시험관법에의한 식물열수추출물의 3-Hydroxy-3-Methylglutaryl CoA Reductase 및 Acyl-CoA:Cholesterol Acyltransferase 활성 저해도 측정 (Measurement of Inhibitory Activities on 3-Hydorxy-3-Methylglutaryl CoA Reductase and Acyl-CoA:Cholesterol Acyltransferase by Various Plant Extracts in vitro)

  • Hydroxy methylglutaryl CoA(HMG CoA) reductase and acyl CoA:cholesterol acyltransferase(ACAT) are two important enzymes that are associated with regulation of cholesterol metabolism. The inhibitors of HMG CoA reductase and ACAT are very effective in lowering serum cholesterol in most animal species. In present study, various plant extracts with hot water were used to examine the inhibitory activities against HMG CoA reductase and ACAT that are involved in cholesterol biosynthesis and cholesterol esterification in tissues, respectively. The extracts of Fagophyrum rotundatum, Rosa multiflora, Rosa rugosa and Alisma orientalis exhibited significant inhibitory activities against the ACAT, 29%, 24%, 19%, and 18%, respectively. However the extracts of Typha augustifolia, Polygonum cuspidatum, Crataegus pinnatifida, Polygonum multiflorum inhibited the HMG CoA reductase activity by 53%, 42%, 37%, and 33% respectively. Results suggest that these plant extracts might play important roles in the regulation of the cholesterol metabolism in vivo.

  • Hot aqueous extracts of 130 herbal medicines were screened for platelet activating factor (PAE) receptor binding antagonistic activity using rabbit platelet. The results suggested that 4 medicinal plants including Biota orientalis are potential sources of PAF antagonists.

Evaluation of New Selective Molecularly Imprinted Polymers for the Extraction of Resveratrol from Polygonum Cuspidatum

  • Four different molecularly imprinted polymers (MIPs) were prepared using resveratrol as the template, methacrylic acid (MAA) or acrylamide (AA) as functional monomers, 2,2-azobisisobutyronitrile (AIBN) as the initiator, and thermo- or photo-induced polymerization. The ability of the different polymers to rebind selectively not only the template but also other phenols was evaluated. In parallel, the influence of the different templates and functional monomers used during polymer syntheses on the performance of the obtained MIPs was also studied through different rebinding experiments. The binding ability and selectivity of the polymer were studied by static balance method and Scatchard analysis. It was concluded that AA-based polymer by photo-induced polymerization presents the best properties to be used as a selective absorbent for the extraction of resveratrol.

호장(虎杖)의 생약학적(生藥學的) 연구(硏究)(I) -한국산(韓國産) 호장(虎杖)의 기원식물(基源植物)- (Pharmacognostical Studies on 'Ho-Jang'(I). -Original Plants of Korean 'Ho-Jang'-)

  • Polygonum cuspidatum $S_{IEB}$. et $Z_{UCC}$. ('Ho-Jang'), P. sachalinense $F_R$. $S_{CHM}$. ('Wang Ho-Jang') and P. ellipticum $M_{IGO} ('Dung Geun Ib Ho-Jang') are the three species growing in Korea. Their rhizomes can be classified into two types: horizontal rhizome type ('Ho-Jang') and erect type ('Wang Ho-Jang' and 'Dung Geun Ib Ho-Jang'). The most of 'Ho-Jang' on Korean markets are the rhizome of 'Wang Ho-Jang' and only limited quantity of the rhizome of 'Ho-Jang' is used. 'Dung Geun Ib Ho-Jang' is a species transplanted from the China continent and is not collected for medicinal use. It is suggested that the name 'Ho-Jang Geun' (Reynoutriae Radix) should be corrected to 'Ho-Jang' (Reynoutriae Rhizoma=Polygoni Rhizoma).

호장(虎杖)의 생약학적(生藥學的) 연구(硏究)(II) -Oxyanthraquinone의 계절적(季節的) 함량(含量) 변화(變化)- (Pharmacognostical Studies on Korean 'Ho-Jang'(II) -Seasonal Variation of Oxyanthraquinones-)

  • The rhizomes of Polygonum cuspidatum, P. sachalinense and P. ellipticum which grow in Korea and which can be used for medicinal purpose were collected monthly from April to October and were analyzed for the contents of the free and combined derivatives of oxyanthraquinones. On the basis of the total oxyanthraquinone contents, the suitable month for collecting the rhizome of the former can be April or May, since it contained 0.46% in May and 0.05% in August. The rhizomes of the latter two species should be collected in June or July, since they contained $0.72{\sim}3%$ and $0.31{\sim}2%$ in April.

제주 자생 식물들에 대한 항비만 효능에 대한 연구 (The Study on Anti-obesity Activity of the Wild Plants of Jeju Island)

  • 본 연구에서는 제주도에서 자생하는 30여 종의 식물추출물에 대한 항비만 효능을 조사하였다. 항비만 효능은 in vitro oil red-O staining 방법을 이용하여 지방전구세포 3T3-L1에서 분화억제력을 측정하였다. 그 결과, 30여 종의 식물 중에서 약도라지, 호장근, 유근피 등을 포함한 6종의 식물이 지방전구세포 3T3-L1의 분화를 억제하였다. 지방전구세포의 분화를 억제하는 물질 중 유근피, 약쑥, 호장근, 후박 4종은 우수한 항산화 효능도 동시에 가지고 있음을 확인하였다. 이러한 결과를 통해, 본 실험에서 확보된 추출물이 항비만 물질로 사용될 수 있는 가능성을 확인하였다.

천연 약용식물 추출물의 구강상피세포암 세포주에 대한 항암효과 (Anticancer Effects of Natural Medicinal Plant Extracts on Oral Carcinoma Cells)

  • The anticancer effect of medicinal plants against two oral carcinoma cells, A253 and SCC-25 were investigated in this study. Methanol extracts from 63 medicinal plants, which have anticancer activities against other cancers such as stomach, hepatocellular or colon carcinomas, were prepared and screened for their anti- oral cancer activity by using MTT assay. Thirty one samples showed anti-oral cancer activity against either cell line used, however, other 32 samples had no anti-oral cancer activity. Among these samples methanol extract of Caesalpinia sappan revealed the strongest anti-oral cancer activity. The $IC_{50}$/ values of this extract against A253 and SCC-25 cells were 16 and 25 $\mu$g/m1, respectively. Fractions of n-hexane, dichloromethane, ethylacetate, n-buthanol and water were prepared from methanol extracts of Caesalpinia sappan, Anthriscus sylvestris, Rhus japonica, Curcuma arowatica, Inula helenium, Sinoarnudinaria reticulata, and Polygonum cuspidatum, respectively. Among these 35 fractions the n-hexane fraction of Inula helenium showed the strongest anti-oral cancer activity, the $IC_{50}$/ value was 1.6$\pm$0.3 $\mu\textrm{g}$/ml. Ten other fractions showed $IC_{50}$/ values lower than 10 $\mu\textrm{g}$/ml.

천연물로부터 $5{\alpha}-Reductase$ 활성억제 및 면포용해효과의 검색 (Screening of $5{\alpha}-Reductase$ Inhibition and Comedolytic Effects from Natural Products)

  • The antibacterial activity against Propionibacterium acnes (P. acnes), $5{\alpha}-reductase$ inhibition and comedolytic effects are the important pharmacological target sites of antiacne drughs. We previously reported on the antibacterial activities against P. acnes by natural products. In the present study the screening of $5{\alpha}-reductase$ inhibition and comedolytic effects from natural products were performed. Seven natural products such as Angelica koreana, Sophora flavescens, Prunus persica, Bombyx mori, Areca catechu, Galla rhois and Gleditschia koraiensis perfectly inhibited the activity of $5{\alpha}-reductase$ at the concentration of 0.01% (w/v). Sixteen natural products which were shown to have the potent antibacterial activities against P.acnes or $5{\alpha}-reductase$ inhibition activities were assayed for the comedolytic test. In the results of comedolytic effects on experimentally-induced comedones (EIC), Sophora flavescens showed the strongest comedolytic effect on EIC, and Polygonum cuspidatum and Angelica koreana showed stronger comedolytic effects on EIC than azelaic acid used for a positive control at the concentration of 3% (w/v). These results suggest that several natural products including Sophora flavescens can be developed as noble antiacne agents.

오배자의 항산화 및 자유라디칼 소거효과 (Antioxidative and Free Radical Scavenging Effects of Rhus javanica Linne)

  • 인구의 수명이 연장되고 노령화 사회로 발전됨에 따라 다양한 성인병이 야기되고 있고, 이들 성인병의 원인이 활성산소에 의한 것으로 밝혀짐에 따라 이들의 다양한 성인병을 예방 치료할 수 있는 안전하고 우수한 항산화 물질을 천연물로부터 개발하기 위하여 약용식물 30종을 대상으로 그들의 MeOH 엑스에 대한 자유라디칼소거효과 및 오배자의 분획에 대한 항산화효과를 연구하여 다음과 같은 결과를 얻었다. 1. DPPH법을 사용하여 약용식물 10종의 MeOH 엑스에 대한 1차 라디칼 소거작용 검색 시험을 한 결과 오배자, 토복령과 호장근의 MeOH 엑스가 가장 강한 항산화 활성 을 나타내었으며 이중에서 오배자의 자유라디칼 소거효과가 가장 높게 나타났다. 2. 1차 실험에서 얻어진 결과를 토대로 가장 강한 활성을 나타낸 오배자에 대하여 오배자의 MeOH 엑스로부터 각 극성별로 분획하여 얻어진 n-Hpxane, EtOAc, BuOH, $H_2O$ 엑스에 대하여 DPPH법을 이용한 라더칼 소거작용을 시험한 결과, n-Hexane, EtOAc, BuOH 엑스가 강한 자유라디칼 소거활성을 나타내었다. 3. Ferric-Thiocyana법을 이용하여 오배자의 n-Hexane, EtOAc, BuOH 및 $H_2O$엑스에 대하여 지질 과산화 억제 작용에 의한 항산화 활성을 시험한 결과 오배자의 n-Hexane, EtOAc, 및 BuOH 엑스는 천연 항산화제인 tocopherol 및 합성 항산화제 인 BHA 보다 우수한 탁월한 항산화 효과를 나타내었고, $H_2O$ 엑스는 tocopherol 보다는 우수하고 BHA와는 유사한 강한 항산화 활성을 나타내었다.

생약재 추출물로부터 분리한 ${\alpha}$-Glucosidase 저해활성물질 탐색 (${\alpha}$-Glucosidase Inhibitory Substances Exploration Isolated from the Herb Extract)

  • 당뇨병에 효과가 있는 식품소재의 탐색과 그 소재를 이용한 기능성 빵제품 개발에 관한 연구의 일환으로 당뇨병에 효과가 있다고 알려진 소장흡수 저해효소인 ${\alpha}$-glucosidase를 target로 시중에 유통되는 다양한 생약재를 대상으로 탐색을 하였다. ${\alpha}$-Glucosidase 저해활성을 조사하고자 생약재 200여종을 대상으로 물층과 메탄올 층으로 구분하여 탐색한 결과 물층에서는 황기(70.9%)가, MeOH층에서는 구인(72.9%)이 70%이상의 강력한 저해효과를 나타났으며 60%이상으로는 물층에서는 육두구(69%), 상백피(66.9%), 오미자(65%), 삼칠(63.9%), 전호(62.9%), 천문동(62.1%), 정공등(60.9%), 호장근(60.7%), 창출(60.2%), 소자(60.2%)등 이었으며, MeOH층으로는 만삼(67.8%), 도인(67.6%), 방기(63.5%), 동규자(61.6%) 순으로 활성효과를 나타내었으며 50%이상의 저해효과를 보인 것으로는 물층에서는 황정(59.8%), 신곡(59.7%), 택사(59.5%), 의인(59.2%), 세신(59.0%), 시호(53.0%)를 포함한 31여종, MeOH층에서는 사군자(58.8%), 구기자(58.3%), 과루인(58.0%), 백자인(55.9%), 백강잠(55.6%), 계내금(55.4%), 독활(55.3%), 구척(52.7%), 대극(52.7%), 백편두(52.5%) 10종이 저해효과를 나타내었다. 따라서 당뇨병 환자의 기능성 소재로 개발하는데 기초 자료가 될 것으로 기대된다.
