Growth Hormone Signaling in the Regulation of Acid Labile Subunit

  • The past decades have seen enormous advances in our understanding of how GH acts. GH is a pituitary-derived polypeptide hormone that has diverse physiological effects including the regulation of bone growth, carbohydrate and lipid metabolism. The effects of GH are mediated directly and indirectly through IGF-I. In addition, GH stimulates the hepatic production of ALS. In postnatal life, IGF-I and -II circulate as 150 kDa ternary complexes consisting of one molecule each of IGFBP-3 or IGFBP-5, IGF-I or IGF-II and ALS. It is now known that ALS increases significantly the half-lives of the IGFs, IGFBP-3 and -5, and therefore is responsible for maintaining a circulating reservoir for each of these proteins.

  • 생물학에서 특정 단백질을 구성하는 아미노산 서열을 결정하는 것은 매우 중요하다. 이를 위해 펩타이드의 단동위질량을 결정하는 것은 매우 중요한 문제이다. 하지만 Selenocysteine(Sec)이 가지고 있는 특이 성질 때문에 기존의 알고리즘으로 Sec를 포함한 펩타이드의 단동위질량을 결정하는 것은 쉽지 않다. J.W.Kim et.al.은 Sec의 특성을 고려하여 새로운 모델을 제시하고 그에 맞는 알고리즘을 보였다. 이 논문에서는 실험을 통해 J.W.Kim et.al.이 제안한 알고리즘을 실제 프로그램으로 구현하여 실행할 때 고려해야 할 주요 파라미터 및 함수에 대한 분석을 수행한다.

닭 태자의 직결장에 대한 조직학적 및 면역조직화학적 연구 (Histological and immunohistochemical studies on the colo-rectum of the chicken embryos)

  • Histological changes and ontogeny, distribution and relative frequency of bovine SP-1/chromogranin(BCG)-, serotonin-, human pancreatic polypeptide(HPP)- and glucagon-immunoreactive cells were investigated in the colo-rectum of the chicken embryos. The pseudostratified columnar-like epithelium was observed in 10 days of incubation to 15 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium and goblet cells were detected for the first time on 19 days of incubation. BCG and serotonin-immunoreactive cells were observed for the first time on 15 days of incubation respectively, thereafter these cells were increased with ages. However no HPP and glucagon-immunoreactive cells were found in this study.

Isolation and Characterization of the C-type Lysozyme Gene from the Common Cutworm Spodoptera litura

  • We have isolated and characterized a new insect chicken type (c-type) lysozyme gene from the common cutworm, Spodoptera litura. The full-length cDNA of Spodoptera lysozyme is cloned by rapid amplification of cDNA ends PCR (RACE-PCR). The isolated cDNA consists of 1039 bp including the coding region for a 142-amino acid residue polypeptide, which included a signal peptide of 21-amino acid residue and a mature protein of 121-amino acid residue. The predicted molecular weight of mature lysozyme and its theoretical isoelectric point from amino acid composition is 13964.8 Da and 9.05, respectively. The deduced amino acid sequence of Spodoptera lysozyme gene shows the highest similarity (96.7%) to Spodoptera exigua lysozyme among other lepidopteran species. Amino acid sequence comparison with other the c-type lysozymes, Spodoptera lysozyme has the completely conserved $Glu^{32}$ and $Asp^{50}$ of the active site and eight Cys residues are completely conserved in the same position as that of other lepidopteran lysozymes.

Enhanced Resistance to Botrytis cinerea Mediated by Transgenic Expression of the Spider Chitinase Gene AvChit in Arabidopsis

  • The AvChit gene encodes for a chitinase from the spider, Araneus ventricosus. This spider, A. ventricosus, is an abundant species in Korea. Arabidopsis thaliana plants were transformed with the AvChit gene using Agrobacterium tumefaciens. Thirteen transgenic lines expressing the AvChit gene were obtained. Functional expression of the AvChit gene in transgenic Arabidopsis was confirmed by Southern, northern and western blot analysis. The AvChit cDNA was expressed as a 61 kDa polypeptide in baculovirus-infected insect Sf9 cells. AvChit protein extracted from transgenic Arabidopsis exhibited high levels of chitinase activity. Phytopathological tests showed that two transgenic Arabidopsis lines expressing the AvChit gene displayed high levels of resistance to gray mold disease (Botrytis cinerea).

  • Computer aided molecular modeling can help to predict the three dimensional structure of the polypeptide without the sample. The study on soybean Bowman-Birk protease inhibitor (SBI) is valuable, because it has been recently known that SBI possesses anticarcinogenic activities and immune-stimulating properties. SBI has several isoinhibitors, whose isolation and characterization were reported in 1990. Among these, DII inhibits trypsin only. The different inhibitory specificities cannot be explained only by their different primary sequences, but is possible with further assistance by the study on their different three dimensional structures. The study on the three dimensional structure of DII using homology method is reported in this paper.

  • Equinechorionic gonadotropin(eCG) is a member of the glycoprotein hormone family which includes FSH, hCG, TSH. These hormone family is characterized by a heterodimeric structure composed a common α-subunit noncovalently linked to a hormone specific β-subunit. To determine a and β-subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-molecule by fusing the carboxyl terminus of the eCG β-subunit to the amino terminus of the α-subunit was constructed and transfected into chinese hamster ovary (CHO-K1) cells.   • Animal reproductive performance could be affected by several regulatory factors, including nutritional, environmental, and genetic factors. Particularly, during the last half of this century, animal reproductive performance has been remarkably successful in improving the efficiency of livestock production. For some traits efficiency gains have been achieved with little or no knowledge of the genes underlying the traits. And, they have depended upon the phenotypic selection by statistical methods to estimate the genetic parameters of some reproductive traits. In spite of these successes, it is clear that recent advances in both developmental biology and molecular biology are set to revolutionize he practice of animal reproductive performance n the 21th century.   • Equine chorionic gonadotropin (eCG), which consists of highly glycosylated α- and β-subunits, is a unique member of the gonadotropin family because it elicits response characteristics of both follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in other species than the horse. To determine whether α and β subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule was constructed and transfected into Chinese hamster ovary (CHO-K1) cells.   • A chymotrpsin gene homologue was cloned from the mulberry longicorn beetle, Apriona germari. The A. germari chymotrypsin cDNA contains an ORF of 950 nucleotides capable of encoding a 283 amino acid polypeptide with a predicted molecular mass of 29151 Da and pI of 9.38. The A. germari chymotrypsin has conserved six cysteine residues and active triad formed by His, Asp and Ser. The deduced amino acid sequence of the A. germari chymotrypsin cDNA was closest in structure to the Anthonomus grandis chymotrypsin. Northern blot analysis revealed that A. germari chymotrypsin showed the midgut-specific expression.
