백색부후균에 의한 염료의 탈색 (Decolorization of Dyes by White Rot Fungi)

  • 백색부후균류에 의한 poly R-478, congo red 및 methylene blue 등 3 가지 염료의 탈색을 여러 가지 배양조건에서 분석한 결과 국내에서 분리한 구름버섯 (Coriolous versicolor KR-11W 와 C. versicolor KR-65W) 2균주가 정치배양시 가장 우수한 효과를 나타내었다. C. versicolor KR-11W는 poly R-478을 13일만에, congo red를 7일만에 100%, methylene blue를 7일만에 90%까지 탈색시켰다. C. versicolor KR-65W는 poly R-478을 15일만에 100%, congo red를 7일만에 85%, methylene blue를 7일만에 100% 까지 탈색시켰다. 대조구로 사용한 Phanerochaete chrysosporium IFO 31249는 poly R-478 을 15일만에 약 35%, congo red를 7일만에 85%, methylene blue를 7일만에 95%까지 탈색시켰다.

구름버섯(Coriolus versicolor IFO 30388)에 의한 Poly R-478 염료의 탈색 (Decolorization of Poly R-478 Dye by Coriolus versicolor IFO 30388)

  • 구름버섯에 의한 Poly R-478염료의 탈색에 미치는 탄소원과 질소원의 영향을 조사하였다. 2.0% glucose, 0.04% ammonium tartrate, 2% $KH_2PO_4$, 0.5% $MgSO_4{\cdot}7H_2O$, 0.1% $CaCl_2{\cdot}2H_2O$, 0.002% thiamine-HCI, 10 mM 2,2-dimethylsuccinate(sodium)와 0.02% Poly R-478 염료 등으로 조성된 질소제한배지에서 균주를 $28^{\circ}C$에서 10일간 정치배양 했을때 염료의 탈색율은 87.2%로 나타났다. 염료의 탈색은 배지내에 질소원이 존재할 때에 일어났으며 탈색율은 배지내에 질소원이 고갈된 직후에 급격히 증가하였다.

Screening of Biodegradable Function of Indigenous Ligno-degrading Mushroom Using Dyes

  • The process of biodegradation in lingo-cellulosic materials is critically relevant to biospheric carbon. The study of this natural process has largely involved laboratory investigations, focused primarily on the biodegradation and recycling of agricultural by-products, generally using basidiomycetes species. In order to collect super white rot fungi and evaluate its ability to degrade lingo-cellulosic material, 35 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye. In the laccase enzymatic analysis chemical test, 33 white rot fungi and 2 brown rot fungi were identified. The degradation ability of polycyclic aromatic hydrocarbons (PAHs) according to the utilized environmental conditions was higher in the mushrooms grown in dead trees and fallen leaves than in the mushrooms grown in humus soil and livestock manure. Using Poly-R 478 dye to assess the PAH-degradation activity of the identified strains, four strains, including Agrocybe pediades, were selected. The activities of laccase, MnP, and Lip of the four strains with PAH-degrading ability were highest in Pleurotus incarnates. 87 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye on solid media. Using Poly-R 478 dye to assess the PAHdegrading activity of the identified strains, it was determined that MKACC 51632 and 52492 strains evidenced superior activity in static and shaken liquid cultures. Subsequent screening on plates containing the polymeric dye poly R-478, the decolorization of which is correlated with lignin degradation, resulted in the selection of a strain of Coriolus versicolor, MKACC52492, for further study, primarily due to its rapid growth rate and profound ability to decolorize poly R-478 on solid media. Considering our findings using Poly-R 478 dye to evaluate the PAH-degrading activity of the identified strains, Coriolus versicolor, MKACC 52492 was selected as a favorable strain. Coriolus versicolor, which was collected from Mt. Yeogi in Suwon, was studied for the production of the lignin-modifying enzymes laccase, manganese-dependent peroxidase (MnP), and lignin peroxidase (LiP).

Decolorization of Dyes by Selected Wood Degradation Fungus

  • The objectives of this study were to select superior fungus for lignin degradation and to decolor dyes by selected fungus. Ligninolytic fungi were screened and isolated from decayed woods. Ten ligninolytic fungi were selected by ligninolytic enzyme activity on the PDA media containing rhemazol brilliant blue R, guaiacol and gallic acid. Their lignin degradation abilities were tested on the extractive-free wood powder of Quercus acutissima and Pinus densiflora. As a result, 8J-28 was selected as superior fungus for lignin degradation. Also, decolorization abilities of dyes were examined by shaking and static culture. And congo red, crystal violet, poly R-478, methylene blue used to investigate decolorization abilities of dyes. As a result, 8J-28 showed over 90% in decolorization of congo red, crystal violet, poly R-478.

정제된 먹물버섯(Coprinus comatus) laccase에 의한 염료 탈색 (Decolorization of dyes by a purified laccase from Coprinus comatus)

  • 먹물버섯 Coprinus comatus를 yeast extract peptone dextrose 액체배지에서 배양하고 laccase의 생성 분비를 확인하였다. 이 때 분비된 laccase를 이온교환수지 방법과 전기영동 분획수집방법으로 순수하게 정제하였다. 정제된 효소단백질의 분자량은 SDS-PAGE로 분석하였을 때 약 67 kDa으로 나타났고 최적산도는 pH 4.3, 최적온도는 $25^{\circ}C$로 확인되었다. 기질 o-tolidine에 대한 Km 값은 0.45 mM, Vmax 값은 0.0132 OD/min/unit로 나타났다. 정제된 laccase를 사용하여 염료 RBBR과 Poly R-478의 탈색을 분석한 결과 배양 12시간 후에 RBBR은 49.3% 탈색되었고 poly R-478은 41.6% 탈색되었다.

Trametes sp. CJ-105에 의한 염료의 색도제거

  • Decolorization of congo red, methyl orange, poly R478, remazol brilliant blue R and crystal violet by white-rot fungus Trametes sp. CJ-105, isolated in Korea, was investigated. Remazol blue and methyl orange were almost completely decolorized after 2 days of culture, but congo red, crystal violet and poly R478 were decolorized by about 80%, 40% and 30% after 10 days of culture, respectively. As a result of determination of cell mass and enzyme activity, it was shown that color removal efficiency was related to cell mass and enzyme activity, and also found that only laccase (E.C.1.10.3.2) activity was existed in the culture broth. The decolorization ratios of remazol blue in the concentrations of 100ppm to 3, 000 ppm were 85% and above after 2 days of culture. In this study, we found that white-rot fungus, Trametes sp. CJ-105, was effective in decolorizing a wide range of structurally different synthetic dyes.

Fungal Growth and Manganese Peroxidase Production in a Deep Tray Solid-State Bioreactor, and In Vitro Decolorization of Poly R-478 by MnP

  • The growth of Irpex lacteus F17 and manganese peroxidase (MnP) production in a selfdesigned tray bioreactor, operating in solid-state conditions at a laboratory scale, were studied. The bioreactor was divided into three layers by three perforated trays. Agroindustrial residues were used both as the carrier of bound mycelia and as a nutrient medium for the growth of I. lacteus F17. The maximum biomass production in the bioreactor was detected at 60 h of fermentation, which was consistent with the CO2 releasing rate by the fungus. During the stationary phase of fungal growth, the maximum MnP activity was observed, reaching 950 U/l at 84 h. Scanning electron microscopy images clearly showed the growth situation of mycelia on the support matrix. Furthermore, the MnP produced by I. lacteus F17 in the bioreactor was isolated and purified, and the internal peptide sequences were also identified with mass spectrometry. The optimal activity of the enzyme was detected at pH 7 and 25℃, with a long half-life time of 9 days. In addition, the MnP exhibited significant stability within a broad pH range of 4-7 and at temperature up to 55℃. Besides this, the MnP showed the ability to decolorize the polymeric model dye Poly R-478 in vitro.

흰구름버섯(Coriolus hirsutus)에 의한 방향족 염료의 탈색 (Decolorization of Aromatic Dyes by White Rot Fungus Coriolus hirsutus)

  • 담자균류 백색부후균의 일종인 흰구름버섯(Coriolus hirsutus)을 실험균주로 하여 수종의 난분해성 방향족염료의 분해능을 측정하였다. 사용된 4종류의 염료 중, triphenyl methane 염료인 bromophenol blue가 탈색율 95% 이상으로 가장 잘 탈색되었으며, Congo red와 Poly R-478은 이보다는 낮은 57%, 55%가 탈색되었다. 그러나, heterocyclic 염료인 methylene blue는 본 균주에 의해 거의 탈색되지 않았으며, UV-visible spectrum상에서의 심색성 이동만 관찰되었다. 세포외 laccase와 peroxidase의 활성은 각 염료의 탈색율과 비례하여 나타났으며, 최대 활성 또한 최대 탈색시기에 관찰되었다. 효소의 활성 염색시 모든 염료의 탈색배지에서 공통적인 laccase와 peroxidase의 활성 띠가 관찰되었다. 이러한 결과로 볼 때, 세포외 laccase와 peroxidase가 난분해성 염료의 탈색에 중요한 역할을 할 것으로 판단된다.

목재 부후균의 리그닌 분해효소 활성과 염료 화합물의 탈색 (Production of Lignin Degrading Enzymes and Decolorization of Various Dye Compounds by Wood-Rot Fungi)

  • 목재부후균은 리그닌 분해효소로 lignin peroxidase (LIP), Mn-peroxidase (MNP) 및 laccase를 생성하는데 균류에 따라 위의 효소중 하나 혹은 둘 이상의 효소를 분비하거나 전혀 생성하지 않는 균도 있다. 본 실험은 이러한 목재 부후균의 효소생성 양상과 몇 종의 염료화합물 탈색과의 상관관계를 조사하고자 하였다. 조사한 23종 36균주 중 MNP 생성균은 30균주였으며 LIP 혹은 laccase 생성균은 각각 11균주와 12균주였다. 또한 같은 종에서도 효소활성은 다양한 양상을 보여 주었다. 리그닌 분해효소 활성과 비교하여 염료 탈색 정도는 세 효소가 모두 분비되는 백색 부후균의 경우 염료 탈색율이 상대적으로 우수하였고 균주에 따라 차이가 있으나 MNP 활성만을 갖는 균주의 경우, poly R-478 polymeric dye 및 anthron-type dye 인 remazol brilliant blue R염료는 효소 활성도와 다소 유연관계를 보였으며 methylene blue, bromophenol blue및 congo red 염료는 위의 효소들과는 직접적인 관련이 없는 것으로 판단되었으며, 오히려 균사의 생장과 비례하여 탈색율을 나타냈다. LIP, MNP 및 laccase 효소활성이 거의 검출되지 않은 갈색 부후균에서는 bromophenol blue를 제외하고는 염료의 탈색이 10%미만 혹은 전혀 탈색이 되지 앓았다.
