Urinary 1-hydroxypyrene glucuronide and genetic polymorphisms of xenobiotic metabolism enzymes in shipbuilding workers using coal tar paint (콜타르가 함유된 페인트 사용 조선업 근로자에서 요중 1-hydroxypyrene glucuronide와 대사효소 유전자 다형성에 관한 연구)

  • Although shipbuilding workers were exposed to a variety of genotoxic compounds including polycyclic aromatic hydrocarbons (PAHs), limited number of studies were conducted to evaluate the biomarkers related to PAH exposure in painting workers in shipbuilding industry. One hundred and thirty three workers including 73 employees using coal tar paints were recruited from a shipbuilding company located in South Korea. Urinary 1-hydroxypyrene glucuronide (1-OHPG), as internal dose of PAH exposure, were measured by synchronous fluorescence spectroscopy after immunoaffinity purification using monoclonal antibody 8E11. Glutathione S-transferase (GST)M1 and GSTT1 genotypes were assessed by multiplex PCR. Information on demographic characteristics, smoking gabit, diet, job title, use of personal protective equipments were collected by self-administered questionnaire. Urinary 1-OHPG were higher in workers using coal tar paints than in workers using general paints, however, the difference was not statistically significant (p=0.20, Mann-Whitney U test). Urinary 1-OHPG levels in smokers were higher than in non-smokers (p<0.05 by Mann-Whitney U test) and there was a significant increase in urinary 1-OHPG levels with the numbers of cigarettes consumed per day (Spearman's correlation coefficient = 0.28, p=0.02). Genetic polymorphisms of GSTM1 and GSTT1 did not influence the level of 1-OHPG in study subjects. Multiple regression analysis show that smoking is the only significant predictor for lon-transformed 1-OHPG (overall model R2=0.1). These results suggest that workers using coal tar paints were exposed to significant amount of PAHs and individual difference in xenobiotic metabolism might affect the levels of internal dose of PAHs.

  • CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydroxyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrchlorodibenzo-p-dioxin) and these inductions were dose-dependent. Recent industrialized society, human hasbeen widely been exposed to widespread environmental contaminants such as PAHs (polycyclic aromatic hydrocarbon) that are originated from the incomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR (aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarket for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounda such as policyclic aromatic hydrocarbon (PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. We examined effects of PAHs on the CYP1B1-lucifrease reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. flvonoids such as genistein decreased B(k)F induced luciferase activity at low concentration. it exhibited stimulatory effect at high concentration.

  • In this study, the methods were developed to measure polycyclic aromatic hydrocarbons(PAHs) in the air, metabolites of pyrene and benzo(a)pyrene via human urine, genetic polymorphisms in human buccal cell for evaluation of the health effects about environmental pollution. We have also performed a preliminary molecular epidemiology study on residents in the metropolitan area and workers in workplace for these method applications.

  • Passive air samplers (PAS) have been developed since the early 2000s and widely used for the atmospheric monitoring of persistent organic pollutants (POPs). PAS are useful especially for the investigation of source-receptor relationship of POPs because they provide higher spatial resolution data. In Korea, however, only a few research groups have conducted POPs monitoring using PAS. One of the reasons for the limited application of PAS might be due to a complicated calculation method for air concentration. In this study, therefore, we introduced the principle of polyurethane foam (PUF)-PAS, which has been most widely used in the world, and provided an example of the calculation of air concentration of polycyclic aromatic hydrocarbons (PAHs). As all data tables and equations for this calculation were provided, this method can be used for the conversion of the amount of POPs (ng) in a PUF disk to air concentration ($ng/m^3$).

Electrokinetic Soil Flushing with Nonionic Surfactant for Removal of Phenanthrene

  • Polycyclic aromatic hydrocarbons (PAHs) are representative hydrophobic organic carbons (HOCs). Surfactant-enhanced electrokinetic (EK) remediation is an innovative in-situ technology that can effectively remove HOCs from low-permeability soils. In this study, the electrokinetic remediation using Tergitol 15-S-12, a nonionic surfactant, was conducted for the removal of phenanthrene from kaolinite. Tergitol 15-S-12 was used at concentrations of 1.5, 2.0, 2.5 and 7.5 g/L to enhance the solubility of phenanthrene. When the surfactant solution was applied to EK system, high electrical potential gradient was maintained and the amount of electroosmotic flow decreased. Removal efficiency of phenanthrene was proportional to the concentration of Tergitol 15-S-12 because the solubility and mobility of phenanthrene was enhanced by surfactant micelle. Therefore, the suitable concentration of nonionic surfactant Tergitol 15-S-12 is expected to improve the removal efficiency of PAHs in EK remediation.

  • A dynamic multimedia model with 2-dimensional spatial resolution, POPsME (Persistent Organic Pollutants in Multimedia Environments), was evaluated by comparing predicted relative concentrations with those measured. A total of 12 polycyclic aromatic hydrocarbons (PAHs) were tested (phenanthrene, anthracene, fluoranthene, pyrene, chrysene, benz (a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenz(a,h)anthracene, benzo(g,h,i)perylene, and indeno(1,2,3-c,d)pyrene). The concentrations were measured in air particulates, water (dissolved phase and suspended solids (ss)), soil, sediment, and leaves of Pinus koraiensis and Prunus serrulata at seven sites in the Seoul and neighboring area (150km x 150km), Korea.

NMR-based metabolic responses of the zebrafish exposed to Benzo[a]pyrene

  • Benzo[a]pyrene (BaP), one of the polycyclic aromatic hydrocarbons (PAHs), is an endocrine disruptor and carcinogenic. This study was conducted to investigate the metabolic changes of zebrafish short-term exposure to BaP using nuclear magnetic resonance (NMR) spectroscopy. In our results, the multivariate analysis showed that the metabolic responses were differed according to the exposure concentration. Also, it was observed that exposure to high concentration of BaP (162 ㎍/L and 1620 ㎍/L) increased the levels of creatine, histidine, and inosine in zebrafish, which means high concentration of BaP exposure affected the energy metabolism and immune function in zebrafish.

Estimation of Gas-particle partitioning Coefficients (Kp) of Carcinogenic polycyclic Aromatic hydrocarbons in Carbonaceous Aerosols Collected at Chiang - Mai, Bangkok and hat-Yai, Thailand

  • To assess environmental contamination with carcinogens, carbonaceous compounds, water-soluble ionic species and trace gaseous species were identified and quantified every three hours for three days st three different atmospheric layer at the heart of chiang-Mai, bangkok and hat-Yai from December 2006 to February 2007. A DRI model 2001 Themal/Optical Carbon Analyzer with the IMPROVE thermal/optical reflectance (TOR) protocol was used to quantify the organic carbon(OC) and elemental carbon content in $PM_{10}$. Diurnal and vertical variability was also carefully investigated. In general, OC and EC contenttration shoeed the highest values at the monitoring period o 21.00-00.00 as consequences of human activities at night bazaar coupled with reduction of mixing layer, decreased wind speed and termination of photolysis nighttime. Morning peaks of carboaceous compounds were observed during the sampling period of 06:00 -09:00, emphasizing the main contribution of traffic emission in the three cities. The estimation of incremental lifetime partculate matter exposure (ILPE) raises concern of high risk of carbonaceous accumulation over workers and residents living close to the observatory sites. The average values of incremental lifrtime particulate matter exposure (ILPE) of total carbon at Baiyoke Suit Hotel and Baiyoke Sky Hotel are approsimately ten time shigher then those air sample collected at prince of songkla University Hat-Yai campus corpse incinerator and fish-can maufacturing factory but only slightly higher than those of rice straw burnig in Songkla province. This indicates a high risk of developing lung cancer and other respiratory diseases across workers and residents living in high buildings located in Pratunam area. Using knowledge of carbonaceous fractions in $PM_{10}$, one can estimate the gas-particle partitioning of polycyclic aromatic hydrocarbons (PAHs). Dachs-Eisenreich model highlights the crucial role of adsorption in gas-particle partitioning of low molecular weight PAHs, whereas both absorption and adsorption tend to account for gas-particle partitioning of high molecular weight PAHs in urban residential zones of Thailand. Interestingly, the absorption mode alone plays a minor role in gas-partcle partitiining of PAHs in Chiang-Mai, Bangkok and hat-Yai.

  • Estimation of concentrations of PAHs [benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, benzo(g,h,i)perylene, indeno(1,2,3-c,d)pyrene] in cereals, pulses, potatoes, and their products available in Korean markets gave mean levels of 0.13, 0.08, 0.06, 0.03, 0.08, 0.15, 0.45, and $0.14{\mu}g/kg$, respectively, with recoveries between 82.6-106.6%. Methodology involved saponification and extraction with n-hexane, purification on Sep-Pak Florisil cartridges, and high performance liquid chromatography using a fluorescence detector.
