전사체와 대사물질 구조분석을 통한 Novosphingobium pentaromativorans US6-1의 dibenzofuran 분해 경로 해석 (Investigation of biodegradation pathway of dibenzofuran by Novosphingobium pentaromativorans US6-1 via transcriptomic and mass-spectrometric analysis)

  • 다환 방향족 탄화수소(polycyclic aromatic hydrocarbon, PAH) 우수 분해균주인 Novosphingobium pentaromativorans US6-1의 dibenzofuran (DBF) 분해경로를 밝히기 위하여 중간대사물질 분석과 전사체 분석을 진행하였다. GC/MS로 중간대사물질을 분석한 결과, 3(2H)-벤조퓨라논이 검출되었는데 이 화합물은 측면 이산소화에 의해 생성된 중간대사산물들의 기본 골격이 되는 물질로써 균주 US6-1에 의한 DBF의 분해가 측면 이산소화로 진행될 가능성을 시사한다. RNA-Seq 분석 결과, 균주 US6-1이 DBF에 노출되었을 때 발현되는 유전자들의 대부분이 lateral dioxygenation과 관련이 있다는 것을 확인하였다. 이상의 결과로부터N. pentaromativorans US6-1에 의해 일어나는 측면 이산소화를통한 DBF 분해경로와 관련 유전자들을 제시하였다.

식·약공용 농·임산물의 다환방향족탄화수소 오염도 조사 및 위해도 결정 (Contamination Investigation and Risk Characterization on the Polycyclic Aromatic Hydrocarbon of Agricultural Products Used for Food and Medicine)

  • Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.

  • Benzo(a)pyrene을 비롯한 16종의 polycyclic aromatic hydrocarbon(PAHs)과 4종의 nitro-PAHs를 reversed-phase liquid chromatography/diode array detection (RPLC/DAD)를 이용하여 기울기용리법으로 분리하고, PAH류의 머무름거동에 영향을 미치는 인자들, 즉 탄소수, F인자, L/B ratio, 물에 대한 용해도 및 방해수소원자쌍의 수와 시료의 머무름과의 상관관계를 고찰하였다. 대부분의 PAH류는 탄소원자수와 F값이 커질수록 logk'이 증가하는 사실을 확인하였다. 그리고 몇 가지 이성체들과 nitro-PAH류들은 용해도가 크고, L/B값이 작으며, 방해수소원자쌍의 수가 많을수록 먼저 용리됨을 확인하였다. 각 PAH류의 검출한계는 100~500ng/mL 범위로 나타났으며, 실제 물시료에 대한 검출한계(method detection limit, MDL)는 0.1~0.5ng/mL 범위였다.

전처리 방법에 따른 커피원두 중 polycyclic aromatic hydrocarbons 함량 분석 (Analysis of Polycyclic Aromatic Hydrocarbon Content in Coffee Beans with Different Preparation Method)

  • 원두커피의 PAHs 분석을 위한 시료 전처리는 Soxhlet 추출법과 액/액 추출법 두 가지로 하였다. 두 방법의 간단한 모식도는 Fig. 2을 통해 나타내었다. Soxhlet 추출법을 이용하여 추출한 후 비누화시켜 SPE cartridge로 silica cartridge를 사용하여 효과적으로 정제하였으며, 액/액 추출법은 먼저 비누화를 시킨 후 n-hexane을 이용하여 추출하고 florisil cartridge로 clean up 시킨 후 각각 HPLC-FLD를 사용하여 분석하였다. 이러한 두 가지 방법으로 진행된 실험의 결과 90% 이상의 회수율을 보인 것은 액/액 추출법의 B[a]A, Chrys이었고, 80% 이상의 회수율을 보인 것은 액/액 추출법의 B[b]F, B[k]F, B[a]P, DB[a,h]A과 Soxhlet 추출법의 B[a]A, Chrys으로 나타났으며, 그 외에는 약 70% 정도의 회수율을 나타내었다. 7종의 총 PAHs 회수율은 각각 Soxhlet 추출법은 74.25%이고 액/액 추출법 77.08%로 많은 차이를 보이진 않았지만 액/액 추출법이 더 높은 회수율을 나타내었다. 두 방법 모두에서 Chrys의 회수율이 가장 높았으며, 두 가지 방법을 비교하여 실험한 결과 많은 차이를 보이지는 않았지만 Soxhlet 추출법보다 액/액 추출법이 전처리에 소비되는 시간을 줄일 수 있었으며 또한 액/액 추출법에서 표준물질의 회수율도 높게 나타났다.

  • We present a comparative study of CO and polycyclic aromatic hydrocarbon (PAH) emission toward a region including the massive star-forming regions of NGC 6334 and NGC 6357. We use the NANTEN $^{12}CO(J=1-0)$ data and the AKARI $9{\mu}m$ All-Sky diffuse map in order to evaluate the calibration accuracy of the AKARI data. We confirm that the overall CO distribution shows a good spatial correspondence with the PAH emission, and their intensities exhibit a good power-law correlation with a spatial resolution down to 4' over the region of $10^{\circ}{\times}10^{\circ}$. We also reveal poorer correlation for small scale structures between the two quantities toward NGC 6357, due to strong UV radiation from local sources. Larger scatter in the correlation toward NGC 6357 indicates higher ionization degree and/or PAH excitation than that of NGC 6334.

  • Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrachloto-p-dioxin). Recent industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. Acenaphthene, anthracene, benzo(b)fluoranthene, fluorene, fluoranthene, anphthanlene, pyrene, phenanthrene and carbazole were weak responders in MCF-7 cells. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA.

Elucidating H/D-Exchange Mechanism of Active Hydrogen in Aniline and Benzene-1,2-dithiol

  • In this study, the hydrogen/deuterium (HDX) exchange mechanism of active hydrogen, nitrogen, and sulfur-containing polycyclic aromatic hydrocarbon (PAH) dissolved in toluene and deuterated methanol by atmospheric pressure photoionization (APPI) is investigated. The comparison of the data obtained using APPI suggests that aniline and benzene-1,2-dithiol contain two exchanging hydrogens. The APPI HDX that best explains the experimental findings was investigated with the use of quantum mechanical calculations. The HDX mechanism is composed of a two-step reaction: in the first step, analyte radical ion gets deuterated, and in the second step, the hydrogen transfer occurs from deuterated analyte to de-deuterated methanol to complete the exchange reaction. The suggested mechanism provides fundamentals for the HDX technique that is important for structural identification with mass spectrometry. Fluorescence Characteristic Spectra of Domestic Fuel Products through Laser Induced Fluorescence Detection

  • Traditional investigation procedures of soil and groundwater contamination are followed by soil gas sampling, soil sampling, groundwater sampling, establishment of monitoring wells, and groundwater monitoring. It often takes several weeks to obtain the analysis reports, and sometimes, it needs supplemental sampling and analysis to delineate the polluted area. Laser induced fluorescence (LIF) system is designed for the detection of free-phase petroleum pollutants, and it is suitable for on-site real-time site investigation when coupling with a direct push testing tool. Petroleum products always contain polycyclic aromatic hydrocarbon (PAH) compounds possessing fluorescence characteristics that make them detectable through LIF detection. In this study, LIF spectroscopy of 5 major fuel products was conducted to establish the databank of LIF fluorescence characteristic spectra, including gasoline, diesel, jet fuel, marine fuel and low-sulfur fuel. Multivariate statistical tools were also applied to distinguish LIF fluorescence characteristic spectra among the mixtures of selected fuel products. This study successfully demonstrated the feasibility of identifying fuel species based on LIF characteristic fluorescence spectra, also LIF seemed to be uncovered its powerful ability of tracing underground petroleum leakages.

  • In mammalian, cytochrome P4501A1 (CYP1A1) is very important for metabolism of xenobiotics such as PAHs(Polycyclic aromatic hydrocarbon) and heterocyclic amine, and it is induced by environmental contaminants such as PAHs, TCDD(2,3,7,8-tetrchlorodibenzo-p-dioxin) and 3-MC (3-methylcholanthrene). In fish, like mammalian, when it is exposed to environmental contaminants, they cause specific and sensitive induction of CYP1A. Therefore, induction of CYP1A in fish and mammalian is widely used as a biomarker for exposure of environmental contaminants. In this study, to compare the function of Cyp1a1 in fish with it in mammalian, we have used rainbow trout(Oncorhynchys mykiss) hepatoma cells (RTH-149) and mouse hepatocyte (Hepa-I). in order to examine induction of Cyp1a1 by TCDD, we have used the bioassay system. We examined effects of TCDD on the Cyp1a1-luciferase reporter gene activity, 7-ethoxyresorufin O-deethylase(EROD) activity and Cypa mRNA level.
