A total of 187 endophytic fungi were isolated from 11 plant species, which were collected from 11 locations in Korea. Their antifungal activities were screened in vivo by antifungal bioassays after they were cultured in potato dextrose broth and rice solid media. Antifungal activity against plant pathogenic fungi such as Magnaporthe grisea(rice blast), Corticium sasaki(rice sheath blight), Botrytis cinerea(tomato gray mold), Phytophthora infestans(tomato late blight), Puccinia recondita(wheat leaf rust), and Blumeria graminis f. sp. hordei(barley powdery mildew) was determined in vivo by observing the inhibition of plant disease development. Twenty(11.7%) endophytic fungi fermentation broths were able to control, by more than 90%, at least one of the six plant diseases tested. Among 187 liquid broths, the F0010 strain isolated from Abies holophylla had the most potent disease control activity; it showed control values of more than 90% against five plant diseases, except for tomato late blight. On the other hand, fourteen(7.5%) solid culture extracts exhibited potent disease control values of more than 90% against one of six plant diseases. The screening results of this study strongly suggested that metabolites of plant endophytic fungi could be good potential sources for screening programs of bioactive natural products.
The main objective of this breeding program is to develop high yielding, disease resistance and good processing potato cultivar. 'Gui Valley' is a clonal selection resulting from a cross between 'ND2471-8' and 'Cona'. It has medium plant height and light green foliage. 'Gui Valley' has medium flowering habit and light pink flowers. 'Gui Valley' is a medium maturing potato cultivar and tubers are smooth, yellow skin, light yellow flesh, long-oval tuber shape, distinct red eyes with medium depth and medium dormancy. It has high level of tuber uniformity and good keeping quality. 'Gui Valley' demonstrates resistance to potato virus Y (PVY), soft rot, but moderately susceptible to late blight and common scab. It is also resistant to most of the internal and external physiological disorders particularly dehiscence, hollow heart and internal brown spot. The specific gravity of 'Gui Valley' is significantly higher (1.097) than that of 'Shepody' (1.078). 'Gui Valley' has suitable for processing mainly French fries and chips. This cultivar has high level of tuber uniformity and capable of yielding 37.6 $t{\cdot}ha^{-1}$, which is 18.2% higher than the control potato cultivar 'Shepody' under optimum agronomical practices.
Commercial formulation of fungicides was studied in vitro for sensitivity against Typhula species causal agents of Typhula blight. Efficacies of fungicides application, spray volume, nozzle types and fungicides applied time (early fall and late fall) were evaluated for their influence on the chemical control of Typhula blight of turfgrass during the winter season in Wisconsin. All fungicides effectively reduced the mycelial growth of eight isolates of Typhula spp. in vitro on potato dextrose agar (PDA) media. For inhibitory effects on mycelial growth of eight isolates, propiconazole was the most effective at $1.0{\mu}g$ active ingredient (a.i) / ml of PDA. Typhula incarnata two isolates were significantly more sensitive to all fungicides of PDA than six isolates of three varieties of T. ishikariensis. For 2 years in field experiment, unsprayed control has significantly more disease severity than seven fungicides were applied to field plots at two locations. Propiconazole was the most effective for controlling Typhula blight, at two locations in both years. The level of disease control was not dependent on fungicides spray volume or nozzle types at two locations. The disease damage treated with triadimefon applied time (early fall and late fall) was not significantly different at two location for two years.
Efficacy of resistance induction by the bacterial isolates Pseudomonas putida (TRL2-3), Micrococcus luteus (TRK2-2) and Flexibacteraceae bacterium (MRL412), which were isolated from the rhizosphere of plants growing in Jeju Mountain, were tested in a greenhouse. The disease severity caused by Phytophthora infestans was effectively reduced in the potato plants pre-inoculated with bacterial isolates compared with those of the untreated control plants growing in a greenhouse. In order to estimate the level of protection by the bacterial isolates, Mancozeb WP (Diesen $M^{(R)}$, Kyong nong) and DL-3-amino butyric acid (BABA) were pre-treated, whereas Dimethomorph WP ($Forum^{(R)}$, Kyong nong) and phosphonic acid ($H_{3}PO_{3}$) were post-treated the challenge inoculation with the pathogen. Disease severities of chemical pre-treated as well as post-treated plants were reduced compare to those of the untreated. The disease reduction in the plants pre-treated with Mancozeb WP was the highest, whereas that of post-treated with Dimethomorph WP was the lowest. The yields of plants pre-inoculated with three bacterial isolates were greatly increased than those of control plants. These results suggest that biological control by bacterial isolates might be an alternative strategy against late blight disease in potato plants growing in greenhouse.
For rapid and secure differentiation of P. infestans from other Phytophthora species, two fragments obtained from randomly amplified polymorphic DNA (RAPD) profiles were selected as markers. Also, primers for in polymerase chain reaction (PCR) to detect P. infestans specifically were developed by analyzing the sequences of ITSII regions in rDNA of Phytophthora species. The primers, PISP-1 and ITS3 amplified a single. Fragment 450 bp of about in P. infestans, but not in other fungal or bacterial isolates. Annealing temperatures and template DNA quantities were varied for the optimization of PCR conditions. From the result of the PCR detection study, species-specific primers were selected under annealing temperatures ranging from 55$^{\circ}C$ to 61$^{\circ}C$, and template DNA levels ranging from 10 pg to 100 ng.
In July, 1963, a virulent outbreak of late blight in the potato field of Daekwanlyung area was studied and it was known as epidemics. Two stocks are $T_1$ and $T_1$ of Phytophthora infestans (Mont.) De Bary which isolated from Irish Cobbler were inoculated at field and green house respectively by cutted leaves method. Two strains have been distinguished in reactions to leaves: $T_1$: Irish Cobbler (r), Morin No. 1 (r) have shown infectivity of disease and Kennebec ($R_1$), 1512-C(16) ($R_2$), Pentland Ace ($R_3$) and Hokkai No. 17 ($R_4$) have not shown infectivity of disease; $T_1$: Irish Cobbler (r), Norin No. I (r) and Kennebec ($R_1$) have shown infectivity of disease and 1512-C (16) ($R_2$), Pentland Ace ($R_3$) and Hokkai No. 17($R_4$) have not shown infectivity of disease. Both are the first record of race O and race 1 of Phytophthora infestans (Mont.) De Bary in Korea.
Potato cv. 'Haryeong' was bred with high solids, resistance to late blight and good culinary quality. It has been registered as new potato variety in 2005. Tuber necrosis symptoms such as severe superficial necrosis, raised surface lesions and ringed necrotic areas were found in tubers of 'Haryeong' during storage of seed potatoes in 2010. Potato virus Y (PVY) was detected from these symptomatic tubers by the analysis of RT-PCR using a primer set specific to coat protein gene of PVY. The nucleotide sequence of RT-PCR product ($PVY^{Hkr}$) was determined and compared to those of other strains, such as $PVY^{Kor}$, $PVY^N$, $PVY^{NTN}$, $PVY^O$, and $PVY^C$ registered in GeneBank. The result showed that $PVY^{Hkr}$ was exactly the same as $PVY^{Kor}$, Korean isolate reported in 2005, except two nucleotides. To verify the PVY was responsible for the tuber necrosis symptoms shown in the tubers of 'Haryeong', a bioassay was done using two viruses (PVY and Potato leafroll virus) and five potato cultivars ('Haryeong', 'Superior', 'Atlantic', 'Dejima', and 'Chubaek'). As expected, the same necrosis symptom appeared in tubers of 'Haryeong' infected with PVY only during the storage period.
The tetraploid Solanum acaule is a wild potato species from Bolivia widely used for potato breeding because of its diverse attractive traits, including resistance to frost, late blight, potato virus X, potato virus Y, potato leafroll virus, potato spindle tuber viroid, and cyst nematode. However, the introgression of useful traits into cultivated potatoes via crossing has been limited by differences in endosperm balance number between species. Somatic fusion could be used to overcome sexual reproduction barriers and the development of molecular markers is essential to select proper fusion products. The chloroplast genome of S. acaule was sequenced using next-generation sequencing technology and specific markers for S. acaule were developed by comparing the obtained sequence with those of seven other Solanum species. The total length of the chloroplast genome is 155,570 bp, and 158 genes were annotated. Structure and gene content were very similar to other Solanum species and maximum likelihood phylogenetic analysis with 12 other species belonging to the Solanaceae family revealed that S. acaule is very closely related to other Solanum species. Sequence alignment with the chloroplast genome of seven other Solanum species revealed four InDels and 79 SNPs specific to S. acaule. Based on these InDel and SNP regions, one SCAR marker and one CAPS marker were developed to discriminate S. acaule from other Solanum species. These results will aid in exploring evolutionary aspects of Solanum species and accelerating potato breeding using S. acaule.
Climate in the year of 2000 was characterized as a long severe drought in tile spring, unusually high and low temperature in summer, two times of typoons, and floods by heavy rains in fall. Rice leaf and panicle blast and bacterial grain rot occurred severely comparing with 1999 and Bipolaris leaf spot spread over tile country. Phytophthora blight and anthracnose in red-pepper became epidemic especially in the late season causing severe yield losses. Tomato fusaruim wilt, CGMMV, powdery mildew, and sudden wilt syndrom of cucurbits and strawberry powdery mildew were also severe in 2000. In garlic, sclerotium rot occurred severely mainly due to the frequent rainfalls in planting time and much snowfalls in 1999's winter. Spring potato had severe infection of viruses due to a long spring drought, and fall potato had high incidence of bacterial soft rot and bacterial wilt due to fall floods by heavy rains. In sweet potato fusarium wilt was the most severe as in other year. Disease incidence of apple and pear trees was rotatively mild compared with previous years. In wheat and barley, Gibberella petch rarely occurred because of spring drought.

In early spring, water-soaked lesions appeared on the petals and leaves of gaenari (Forsythia koreana), and the tissues were necrotic and dry. Cankers appeared on the infected branches around late spring and the above part of a branch withered and died. However, it was very rare that the base of the cankered-branch died. The identical fungi were isolated from the lesions on various tissues, and they grew with white colonies on potato dextrose agar medium. The fungus grew most actively at 23℃ and produced many sclerotia of various sizes. In a pathogenicity assay in which mycelial and sclerotial suspensions were inoculated on each organ of forsythia, it was found that the pathogen infects the flower only, but not the leaves or branches. Symptoms on the flowers spread to the next leaves and branches over time and the infected branches were eventually withered. To identify the isolates, DNA sequences of four phylogenetic markers including ITS, LSU, Tub2, and CAL were analyzed and all isolates were identified as a species in the genus Septotinia. This is not only the first report of gaenari (forsythia) shoot blight caused by the fungus Septotinia sp., but also the first report on the genus Septotinia as a plant pathogen in Korea.
