기주 식물체 내에서 식물바이러스의 증식과 이동 여부는 바이러스 게놈과 기주 간의 상호작용에 의해 결정된다. 바이러스는 기주 내에서 바이러스가 증식하고 이동하기 위해서는 기주의 요소들을 이용해야 하며, 이러한 기주 요소들은 바이러스의 기주내 침입(entry),바이러스 유전자의 발현, 그리고 바이러스 입자형성(virion assembly) 등 모든 과정에서 직접적으로 관여를 하거나, 또는 기주 단백질 발현과 저항성을 조절하여 바이러스 증식에 간접적으로 관여를 한다. 기주 요소들과 상호작용을 통해서 바이러스 증식에 관여함으로써, 기주 특이성 및 바이러스의 병 발생에 관여를 할 것으로 보고 있다.
Potato virus X (PVX) is a flexuous rod-shaped virus containing a single plus-strand RNA. Viral RNA synthesis is precisely regulated by regulatory viral sequences and by viral and/or host proteins. RNA sequence element as well as stable RNA stem-loop structure in the 5' end of the genome affect accumulation of genomic RNA and subgenomic RNA (sgRNA). The putative sgRNA promoter regions upstream of the PVX triple gene block (TB) and coat protein (CP) gene were critical for both TB and CP sgRNA accumulation. Mutations that disrupted complementarity between a region at the 5' end of the genomic RNA and the sequences located upstream of each sgRNA initiation site is important for PVX RNA accumulation. Compensatory mutations that restore complementarity restored sgRNA accumulation levels. However, the extent of reductions in RNA levels did not directly correlate with the degree of complementarity, suggesting that the sequences of these elements are also important. Gel-retardation assays showed that the 5' end of the positive-strand RNA formed an RNA-protein complex with cellular proteins, suggesting possible involvement of cellular proteins for PVX replication. Future studies on cellular protein binding to the PVX RNA and their role in virus replication will bring a fresh understanding of PVX RNA replication.
Turnip yellow mosaic virus (TYMV) is a spherical plant virus that has a single 6.3 kb positive strand RNA as a genome. In this study, RNA1 sequence of Flock house virus (FHV) was inserted into the TYMV genome to test whether TYMV can accommodate and express another viral entity. In the resulting construct, designated TY-FHV, the FHV RNA1 sequence was expressed as a TYMV subgenomic RNA. Northern analysis of the Nicotiana benthamiana leaves agroinfiltrated with the TY-FHV showed that both genomic and subgenomic FHV RNAs were abundantly produced. This indicates that the FHV RNA1 sequence was correctly expressed and translated to produce a functional FHV replicase. Although these FHV RNAs were not encapsidated, the FHV RNA having a TYMV CP sequence at the 3'-end was efficiently encapsidated. When an eGFP gene was inserted into the B2 ORF of the FHV sequence, a fusion protein of B2-eGFP was produced as expected.
Turnip yellow mosaic virus (TYMV) is a positive strand RNA virus that infects mainly Cruciferae plants. In this study, the TYMV genome was modified by inserting an extra subgenomic RNA promoter and a multiple cloning site. This modified TYMV was introduced into Nicotiana benthamiana using a Agrobacterium-mediated T-DNA transfer system (agroinfiltration). When a gene encoding $\beta$-glucuronidase or green fluorescent protein was expressed using this modified TYMV as a vector, replication of the recombinant viruses, especially the virus containing $\beta$-glucuronidase gene, was severely inhibited. The suppression of replication was reduced by co-expression of viral silencing suppressor genes, such as tombusviral p19, closteroviral p21 or potyviral HC-Pro. As expected, two subgenomic RNAs were produced from the recombinant TYMV, where the larger one contained the foreign gene. An RNase protection assay revealed that the recombinant subgenomic RNA was encapsidated as efficiently as the genuine subgenomic RNA.
Interplay between Cymbidium mosaic virus (CymMV)/Odontoglossum ringspot virus (ORSV) and its host plant Phalaenopsis equestris remain largely unknown, which led to deficiency of effective measures to control disease of P. equestris caused by infecting viruses. In this study, for the first time, we characterized viral small interfering RNAs (vsiRNAs) profiles in P. equestris co-infected with CymMV and ORSV through small RNA sequencing technology. CymMV and ORSV small interfering RNAs (siRNAs) demonstrated several general and specific/new characteristics. vsiRNAs, with A/U bias at the first nucleotide, were predominantly 21-nt long and they were derived predominantly (90%) from viral positive-strand RNA. 21-nt siRNA duplexes with 0-nt overhangs were the most abundant 21-nt duplexes, followed by 2-nt overhangs and then 1-nt overhangs 21-nt duplexes in infected P. equestris. Continuous but heterogeneous distribution and secondary structures prediction implied that vsiRNAs originate predominantly by direct Dicer-like enzymes cleavage of imperfect duplexes in the most folded regions of the positive strand of both viruses RNA molecular. Furthermore, we totally predicted 54 target genes by vsiRNAs with psRNATarget server, including disease/stress response-related genes, RNA interference core components, cytoskeleton-related genes, photosynthesis or energy supply related genes. Gene Ontology classification showed that a majority of the predicted targets were related to cellular components and cellular processes and performed a certain function. All target genes were down-regulated with different degree by vsiRNAs as shown by real-time reverse transcription polymerase chain reaction. Taken together, CymMV and ORSV siRNAs played important roles in interplay with P. equestris by down modulating the expression levels of endogenous genes in host plant.
Replication of positive-strand RNA virus is mediated by a virus-encoded RNA-dependent RNA polymerase (RdRp). To study the replication of Ectropis obliqua virus (EoV), a newly identified insect virus belonging to the family Iflaviradae, we expressed the RNA polymerase domain in Escherichia coli and purified it on a Ni-chelating HisTrap affinity column. It is demonstrated that EoV RdRp initiated RNA synthesis in a primer and poly (A)-dependent manner in vitro. Furthermore, the effect of primer concentration, temperature, metal ions ($Mg^{2+}$, $Mn^{2+}$, and $K^+$) on enzymatic activity were determined. Our study represented a first step towards understanding the mechanism of EoV replication.
폴리오바이러스는 바이러스들 중에서도 특히 커기가 작은 바이러스로서 피막(coat)을 둘러싸는 막(envelop) 이 없다. 폴리오바이러스는 (+) 가닥의 단일 RNA 게놈을 갖는데 이는 한 개의 해독판 (open reading frame)을 이용하여 다단백전구체를 만든 후 바이러스 자체의 단백질분해효소에 의해 스스로 잘라져서 궁극적으로느 특이한 기능을 갖는 여러개의 단백질이 된다. P1 다단백질전구체로부터 만들어지는 단백질들은 바이러스의 피막을 구성하는 성분이다. 단백질분해효소인 2A에 의한 최초의 절단은 구조단백질 P1 전구체와 구조단백질이 아닌 P2-P3간을 분리시켜준다. 단백질분해효소 2A는 진핵세포 판독개시인자(translation initiation factor) 4F의 한 subunit인 숙주단백질 p220의 절단에 간접으로 참여한다. 이 단백질의 절단은 캡(cap)에 의존하는 숙주세포의 대부분의 판독을 차단하게 되며 이는 판독에 사용되는 숙주세포의 모든 기구들을 캡에 의존하지 않는 폴리오바이러스 NA 특유의 판독을 위해 전적으로 사용할 수 있게 해준다. 2B, 2C, 2BC 단백질의 기능에 대해서는 많이 알려져 있지 않다. 2B, 2C, 2BC와 3CD 단백질들은 바이러스로 인해 만들어지는 소낭(vesicle)의 복제복합체에 함유되어 있으므로 바이러스의 RNA 복제시 중요한 역할을 함을 암시해준다. 새로이 만들어진 모든 바이러스 RNA는 VPg와 공유결합으로 연결되어 있다. VPg는 3AB로부터 만들어진 아미노산 22개 짜리의 폴리펩타이드이다. 3C와 3CD는 단백질분해소로 다단백질 전구체의 대부분의 절단부위를 잘라준다. 3C단백질은 숙주의 전사인자를 불활성화 시킴으로써 RNA polymer II와 III에 의한 전사를 저해한다. 3D는 RNA의존선RNA 중합효소이다. 폴리오바이러스는 (+)가닥 RNA 바이러스의 일반적인 복제양식을 따른다. 즉 (+) 가닥 RNA는 이와 상보적인 (-)가닥 RNA로 전사되고 이는 다시 (+)가닥 RNA의 합성을 위한 주형으로 사용된다. 폴리오바이러스의 RNA 합성은 세포내막에서 일어나는 데 RNA 복제에 요구되는 주형 RNA와 이때 필요한 단백질들이 어떤 방법으로 세포내막에서 모일 수 있는지는 아직 밝혀진 것이 적다. 바이러스입자의 형성은 세포막의 RNA 복제가 들어가는 데 피막단백질이 (+)가닥 RNA을 인식하는 표지 즉 packaging singal에 대해서는 거의 알려져 있지 않다. 폴리오바이러스 감염 후 첫 바이러스입자가 만들어지기 까는 약 6시간이 소요된다.
Cowpea chlorotic mottle virus (CCMV)는 Group IV positive sense single strand RNA virus, Bromoviridae과, Bromovirus속으로 분류하는 식물병원성 바이러스로, 강낭콩(Phaseolus vulgaris), 나비완두(Clitoria ternatea), 담배(Nicotiana tabaccum), 대두(Glycine max), 동부(Vigna unguiculata, Vigna siensis) 및 땅콩(Arachis hypogaea)이 국내로 수입될 경우, 검사를 수행하는 관리급 검역바이러스이다. 본 연구에서는, RT-PCR, nested PCR 및 유전자-삽입 양성대조구를 개발하여, CCMV를 현장에서 신속, 정확하게 진단할 수 있는 정밀검정 시스템을 구현하였다. 본 연구에서 개발한 방법은 지속적으로 현장에서 활용되어 식물검역에 기여할 것이라고 기대된다.
Turnip yellow mosaic virus (TYMV) is a positive strand RNA virus. We have modified TYMV coat protein (CP) by inserting a c-Myc epitope peptide at the N- or C-terminus of the CP, and have examined its effect on assembly. We introduced the recombinant CP constructs into Nicotiana benthamiana leaves by agroinfiltration. Examination of the leaf extracts by agarose gel electrophoresis and Western blot analysis showed that the CP modified at the N-terminus produced a band co-migrating with wild-type virions. With C-terminal modification, however, the detected bands moved faster than the wild-type virions. To further examine the effect, TYMV constructs producing the modified CPs were prepared. With N-terminal modification, viral RNAs were protected from RNase A. In contrast, the viral RNAs were not protected with C-terminal modification. Overall, the results suggest that virion assembly and RNA packaging occur properly when the N-terminus of CP is modified, but not when the C-terminus is modified.
Coxsackievirus B3 (CVB3) is the main cause of acute myocarditis and dilated cardiomyopathy. Plant extracts are considered as useful materials to develop new antiviral drugs. We had previously selected candidate plant extracts, which showed anti-inflammatory effects. We examined the antiviral effects by using a HeLa cell survival assay. Among these extracts, we chose the Amomi Cardamomi (Amomi) extract, which showed strong antiviral effect and preserved cell survival in CVB3 infection. We investigated the mechanisms underlying the ability of Amomi extract to inhibit CVB3 infection and replication. HeLa cells were infected by CVB3 with or without Amomi extract. Erk and Akt activities, and their correlation with virus replication were observed. Live virus titers in cell supernatants and viral positive- and negative-strand RNA amplification were measured. Amomi extract significantly increased HeLa cell survival in different concentrations ($100-10{\mu}g/ml$). CVB3 capsid protein VP1 expression (76%) and viral protease 2A-induced eIF4G1 cleavage (70%) were significantly decreased in Amomi extract ($100{\mu}g/ml$) treated cells. The levels of positive- (20%) and negative-strand (80%) RNA were dramatically decreased compared with the control, as revealed by reverse transcription-PCR. In addition, Amomi extract improved mice survival (51% vs 26%) and dramatically reduced heart inflammation in a CVB3-induced myocarditis mouse model. These results suggested that Amomi extract significantly inhibited Enterovirus replication and myocarditis damage. Amomi may be developed as a therapeutic drug for Enterovirus.
