In more than half of human tumors, the p53 tumor suppressor gene is mutated. Thus, restoration of wild-type p53 activity by repair of mutant RNA could be a potentially promissing approach to cancer treatment. To explore the potential use of RNA repair for cancer therapy, trans-splicing group I ribozymes were developed that could replace mutant p53 RNA with RNA sequence attached to the 3'end of ribozymes. By employing a mapping library of ribozymes, we first determined which regions of the p53 RNA are accessible to ribozymes, and found that the leader sequences upstream of the AUG start codon appeared to be particularly accessible. Next, trans-splicing ribozymes were generated that specifically recognized the sequences around these accessible regions. Subsequently, the ribozymes reacted with and altered the p53 transcripts by transferring a 3'exon tag sequence onto the targeted p53 RNA with high fidelity. Thus, these ribozymes could be utilized to repair mutant p53 in tumors, which would revert the neoplastic phenotype.
The Honam-Jeju, Korea-Japan, and Korea-China subsea tunnel construction projects have drawn significant attention since the early 2000s. These subsea tunnels are much deeper than most existing natural shallow sea tunnels linking coastal areas. Thus, the need for developing new technologies for the site selection and construction of deep subsea tunnels has recently emerged, with the launch of a research project titled "Development of Key Subsea Tunnelling Technology" in 2013. A component of this research, an analysis of deep subsea geological structure, is currently underway. A ground investigation, such as a borehole or geophysical investigation, is generally carried out for tunnel design. However, when investigating a potential site for a deep subsea tunnel, borehole drilling requires equipment at the scale of offshore oil drilling. The huge cost of such an undertaking has raised the urgent need for methods to indirectly assess the local geological structure as much as possible to limit the need for repeated borehole investigations. This study introduces an indirect approach for assessing the geological structure of the seafloor through a submarine bathymetry analysis. The ultimate goal here is to develop an automated approach to the analysis of submarine geological structures, which may prove useful in the selection of future deep subsea tunnel sites.

The size of the superconducting domains and the critical current density inside these domains have to be enhanced for most of cryomagnetic applications of melt-textured YBCO bulks. To enlarge the size of the domains we studied the multi-seeding technique based on a well-established procedure for preparing high quality YBCO monoliths using self-made SmBCO seeds. The distance between the seeds was optimised as a result of the investigation of the effects of various seed distances on the characteristics of the grain boundary Junctions. The influences of a-b plane intersections and c-axis misalignments were researched. Thereby, a small range of tolerance of the misorientations between the seed crystals was found. Field mapping was applied to control the materials quality and the superconductor's grain structure was investigated using polarisation microscopy. YBCO function elements with iou. seeds in a line and an arrangement of making type (100)/(100) and (110)/(110) boundary junctions, respectively, were processed. The trapped field profile in both sample types shows single domain behaviour. To demonstrate the potential of the multi-seeding method a ring-shaped sample was processed by placing sixteen seeds in a way to make both (100)/(100) and (110)/(110) grain junctions at the same time. The results up to now are very promising to prepare large single domain melt-textured YBCO semi-finished products in complex shapes.

Remote sensing(RS)- and geographic information system(GIS)-based information management to measure and assess agri-environment schemes, and to quantify and map environment indicators for nature and land use, climate change, air, water and energy balance, waste and material flow is in high demand because it is very helpful in assisting decision making activities of farmers, government, researchers, and consumers. The versatility and ability of RS and GIS containing huge soil database to assess agricultural environment spatially and temporally at various spatial scales were investigated. Spectral and microwave observations were carried out to characterize crop variables and soil properties. Multiple sources RS data from ground sensors, airborne sensors, and also satellite sensors were collected and analyzed to extract features and land cover/use for soils, crops, and vegetation for support precision agriculture, soil/land suitability, soil property estimation, crop growth estimation, runoff potential estimation, irrigated and the estimation of flooded areas in paddy rice fields. RS and GIS play essential roles in a management and monitoring information system. Biosphere-atmosphere interection should also be further studied to improve synergistic modeling for environment and sustainability in agri-environment schemes.
Augmenting-reducing evoked potentials(AREP) were studied in 38 college students to explore the topographic distribution between AR slope and personality. The Zuckerman Seeking Scale(SSS) and Eysenck Personality Questionnaire(EPQ) assessed personality. There was a significant positive correlation between AR slope and Extraversion-Introversion(E) in the frontocentral area ; the right posterior area showed a significant negative correlation with E. The Thrill and Adventure Seeking(TAS) subscale showed a significant negative correlation with slope in the right posterior temporal area. The average slope map of all subjects revealed a distribution showing more augmenting in frontocentral areas and more reducing in posterior areas.

This paper deals with a new mathematical formulation of nonlinear wave profile based on Banach fixed point theorem. As application of the formulation and its solution procedure, some numerical solutions was presented in this paper and nonlinear equation was derived. Also we introduce a new operator for iteration and getting solution. A numerical study was accomplished with Stokes' first-order solution and iteration scheme, and then we can know the nonlinear characteristic of Stokes' high-order solution. That is, using only Stokes' first-oder(linear) velocity potential and an initial guess of wave profile, it is possible to realize the corresponding high-oder Stokian wave profile with tile new numerical scheme which is the method of iteration. We proved the mathematical convergence of tile proposed scheme. The nonlinear strategy of iterations has very fast convergence rate, that is, only about 6-10 iterations arc required to obtain a numerically converged solution.
Some computational approaches are needed for clarifying RNAi sequences, because it takes much time and endeavor that almost of RNAi sequences are verified by experimental data. Incorrectness of RNAi mechanism and other unaware factors in organism system are frequently faced with questions regarding potential use of RNAi as therapeutic applications. Our massive parallelized pair alignment scoring between dsRNA in Genebank and expressed sequence tags (ESTs) in Caenorhabditis elegans Genome Sequencing Projects revealed that this provides a useful tool for the prediction of RNAi induced cosuppression details for practical use. This pair alignment scoring method using high performance computing exhibited some possibility that numerous unwanted gene silencing and cosuppression exist even at high matching scores each other. The classifying the relative higher matching score of them based on GO (Gene Ontology) system could present mapping dsRNA of C. elegans and functional roles in an applied system. Our prediction also exhibited that more than 78% of the predicted co-suppressible genes are located in the ribosomal spot of C. elegans.

US transportation agencies are dealing with shrinking budgets, limited work forces, and deteriorating infrastructure. In order to cope with funding uncertainty, state highway agencies are now looking into their own organizations and identifying programs, practices, and processes that have potential for cost saving. A quality assurance (QA) program is an integral part of highway construction and ensures a project's contracted level of quality. The cost of quality (conforming and nonconforming) can constitute a sizable part of total construction cost. As the quality assurance programs evolved, various practices and processes were developed over time and later adopted by state highway agencies. These practices and processes include different QA standards and specifications, varying testing methods, central testing lab vs. on site testing, performance based vs. prescribed quality assurance practices, implementation of innovative quality assurance practices, etc. Therefore, there is an opportunity to assess different QA strategies and recommend those practices that are effective and cost efficient. A national survey was conducted by the authors, which provided a detailed mapping of various QA practices and processes used as part of QA programs and identified areas where agencies can focus on for cost savings. The survey found that QA sampling and testing plans, optimization of sampling plans, optimization of QA standards and specifications, and implementation of innovative test methods and processes are the main areas the agencies should focus to lean the current QA programs.
A simple array of metal-insulator-metal capacitive elements was proposed for a potential application in humidity sensing platforms. We fabricated meso-scale sensors with different sizes(large-size: $2.7{\times}2.7mm^2$ ; mid-size: $1.5{\times}1.5mm^2$ ; small-size: $0.7{\times}0.7mm^2$) and characterized the performance of each design. Polyimide films were utilized as a humidity-sensitive layer. Capacitance changes of the polyimide layer were measured with respect to water absorption. The device showed sensitivity in the full range of relative humidity (RH) with excellent linearity(correlation coefficient > 0.994). This array structure exhibits unique advantages including easy fabrication process, high batch productivity, and high structural compatibility with various substrate materials. It is anticipated that this device structure will be potentially useful in unique applications including mapping spatial humidity variations over a meso-scale area and implementing flexible humidity sensing element arrays.
The extraction of important features in cancer cell image analysis is a key process in grading renal cell carcinoma. In this study, we applied three-dimensional (3D) texture feature extraction methods to cell nuclei images and evaluated the validity of them for computerized cell nuclei grading. Individual images of 2,423 cell nuclei were extracted from 80 renal cell carcinomas (RCCs) using confocal laser scanning microscopy (CLSM). First, we applied the 3D texture mapping method to render the volume of entire tissue sections. Then, we determined the chromatin texture quantitatively by calculating 3D gray-level co-occurrence matrices (3D GLCM) and 3D run length matrices (3D GLRLM). Finally, to demonstrate the suitability of 3D texture features for grading, we performed a discriminant analysis. In addition, we conducted a principal component analysis to obtain optimized texture features. Automatic grading of cell nuclei using 3D texture features had an accuracy of 78.30%. Combining 3D textural and 3D morphological features improved the accuracy to 82.19%. As a comparative study, we also performed a stepwise feature selection. Using the 4 optimized features, we could obtain more improved accuracy of 84.32%. Three dimensional texture features have potential for use as fundamental elements in developing a new nuclear grading system with accurate diagnosis and predicting prognosis.
