STZ으로 유도된 당뇨 마우스에서 오디열매추출물의 식후 고혈당 완화 효과 (Alleviating Effects of Mulberry Fruit Extract on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Mice)

  • 식후 고혈당은 제2형 당뇨병 초기 진단과 항당뇨병의 중요한 바이오마커 중 하나이다. α-글루코시다아제 억제제는 소장에서 탄수화물 소화의 속도를 방해함으로써 식후 고혈당을 조절한다. 본 연구에서는 오디열매추출물이 α-글루코시다아제와 α-아밀라아제에 미치는 억제효과 및 스트렙토조토신(STZ)이 유발하는 당뇨병 생쥐의 식후고혈당에 미치는 완화 효과를 조사하였다. α-글루코시다아제와 α-아밀라아제에 대한 오디열매추출물(MFE)의 IC50 값은 각각 0.16과 0.14 mg/ml의 결과값을 나타내어, 양성대조군인 acarbose보다 더 효과적이었다. STZ으로 유발된 당뇨병 흰쥐의 식후 혈당 수치는 정상 대조군에 비해 오디열매추출물에서 유의적으로 더 낮았다. 더욱이, 오디열매추출물 투여는 당뇨병 흰쥐에서 포도당 반응에 대한 곡선하면적 감소와 관련이 있었다. 결론적으로, 오디열매추출물은 α-글루코시다아제와 α-아밀라아제 활성의 강력한 억제제이며 식후 고혈당을 완화할 수 있다는 내용이다. 따라서 오디열매추출물은 당뇨병 치료제 및 기능성식품의 소재로 가치가 높다고 사료된다.

작약(Paeoniae radix) 추출물의 식후 과혈당 억제작용 (Inhibitory Effect of Extracts from Paeoniae radix on Postprandial Hyperglycemia)

  • 본 연구에서는 작약 추출물의 식후 과혈당 억제작용을 조사하였다. 작약 유기용매(hexane, ethyl acetate, butanol, water) 추출물들을 high performance liquid chromatogrphy를 이용하여 분획한 분획물(fraction)들으 $\alpha$-glucosidase(EC 3.2.1.20) 저해제를 탐색하였다. 작약 ethyl acetate 추출물의 11, 12, 18, 19번 분획물들은 20 $\mu\textrm{g}$/mL에서 각각 85%, 84%, 77%, 77%의 강력한 저해 활성을 나타내었으며, 작약 ethyl acetate 추출물의 10~19번 분획을 경구투여한 in vivo당 (maltose) 부하 실험에서도 양성대조군과 비교하여 유의성 있게 22%의 혈당을 낮추었다. 따라서 본 연구의 in vitro와 in vivo 실험을 통하여 확인 된 $\alpha$-glucosidase 저해 활성을 갖는 작약 분획물들은 식후의 혈당상승 작용을 억제하는 새로운 nutraceutical 소재와 항당뇨 신약개발을 위한 탐색자원으로서 매우 가치있는 자원으로 기대된다.

Effect of Paeonia lactiflora Extracts on ${\alpha}-Glucosidase$

  • This study was carried out to investigate inhibitory effect of extracts from the root of Paeonia lactiflora on postprandial hyperglycemia. Organic solvent (hexane, ethyl acetate, butanol, aqueous) extracts from the crude drug were fractionated by high performance liquid chromatography. These fractions were examined to evaluate ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) inhibition by microplate colorimetric assay. Among the fractions examined, the ethyl acetate fraction from the roots of Paeonia lactiflora showed potent inhibitory effects on ${\alpha}-glucosidase$. Therefore, further fractionation of the fraction was carried out to isolate the active principles. Finally, we isolated and Purified 1,2,3,4,6-penta-O-galloyl-beta-D-glucose (PGG) as a active principle by activity-guided fractionation. These results suggest that the extract from the root of Paeonia lactiflora can be used as a new nutraceutial for inhibition on postprandial hyperglycemia and PGG might be a candidate for developing an ${\alpha}-glucosidase$ inhibitor.

Growth, Feed Utilization and Blood Metabolic Responses to Different Amylose-amylopectin Ratio Fed Diets in Tilapia (Oreochromis niloticus)

  • A feeding trial was conducted in tilapia to determine the growth performance, nutrient digestibility, digestive enzymes, and postprandial blood metabolites in response to different dietary amylose-amylopectin ratios. Five isonitrogenous and isolipidic diets containing an equal starch level with different amylose-amylopectin ratios of 0.11 (diet 1), 0.24 (diet 2), 0.47 (diet 3), 0.76 (diet 4) and 0.98 (diet 5) were formulated using high-amylose corn starch (as the amylose source) and waxy rice (as the amylopectin source). Each diet was hand-fed to six tanks of 15 fish each, three times a day over a 6-wk period. After the growth trial, a postprandial blood metabolic test was carried out. Fish fed diet 2 exhibited the highest percent weight gain and feed efficiency and protein efficiency ratio, whereas fish fed with diet 5 showed the lowest growth and feed utilization among treatments. The digestibility for starch in fish fed diet 1 and 2 was higher than those in fish fed with other diets (p<0.05). The highest activities for protease, lipase and amylase were found in fish fed the diet 2, diet 1, and diet 1 respectively among dietary treatments, while the lowest values for these indexes were observed in fish fed the diet 3, diet 5 and diet 4, respectively. The liver glycogen concentrations in fish fed diets 4 and 5 were found higher than in fish fed other diets (p<0.05). The feeding rate, hepatosomatic index, condition factor, and plasma parmeters (glucose, triglyceride, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol) did not differ across treatments. In terms of postprandial blood responses, peak blood glucose and triglycerides were lower after 3 or 6 h in the fish fed with diets 3-5 than in the fish fed diet 1, but delayed peak blood total amino acid time was observed in fish fed with the diets 1 or 2. The lowest peak values for each of the three blood metabolites were observed in fish fed diet 5. The results indicate that high-dietary amylose-amylopectin ratio could compromise growth, but help in reducing the blood glucose stress on fish caused by postprandial starch load.

당뇨 모델을 이용한 벼메뚜기(O. Mistshenk) 추출물의 식후 고혈당 완화 효과 (Oxya Chinensis Sinuosa Mishchenko Extract: Potent Glycosidase Inhibitor Alleviates Postprandial Hyperglycemia in Diabetic Mice)

  • 벼메뚜기는 메뚜기과에 속하는 불완전 변태 곤충으로, 국내에서 오랫동안 식용으로 이용되어 왔다. 현재까지 벼메뚜기의 항산화 및 항염증 효과 등의 기능들이 연구되었으나, 탄수화물 소화 효소나 식후 혈당 수치에 미치는 영향에 관한 연구는 부족한 실정이다. 이에 본 연구는 벼메뚜기 에탄올 추출물(OEE)와 물 추출물(OWE)이 탄수화물 소화 효소를 저해하고, streptozotocin (STZ)으로 유도된 당뇨병 마우스에서 식후 고혈당을 강하시키는 효과에 대해 조사하였다. 그 결과 OEE에 의한 α-glucosidase와 α-amylase 저해 효과가 OWE 보다 더 효과적이었다. 당뇨병 마우스에 전분(2 g/kg)을 투여한 후의 혈당 증가는 15, 30, 60, 120분에 각각 406.50, 431.50, 473.50 and 460.00 mg/dl로 나타났고, 전분(2 g/kg)과 OEE 추출물(300 mg/kg)을 투여한 후 혈당 증가는 15, 30, 60, 120분에 각각 327.00, 361.00, 358.67 and 291.00 mg/dl로 나타나, OEE 추출물 투여군이 대조군에 비해 식후 혈당 강하가 효과적으로 나타남을 알 수 있었다. 이러한 결과는 벼메뚜기 추출물이 α-glucosidase와 α-amylas를 저해함으로써 식후 고혈당을 완화시키고, 특히 벼메뚜기 에탄올 추출물(OEE)이 벼메뚜기 물 추출물(OWE) 보다 식후 고혈당을 완화시키는데 더욱 효과가 있는 것으로 나타났다. 따라서 OEE가 탄수화물 소화 효소 저해 효과로 식후 고혈당을 완화시키는 유용한 천연 기능성 식품이 될 것으로 사료된다.

제주산 녹차, 발효차 추출물과 바나듐의 식후 혈당강하 및 항고혈압 기능에 미치는 In Vitro 효과 (Inhibitory Effect of Jeju Tea Extracts and Vanadate on Postprandial Hyperglycemia and Hypertension, and In Vitro Study)

  • 현대인의 기호식품인 차를 이용하여 비 발효차인 녹차와 3가지 발효차로 구분하여 식후 고혈당의 지표가 되는 알파글루코시데이스(α-glucosidase) 활성과 고혈압활성의 지표로 사용되는 안지오텐신전환효소(angiotensin converting enzyme, ACE)의 억제작용을 확인하였다. α-glucosidase 억제 활성의 상승효과를 알아보기 위해 녹차추출물에 인슐린과 유사기능이 있다고 밝혀진 바나듐을 미량(50 ㎍/mL)으로 혼합하여 α-glucosidase 활성 억제를 위한 상승효과 여부를 조사하였다. 또한 녹차의 기능성물질로 알려진 에피갈로카테킨 갈레이트(epigallocatechin gallate, EGCG)의 농도와 카페인(caffeine)의 농도를 측정하여 녹차와 발효차 효능을 분석하고자 하였다. 녹차와 발효차 추출물로부터 식후 혈당을 증가시키는 α-glucosidase 활성이 크게 억제되어 차의 식후 혈당저해 효과가 있음을 확인하였으며 차 추출물 중에서도 20% 발효차가 가장 우수한 기능을 보였다. 그러나 차 추출물만을 단독으로 처리한 경우와 바나듐과 함께 처리한 경우 서로 다른 결과를 보였다. α-glucosidase 저해효과는 저 농도(50 ㎍/mL)의 녹차와 바나듐 혼합 처리에서 단독 처리에 비해 저해활성이 2배나 증가되어 녹차가 바나듐과 가장 큰 상승효과를 보임을 알 수 있었다. ACE 저해활성은 모든 차 추출물에서 확인되었으나 녹차에서 가장 높게 조사되어 녹차가 다른 차에 비해 가장 높게 측정된 EGCG 농도의 효과로 생각되며 발효차의 식후 혈당강하 및 ACE 저해효과는 EGCG 외에 또 다른 기능성물질이 관여한 것임을 알 수 있었다.

Effects of soybean supplementation on blood glucose, plasma lipid levels, and erythrocyte antioxidant enzyme activity in type 2 diabetes mellitus patients

  • The purpose of this study was to investigate the effect of soybean on blood glucose and lipid concentrations, and antioxidant enzyme activity in type 2 diabetes mellitus(DM) patients. We divided patients into two groups and fed them, respectively, a basal diet(control group) and a basal diet with 69 g/d of soybean(soybean group) for 4 weeks. Pills with roasted soybean powder were provided to the soybean supplementation group three times a day. Macronutrients intake except dietary fiber was similar between the two groups. No significant differences were observed in dietary intakes or body weight before and after the supplementation. Energy composition ratio of C:F:P was 65:19:16 in the control group, 64:20:16 in the soybean group. The blood parameters of subjects before supplementation, such as fasting blood glucose, postprandial glucose level, total cholesterol, triglyceride, LDL-cholesterol and $HbA_1C$ were not different between the two groups. After supplementation, fasting blood glucose(p<0.001), postprandial glucose level(p<0.001) and serum triglyceride level(p<0.05) were significantly reduced in the soybean group in comparison with the control group. The total cholesterol level was not significantly different between the control and the supplemented group after 4 weeks of treatment. TBARS levels of the soybean group were not significantly different from those of the control group. The activities of catalase(p<0.01) and glutathione peroxidase(p<0.05) were significantly higher in the soybean group compared to the control group. The results of this study suggest that soybean supplementation would be helpful to control blood glucose and serum lipid in diabetic patients. Also, soybean showed an antioxidant activity that may contribute to enhance the effect of antioxidant defense. This activity contributes to protection against oxidative damage in type 2 DM patients. Soybean may have potential use in the disease management of patients with DM.

The Inhibitory Activity of Polygonum Multiflorum Thunberg and its Effect on Postprandial Hyperglycemia in Streptozotocin-induced Diabetic Rats

  • To keep blood glucose levels as close to normal as possible is the major goal of diabetes mellitus treatment $\alpha$-Glucosidase is the enzyme that digests die1my carbohydrate and inhibition of this enzyme may suppress postprandial hyperglycemia. The methanol extract of Polygonum multiflorum Thunberg was tested for inhibitoty activity against $\alpha$-glucosidase in vitro and in vivo. Polygonum multiflorum Thunberg extract inhibited yeast $\alpha$-glucosidase activity in a concentration-dependent manner. Polygonum multiflorum Thunberg showed an $IC_{50}$ value of 0.48 mg/mL. The ability of Polygonum multiflorum Thunberg extract to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without the methanol extract of Polygonum multiflorum Thunberg extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast A single oral dose of Polygonum multiflorum Thunberg extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that Polygonum multiflorum Thunberg may have an antihyperglycemic effect by inhibiting $\alpha$-glucosidase activity in the animal model of diabetes mellitus.

Acarbose 제제의 약력학적 평가 및 생물학적동등성 시험법에 대한 연구 (Methods for Pharmacodynamic Analysis and Proposed Protocols for Bioequivalence Study of Acarbose)

  • Arcabose is a competitive inhibitor of the intestinal ${\alpha}$-glucosidases and reduces the postprandial digestion and absorption of carbohydrate and disaccharides. Due to its negligible oral absorption, measuring drug concentration in the plasma is impractical. Thus, the common pharmacokinetic study is not available to determine the bioequivalence of the generic acarbose preparations. The aim of this study is the establishment of pharmacodynamic assessment method for the bioequivalence test of the generic acarbose preparations. Placebo-controlled cross-over ($3{\times}3$) clinical study was conducted in 23 healthy volunteers. Volunteers received a single oral dose of placebo, reference drug ($Glucoby^{(R)}$ 100 mg, Lot # D043) or test drug ($Glucoby^{(R)}$ 100 mg, Lot # E005) just before breakfast, then blood samples for evaluation of serum glucose and insulin levels were taken during for 4 hours. $C_{max},\;AUC_{0-2},\;AUC_{0-4},\;{\Delta}C_{max},\;{\Delta}AUC_{0-2}\;and\;{\Delta}AUC_{0-4}$ of the postprandial plasma glucose level significantly decreased when a single dose of acarbose 100 mg preparations was administered. However, any significant difference was not detected between the groups taken the reference drug and the test drug. These results proposed that the pharmacodynamic protocols of this study is suitable to use for bioequivalence test of acarbose preparations. On the basis of the results of this study and the data of literature on this subject, the standard protocols of bioequivalence study of acarbose preparation are proposed.
