Antioxidant effect of ethanol extract from Poria cocos depending on cultivation methods (재배방법을 달리한 복령 에탄올추출물의 항산화 효과)

  • Objectives : Poria cocos Wolf has been widely used in Korean medicine as a medicinal fungus. In this study, we investigated that comparative anti-oxidant effects of ethanol extract from wild Poria cocos (WP) and plastic bag-cultivated Poria cocos (PBP).Methods : Total polyphenol and flavonoid contents in WP and PBP extract were monitored. And DPPH, ABTS, hydroxyl (·OH) free radical scavenging capacity, reducing power and xanthine oxidase inhibition activities of them were determined at 5, 1, 0.5 mg/ml concentrations.Results : Higher total polyphenol contents were found in the PBP extract (52.07±0.6 mg/TAEg) than in the WP extract (28.44±0.26 mg/TAEg). The flavonoid contents in WP and PBP extract were 17.29±0.30 mg/ RUEg, 21.36±0.40 mg/ RUEg, respectively. Also, DPPH, ABTS and ·OH free radical scavenging capacity of PBP extract in treated concentrations (5, 1, 0.5 mg/ml) significantly increased compared to those of WP-treated group. In particular, DPPH, ABTS free radical scavenging capacity and reducing power of PBP extract at 5 mg/ml concentration were similar to positive control (BHA or vit. C). Xanthine oxidase (XO) inhibition rate in both extract increased dose dependently. But it was significantly increased in PBP-treated group, only at 5 mg/ml, compared to those of WP-treated group. Then, their inhibition rate by PBP was similar to positive control (BHA).Conclusions : These results suggest that PBP extract is superior to WP extract in anti-oxidant capacity thus PBP can be applied in variable antioxidative products as a substitute for WP.

  • Extract of drugs in commerce was administered to mice for 3 days and its fatigue effects were compared with those of P. ginseng extract and A. sessiliflorum extract. Mice were used for these experiments divided into ethyl alcohol and water extract of P. cocos, S. japonica, P. multiflorum, L. chinense, A. cochinchinensis, S. indicum, N. nucifera were given ordinary dose orally. For antifatigue test, swimming performance were determined after administration of the extract The group of P. cocos gave high improvement of the swimming performance in ethyl alcohol extract group. And the group of S. japonica an improvement was also observed in water extract group.

  • The present genus, once applied as an all-round taxon on resupinate polypores, was considered on account of its common occurrence throughout the world and outstanding wood-rotting nature. In Korea two species, P. vaporaria(but even this seems misjudged) and P. cocos have been known. The authors used to examine tentatively the materials formerly preserved in dried state. Among about 40 collections half of them was recognized as P. cocos and 4 unrecorded species, P. versipora, P. subacida, P. medulla-panis, and P. eupora, which are newly registered to Korea in this article.

Effect of Rice Fermented using Poria cocos (a Wood-decay Fungus) Mycelium on Fermentation of Doenjang (Soybean Paste) (복령(Poria cocos) 균사체 발효쌀의 첨가가 된장의 품질에 미치는 영향)

  • We investigated changes in doenjang quality upon addition of rice fermented with Poria cocos mycelium. Sensory evaluation showed that rice addition to 15% (w/w) was optimal. The content of free amino acids was 1,899.17 mg% in doenjang fermented for 90 days with P. cocos mycelium. The rice contained seven essential amino acids: leucine, valine, phenylalanine, isoleucine, lysine, threonine, and methionine. The levels of essential amino acids increased after fermentation with P. cocos mycelium. Vitamin A (7.47 RE/100 g), Vitamin D (0.45 mg/100 g), and Vitamin E (5.73 mg/100 g) were detected in the experimental preparation but not in the control. In terms of electron-donating ability, the highest scores were 73.8% (in a water extract) and 76% (in an ethanolic extract) of doenjang fermented with P. cocos mycelium. Nitrite-scavenging ability was higher in the ethanolic extract than in the water extract of rice fermented with P. cocos mycelium.

  • The qualitative and quantitative analysis of fatty acids in the Korean White Poria cocos(Schw.) Wolf were performed. Dry powder of sample was extracted with chloroform-methanol(2 : 1, v/v) mixed solvent. The extract was saponified and methylated, and then analyzed by gas chromatography. Total 24 peaks were observed in the gas chromatogrom and 5 peaks i.e, $C_{8:0}\:;\:caprylic\:acid$, $C_{11:0}\:;\:undecanoic\:acid$, $C_{12:0}\:;\:lauric\:acid$. $C_{12:1}\:;\:dodecenoic\:acid$ and $C_{16:0}\:;\:palmitic\:acid$ were identified. Major components were $C_{8:0}\:and\:C_{12:1}$ were determined as $6.657\:{\mu}g/g\:and\:10.176{\mu}g/g$ respectively.

Cytotoxicity and DNA Topoisomerases Inhibitory Activity of Constituents from the Sclerotium of Poria cocos

  • The bioactivity-guided fractionation of the methylene chloride extract of the sclerotium of Poria cocos led to the isolation of (S)-(+)-turmerone (1), ergosterol peroxide (2), polyporenic acid C (3), dehydropachymic acid (4), pachymic acid (5), and tumulosic acid (6). Compounds 4-6 exhibited moderate cytotoxicities, with $IC_{50}$ values of 20.5, 29.1, and $10.4{\;}\mu\textrm{m}$, respectively, against a human colon carcinoma cell line. However, 3-6 not only showed inhibitory activities as potent as etoposide used as a positive control on DNA topoisomerase II (36.1, 36.2, 43.9 and 66.7% inhibition at a concentration of $20{\;}\mu\textrm{m}$, respectively), but also inhibition of DNA topoisomerase I (55.8, 60.7, 43.5, and 83.3% inhibition at a concentration of $100{\;}\mu\textrm{m}$, respec-tively).

  • Total ethanol extract of chinese drugs was administered to mice for 4 weeks (small dosage group) and 3 days (large dosage group), and its antifatigue effects and hepatonic activities were compared with those of P. ginseng and A. sessiliflorum. 1. In the long-term administration of extracts, the swimming performance peaked at 2 weeks, and the recovery effects on the intoxicated liver appeared to be significant in the extracts of P. officinale, P. cocos and A. cochinchinensis among tested chinese drugs. 2. The large dose administration of extracts gave high improvement of recovery effect on the intoxicated liver.

  • The antibacterial activity of Poria cocos against the human pathogenic bacteria and general bacteria was observed. The results obtained were as follows; 1. There wasn't any anti­bacterial activity at the Poria cocos powders, pieces and its extracts in every different kinds and places of origin. 2. Hyphae of Poria cocos in every different kinds and places of origin had the antibacterial activity against four Gram positive bacteria, i.e., Staphylococcus aureus, S. epidermidis, Bacillus subtilis and Streptococcus faecalis, on the other hand, hadn't the antibacterial activity against six Gram negative bacteria, Salmonella typhi, Shigella dysenteriae, Pseudomonas aeruginosa, Escherichia coli, Proteus vulgaria and Klebsiella pneumoniae. 3. The average inhibitory zone was 13.6mm for S. aureus, 14.7mm for S. epidermidis, 12.7mm for B. subtilis, and 13.2mm for S. faecalis at the concentration of $335{\mu}g$ of hyphae of Poria cocos.

Tyrosinase Inhibition Activity and Antioxidant Capacity by Fermented Products of Some Medicinal Plants (한방 생약재 발효액의 항산화 활성 및 tyrosinase 저해 활성)

  • The effects of fermented products from 40 medicinal herbals commonly available in Korea were examined according to concentrations of polyphenolic compound and kojic acid, and the activities of DPPH ($\alpha,\alpha$'-diphenyl-$\beta$-picrylhydrazyl) free radical scavenging and tyrosinase. The polyphenolic compound concentrations were 0.24 by Corydalis turtschaminovill ~ 11.42% (dry matter basis) by Syringa velutina in the extracts and 0.18 by Poria cocos ~ 12.27% by S. velutina in the fermented products. Kojic acid concentrations were 0.02 by Poria cocos Sclerotium ~ 9.67 mM by S. velutina in the extracts and 0.33 by P. cocos ~ 10.32 mM by S. velutina in the fermented products. Syringa velutina contained the highest polyphenolic compound and kojic acid concentrations, which were higher in the fermented product than in the extract. Higher DPPH free radical scavenging activity (>60%) was observed in the extracts of A. sessiliflorum, Citrus nobillis, and Angelica gigas and the fermented product of A. sessiliflorum compared to the other medicinal plants. Higher tyrosinase inhibition activity (>50%) was observed in the extracts of Morus alba, Glycyrrhiza glabra, and Rubus coreanus and the fermented products of G. glabra, Cnidium officinale, and S. velutina. Based on the above results, G. glabra, C. officinale, and S. velutina possessed high tyrosinase-inhibitive activities and kojic acid concentrations, which could be definitely enhanced by the fermentation of Phenillus linteus mycelium.

Effects of Poria cocos and Panax ginseng Extracts on Hemogram of Sarcoma-180 Mouse (복령 및 인삼추출물이 Sarcoma -180 Mouse의 혈액상(血液象)에 미치는 영향(影響))

  • This study was made to observe changes in hemograms of the Sarcoma-180 mouse treated with extracts of Poria cocos and Panax ginseng. The life span of the Sarcoma-180 mouse administered with the mixture of the extract of Panax ginseng with the extract of P. cocos and the extract of P. cocos was extended more than the non-treated group. In the body weight of the Sarcoma-180 mouse, the non-treated group was increased more than the normal group and the groups administered with the mixture of ginseng extract with hoelen extract and hoelen extract. In the numbers of white blood cell especially lymphocyte, the groups administered with the mixture of ginseng extract with hoelen extract and hoelen extract were similar to the normal group, but the non-treated group was decreased. The Hematocrit, mean corpuscular volume and mean corpuscular hemoglobin in the mice infected with Sarcoma-180 were not significantly different between the group treated with extracts of them and the non-treated group. In the distributions and sizes of white blood cell and platelet in the mice, the group administered with the Hoelen extract was similar to the normal group comparing to the non-treated group.
