. + Advb. + $Pr\"{a}d$. Auch im Deutschen kann man ahnlich wie im Koreanischen didaktisch geeignete Satzmodelle herausarbeiten. Auch dabei wurden bisherige Untersuchungen zu den deutschen Satzmodellen herangewgen. Zu der $d\"{a}fur$ $ausgew\"{a}hlten$ Terminologie $geh\"{o}ren$ Subjekt(S), $Pr\"{a}dikat(P)$, $Pr\"{a}dkativerganzung(PE)$, $Objekterg\"{a}nzung(OE)$, $Adverbialerg\"{a}nzung(AE)$. In der deutschen Sprache werden nach den $Valenzm\"{o}glich­keiten$ des $Pr\"{a}dikativteils$ folgende Modelle unterschieden: S + P, S + P + PE, S + P + OE, S + P + AE, S + P + OE + OE/PE/AE. Unter die OE werden okk. OE, dat. OE, $pr\"{a}d$. OE, refl. OE usw. subsumiert. Um die Stichhaltigkeit der in dieser Schrift neu festgestellten Satzmodelle zu $\"{u}berpr\"{u}fen$, sollten sie anhand eines Textkorpus heutiger Texte in weiteren Untersuchungen validiert werden. Ferner ist es zu empfehlen, die Anwendbarkeit der hier vorgestellten Satzrnodelle im Deutschunterricht $f\"{u}r$ Koreaner $gr\"{u}ndlich$ zu testen

Studies on the Bacteriophages of Brevibacterium lactofermentum (L-글루타민산 생산균 Brevibacterium lactofermentum의 Bacteriophag에 관한 연구)

  • Many industrial processes those employ bacteria are subjected to phage infestations. In L-glutamic acid fermentions using acetic acid, the phage infestations of the organisms have been recently recognized. In efforts to elucidate the sources of phage contamination involved in the abnormal fermentation, a series of study was conducted to isolate the phages both from the contents of abnormally fermented tanks and the soil or sewage samples from the surroundings of a fermentation factory, to define major charateristics of the phage isolates, and finally to determine the correlation between the phage isolates and temperate phages originating from the miscellaneous bacterial species isolated from the soil or sewage samples. The results are summarized as follows; 1) All phages were isolated from the irregular fermentation tanks and soil or sewage samples, and they were designated as phage PR-1, PR-2, PR-3, PR-4, PR-5, PR-6, and PR-7, in the order of isolation. These PR-series phages were proved to be highly specific for the variant strains of Br. lactofermentum only, namely, phage PR-1 and PR-2 for Br. lactofermentum No. 468-5 and phage PR-3~PR-7 for Br. lactofemrentum No. 2256. By cross-neutralization test, the 7 phagescould be subdivided into 3 groups, i. e., phage PR-I and PR-2 the first, phage PR-3, PR-4, PR-5, PR-6 the second, and the phage PR-7 the third. 2) The 7 phages were virulent under the experimental conditions. They produced plaques with clear and relatively sharp margins without distinct halo. The mean sizes of plaques were 1.5mm in diameter for phage PR-1 and PR-2, and 1. Omm for phages PR-3~PR-7. Double layer technique modified by Hongo and described by Adams, was applied to assay of the PR-series phages. The factors influencing the plaques were as follows;young age cells of host bacteria cultured for 3-6 hours represented the largest number and size, optimum was pH 7.0, incubation temperature was $30^{\circ}C$, and agar concentration and amount of overlayer medium were 0.6% and 0.2ml, respectively. 3) PR-series phages were stable in 0.05M tris buffer and 0.1M ammonium acetate buffer solution. The addition of $5{\times}10^{-3}M$ magnesium ion effectively increased the stability. Thermostability experiments indicated that PR-series phages were stable at the teinperture between $50^{\circ}{\sim}55^{\circ}C$ in nutrient medium, $45^{\circ}{\sim}50^{\circ}C$ in buffer solution. However, the phages mere completely inactivated at 603C and 65$^{\circ}$C within 10 minutes. The phages were stable at the range of pH6~9 in nutrient medium and of pH 8-9 in buffer solution, respectively. Exposure of the phages to UV for 25, 60 and 100 seconds resulted in the complete loss of infectivily, respectively. 4) Electron microscopy showed that PR-series phage particles exhibited rather similar morphology, differing in the size All of PR-series phages had a multilateral head and had a simple long tiil about three to five times long as compared with head. By the size, phage PR-1 and PR-2, PR-3, PR-4, PR-5, and PR-6 and PR-7 were classified into same groups, respectively. The head and tail size of phage PR-1, PR-5, PR-5(T) and PR-7 were 85nm, 74nm and 235nm and 350mm, and 72nm and 210nm, respectively. 5) Nucleic acids of PR-series phages were double stranded DNA. The G+C contents of phage PR-1, PR-5 and PR-7 were 56.1, 52.9 and 53.7, respectively. The values of G+C contents derived from the $T_m$ were in agreement with the chemically determined values. 6) PR-series phages effectively adsorbed on their host bacteria at the rate of more than 90% during 5 min. K value for phage PR-1, PR-5 and PR-7 were calculated to be $6{\times}10^9 ml$ per minute, respectiveky. The pH of the medium did effect adsorption rate, but both temperature and age of host cells did not. Generally, optimum adsorption condition of phages seemed to be almost same as optimum growth conditions of host bacteria. 7) In one-step growth experiments, the latent periods at $30^{\circ}C$ for PR-1, and PR-7 were about 70, 50 and 55 min, respectively. The corresponding average burst size was 200, 70 and 90, respectively. Lpsis period according to the multiplicity of infection and a phage series. In case of m. o. i. 100, strain No. 2256 (PR-5) and No. 468-5(PR-1) failed to grow and turbidity decreased after 50 and 70min, respectively. 8) In the lysate of a plaque purified phage PR-5 infected bacteria, there observed 2 types ofphage particles, i. e., phage PR-5 and PR-5 (T) of similar morphology but differing at the length of phage tail, and phage tail like particles. The phage taillike particles could be divided into 4 types by the length. Induction experiments of Br. lactofermentum with UV irradiation, mitomycin C or bacitracin treatment produced neither phage PR-5 (T) or phage tail-like particles. 9) No lysis occured when the growth of 7 strains of miscellaneous bacteria, isolated from soil and sewage samples, were inoculated with either phage PR-5 (T) or phage tail-like particles the inoculation of phage PR-5 pellet resulted in the growth inhibition of the orgainsms in the spot test. The lysates obtained from 3 miscellaneous soil derived bacteria following mitomycin C treatment the growth of Br. lactofermentum, but did not lyze the bacterium.

  • Let R be a G-graded commutative ring with a nonzero unity and P be a proper graded ideal of R. Then P is said to be a graded uniformly pr-ideal of R if there exists n ∈ ℕ such that whenever a, b ∈ h(R) with ab ∈ P and Ann(a) = {0}, then bn ∈ P. The smallest such n is called the order of P and is denoted by ordR(P). In this article, we study the characterizations on this new class of graded ideals, and investigate the behaviour of graded uniformly pr-ideals in graded factor rings and in direct product of graded rings.

Synthesis and Reactions of Organoruthenium(Ⅲ) Complexes (새로운 3가 유기루테늄 착물의 합성과 반응)

  • The paramagnetic organoruthenium(III) complexes $({\eta}^5-C_5Me_5)RuCl_2(PR_3) (PR_3 = PMe_3,\;PEt_3,\;PiPr_3,\;PCy_3,\;PMe_2Ph,\;PMePh_2,\;PPh_3,\;P(p-C_6H_4CH_3)_3$, DPPE, DPPB, Py) (2a∼2k) were synthesized by the reaction of $[({\eta}^5-C_5Me_5)RuCl_2]_2$ (1) with 1 equivalent of the corresponding phosphines $(PR_3)$. The effective magnetic moment ((${\mu}_{eff} = 1.65∼2.07 B.M.$)) derived from the magnetic susceptibility measurements of the complexes (2a∼2k) were consistent with the presence of a "single" unpaired electron in the molecule. Treatment of dichlororuthenium (III) complex ({\eta}^5-C_5Me_5)RuCl_2(PR_3)$ (2) (i) with KBr in acetone afforded the dibromoruthenium (III) complex $({\eta}^5-C_5Me_5)RuBr_2(PR_3) (PR_3 = PPh_3)$, (ii) with sodium amalgam in diethylether led to the bis(phosphine) derivatives $({eta}^5-C_5Me_5)RuCl(PR_3)_2 (PR_3 = PMe_3,\;PMePh_2)$, and (iii) with carbonmonoxide gave to the carbonyl derivatives $({\eta}^5-C_5Me_5)RuCl(PR_3)(CO) (PR_3 = PMe_3,\;PPh_3)$.

Synthesis and characterization of CaZrO3:Pr3+ phosphor by skull melting method (스컬용융법에 의한 CaZrO3:Pr3+ 형광체 합성 및 특성분석)

  • Ca1-xZrO3:xPr phosphor with perovskite structure was successfully synthesized by using skull melting method. The crystal structure, morphology and optical properties of synthesized phosphor were characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), ultraviolet fluorescence reaction and photoluminescence. The XRD results indicated that single crystals of CaZrO3:Pr3+ belongs to orthorhombic perovskite system. The synthesized phosphor could be excited by UV light (254 nm) and the emission spectra results indicated that green luminescence of CaZrO3:Pr3+ due to charge transfer transition 3P03H4, 3P13H5 and 3P03H5 at 506, 536 and 548 nm was dominant.

Solution Structure of a Prion Protein: Implications for Infectivity

  • Prions cause neurodegenerative diseases in animals and humans. The scrapie prion protein (PrPSc) is the major-possibly only-component of the infectious prion and is generated from the cellular isoform (PrPC) by a conformational change. Limited proteolysis of PrPSc produces an polypeptide comprised primarily of residues 90 to 231, which retains infectivity. The three-dimensional structure of rPrP(90-231), a recombinant protein resembling PrPC with the Syrian hamster (SHa) sequence, was solved using multidimensional NMR. Low-resolution structures of rPrP(90-231), synthetic peptides up to 56 residues, a longer (29-231, full-length) protein with SHa sequence, and a short here further structure refinement of rPrP(90-231) and dynamic features of the protein. Consideration of these features in the context of published data suggests regions of conformational heterogeneity, structural elements involved in the PrPC\longrightarrowPrPSc transformation, and possible structural features related to a species barrier to transmission of prion diseases.

Comparision for Hospital P. R. Activities among Large General Hospitals (병원홍보실태에 대한 비교연구)

  • This study is purposed to compare PR activities among the 5 large hospitals in Seoul and Kyonggido. The research methods are used in interview survey for employees and Questionnaires survey for patients or their relatives. The 427 patients or their relatives(response rate: 85.4%) were surveyed. The hospital which is below, is more active than any others in special PR activities. 1. Distribution of hospital information books: E, P, A 2. Distribution of calendar: P 3. Broadcast in hospital: E 4. Education of employees about PR : E, A 5. Education of employees about service: all hospitals are active. The PR sectors are organized under the Department of planning(S,J,E) or CEO(P,A). And the budget of PR activities is almost a similar level among all. The degree of recognition and utilization for PR activities is not different among hopitals by the sociodemographic variables of patients. And the average of correlation ratio between recognition of each hopital's PR activities and its utilization is 62.01%(range:51.29%-67.98%) average.

Effects of Puerariae Radix extract on Cisplatin-Induced Apoptosis of Rat Mesangial Cells (갈근(葛根) 추출물이 cisplatin에 의해 유도된 rat mesangial cell의 apoptosis에 미치는 영향)

  • Puerariae radix (PR) is a popular natural herb and a traditional food in Asia, which has antithrombotic and anti-allergic properties and stimulates estrogenic activity. One of the major side effects of cisplatin is nephrotoxicity, leading to acute renal failure. Recent study has suggested a role of ROS and p53 in renal cell injury by cisplatin. We studied that protective effects of PR on cisplatin-induced apoptosis in rat mesangial cell. Rat mesangial cell was preincubated with PR (50, 100, 150 and 200 ${\mu}g/m{\ell}$) for 12 hr and then treated with 30 ${\mu}M$ cisplatin for 24 hr. Protective effect of PR on cisplatin-induced apoptosis in ECV304 cells was determined using MTT assay, FDA-PI staining, flow cytometric analysis, caspase-3 activity assay, ROS assay and western blot. Our results showed that PR inhibited in cisplatin-induced apoptosis and ROS production in ECV304 cells. Moreover, PR reduced ERK, p38 and JNK activation that increased in cisplatin-treated rat mesangial cell. Furthermore, activation of p53 by cisplatin in rat mesangial cell was inhibited by PR treatment. These results suggest that protective effect of PR on cisplatin-induced apoptosis in rat mesangial cell may be associated with reduction of ERK, p38, JNK, p53 activation.

Cytosolic prion protein induces apoptosis in human neuronal cell SH-SY5Y via mitochondrial disruption pathway

  • Different neurodegenerative disorders like prion disease, is caused by protein misfolding conformers. Reverse-transfected cytosolic prion protein (PrP) and PrP expressed in the cytosol have been shown to be neurotoxic. To investigate the possible mechanism of neurotoxicity due to accumulation of PrP in cytosol, a PrP mutant lacking the signal and GPI (CytoPrP) was introduced into the SH-SY5Y cell. MTT and trypan blue assays indicated that the viability of cells expressing CytoPrP was remarkably reduced after treatment of MG-132. Obvious apoptosis phenomena were detected in the cells accumulated with CytoPrP, including loss of mitochondrial transmembrane potential, increase of caspase-3 activity, more annexin V/PI-double positive-stained cells and reduced Bcl-2 level. Moreover, DNA fragmentation and TUNEL assays also revealed clear evidences of late apoptosis in the cells accumulated CytoPrP. These data suggest that the accumulation of CytoPrP in cytoplasm may trigger cell apoptosis, in which mitochondrial relative apoptosis pathway seems to play critical role.
