Artesunate inhibits collagen-induced human platelets aggregation through regulation of PI3K/Akt and MAPK pathway (PI3K/Akt 및 MAPK 기전 조절을 통한 Artesunate의 콜라겐 유도의 사람 혈소판 응집 억제효과)

  • Excessive activation and aggregation of platelets is a major cause of cardiovascular disease. Therefore, inhibition of platelet activation and aggregation is considered an attractive therapeutic target in preventing and treating cardiovascular diseases. In particular, strong platelet activation and aggregation by collagen secreted from the vascular endothelium are characteristic of vascular diseases. Artesunate is a compound extracted from the plant roots of Artemisia or Scopolia species, and has been reported to be effective in anticancer and Alzheimer's disease fields. However, the effect and mechanism of artesunate on collagen-induced platelet activation and aggregation have not been elucidated. In this study, the effect of artesunate on collagen-induced human platelet aggregation was confirmed and the mechanism of action of artesunate was clarified. Artesunate inhibited the phosphorylation of PI3K/Akt and Mitogen-activated protein kinases, which are phosphoproteins that are known to act in the signal transduction process when platelets are activated. In addition, artesunate decreased TXA2 production and decreased granule secretion in platelets such as ATP and serotonin release. As a result, artesunate strongly inhibited platelet aggregation induced by collagen, a strong aggregation inducer secreted from vascular endothelial cells, with an IC50 of 106.41 µM. These results suggest that artesunate has value as an effective antithrombotic agent for inhibiting the activation and aggregation of human platelets through vascular injury.

  • Cardiovascular disease (CVD) is increasingly increasing as the main cause of death worldwide, and activation of platelet in vascular damage is one of the important causes of CVD. In recent, there is a growing interest in anti-thrombotic materials through platelet suppression, and efforts are being made to reduce side effects by using natural bioactive compounds. Known as one of the Flavonoids, hydroxygenkwanin (HGK) is a purified substance in Daphne Genkwa, which is known to have antibacterial, anti-inflammatory and anti-cancer effects, and has been reported to serve as an inhibitor of tissue factor that prevents thrombosis, but its anti-platelet effects and the action mechanisms is not known. In this study, we confirmed that the effects of HGK on the collagen-induced human platelets activation. HGK suppressed phosphorylation of PI3K/AKT and mitogen-activated protein kinases during platelet signaling, and reduced granule secretion in platelets such as ATP and serotonin. In addition, HGK inhibited the phosphorylation of cPLA2 and strongly undermined the production of TXA2, which is a powerful aggregation amplifier. As a result, the platelet aggregation derived by Collagen, a cohesive induced substance, was strongly suppressed by HGK to an IC50 of 86.36 µM. Therefore, HGK might be worth the antithrombotic substance that inhibits the activation and aggregation of human platelets that occur through blood vessel damage.

Geometrical imperfection and thermal effects on nonlinear stability of microbeams made of graphene-reinforced nano-composites

  • This research is related to nonlinear stability analysis of advanced microbeams reinforced by Graphene Platelets (GPLs) considering generic geometrical imperfections and thermal loading effect. Uniform, linear and nonlinear distributions of GPLs in transverse direction have been considered. Imperfection sensitivity of post-bucking behaviors of the microbeam to different kinds of geometric imperfections have been examined. Geometric imperfection is first considered to be identical as the first buckling mode, then a generic function is employed to consider sine-type, local-type and global-type imperfectness. Modified couple stress theory is adopted to incorporate size-dependent behaviors of the beam at micro scale. The post-buckling problem is solved analytically to derive load-amplitude curves. It is shown that post-buckling behavior of microbeam is dependent on the type geometric imperfection and its magnitude. Also, post-buckling load can be enhanced by adding more GPLs or selecting a suitable distribution for GPLs.

An Experimental Study On The Effect of Herba Segesbeckiae on the Intravascular Coagulation Induced by Endotoxin Rats (희첨이 Endotoxin으로 유발(誘發)된 흰쥐의 혈전(血栓)에 미치는 영향(影響))

  • In order to investigate the effect of the Herba Segesbeckiae on the thrombosis induced by endotoxin in rats. The rats were treated with solid extract of Herba Siegesbeckiae 41.9mg /200g(Sample) which were administered orally. And the thrombosis was induced by injecting endotoxin in the caudal vein rats. And then the number of platelets, concentration of fibrinogen, prothrombin time and FDP(fibrinogen degradation product) were measured. The results were as follows; 1. Platelet was increased significantly in sample group compared with control group. 2. Prothrombin time was shortened significantly in sample group compared with control group. 3. Fibrinogen was increased significantly in sample group compared with control group. 4. FDP was decreased significantly in sample group compared with control group. According to the above results, Herba Siegesbeckiae had significant effects on number of Platelets, Prothrombin time, Fibrinogen and FDP. therefore, it is considered that Herba Siegesbeckiae can be applied to diseases related to thrombosis.

  • Poly(3-hydroxybutyrate-co-3-hydroxyvalerate), poly(3HB-co-3HV), copolyesters, with 3-hydroxyvalerate (3HV) contents ranging from 17 to 60 mol%, were produced by Alcaligenes sp. MT-16, and their biocompatibility evaluated by the growth of Chinese hamster ovary (CHO) cells and the adsorption of blood proteins and platelets onto their film surfaces. The number of CHO cells that adhered to and grew on these films was higher with increasing 3HV content. In contrast, the tendency for blood proteins and platelets to adhere to the copolyester surfaces significantly decreased with increasing 3HV content. Examination of the surface morphology using atomic force microscopy revealed that the surface roughness was an important factor in determining the biocompatibility of theses copolyesters. The results obtained in this study suggest that poly(3HB-co-3HV) copolyesters, with >30 mol% 3HV, may be useful in biocompatible biomedical applications.

  • The intercalation chemistry of graphite presents an attractive route to obtain few-layered graphene platelets based on the expanded interlayer spacing. We report that the lithium can be intercalated into the graphite in a controllable manner by adjusting the variables such as temperature, pressure, and reaction time. From the X-ray diffraction experiments, the lithium-graphite intercalaltion compounds (Li-GICs) can be produced as the first stage compounds ($LiC_6$), the second-stage compounds ($LiC_{12}$), and the mixtures, which is most likely to be dependent on the temperature and reaction time. Since these Li-GICs are expected to facilitate the exfoliation of graphite, we investigated the feasibility of Li-GICs as a effective precursors for the generation of single-or few-layered graphite nano-platelets.

  • From in vivo and in vitro experiments using Korean Red Ginseng and its main components. ginsenoside saponins, it was found that ginesenoside may exerts its anti-thrombotic action by decreasing $TXA_2$ formation in platelets and increasing $PGI_2$ formation in vascular walls. This may certainly contribute to prevention and development of atherosclerosis and thrombosis by administration of Korean Red Ginsengs.

  • When three systems commonly used for screening(washed platelets suspended in uffer containing Ca2+, washed platelets in buffer without Ca2+and platelet rich plasma) are compared by studying the anti-aggregating capacity of garlic extract, platelet rich plasma was the least sensitive system. The most sensitive assay system, based on the IC50s of garlic extract and 2 other platelet aggregation-inhibiting agents(vitamin K3 and propranolol), was the platelet preparation without Ca2+in the suspension buffer. This system was confirmed as the most sensitive during subsequent investigation of garlic extract's capacity to inhibit platelet ATP release. These results suggest that applying the system with washed platelet without Ca2+is most effective to screen for the anti-affregating capacity of natural food.

  • Platelets serve many biological functions, including a major role in the haemostatic process. But platelets also play a crucial role in the formation of arterial thrombosis, arteriosclerosis and other pathologic processes. Thus, there have been many studies to develop new antiplatelet agents from foods and plants for decades. Inthis study, inhibitory effects of the oriental onion (Allium fistulosum) on platelet aggregation were investigated using platelet rich plasma (PRP). Water extracts of oriental onion was separated into two fractions (Fraction I and Fraction II) by Sephadex G-150 column. Platelet aggregations were inhibited by total water extracts as well as Fraction I and II. IC50 value of Fraction I was much lower than that of Fraction II. Inhibitory effects of total water extracts of oriental onion on ATP release by PRP were also observed.

Inhibition of Cyclooxygenase/Lipoxygenase from Human Platelets by Polyhydroxylated/Methoxylated Flavonoids Isolated from Medicinal Plants

  • Various flavonoid derivatives were previously reported to possess the inhibitory activity on cyclooxygenase/lipoxygenase. And these properties of flavonoids might contribute to their anti-inflammatory activity in vivo. In this study, several polyhydroxylated/methoxylated flavonoid derivatives such as oroxylin A. wogonin, skullcapflavone II, tectorigenin and iristectorigenin A were isolated from the medicinal plants. these compounds were evaluated fro their inhibitory effects on cyclooxygenase/lipoxygenase from the homogenate of human platelets in vitro. It was found that isoflavones including daidzein and tectorigenin possessed the inhibitory activity on cycloooxygenase, although the potency of inhibition was far less than that of indomethacin. In addition, oroxylin A, baicalein and wogonin inhibited 12-lipoxygenase activity without affecting cyclooxygenase, which suggested that 5,6,7- or 5,7,8-trisubstitutions of A-ring of flavone gave favorable results. The IC50 values of oroxylin A and NDGA against 12-lipoxygenase were found to be 100 and 1.5 uM, respectively.
