Psychrophilic Extremophiles from Antarctica: Biodiversity and Biotechnological Potential

  • Recently there has been a rapid accumulation of knowledge of microbial life in cold and frozen ecosystems. This understanding has revealed the extensive diversity of psychrophilic prokaryotes. Cultivation-based and molecular-based surveys have been performed in Antarctic habitats ranging from glacial ice to continental shelf sediments. Results indicate that psychrophilic taxa permeate throughout the Bacteria while they represent a more mysterious element of diversity in the Archaea owing to a notable lack of cultured strains. In certain cold climate ecosystems the diversity of psychrophilic populations reach levels comparable to the richest temperate equivalents. Within these communities must exist tremendous genetic diversify that is potentially of fundamental and of practical value. So far this genetic pool has been hardly explored. Only recently have genomic data become available for various psychrophilic prokaryotes and more is required. This owes to the fact that psychrophilic microbes possess manifold mechanisms for cold adaptations, which not only Provide enhanced survival and Persistence but Probably also contributes to niche specialisation. These mechanisms, including cold-active and ice-active proteins, polyunsaturated lipids and exopolysaccharides also have a great interest to biotechnologists.

Quantification of Reproductive Output of the Butter Clam, Saxidomus purpuratus(Sowerby, 1852) Using Enzyme-Linked Immunosorbent Assay (ELISA)

  • An immunological method was developed in this study to quantify reproductive output of the female butter clam, Saxidomus purpuratus. A clam egg-specific polyclonal antibody was developed using the purified butter clam egg as an antigen. An indirect Enzyme-Linked Immunosorbent Assay (ELISA) was used in quantitative measurement of the eggs. Size of the butter clam eggs ranged from $70.81{\pm}7.52{\mu}m$ in histology or $88.56{\pm}11.31{\mu}m$ in intact eggs. The predominant egg constituent was protein (37.44%), followed by lipids (11.40%) and carbohydrates (9.68%). The SDS-PAGE showed that the egg proteins are composed of several peptides of molecular weights consisting of 247, 200, 99, 91, 54 and 47 kDa. ELISA indicated that the clams collected from Geoje Island in May 2002 produced 8.2 to 26.8% of their body weight as eggs or 9,307,309 to 31,156,333 with a mean of 16,931,893 eggs per individual clam. The results of this study thus suggest that indirect ELISA using rabbit anti-clam egg IgG as a primary antibody is a rapid, affordable and sensitive method to assess reproductive output of 5. purpuratus and possibly other bivalves using a small amount of eggs.

Characterization of the Thermophilic Bacterium Geobacillus sp. Strain GWE1 Isolated from a Sterilization Oven

  • A gram-positive, rod-shaped, spore-forming, motile thermophilic bacterium was isolated from a sterilization oven. The microorganism GWE1, formally named Geobacillus wiegelii identified as a member of the genus Geobacillus. GWE1 grew under aerobic conditions of between $60-80^{\circ}C$ (optimum $670^{\circ}C$), in a pH range of 3.0-8.0 (optimum $pH^{70^{\circ}C}$ 5.8), and between 0 and 2 M NaCl (optimum 0.3 M). The membrane polar lipids were dominated by branched saturated fatty acids, which included as the major constituents; iso-15:0 (13.3%), 16:1(${\omega}7$) (12.8%), 16:0 (28.5%), iso-17:0 (13.5%) and anteiso-17:0 (12.3%). The DNA G+C content was 47.2 mol% (determined by HPLC). The 16S rRNA gene sequence of GWE1 showed a high similarity with Geobacillus caldoxylosilyticus (97%). However, the level of DNA-DNA relatedness was only 58%. These data suggest that GWE1 is probably a novel specie of the genus Geobacillus.

Temporal Variation in the Reproductive Effort and Tissue Biochemical Composition in Manila Clam, Ruditapes philippinarum from a Sand Flat on the East Coast of Jeju Island Korea

  • In the present study, we investigated temporal variation in the reproductive effort and biochemical contents in adult Manila clam Ruditapes philippinarum at Shi-Heung-Ri beach on the east coast of Jeju Island. Gonad-somatic index (GSI), a ratio of the egg mass to the total biomass determined using ELISA increased dramatically from late May to June (7.1% to 19.5%). In mid-July, GSI dropped to 15.1%, indicating spawning on a small scale during this period. GSI also declined dramatically from late August (15.5%) to September (4.3%), suggesting that massive spawning occurred during this period. A positive correlation was observed between clam size and potential fecundity, while potential fecundity ranged between 1.19-8.40 million eggs/clam. Total proteins and lipids in the tissue increased from late spring to mid-summer, coinciding with an increase in GSI. Protein, lipid and carbohydrate contents in the tissue were lowest in January, indicating that clams were suffering from poor nutritional circumstances, which may be associated with poor food supply from the environment. Monthly increase and/or decrease in the body weight were closely related to gonad maturation and subsequent spawning, which was also linked to temporal changes in the contents of protein, lipid and carbohydrates in the tissue.

Chitinimonas naiadis sp. nov., Isolated from a Freshwater River

  • A rod shaped, aerobic, Gram-stain-negative, and motile bacterium, strain $AR2^T$, was isolated from a water sample of Yeongsan river, Republic of Korea. Strain $AR2^T$ clustered closely with the members of the genus Chitinimonas and showed the highest 16S rRNA gene sequence similarity with Chitinimonas prasina $LY03^T$ (96.4%), Chitinimonas viridis $HMD2169^T$ (96.4%), Chitinimonas taiwanensis $cf^T$ (96.2%), and Chitinimonas koreensis $R2A43-10^T$ (94.2%). The predominant fatty acids of strain $AR2^T$ were identified to be summed feature 3 (comprising $C_{16:1}{\omega}7c$ and/or $C_{16:1}{\omega}6c$), $C_{16:0}$, and $C_{10:0}3-OH$. Diphosphatidylglycerol, phosphatidylglycerol, and phosphatidylethanolamine were found to be the major polar lipids. The genomic DNA G+C content was 60.4 mol%. Based on the polyphasic characterization, the isolated strain $AR2^T$ is described as a representative of a novel species in the genus Chitinimonas, for which the name Chitinimonas naiadis sp. nov. (type strain =$AR2^T$ =KCTC $42755^T$ =JCM $31504^T$) is proposed.

  • Monolayers of lipids on a water surface have attracted much interest as models of biological membranes, but also as precursors of multilayer systems promising many technical applications. Until now, many methodologies have been developed in order to gain a better understanding of the relationship between the structure and function of the monolayers. Maxwell displacement current (MDC) measurement has been employed to study the dielectric property of Langmuir-films. MDC flowing across monolayers is analyzed using a rod-like molecular model. It is revealed that the dielectric relaxation time $\tau$ of mono layers in the isotropic polar orientational phase is determined using a liner relationship between the monolayer compression speed a and the molecular area $A_m$. Compression speed a was about 30, 40, 50mm/min. also, LB layers of Arachidic acid deposited by LB method were deposited onto slide glass as Y-type film. The structure of manufactured device is Au/Arachidic acid/Al, the number of accumulated layers are 9 ~ 21 and we then examined of the Metal-Insulator-Metal(MIM) device by means of I-V.

Aureivirga callyspongiae sp. nov., Isolated from Marine Sponge Callyspongia elegans

  • A Gram-negative, aerobic, motile by gliding, and rod-shaped marine bacterium, designated CE67T was isolated from the marine sponge Callyspongia elegans on Biyang-do in Jeju Island. The CE67T strain grew optimally at 25℃, pH 7.5, and in the presence of 2-3% (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain CE67T was related to the genus Aureivirga and had the highest 16S rRNA gene sequence similarity to the Aureivirga marina VIII.04T type strain (96.3%). The primary fatty acids (>10%) of strain CE67T were iso-C15:0 (35.3%) and iso-C17:0 3OH (21.8%). The polar lipid profile of strain CE67T contained phosphatidylethanolamine, unidentified aminolipids, and unidentified lipids. The predominant menaquinone was MK-6. The DNA G+C content was 29.1 mol%. Based on the polyphasic taxonomic analysis, strain CE67T was determined to be a representative novel species of the genus Aureivirga for which we propose the name Aureivirga callyspongiae sp. nov., whose strain type is CE67T (=KCTC 42847T=JCM 34566T).

  • 신미종(辛味種)고추의 추숙(追熟)에 따른 지질(脂質)의 변화(變化)를 중성지질(中性脂質)과 극성지질(極性脂質)로 분획(分劃)하여 실험(實驗)한 결과(結果)는 다음과 같다. 1. 중성지질(中性脂質)의 함량(含量)은 cli. stage에서 최저치(最低値)가되고 극성지질(極性脂質)의 함량(含量)은 post-cli. stage에서 격감(激減)하고있다. 2. 중성지질(中性脂質)의 지방산(脂肪酸)으로서 주(主)가 되는 것은 linoleic acid이고 이것은 추숙(追熟)에 따라 점감(漸減)하고 있다. 3. 중성지질(中性脂質) fatty acid의 saturates/unsaturates치(値)가 cli. stage에서 최저(最低)가 되는데, 이것을 중성지질함량(中性脂質含量)도 cli.stage에서 최저(最低)를 가리키는 것과 아울러 생각할때 climacteric rise가중성지질(中性脂質)의 대사(代謝)와 관계(關係)가 있는것 같다. 4. 극성지질(極性脂質)의 지방산(脂肪酸) 조성(組成)은 추숙(追熟)에 따라 linoleic acid가 줄곧 감소(減少)하고 linoleic acid가 증가(增加)하여 cli. stage에서 역전(逆轉)하고 있는데, 이것은 고추의 climacteric rise에 따라 일어나는 막성분(膜成分)의 변화(變化)를 가리키는 한 지표(指標)가 될 수 있을 것 같다.

피조개 지질 및 지방산 조성에 관한 연구 (Studies on the Lipid and Fatty Acid Compositions of Ark-Shell, Anadara broughtonii)

  • 피조개 월별 생산량이 8~9월에 가장 많고, 피조개 소비는 여름철에 집중되고 있어, 여름산 피조개의 식품학적 특성을 구명하기위하여 불검화물함량, 요오드가, 지질조성 및 지방산조성을 분석하였다. 조지질함량은 0.83%, 불검화물함량은 20.19%였으며, 요오드가는 156.13이었다. 지질조성은 triglyceride가 59.46% 로 가장 높았고, 그 다음으로 diglyceride, hydrocarbon, sterol ester, free fatty acid, phospholipids, free sterol, monoglyceride의 순이었다. 총지질의 지방산조성의 경우, 함량이 특히 많은 것은 $C_{17 : 0}$, $C_{16 : 0}$, $C_{18 : 1}$ and $CT_{16 : 2}$, $C_{16:1}$로 이들 5종의 지방산이 전지방산의 66.55%를 차지하였다. 중성지질의 지방산조성의 경우, 함량이 특히 많은 것은 $C_{16 : 0}$, $C_{18 : 1}$, $C_{22 : 1}$로 이들 3종의 지방산이 전지방산의 52.93%를 차지하였으며, 극성지질의 경우는 $C_{16 : 0}$, $C_{18 : 2}$, $C_{20 : 5}$, $C_{22 : 6}$ 등의 4종의 지방산이 전지방산의 58.16%를 차지하였다. 구성지방산의 조성비를 비교하면, 총지질은 포화지방산> rnonoene 산>polyene산의 순으로 포화지방산이 가장 높았고, 중성지질은 monoene산>포화지방산> polyene산의 순으로 monoene산이 가장 높았고, 극성지질은 포화지방산>polyene산>monoene산의 순으로 포화지방산이 가장 높았다.e산의 순으로 포화지방산이 가장 높았다.

10주간 스마트머신 순환운동이 비만 중년여성의 체조성, 폐기능, 혈중지질 및 인슐린 저항성에 미치는 영향 (Effect of 10 Weeks Smart Machine Circulation Exercise on Body Composition, Lung Function, Blood Lipids and Insulin Resistance in Obesity Middle-aged Women)

  • 본 연구는 만 40-60세 비만 중년여성을 대상으로 10주간 스마트머신 순환운동이 체조성, 폐기능, 혈중지질 및 인슐린 저항성에 미치는 영향을 구명하기 위하여 운동군(n=8), 대조군(n=6)으로 구분하여 실시하였다. 스마트머신 순환운동은 주 3회, 회당 55분으로 유산소 운동의 강도는 스마트머신과 POLAR T31이 연동 되어 스마트머신에 적용되며, 1-4주차는 40-50%HRR, 5-8주차는 50-60%HRR, 9-10주차는 60-70%HRR을 적용하였고, 저항성 운동의 강도는 스마트머신을 이용하여 등속성 운동 기반으로 대상자들의 1-RM test의 데이터 값을 이용하여 1-4주차는 1-RM의 40%, 5-8주차는 1-RM의 60%, 9-10주차는 1-RM의 80%를 적용하여 실시하였다. 그 결과 체중, 체질량지수, 체지방율, 허리-엉덩이둘레비율에서 그룹×시기 간 상호작용 효과가 나타났다. 폐기능의 FVC는 시기 간 주 효과와 사후 그룹 간 유의한 차이가 나타났으며, FVC 및 FEV1은 그룹×시기 간 상호작용 효과가 나타났다. TC 및 TG는 시기 간 주 효과가 나타났으며, TC, TG 및 HDL-C는 그룹×시기 간 상호작용 효과가 나타났다. Insulin, Glucose 및 HOMA-IR은 운동 전·후 시기 간 차이에서 운동군이 유의하게 감소한 것으로 나타났다. 따라서 10주간 스마트머신 순환운동 프로그램이 비만 중년여성의 체조성, 폐기능, 혈중지질 및 인슐린 저항성에 긍정적인 영향을 미쳤으며 이는 중년여성의 비만을 개선하거나 비만을 예방할 수 있는 운동 프로그램이라고 사료된다.
