Study on the Antioxidant Effect and Total Phenolics Content in Rosaceae Plant Stem (장미과 식물 줄기의 항산화 효과와 총 페놀류 함량에 관한 연구)

  • The antioxidant activities and total phenolics of four Rosaceae species Pourthiaea villosa (Thunb.) Decne, Sorbus commixta Hedlund, Sorbaria sorbifolia var. stellipila Maxim and Pyrus pyrifolia (Burm.f.) Nakai were determined. Phenolic content (polyphenol and flavonoid), radical scavenging activities [2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylnezthiazoline-6-sulfoic acid) (ABTS)] and ferrous ion chelating effect were evaluated. Total polyphenol and flavonoid contents were highest in Pourthiaea villosa (Thunb.) Decne and lowest in Pyrus pyrifolia (Burm.f.) Nakai. Phenolic contents of Pourthiaea villosa (Thunb.) Decne was $331.45{\pm}7.78$ and $90.4{\pm}3.5mg{\cdot}g^{-1}$. DPPH and ABTS radical scavenging activities were found to be lowest in Sorbus commixta Hedlund whereas Sorbaria sorbifolia var. stellipila Maxim and Pourthiaea villosa (Thunb.) Decne showed relatively good DPPH and ABTS radical scavenging activities. Ferrous ion chelating effect was highest in Pyrus pyrifolia (Burm.f.) Nakai ($1.05{\pm}0.04mg{\cdot}ml^{-1}$) and lowest in Sorbus commixta Hundlund ($4.22{\pm}0.71mg{\cdot}ml^{-1}$).

  • The fruits of Pourthiaea villosa were extracted with methanol and its extract was fractionated with n-hexane, methylene chloride, ethyl acetate and n-butanol. Repeated column chromatography of silica gel, sephadex LH-20 and HPLC led to the isolation of nine phenolic compounds from ethyl acetate soluble fraction. The chemical structures were elucidated as kaemferol-3-O-${\beta}$-D-glucopyranoside (astragalin) (1), isorhamnetin-3-O-${\beta}$-D-glucopyranoside (2), kaempferol-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\alpha}$-L-rhamnopyranoside (3), caffeic acid (4), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-a-L-rhamnopyranoside (5), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\beta}$-D-glucopyranoside (6), quercetin-3-O-${\beta}$-D-xylopyranosyl($1{\rightarrow}2$)-${\beta}$-D-galactopyranoside (7), quercetin-3-O-${\alpha}$-L-rhamnopyranoside (quercitrin) (8), and kaempferol-3-O-${\alpha}$-L-rhamnopyranoside (afzelin) (9) by spectroscopic techniques. These compounds were isolated from this plant for the first time.

Quantitative Analysis and Anti-inflammatory of Lyoniside from the Pourthiaea villosa var. brunnea (떡윤노리나무로부터 분리된 Lyoniside의 함량분석과 항염증 효과)

  • In this study, the purification of the MeOH extract from the stems and leaves of Pourthiaea villosa var. brunnea using column chromatography furnished a main compound, lyoniside. The structure was elucidated on the basis of $^1H$ and $^{13}C$ NMR spectroscopic data. Quantitative analysis of lyoniside was conducted by HPLC method and the highest content of lyoniside was found in 50% MeOH reflux extraction. To investigate the anti-inflammatory effect of the lyoniside, we measured nitric oxide and pro-inflammatory cytokines such as TNF-${\alpha}$, IL-$1{\beta}$, IL-6 levels in lipopolysaccharide-induced murine macrophage cell line RAW 264.7. As a results, lyoniside decreased the level of nitric oxide and IL-6 in concentration dose dependent manner in RAW 264.7 cells.

Analysis of Morphological Characteristics and Variation among Six Populations of Pourthiaea villosa (Thunb.) Decne. var. villosa in Korea (국내 윤노리나무 6개 집단의 형태적 특징과 변이조사)

  • This study was conducted to examine the morphological characteristics and variations of the 6 populations of Pourthiaea. villosa (Thunb.) Decne. var. villosa in Korea. The ANOVA results showed that the populations were significantly different in 18 of the 21 quantitative characteristics that were analyzed. In the results of principal component analysis, 6 principal components (PC) represented 68.28% for the total variations. 'Petiole length' (0.764), 'Leaf shape' (0.834), 'Leaf length' (0.753), 'Crown diameter' (0.663), 'Inflorescence width' (0.492), and 'Leaf base shape' (0.721) showed the highest contribution to PC1, PC2, PC3, PC4, PC5, and PC6, respectively. According to the results of cluster analysis, one cluster comprised only 'Goesan-gun Ssanggok Valley' population. 'Namhae-gun Mijori Evergreen Forest' population was grouped with 'Hamyang-gun Sang Forest' population, which was the nearest subgroup. Further, 'Namhae-gun Geum Mountain' population, 'Jindo-gun Cheomchal Mountain' population, and 'Jeju-si Barimae Parasitic volcano' population were placed in the same cluster.

Phytosociological Studies of Pinus densiflora Forest in Islets of Southwestern Coast, Korea (한국 서남해 도서의 소나무 (Pinus densiflora S. et Z.)림에 대한 식물사회학적 연구)

  • This study was carried out to analyze Pinus densiflora forest in islets of southwestern coast of the Korean peninsula by the phytosociological methods. As a result of the investigation of 26 quadrats, the pine forest was classified two communities and six subcommunities : Ardisia japonica - Pinus densiflora community and lsodon japonicus - Pinus densiflora community, and Farfugium japonicum subcommunity, Quercus serrata subcommunity, Typical subcommunity of Ardisia japonica - P. densiflora community, Juniperus rigida subcommunity, Pourthiaea villosa subcommunity and Typical subcommunity of lsodon japonicus - P. densiflora community. The subsidiary knowledges from this study provide practical information to clarify the syntaxonomy of natural pine forest of the Korean peninsula by the phytosociological methods.

Antioxidant Activity of 11 Species in Korean Native Forest Plants (국내 산림식물 11종의 항산화 활성)

  • This study aimed to investigate antioxidant activities from 11 forest plants, and determine their total phenolics, flavonoids and proantocyanidins contents. In addition, the antioxidant activities were correlated with antioxidant compounds. Among the samples, Cornus officinalis, Castanea crenata, Lindera erythrocarpa, Carpinus laxiflora and Pourthiaea villosa showed significantly higher 2,2-diphenyl-1-picrylhydrazyl (DPPH) ($IC_{50}=21.12{\sim}28.93{\mu}g/mL$) and 2,2'-azino-bis(3-ethylbenzothia zoline-6-sulfonic acid) (ABTS) ($IC_{50}=28.18{\sim}52.55{\mu}g/mL$) radical scavenging ability with reducing power ($IC_{50}=59.91{\sim}93.64{\mu}g/mL$) than other plants; and C. crenata, L. erythrocarpa and Rubus coreanus showed strong nitric oxide (NO) inhibition activity (${\geq}60%$). In addition, L. erythrocarpa, C. laxiflora and P. villosa showed higher oxygen radical absorbance capacity (ORAC) values (${\geq}1,100{\mu}M$ TE/g sample) than other samples. High total phenolic contents were observed in C. crenata (429.11 mg GAE/g), L. erythrocarpa (437.11 mg GAE/g), C. laxiflora (408.67 mg GAE/g) and P. villosa (404.11 mg GAE/g). The DPPH and ABTS radical scavenging activity with reducing power were significantly correlated with total phenolic contents ($R^2=0.71{\sim}0.79$), but total phenolic contents were not correlated with NO inhibition and ORAC ($R^2=0.35{\sim}0.43$). Therefore, these results suggested that C. officinalis, C. crenata, L. erythrocarpa, C. laxiflora and P. villosa are potential natural antioxidative candidate ingredients.

  • The regulation of plasma lipid level, particularly LDL cholesterol, represents the focus of current therapy for atherosclerosis. And $Lp-PLA_2$ is able to hydrolyse oxidized phosphatidylcholine within LDL into lyso-PC and oxidized fatty acids. $Lp-PLA_2$ is a potential biomarker of coronary heart disease and plays an important proinflammatory role in the progression of atherosclerosis. We investigated the inhibitory effects of methanol extracts of 224 natural plants on $Lp-PLA_2$ activity. Seven kinds of methanol extracts of tested plants showed above 50% inhibitory effect with the concentration of $100\;{\mu}g/ml$. The concentrated aqueous suspensions of each methanol extract were partitioned with n-hexane, $CHCl_3$, and EtOAc. Among them, EtOAc extracts of Astilbe chinensis var. davidii (root) and Pourthiaea villosa var. brunnea (leaf) significantly inhibited $Lp-PLA_2$ activity at the same concentration.

Community Classification for Actual Vegetation of Anmyeon Island in Chungcheongnam-do Province, Korea (안면도 현존식생에 대한 군락분류)

  • The purpose of this study was to explain community structure for actual vegetation and their environment variables in Anmyeon Island. Samples were collected for 106 plots using ZM phytosociology method and coincidence method. Actual vegetation were classified into three vegetations types(forest vegetation type, maritime vegetation type, lake vegetation type) and eleven community units. Pourthiaea villosa community in forest vegetation type was divided into 5 groups such as Carpinus coreana group, Pinus rigida group, Chamaecyparis obtusa group, Castanea crenata group and Typical group. Maritime vegetation type was divided into 3 communities, such as Vitex rotundifolia community, Koelreuteria paniculata community and Suaeda japonica community. V. rotundifolia community was subdivided into 2 groups, Rosa rugosa group and Diodia teres group. K. paniculata community was subdivided into 2 groups, Grewia biloba var. parviflora group and Typical group. Lake vegetation type was divided into 1 community, Nelumbo nucifera community. And it was entirely classified into 11 community units.
