Zeolite/Zinc-polypeptide를 코팅한 폴리프로필렌필름의 항균 및 항진균 특성에 관한 연구 (A Study on the Antibacterial and Antifungal Properties of Zeolite/Zinc-polypeptide Coated Polypropylene Film)

  • 본 연구는 기능성 항균 포장재를 신선식품에 적용하기 위한 연구로서 Zeolite/Zinc-polypeptide를 식품용 포장재 중 하나인 PP 필름에 코팅하여 항균 및 항진균성을 확인하였다. 이를 검증하기 위하여 Zeolite/Zinc-polypeptide를 PP 필름에 각각 5%, 10%, 15% 농도로 코팅하였으며, FESEM과 FT-IR 분석을 통해 분산된 Zeolite/Zinc-polypeptide의 코팅 여부를 확인하였다. 또한 항균 효과를 검증하기 위하여 대조군과 코팅 물질의 농도에 따른 필름의 항균 및 항진균성을 분석하였다. 코팅 필름의 표면 분석 결과 PP 필름의 표면에서 Zeolite/Zinc-polypeptide의 농도에 따라 코팅 액의 분포 크기가 불규칙하지만 넓게 나타났다. 또한 항균 효과에 따른 분석 결과 박테리아 균 중 하나인 E. coli에서 99.9%의 항균 효과를 확인하였으며, R. oryzae의 성장을 약 70% 억제하여 PP 필름 코팅 적용 후에도 Zeolite/Zinc-polypeptide가 E. coli와 R. oryzae에 대하여 항균 및 항진균 특성을 갖고 있음을 확인하였다. 이에 따라 Zeolite/Zinc-polypeptide 코팅 필름은 기능성 포장재로서 신선식품의 부패 방지와 보관수명 향상에 효과적일 것으로 보이며, 향후 다양한 신선식품에 적용하기 위한 저장 실험이 요구되어진다.

소의 테이레리아병 치료시 Theileria sergenti의 32kDa Polypeptide 검출의 유용성 (Usefulness of 32kDa Polypeptide Detection of Theileria sergenti in Monitoring Treatment Progress of Bovine Theileriosis)

  • Bovine piroplasmosis caused by Theileria sergenti is a major cause of economic loss in livestock industry. Five cattle infected with Theileria sergenti showing severe and fatal anemia, confirmed by indirect immunofluorescent assay(IFA), were used in this study. Four cattle were treated with diminazene aceturate and one was not treated as the control. The therapeutic effect of diminazene aceturate against Theileria sergenti infection was monitored by detecting the 32kDa polypeptide specific for Theileria sergenti by the western blotting with both polyclonal and monoclonal antibodies. The 32kDa polypeptide detected at the beginning of diminazene aceturate treatment was not detectable after the treatment. It is postulated that the detection of the 32kDa polypeptide specific for Theileria sergenti may be a good tool for the diagnosis and monitoring the treatment progress of Theileria sergenti infection.

Genetic Engineering for Detection of Endocrine Disruption using I-18 C Gene Expression in Chironomus riparius

  • The 2D/E gel analysis for polypeptide expression reflecting I-18 C gene (early-ecdysterone inducible gene) has conducted the emerged C. riparius adults from larval phase exposure to tebufenozide acting as an ecdysteroidal molting hormone. Control group, the amount of ORE II of the I-18 C gene was larger than that of ORE I of this gene. After treatments, ORE I of the I-18 C gene was overexpressed as the polypeptide, whereas ORF II of this gene was expressed as the polypeptide and was clearly reduced expression. Accordingly, we consider that tebufenozide exhibited endocrine disruptions related processing of ecdysteroid receptor protein reflecting ORF II of I-18 C gene. Also, earlier emergence day was related overexpressed polypeptide reflecting ORE I of I-18 C gene. In this study result, tebufenozide induced changing of physiological condition, and then polypeptide expression reflecting early-ecdysterone inducible I-18 C gene was different between control group and exposure group.

Theileria sergenti merozoite의 합성 polypeptide 백신의 면역원성에 관한 연구 (Study on the immunogenicity of synthetic polypeptide vaccine derived from Theileria sergenti merozoite)

  • Western immunoblot analysis of antigen of T sergenti merozoite revealed that the immunodominant proteins of this organism were characterized as the 18KD, 29KD, 34KD, 45KD and 105KD in Korea. The 34KD and 45KD among those immunodominant proteins of the parasite were isolated and their amino acid sequences from the $NH_2$-terminus were determined and synthesized. They respective polypeptides were cationized to enhance their antigenicity, fortified with Freund's adjuvant and tested for immunogenicity in rabbits and cattle. The results obtained were as follows; 1. Theileria sergenti merozoite antigen was shown in 120KD, 100KD, 66KD, 45KD, 34KD and 30KD in western immunoblot using serum of rabbits immunized with 34KD synthetic polypeptide and 70KD, 58KD, 55KD and 45KD using bovine serum. In western immunoblot, 45KD, 34KD and 30KD were recognized by immunized rabbits, and 50KD and 45KD by cattle sera immunized with 45KD synthetic polypeptide, respectively. 2. The ELISA utilizing the synthetic polypeptides demonstrated significant antibody response to the respective peptides. After the 2nd booster injection, an OD of 0.760(preimmunization 0.132) in rabbits and an OD of 0.645(preimmunization 0.488) to 34KD synthetic polypeptide in cattle were observed. In animals immunized with 45KD synthetic polypeptide, after the 2nd booster injection, an OD of 0.640(preimmunization 0.144) in rabbit, and an OD of 0.776 (preimmunization 0.477) in cattle were measured. 3. After the 2nd booster the reciprocal IFA titer was 1:64 in rabbits and 1:512 in cattle immunized with the 34KD synthetic polypeptide. The IFA titre was observed as 1:512 in rabbit and 1:1,024 in cattle in immunized with the 45KD synthetic polypeptide.

  • Pancreatic endocrine cells of the native Korean goat were investigated immunocytochemically at electron microscopic level. All glucagon-, insulin-, somatostatin- and pancreatic polypeptide(PP)-immunoreactive cells were showed chromogranin(CG) immunoreactivity in the secretory granules of each cells. In addition, bovine pancreatic polypeptide immunoreactivity was found to be colocalized in the secretory granules of the glucagon and insulin cells. These observations support that chromogranin is available as the marker of pancreatic endocrine cells on the native Korean goat and BPP colocalized in the secretory granules of glucagon and insulin cells.

Synthesis of $\alpha$-L-Aspartyl-L-phenylalanine Methyl Ester from an Artificial Polypeptide

  • The aspartame, $\alpha$-L-aspartyl-L-phenylalanine methylester, is an artificial sweetener. Taking advantage of the fact that the aspartame is a derivative of dipeptide, synthesis of aspartame from the artificial polypeptide made by an artificial gene has been attempted. The artificial polypeptide (LAP32), a polymer of tripeptide (aspartyl-phenylalanyl-lysine), was purified from the E. coli cells harboring a recombinant plasmid containing the artificial gene. This polypeptide was then digested with trypsin and carboxypeptidase B to produce dipeptide (Asp-Phe). Using the esterase activity of $\alpha$-chymotrypsin, the dipeptide was directly converted into Asp-Phe methylester in a water-methanol system. When the methanol concentration in reaction mixture was 25%, 50% of dipeptide was converted to the dipeptide methylester without producing any by-products.

Theileria sergenti merozoite부터 합성한 polypeptide vaccine의 예방효과 연구 (Immunoprophylactic effect of synthetic polypeptide vaccine derived from Theileria sergenti merozoite)

  • Eighteen holstein-calves(4~5 months old) in a divided groups including the matched control were immunized with $100{\mu}g/dose$ of 34kDa, 45kDa polypeptide and T sergenti merozoite vaccine(protein content $100{\mu}g/dose$) respectively, previously mixed with aluminium hydroxide to elicit antibodies. All groups of calves were boosted with same dose and intervals. The animals were challenged by tick infestations in the endemic pasture of theileriosis from March to September 1994. The animals were monitored for the erythrocyte count, parasitemia, hematocrit and the specific antibody reactions elicited by immunization. The immunological responses demonstrated that vaccination with 34kDa polypeptide and T sergenti merozoite derived vaccine inhibited to produce the 75kDa band immunological responds even in the vaccinated calves after being challenged by tick infestations in the pasture. However, the specific antibody reactions were detected at the 32kDa band in the nonimmunized calves and T sergenti merozoite derived vaccine by the western blot. The 34kDa polypeptide vaccine and T sergenti merozoite derived vaccine were evaluated to be able to protect inducing anemia and to decrease parasitemias level. These vaccines have the efficacy of inhibition to produce a certain antigen corresponding 75kDa band antigen of parasite in the calves as challenged with tick infestations.

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

  • Pancreatic endocrine cells containing glucagon, insulin, somatostatin and pancreatic polypeptide were identified in the pancreas of the Korean native goat by using immunohistochemical method. Glucagon immunoreative cells were oval or fusiform in shape and located at the periphery of the pancreatic islets. Insulin immunoreactive cells were round or oval in shape and occupied throughout the pancreatic islets except the small area of the periphery. Somatostatin immunoreative cells were oval and elliptical, and mainly located at the periphery of the pancreatic islets. Some of these cells had a cytoplasmic process. Pancreatic polypeptide immunoreactive cells were elliptical or polyhedral and located at the periphery of the pancratic islets where two or more cells formed a cell cluster. The distribution rates of glucagon, insulin, somatostatin and pancreatic polypeptide immunoreactive cells were 24.4%, 44.3%, 13.2% and 18.1% respectively.
