Volatile Components of Oyster Mushrooms (Pleurotus sp.) Cultivated in Korea (한국산 느타리 버섯류 (Pleurotus sp.)의 휘발성 향기성분)

  • Volatile aroma concentrates of four oyster mushrooms cultivated in Korea were obtained by the simultaneous distillation-extraction method. The volatile components were identified by the combined capillary gas chromatography-mass spectrometry and compared with the of retention data on GC of those authentic compounds. About 54 volatile compounds were identified in each of the four edible oyster mushrooms. The main compounds in Pleurotus ostreatus ASI 201 were 1-octen-3-ol, 3-octanone, 3-octanol, in Pleurotus sajor-caju, 1-octen-3-ol, 3-octanone, 1,5-octadien-3-one, 3-octanol, in Pleurotus florida-ostreatus-ostreatus, 1-octen-3-ol, n-hexanol, 3-octanone, 3-octanol, phenol, and in Pleurotus ostreatus (Ae-Neutari), 1-octen-3-ol, n-pentanal, n-hexanol, n-pentanol, 3-octanone, 3-octanol, 1,5-octadien-3-one, respectively. The mushrooms worked were rich in an alcohols and carbonyl compounds containing $C_8$ compounds. The peak area ratio of $C_8$ compounds in aroma concentrates were 56.60% in Pleurotus ostreatus ASI 201, 72.46% in Pleurotus sajor-caju, 54.84% in Pleurotus florida-ostreatus-ostreatus and 35.85% in Pleurotus ostreatus (Ae-Neutari ), respectively.

Comparisons of Soluble Protein Bands for Pleurotus Species and Interspecific Crosses of Pleurotus Species (느타리버섯속균과 교잡종에 대한 단백질 밴드에 의한 분류)

  • In comparison of three Pleurotus species and their selfed and crossed isolates using SDS-polyacrylamide gel electrophoresis of total soluble proteins, Pleurotus ostreatus 201 showed low similarity to selfed or P. ostreatus 201 crossed ones. Pleurotus ostreatus 2042 showed low similarity to selfed or P. ostreatus 2042 crossed ones. However, P. ostreatus $2042{\times}P$. ostreatus 202, P. ostreatus $2042{\times}P$. sajor-caju, and P. ostreatus $2042{\times}P$. ostreatus 900 showed high similarity. Pleurotus ostreatus 202 showed low similarity to selfed or crossed ones. Pleurotus sajorcaju showed low similarity to selfed or crossed ones. Pleurotus ostreatus 900 showed low similarity to selfed or crossed ones. However, selfed P. ostreatus and P. $ostreatus{\times}P$. florida showed high similarity. Pleurotus florida and selfed P. florida showed high similarity, too.

Cultural Conditions of Pleurotus ostreatus 201 for the Production of Cellulolytic Enzymes in Synthetic Medium (합성배지에서 저온성 느타리(Pleurotus ostreatus 201)의 섬유소분해효소 생산조건)

  • Cultural conditions and carbon sources affecting the productivity of cellulolytic enzymes of Pleurotus ostreatus 201 were examined in synthetic media. The optimum cultural temperature and initial pH for the production of enzymes were $25^{\circ}C$ and 5.5 in avicelase, and 3$0^{\circ}C$ and 5.0 In CMCase, and 3$0^{\circ}C$ and 6.5 in B-glucosidase. Among carbon sources, cellulose powder was the best for the production of avicelase, and Na-CMC for CMCase, and cellobiose for $\beta$-glucosidase. The optimum concentration of cellulose powder was 1.0% (w/v), and glucose depressed the production of enzymes remarkably.

  • Chemical composition of the oyster mushroom(Pleurotus spp.) at different cropping stage were analyzed. The contents of crude protein, soluble protein, reducing sugar, total sugar and crude fat in Pleurotus ostreatus 201 were a little higher than those of Pleurotus sajorcaju. The Component contents were not different from the first to the second flush, while the contents were slightly decreased from the third flush. A11 component contents were highest in $3{\sim}5cm$ pileus size, the contents were slightly decreased as pileus size increase to over $3{\sim}5cm$, except for reducing sugar. Crude fiber content in Pleurotus sajor-caju were a little more than Pleurotus ostreatus 201, and in course of increasing cropping time, ash contens of two strains were simmilar, and were decreased after the second flush. These component contents mere increased by the growth of the pileus. The sugars were identified as xylose, glucose, mannitol and trehalose in Pleurotus sajor-caju, and xylose, fructose, glucose, mannitol and trehalose in Pleurotus ostreatus 201, respectively. Mannitol and trehalose contents in Pleurotus sajor-caju were a little more than that in Pleurotus ostreatus 201. Glucose content of Pleurotus sajor-caju and Pleurotus ostreatus 201 was highest in $1{\sim}3cm$ pileus size, while other sugars in $3{\sim}5cm$ pileus size, and the sugar contents were decreased at over the above size of the pileus.

  • This study was performed to investigate the high yielding potential of Pleurotus ostreatus (Fr.) Quel derived from intermating and intramating of three strains: Local strain, Nonggi 201, and Pleurotus florida Eger. All the strains tested were tetrapola mating systems, and hybridable. Intermating system resulted in more fresh sporophores than those from intramating. The highest yield of fresh sporophores of P. ostreatus was obtained from Nonggi 201 crossed with P. florida, when cultivated on the rice straw substrates.

  • For 2 years $(1993{\sim}1994)$, study on development of cheap medium for Pleurotus ostreatus revealed that paper sludge contain more CaO and similar T-C, T-N, $P_2O_5$, MgO but less $K_2O$ than any other medium material in chemical property. Mixed treatment (rice straw + paper sludge 10, 30, 50%, cotton waste + paper sludge 10, 30, 50%, cotton waste + rice hull 20 + paper sludge 10, 30, 50, 70%) is similar or fast a little in mycelial growth and is similar or fast $1{\sim}2$ day in period of primordia formation than cotton waste medium, and in the yield to each medium type also increased but excepted in rice hull 20% + paper sludge 70%, especially mixed medium at 7:3 ratio of cotton waste and paper sludge is best treatment because it is increased to 21%. In economical analysis, mixed medium at 7:3 ratio of cotton waste and paper sludge is increased to 50% compared to cotton waste medium in relative income.

  • During cultivation of Pluerotus ostreatus, there were happened several problems by mushroom flies. Many mushroom farms were faced with the problem that mushroom production periods became shorter by mushroom fly. In this study, we examined effect of Lycoriella ingenua on the production of P. ostreatus. When we used 10 adult flies at the time of inoculation, mushroom production decreased by 10.8% as compared to that of the control. And when L. ingenua used more than 20 adult flies, contamination occurred in all treatments. And when L. ingenua used at the time of P. ostreatus mycelium cultured during 14 days, it was confirmed that the yield of P. ostreatus decreased in proportion to the number of L. ingenua.

  • Nutritional characteristics and cultural conditions for trehalose synthesis and mycelial yield of Pleurotus sajor-caju and Pleurotus ostreatus 201 in submerged culture were investigated. The results were as follows: Among the carbon sources, glucose was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of glucose was 1%. Among the nitrogen sources, peptone was most excellent for trehalose synthesis and mycelial yield. The optimal concentration of peptone was 0.05%. The optimal concentration of $KH_2PO_4$ and $MgSO_47H_2O$ for trehalose synthesis and mycelial yield was 0.1%,0.04% and 0.2%,0.04-0.08%, respective­ly. The optimal temperature and pH for trehalose synthesis were $25^{\circ}C$ and pH 5.5. but optimal temperature and pH for mycelial yield were $30^{\circ}C$ and pH 5.5. The maximum yield of trehalose was obtanined after 10 day cultivation.

Genetic Analysis of Cultivars in Pleurotus spp. of Korea by URP-PCR Polymorphism (URP-PCR 다형성에 의한 국내 느타리버섯 품종의 유전적 특성 분석)

  • Twelve URP primers were used to assess genetic characteristics of oyster mushroom including 59 Pleurotsu ostreatus cultivars, two of P. florida cultivars, one P. sajor-caju cultivars, one P. abalonus cultivar and two P. eryngii cultivars registered in Korea. Six URP primers produced PCR polymorphic bands within and between the Pleurotus species. Primer URP2F produced distinct cultivar specific PCR polymorphic bands that profiled to 15 cultivar types. PCR polymorphic bands amplified by URP2F, URP6R, URP4R and URP2R were used for UPGMA cluster analysis. Fifty nine cultivars of Pleurotus ostreatus are genetically clustered into 5 groups, showing genetic similarity over 70% among them and P. abalonus. P. eryngii and P. sajor-caju, were involved in outside groups.

Characteristics and breeding of a cultivar Pleurotus citrinopileatus 'Jangdari' (노랑느타리 품종 '장다리'의 육성 및 자실체 특성)

  • In Korea, oyster mushroom is one of the commonly cultivated mushrooms. In 2013, the cultivation areas and products of oyster mushrooms were 60,039 M/T and 201 ha, respectively. Many species of oyster mushrooms are cultivated in various countries. These include Pleurotus ostreatus, P. florida, P. sajor-caju, P. eryngii, P. citrinopileatus, P. salmoneostramineus and P. cystidiosus. P. citrinopileatus is a yellow oyster mushroom famous for its health benefits such as anti-cancer and anti-oxidant effects. Therefore, a cultivar P. citrinopileatus 'Jangdari' was developed to improve yield and the ability to grow well at lower temperatures. Two parent strains 'Gumbit (KMCC02150)' and 'KMCC02145' were selected based on their morphological characteristics. 'Jangdari' was developed by the method of Mon-Mon crossing between monokaryons derived from 'Gumbit' and 'KMCC02145', and finally selected through continuous cultivation tests. The optimum temperature for mycelial growth was $30^{\circ}C$. The cultivar could grow well at high temperatures, especially $16{\sim}24^{\circ}C$. Fruiting body production per bottle (850 mL) was about 90.0 g. Stipe length and thickness of 'Jangdari' were similar to those of 'Gumbit'. 'Jangdari' was more resistant to low temperature than 'Gumbit', and thus it could be cultivated with oyster mushrooms (P. ostreatus). In addition, while cultivating 'Jangdari', it is not required to scrape out the upper side of bottle's sawdust medium; hence, its cultivation is expected to save energy and time.
