Objectives : The purpose of this study was verification of the anti-inflammation and anti-oxidant effect of Cheongungdajosan-gamibang extract (CG) in mouse macrophage, RAW 264.7 cells. Methods : We have basically using LPS-stimulated RAW 264.7 cells. The cell toxicity was determined by MTT assay. To evaluate the anti-inflammatory effect of Cheongungdajosan-gamibang, amount of nitric oxide(NO) was measured using the NO detection kit and the IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). Also, free radical scavenging assay has tested for DPPH and ABTS radical activity as well as the contents of total polyphenol. Results : In this study, 96.6% or higher cell viability was observed in all tested groups from 1, 10, $100{\mu}/m{\ell}$ in RAW 264.7 cells. The RAW 264.7 cells were induced by lipopolysaccharide (LPS) and CG 1, 10, $100{\mu}/m{\ell}$. The CG decreased nitric oxide (NO) production activity dose dependently, especially at $100{\mu}/m{\ell}$ of 55%. The production of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ were decreased by 51%, 78% and 35% in CG treated $100{\mu}g/m{\ell}$. CG showed dose-dependent suppression activity of reactive oxygen species (ROS) production, especially at $100{\mu}g/m{\ell}$ of 37%. DPPH radical scavenging activity and ABTS cation decolorization were activated over 86% and 88% in CG at $1,000{\mu}g/m{\ell}$ concentration. Conclusions : According to the results, we thought that CG showed anti-inflammatory and antioxidant activities on the RAW 264.7 cells in mouse macrophage. Therefore, this research is expected to provide the fundamental data about the natural material analysis of relating to the anti-inflammation and antioxidant.
Objective : Reflux esophagitis (RE) is a disease that caused gastric acid reflux and inflammation due to unstable gastroesophageal sphincter, as increasing worldwide respectively. This study was conducted to evaluate the effect of Evodiae Fructus (EF) extract on chronic reflux esophagitis in rats. Methods : The EF was measured antioxidant activity, such as total polyphenol and total flavonoid contents, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-enzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity. Rats were divided into 3 groups; Nor (normal group), Con (chronic acid reflux esophagitis rats treatment with water), EF (chronic acid reflux esophagitis rat treatment with EF 200 mg/kg body weight group). A surgically-induced chronic acid reflux esophagitis (CARE) model was established in SD rats, and treated with water or EF 200 mg/kg body weight for 14 consecutive days. Results : Administration of EF to rats of induction of chronic acid reflux esophagitis was found to reduce esophagus tissues injury. Reactive oxygen species (ROS) and produces peroxynitrite ($ONOO^-$) levels of esophagus tissues were significantly decreased in EF compared to Con group. As results of esophagus protein analyses, EF effectively reduce inflammatory-related factors ($NF-{\kappa}Bp65$, $p-I{\kappa}B{\alpha}$, iNOS, $TNF-{\alpha}$, IL-6), and increase anti-oxidant enzyme (Nrf2, HO-1, SOD, catalase, GPx-1/2). Conclusions : These results suggest that EF administration comfirmed that decreased esophagus tissues injury, oxidantive stress, anti-inflammation effect, and increased anti-oxidant effect. Therefore, EF was the potential to be used as a natural therapeutic drug.
Objective : Gastritis is a major complication of gastrointestinal disease. Lonicera japonica is used in folk medicine to treat different diseases such as exopathogenic wind-heat, epidemic febrile diseases, sores, carbuncles and some infectious diseases. Therefore, this study examined the effects of Lonicera japonica water extract (LJE) on HCl/ethano-linduced acute gastric ulceration and anti-oxidants properties. Methods : LC-ESI-IT-TOF MS was employed for rapid identification of major compound from LJE. The antioxidant activities were evaluated through total polyphenol and flavonoid contents and radical scavenging assays and superoxide dismutase (SOD)-like activity. SD rats were randomly divided into five different groups including the normal group, ulcer group, positive group (20 kg/mg of omeprazole, ip), and experimental groups (100 kg/mg and 500 kg/mg of LJE, ip). Results : 4,5-Dicaffeoyl quinic acid, loganic acid, secologanic acid, sweroside, loganin, vogeloside were identified based on the detection of the molecular ion with those of literature data. The LJE was possessed free radical scavenging activities such as DPPH (IC50=189.7 ㎍/㎖), ABTS (IC50=164.5 ㎍/㎖), and SOD-like activity (IC50=405.02 ㎍/㎖). Macroscopic and histological analyses showed LJE treated group were significantly reduced to an extent that it allowed leukocytes penetration of the gastric walls compared with the ulcer group. In addition, an ulcer inhibition rate and prostaglandin E2 levels were increased in rats treated with LJE. Conclusion : The present study has demonstrated the antioxidantive and gastroprotective effect of LJE, these findings suggested that LJE has the potential for use in treatment of gastric disorders.
본 연구는 미역(Undaria pinnatifida) 추출물과 쇠미역(Costaria costata) 추출물의 항산화 활성 및 HepG2 세포의 알코올로 인한 산화 손상에 대한 보호 효과를 연구하였다. 미역과 쇠미역 추출물의 총 폴리페놀과 플라보노이드 함량은 70% 에탄올 추출물에서 가장 높았다. 또한 미역과 쇠미역 70% 에탄올 추출물의 DPPH (IC50 0.33±0.21, 0.48±0.47 mg/ml), ABTS (IC50 0.34±0.30, 0.47±0.17 mg/ml) 라디칼 소거 활성이 열수 추출물 및 10% 에탄올 추출물 보다 높았다. 추출물의 간 보호 효과를 확인하기 위하여 HepG2 세포에 알코올 산화 스트레스를 유발하여 MTT 분석을 이용하여 세포 생존력을 측정하였다. 미역 및 쇠미역 열수 추출물은 알코올 처리군(73.95%) 대비 세포 생존율은 각각 89.91~97.63% 및 84.99~90.54%로 농도 의존적으로 증가시켰다. 본 연구는 미역과 쇠미역 추출물이 간 보호 및 항산화 효과를 나타냄을 확인하였고 알코올 산화 손상에 대한 간 보호 소재로 활용될 수 있을 것으로 사료된다.
Objective : Gastritis refers to an inflammatory disease of the gastric mucosa. Alcohol is one of the main aggression factors, causing bleeding and inflammation in the gastric mucosa and it is known to not only increase lipid peroxide levels, but also deplete key antioxidant factors. The purpose of this study was to determine the effect of Uncariae Ramulus et Uncus water extract (URW) in alcohol-induced gastritis. Methods : The total polyphenol and flavonoid contents of URW were confirmed through an in vitro experiment. Also, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) activity were confirmed. For in vivo experiments, mice were divided into 4 groups (n=8). Also, 1 hr after oral administration of each drug, 50% ethanol was orally administered to induce gastritis. Results : As a result of in vitro experiments, URW showed excellent antioxidant activity. In alcohol-induced gastritis, URW alleviated the damage to the gastric mucosa caused by alcohol. Also, URW decreased reactive oxygen species (ROS) and malondialdehyde (MDA) levels in serum and gastric tissues, and significantly decreased the expression of NADPH oxidases in gastric tissues. In addition, it significantly modulated the nuclear factor erythroid-derived 2-related factor 2 (Nrf2) and nuclear factor-𝜅B p65 (NF-𝜅B) pathways as well as significantly increased the expression of anti-inflammatory proteins. Conclusions : These results suggest that URW not only reduces oxidative stress through excellent antioxidant activity but also relieves gastric mucosal inflammation as a regulator of Nrf2 and NF-𝜅B pathways.
본 연구는 생김치, 적숙기 및 과숙기 상태의 세 가지 발효단계별 김치의 이화학적 특성과 항산화 활성에 대해 알아보았다. pH 및 산도 측정을 통해 생김치(pH 5.6, 산도 0.3%), 적숙기 김치(pH 4.3, 산도 0.64%), 과숙기 김치(pH 3.7, 산도 1.24%)를 시료로 사용하였다. 발효가 진행될수록 glucosinolate의 함량이 증가하였으며, 특히 생김치, 적숙기 김치, 과숙기 김치의 glucoraphanin 함량이 가장 높았다. 뿐만 아니라, 발효가 진행되면서 항산화 물질로 알려진 glucoraphanin, sulforaphane, 및 총 폴리페놀 함량이 증가하였다. 발효단계별 김치 시료의 항산화 활성을 측정한 결과, 100, 250, 500 ㎍/mL의 농도에서 생김치에 비해 적숙기와 과숙기 김치의 DPPH radical 소거능이 우수하게 나타났으며, ·OH radical 소거능을 측정하였을 때 모든 농도(100, 250, 500, 1000 ㎍/mL)에서 적숙기와 과숙기 김치가 생김치에 비해 우수한 소거능을 나타낸 것으로 보아 김치의 발효가 진행될수록 radical 소거능이 증가함을 알 수 있었다. 이는 생김치에 비해 발효과정을 거친 적숙기 및 과숙기 김치가 glucosinolate, sulforaphane, 총 폴리페놀 함량이 증가하여 항산화 활성이 높게 나타난 것으로 사료된다.
어성초(Houttuynia cordata)는 삼백초과(Saururacease)에 속하는 다년생 초본으로 잎, 줄기 및 뿌리의 전초를 약제로 사용하고 있으며, 민간에서는 혈행개선, 폐렴, 급만성기관지염, 장염, 요로감염증, 종기 치료에 주로 이용되고 있다. 최근에는 항산화, 항염증, 항균 및 암세포 성장억제, 간손상 및 신경세포 사멸 억제, 카드뮴 독성 감소효과 등이 보고되고 있다. 본 연구에서는 어성초 지상부의 에탄올 추출물 및 이의 헥센 분획물, 에틸아세테이트 분획물, 부탄올 분획물 및 물 잔류물을 조제하여 항혈전, 항당뇨, 항산화 및 적혈구용혈활성을 평가하였다. 어성초의 총 폴리페놀 및 총 플라보노이드 함량은 에틸아세테이트 분획물에서 가장 높았으며, 상기 분획물은 5 mg/ml 농도에서 용매 대조구보다 2.09배, 2.19배 및 1.69배 연장된 TT, PT, aPTT를 보여 강력한 항혈전 활성을 나타내었으며, DPPH, ABTS 및 nitrite radical에 대해 각각 30.9, 19.9 및 49.6 ㎍/ml의 RC50을 보여, 9.8, 5.2 및 11,8 ㎍/ml의 RC50를 나타낸 vitamin C의 항산화력에 비교되었다. 트롬빈 저해활성이 강력한 헥센 분획물이 강력한 적혈구 용혈활성을 나타내는 바와 달리, 상기 에틸아세테이트 분획물은 1 mg/ml 농도까지 전혀 용혈활성이 나타나지 않았다. 본 연구결과는 어성초의 항혈전 활성에 대한 최초의 보고로, 어성초의 quercitirin, hyperoside, orientin 및 isoquercitrin이 항혈전 및 항산화 활성에 관련되어 제시하고 있으며, 향후 어성초를 이용한 기능성 항혈전 식품소재 개발 가능함을 제시하고 있다.
Objectives : A persistent inflammatory response can cause diseases such as fibrosis, cancer, and allergies. This study aimed to investigate the anti-inflammatory activity of Curcumae longae Rhizoma and Cinnamomi Ramulus Mixture (CCM) in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Methods : The total polyphenol and flavonoid contents of CCM were confirmed through an in vitro experiment. Also, radical scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and Hydroxyl were confirmed. Moreover, ferric reducing antioxidant power (FRAP) activity were confirmed. After, CCM (50, 100, and 200 ㎍/mL) were applied to 0.1 ㎍/mL LPS-stimulated RAW264.7 cells. The levels of nitric oxide (NO) and pro-inflammatory cytokines in the supernatant fraction were determined. Also, the expressions of mitogen-activated protein kinase (MAPK) and nuclear factor-κB (NF-κB) pathways were detected using Western blot. Results : As a result of in vitro experiments, there was an excellent antioxidant activity in CCM-treated cells. In addition, in RAW264.7 cells stimulated with LPS, the increased NO level was inhibited in a concentration-dependent manner by the treatment of CCM. In addition, inflammatory cytokines production were significantly inhibited in a concentration-dependent manner in CCM-treated group. CCM treatment significantly decreased the protein expressions of MAPKs. Moreover, the expressions of NF-κBp65 and cyclooxygenase-2 (COX-2) were significantly decreased when 200 mg/kg of CCM was applied, and phospho-inhibitor of nuclear factor kappa B-α (p-IκBα) and inducible nitric oxide synthase (iNOS) were significantly decreased at all concentrations treated with CCM. Conclusion : Our findings show that CCM exhibited excellent antioxidant activity and exhibited superior anti-inflammatory effect through the MAPKs and NF-κB pathways in LPS-stimulated RAW 264.7 macrophages.
Objectives We evaluated the wound healing effects of Dangguijakyak-san (DJ) using C57BL/6 mice that were generated open wound. Methods The study was conducted with seven C57BL/6 mice assigned to each group, divided into the normal group, control group, vitamin E group, DJ low-dose group, DJ high-dose group. We measured total polyphenol, flavonoid contents, the size of the wound, liver function, pro-inflammatory cytokine activity in serum, inflammation-related proteins, adhesion molecules and chemokine proteins, collagen-related proteins in skin tissue and histopathological changes by H&E and Masson's staining. Results DJ treatment significantly reduced the area of the wound compared to the control group. Also, inflammatory cytokines were reduced and the expression of anti-inflammatory-related factors (interleukin-4 [IL-4] and IL-10) was significantly increased in the DJ treatment group. We identified that DJ treatment inhibits both pathways of inflammation, the mitogen-activated protein kinases and nuclear factor-κB pathway. Moreover, the protein expressions of Sirt1 (sirtuin 1), MCP-1 (monocyte chemoattractant protein 1), ICAM-1 (intercellular adhesion molecule 1), and VCAM-1 (vascular cell adhesion molecule 1) were decreased by DJ administration. Also, the expression of α-smooth muscle actin and collagen type I alpha 1, collagen-related proteins, that help skin recovery was significantly increased in the DJ treatment group. Histopathologically, a relatively thin epithelial layer could be observed in the DJ administration group, as well as an increase in fibroblasts and collagen fibers. Conclusions These data suggest that DJ treatment is effective in wound healing, suppressing inflammatory proteins, increasing skin repair factors and improving histopathological changes caused by wounds.
본 연구에서는 화살나무 날개를 이용하여 물, 에탄올 및 메탄올을 이용하여 얻은 추출물을 이용하여 항산화 활성, 항당뇨 활성, 총폴리페놀 및 플라보노이드 함량, 암세포 성장 억제능을 평가하였다. 화살나무 날개 추출물은 DPPH radical 소거능 결과에서는 에탄올과 메탄올 추출물에서 positive control과 유사한 radical 소거능을 나타내었으며 ABTS·+ radical 소거능에서는 모든 군의 1,000 ㎍/mL의 농도에서 80% 이상의 소거능을 보여주었다. 또한, 환원력과 hydrogen peroxide 소거능 및 α-glucosidase 저해활성에서 에탄올과 메탄올 추출물은 positive control과 비슷하거나 높은 활성을 나타내었다. 화살나무 날개 추출물 내 총폴리페놀 및 플라보노이드 함량은 에탄올 추출물에서 867.8 mg% 및 551.7 mg%로 물 및 메탄올 추출물과 비교하였을 때 보다 높은 총폴리페놀 및 플라보노이드 함량이 확인되었다. 화살나무 날개 추출물의 암세포 증식 억제 활성은 흑색종 피부암(B16)에서는 큰 효과를 확인할 수 없었지만 유방암(MDA-MB-231), 대장암(HT-29) 및 전립선암(LNCaP)에서는 유의적인 세포 생존 억제능을 확인하였고 특히 LNCaP에 에탄올 추출물을 처리하였을 때 19.1%의 세포 생존율을 보이며 가장 우수한 세포 성장 억제 활성을 확인하였다. 또한, 전립선 정상 세포인 RWPE-1을 화살나무 날개 추출물로 처리하였을 때 세포 생존율에 큰 영향을 받지 않았기 때문에 암세포 특이적인 성장 억제 활성을 확인할 수 있었다. 이러한 결과를 통해 풍부한 폴리페놀 및 플라보노이드 함량을 가진 화살나무 추출물은 항산화, α- glucosidase 억제, 항암 효과를 나타내어 뛰어난 생리활성효능을 가지고 있고, 이는 효과적인 천연물 유래 기능성 원료 및 식품 소재 발굴을 위한 기초자료로 이용할 수 있을것으로 사료된다.

② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
