Biodegradation of Polycyclic Aromatic Hydrocarbons by White Rot Fungi (백색부후균을 이용한 다환방향족 탄화수소(PAHs) 의 분해)

  • The white rot fungi Phanerochaete chrysosporium(IFO 31249) Trametes sp and Pleurotus sp. were studied for their ability to degrade Polycyclic Aromatic Hydrocarbons(PAHs) using anthracene and pyrene as model compounds. The disapperarance anthracene and pyrene of from cultures of wild type strains. P chrysosporium Trametes sp. and Pleurotus sp was observed However the activities of ligninolytic enzymes were not detected in P chrysosporium cultures during degradation while ligninolytic enzymes were detected in both culture of Trametes sp. and Pleurotus sp. Therefore our results showed that PAHs was degraded under ligninolytic as well as nonligninolytic conditions. The results also indicate that lignin peroxidase(LiP) mananese peroxidase(MnP) and laccase are not essential for the biodegradation of PAHs by white rot fungi.

Simultaneous Determination of Polycyclic Aromatic Hydrocarbons by Synchronous Spectrofluorimetry (형광분광법에 의한 방향족 탄화수소의 동시정량)

  • Synchronous spectrofluorimetry was carried out for the simultaneous determination of various polycyclic aromatic hydrocarbons(PAHs) in aqueous solution by fluorescence spectrometry have been studied. The optimal wavelength interval(${\triangle}{\lambda}$) for synchronous spectra of acenaphthene, anthracene, benzo[a]anthracene, fluorene and pyrene were investigated in the presence of surfactants. The great enhancement of the fluorescence of these PAHs in Triton X-100 was obtained and optimal wavelength was 50 nm. The calibration curves in synthetic mixture solution of 5 PAHs were linear over the range from $1.0{\times}10^{-8}M$ to $1.0{\tiems}10^{-4}M$. Under the optimal experimental conditions, the detection limits were $4.9{\tiems}10^{-9}M$,\;7.0{\times}10^{-9}M,\;4.7{\tiems}10^{-9}M,\;1.6{\tiems}10^{-9}M$ and $3.2{\tiems}10^{-9}M$ for acenaphthene, anthracene, benzo[a]anthracene, fluorene and pyrene, respectively.

  • Thermal desorption/gas chromatography/mass selective detection method using Tenax cartridges for the determination of gaseous polycyclic aromatic hydrocarbons(PAH) is described. Glass fiber filter can collect only PAH in particulate. Gaseous PAH may penetrate the filter. Glass cartridge packed Tenax-GC was uses fur adsorption of gaseous PAH. The air of inhalation zone was collected fur 2-10 hours. Cartridges were thermally desorbed in the reverse direction to sample flow. The desorption conditions were as follows; desorption temperature; $300^{\circ}C$; desorption time; 20min; column head pressure; 30psi; inlet split vent; closed during desorption.

  • Polycyclic aromatic hydrocarbons (PAHs), which are ubiquitous in the air, are present as volatile and particulate pollutants that result from incomplete combustion. Most PAHs have toxic, mutagenic, and/or carcinogenic properties. Among PAHs, benzo[a]pyrene (B[a]P) and dimethylbenz[a]anthracene (DMBA) are suspected endocrine disruptors. The testis is an important target for PAHs, yet effects on steroidogenesis in Leydig cells are yet to be ascertained. Particularly, disruption of testosterone production by these chemicals can result in serious defects in male reproduction. Exposure to B[a]P reduced serum and intratesticular fluid testosterone levels in rats. Of note, the testosterone level reductions were accompanied by decreased steroidogenic acute regulatory protein (StAR) and $3{\beta}$-hydroxysteroid dehydrogenase isomerase ($3{\beta}$-HSD) expression in Leydig cells. B[a]P exposure can decrease epididymal sperm quality, possibly by disturbing the testosterone level. StAR may be a key steroidogenic protein that is targeted by B[a]P or other PAHs.

A Comparative Study on the Analytical Methods of Polycyclic Aromatic Hydrocarbons in the Atmosphere (대기 중 다환방향조탄화수소류의 분석 방법간 비교연구)

  • In order to compare analytical conditions for measuring PAHs(polycyclic aromatic hydrocarbons) in ambient air, a series of experiment were conducted using three different types of extraction methods that include:(1) accelerated solvent extraction (ASE), (2) soxhlet, and (3) sonification methods. Efficiency and relevant analytixal parameters for each of these methods were examined on five different types of solvents that include:(1) 10% diethyl ether in n-hexane(v/v), (2) dichloromethane, (3)cyclohexane.(4) toluene, and (5) Benzene: ethanol (4:1). Comparison of extraction efficiency for various combinations between methods and solvents was made through an application of both internal and external standard calibration techniques. The extraction efficiency tests, when checked using both internal and external standards, allowed us to derive meaningful comparison among different techniques and among solvents.

Polycyclic aromatic hydrocarbons (PAHs) in surface water from the coastal area of Bangladesh

  • Sixteen USEPA priority polycyclic aromatic hydrocarbons (PAHs) in the surface water from the coastal areas of Bangladesh were analyzed by GC-MS/MS. Samples were collected in winter and summer, 2015. The total concentration of PAHs (${\Sigma}PAHs$) showed a slight variation in the two seasons, which varied from 855.4 to 9653.7 ng/L in winter and 679.4 to 12639.3 ng/L in summer, respectively. The levels of ${\Sigma}PAHs$ were comparable to or relatively higher than other coastal areas around the world. The areas with recent urbanization and industrialization (Chittagong, Cox's Bazar and Sundarbans) were more contaminated with PAHs than the unindustrialized area (Meghna Estuary). Generally, 2-3-ring PAHs were the dominant compounds. Molecular ratios suggested that PAHs in the study areas could be originated from both pyrogenic and petrogenic sources. The risk assessment revealed the extremely high ecological risk of PAHs, indicating an intense attention should be paid to PAHs pollution in the coastal areas of Bangladesh.

Inhibitory effects of dietary antioxidants on the formation of carcinogenic polycyclic aromatic hydrocarbons in grilled pork

  • Objective: The inhibitory effects of dietary antioxidants, diallyl disulfide (DADS) and quercetin, in marinade were investigated on the formation of carcinogenic polycyclic aromatic hydrocarbons (EPA priority 16 PAHs) in grilled pork. Methods: The formation of PAHs in grilled sirloin pork with different marinades after charcoal-grilling for 2 min/side were evaluated using high performance liquid chromatography with a photodiode array detector (HPLC-DAD). Results: Compared with the control marinade treatment (without antioxidant), the addition of DADS (500 mg/kg meat sample) in marinade significantly decreased benzo[a]pyrene (BaP) (100%) and heavy PAHs (84%) in charcoal-grilled pork, while the addition of quercetin at the same concentration could reduce 23% and 55% of BaP and heavy PAHs, respectively. Conclusion: The results of this study suggested that the addition of DADS in the marinade could be important in decreasing the levels of PAHs in grilled meat.

  • This study was proceeded the analytical methods using various analytical instruments for polycyclic aromatic hydrocarbons (PAHs) in food products. Various analytical methods were developed to determine levels of PAHs including benzo[a]pyrene, benzo[a]anthracene, benzo[b]fluoranthene, and chrysene formed in various food products using gas chromatography-mass spectrometry (GC-MS), enzyme-linked immunosorbent assay (ELISA) and raman spectroscopy. Recently, the rapid on-site response for the detection of hazardous substances in food aims to develop an onsite rapid detection of a simplified technical analysis method to reduce the time and cost required for analysis of PAHs. Current PAHs detection methods have been reviewed along with new raman spectroscopy analytical method.

Distribution and Source of Atmospheric Polycyclic Aromatic Hydrocarbons and Nitropolycyclic Aromatic Hydrocarbons in Tieling City, Liaoning Province, a Typical Local City in Northeast China

  • Airborne particulates were collected in three different size fractions (>7 ${\mu}m$; 2.1-7 ${\mu}m$; < 2.1 ${\mu}m$) by using Andersen low-volume air samplers at three sites in Tieling city, Liaoning Province, a typical local city in northeast China, in every season during the period from July 2003 to May 2004. Nine polycyclic aromatic hydrocarbons (PAHs) and seven nitropolycyclic aromatic hydrocarbons (NPAHs) in the airborne particulates were determined by HPLC with fluorescence and chemiluminescence detection, respectively. The mean total concentrations of the nine PAHs and seven NPAHs were highest at The mixed residential and light industrial site, and lowest at the residential site near the suburbs. At all sites, more than 70% of the total PAHs and more than 60% of the total NPAHs were found in the finest particulate (< 2.1 ${\mu}m$) fraction. Both cluster analysis and several diagnostic ratios showed that the major contributors of PAHs and NPAHs in airborne particulates were coal combustion systems such as domestic stoves and coal boilers in all seasons in Tieling city.

Atmospheric Polycyclic and Nitropolycyclic Aromatic Hydrocarbons in an Iron-manufacturing City

  • Total suspended particulates (TSP) in the atmosphere were collected for 2 weeks during winter in Muroran, Hokkaido, Japan, a typical iron-manufacturing city. The concentrations of polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs) in TSP were determined by high-performance liquid chromatography (HPLC) using fluorescence and chemiluminescence detectors, respectively. No relationship was observed between the atmospheric PAH and NPAH concentration, or the atmospheric PAH and TSP concentration. However, there was a tendency that the atmospheric PAH concentration was higher when the wind blew from the coke-oven plant. Furthermore, the concentration ratios of 1-nitropyrene to pyrene, which is a suitable indicator of the contribution made by automobiles and coal combustion systems to urban air particulates, were smaller in Muroran and the values were close to those observed in particulates from coal combustion systems. Therefore, these results show that the PAH and NPAH compositions for Muroran are characteristic of an iron-manufacturing city.
