Urokinase의 적출심장의 심근허혈에 대한 보호작용과 invitro 및 exvivo항혈전작용 실험 (Protective Effect of Urokinase on Reperfusion Function in Isolated Perfused Rat Heart, and Anti-platelet Aggregation Effect Invitro and Exvivo)

  • Protective effect of urokinase on reperfusion were studied followed by global ischemia in the isolated perfused rat heart. Separately, anti-platelet aggregation effect of urokinase also investigated. Urokinase exhibited positive effect for the protection of rat heart function by increasing the LV dp/dt, coronary flow(CF) and the Tate pressure product(RPP), and by decreasing the LVEDP on reperfusion. Urokinase also decreased arrhythmia by $74.7\%(P<0.05) induced by global ischemia in the rat heart. In the platelet aggregation study, urokinase did not show the inhibitory effect of ADP or collagen induced platelet aggregation inviuo and exvivo.

Inhibitory Effect of Ginkgolide B on Platelet Aggregation in a cAMP- and cGMP-dependent Manner by Activated MMP-9

  • Extracts from the leaves of the Ginkgo biloba are becoming increasingly popular as a treatment that is claimed to reduce atherosclerosis, coronary artery disease, and thrombosis. In this study, the effect of ginkgolide B (GB) from Ginkgo biloba leaves in collagen (10 ${\mu}g/ml$)-stimulated platelet aggregation was investigated. It has been known that human platelets release matrix metallo-proteinase-9 (MMP-9), and that it significantly inhibited platelet aggregation stimulated by collagen. Zymographic analysis confirmed that pro-MMP-9 (92-kDa) was activated by GB to form an MMP-9 (86-kDa) on gelatinolytic activities. And then, activated MMP-9 by GB dose-dependently inhibited platelet aggregation, intracellular $Ca^{2+}$ mobilization, and thromboxane $A_2$ ($TXA_2$) formation in collagen-stimulated platelets. Activated MMP-9 by GB directly affects down-regulations of cyclooxygenase-1 (COX-1) or $TXA_2$ synthase in a cell free system. In addition, activated MMP-9 significantly increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), which have the anti-platelet function in resting and collagen-stimulated platelets. Therefore, we suggest that activated MMP-9 by GB may increase the intracellular cAMP and cGMP production, inhibit the intracellular $Ca^{2+}$ mobilization and $TXA_2$ production, thereby leading to inhibition of platelet aggregation. These results strongly indicate that activated MMP-9 is a potent inhibitor of collagen-stimulated platelet aggregation. It may act a crucial role as a negative regulator during platelet activation.

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that ethyl acetate layer of RVS had strong anti-aggregatory activity. In this study. to investigate the anti-aggregatory activity and antioxidative effects of RVS ethyl acetate layer. the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH). As a result. the fraction 3 was most inhibited the aggregation of platelet in rat whole blood induced by thrombin and all fraction of RVS was detected strong antioxidative effect. (omitted)

  • The extract of Cudrania tricuspidata is used in ethnomedicine throughout Eastern Asia in China, Korea and Japan. In Korean traditional medicine, Cudrania tricuspidata has been used to treat eczema, mumps, tuberculosis, contusions, insomnia and acute arthritis. In addition, it has been reported that root extract of Cudrania tricuspidata has anti-platelet effects. Therefore, we investigated which compound in Cudrania tricuspidata has inhibitory effect on platelet aggregation. In this study, we tried to explain the inhibitory mechanism of steppogenin from Cudrania tricuspidata on human platelet aggregation. Collagen-induced human platelet aggregation and [Ca2+]i mobilization were dose-dependently inhibited by steppogenin and we determined the inhibition by steppogenin is due to the down regulation of extracellular-signal-regulated kinase(ERK) and inositol-1,4,5-triphosphate receptor type I(IP3RI) phosphorylation. In addition, steppogenin inhibited collagen-induced fibronectin adhesion to αIIb/β3 and thromboxane A2 generation. Thus, in the present study, steppogenin showed an inhibitory effect on human platelet aggregation, suggesting its potential use for preventing platelet-induced cardiovascular disease.

양파와 알로에 복합물이 혈소판 응집에 미치는 영향 (Anti-platelet Effects of Mixtures of Onion and Aloe Extract)

  • In this study, we investigated whether the mixtures of onion water extract and aloe ethanol extracts have antiplatelet activities. The mixtures inhibited collagen- and thrombin-induced rat platelet aggregation in vitro. Additionally, the oral administration of the mixtures inhibited platelet aggregation induced by collagen ex vivo but not prolonged mouse tail vein bleeding time in vivo. These results suggest that the combination of onion and aloe extracts has a potential to be a preventive agent against platelet-mediated disorders.

Anti-Thrombotic Effects of Egg Yolk Lipids In Vivo

  • In this study, we investigated the effect of egg yolk lipids (EYL) on collagen ($10\;{\mu}g/ml$)-stimulated platelet aggregation in vivo. Dietary EYL significantly inhibited collagen-induced platelet aggregation, in addition, increased the formation of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), intracellular $Ca^{2+}$-antagonist as aggregation-inhibiting molecules, in collagen-stimulated platelets. These results suggest that EYL inhibits the collagen-induced platelet aggregation by up-regulating the cAMP and cGMP production. On the other hands, prothrombin time (PT) on extrinsic pathway of blood coagulation was potently prolonged by dietary EYL in vivo. These findings suggest that EYL prolongs the internal time between the conversion of fibrinogen to fibrin. Accordingly, our data demonstrate that EYL may be a crucial tool for a negative regulator during platelet activation and blood coagulation on thrombotic diseases.

모링가 지하부의 항혈전 활성 (Anti-thrombosis Activities of the Root Extract of Moringa oleifera Lam)

  • 열매, 잎, 꽃, 봉우리, 꼬투리 등의 지상부를 약용 및 식용으로 사용하고 있는 모링가는 miracle tree라고 불리고 있다. 그러나, 현재까지 주로 잎과 씨에 대한 연구가 진행된 바, 지하부에 대한 성분 및 효능 연구는 초보적인 상태이다. 본 연구에서는 모링가 지하부의 유용 생리활성을 평가하기 위해 지하부의 ethanol 추출물 및 이의 순차적 유기 용매 분획물을 조제하고 이의 혈액응고 저해 및 혈소판 응집저해 활성을 평가하였다. 그 결과, 모링가 지하부 ethanol 추출물은 항응고 활성이 미약하고, 혈소판 응집 촉진효과를 나타내어 지혈작용을 나타내었으나, 추출물의 hexane 및 ethylacetate 분획에서는 우수한 트롬빈, 프로트롬빈, 혈액응고인자 저해와 함께 혈소판 응집저해 활성을 나타내었다. 또한 모링가 지하부 추출물 및 분획물들은 1.0 mg/ml 농도까지 인간 적혈구 용혈활성이 없음을 확인하여, 상기 분획물들이 신규의 항혈전제로 사용 가능함을 제시하였다.

Platelet Shape Changes and Cytoskeleton Dynamics as Novel Therapeutic Targets for Anti-Thrombotic Drugs

  • Platelets play an essential role in hemostasis through aggregation and adhesion to vascular injury sites but their unnecessary activation can often lead to thrombotic diseases. Upon exposure to physical or biochemical stimuli, remarkable platelet shape changes precede aggregation or adhesion. Platelets shape changes facilitate the formation and adhesion of platelet aggregates, but are readily reversible in contrast to the irrevocable characteristics of aggregation and adhesion. In this dynamic phenomenon, complex molecular signaling pathways and a host of diverse cytoskeleton proteins are involved. Platelet shape change is easily primed by diverse pro-thrombotic xenobiotics and stimuli, and its inhibition can modulate thrombosis, which can ultimately contribute to the development or prevention of thrombotic diseases. In this review, we discussed the current knowledge on the mechanisms of platelet shape change and also pathological implications and therapeutic opportunities for regulating the related cytoskeleton dynamics.

흑생강(Kaempferia parviflora)의 항응고 및 혈소판 응집저해 활성 (Anti-coagulation and Anti-platelet Aggregation Activities of Black Ginger (Kaempferia parviflora))

  • 흑생강은 생강과 초본과 뿌리식물로 태국 및 라오스에서는 크라차이담으로 불리며, 뿌리는 향신료와 차로 이용되어 왔다. 말린 뿌리는 위장장애, 통풍, 이질, 알러지 치료 및 강장용으로 전통적으로 이용되어 왔으며, 최근에는 항비만, 항산화, 항염증, 혈전 용해 활성 등의 다양한 유용 생리활성이 보고되어 있다. 본 연구에서는 흑생강의 혈액순환 개선 활성을 확인하기 위해, 흑생강 지하부의 ethanol 추출물 및 열수 추출물을 조제하고 각각 이의 순차적 유기용매 분획물인 hexane 분획물, ethylacetate (EA) 분획물, butanol 분획물 및 물 잔류물을 조제하여 각각의 성분을 분석하였고, 혈전 생성과 관련된 항응고 활성, 혈소판 응집저해 활성 및 적혈구 용혈활성을 평가하였다. 그 결과, ethanol 추출물의 EA 분획물 5 mg/ml 농도에서 TT, PT, aPTT를 각각 1.22, 1.49 및 15배 이상 연장시켜 강력한 항응고 활성을 나타냄을 확인하였으며, ethanol 및 열수 추출물의 EA 분획물 모두에서 동량의 아스피린보다 강력한 혈소판 응집저해 활성을 확인하였다. 또한 상기 EA 분획물들은 0.5 mg/ml 농도까지 인간 적혈구에 대한 용혈활성을 나타내지 않았다. 본 연구 결과는 흑생강의 혈전 생성 억제 활성에 대한 최초의 보고이며, 흑생강의 EA 분획물이 인간 적혈구 용혈활성 없이 강력한 항응고 및 혈소판 응집저해 활성을 나타내어 신규의 항혈전제로 이용 가능함을 제시하고 있다.

꾸지뽕나무 뿌리 추출물의 cGMP에 의한 VASP 인산화 기전을 통한 항혈소판 효과 (Cudrania Tricuspidata root extract (CTE) has an anti-platelet effect via cGMP-dependent VASP phosphorylation in human platelets)

  • 꾸지뽕나무 뿌리 추출물은 항염, 항암, 항산화와 같은 효과를 갖는 많은 생리활성 물질을 포함하고 있다고 알려져 있다. 그러나 꾸지뽕나무 뿌리 추출물(이하 CTE)의 사람 혈소판 응집 억제 기전에 관하여는 알려진 바 없다. 본 연구에서는 CTE가 혈소판에 미치는 영향을 확인하고자 하였다. CTE는 collagen으로 유도한 혈소판 응집 반응에서 cyclooxygenase-1 활성을 억제하고, 세포 내 칼슘 농도를 낮추는 방식으로 thromboxane A2 생성을 억제하였다. 또한, phospholipase Cγ2와 syk의 인산화 반응을 억제하였으며, guanosine monophosphate (cGMP)에 의존적인 방식으로 vasodilator-stimulated phosphoprotein(VASP)의 Ser239 위치를 인산화하여 항혈소판 효과를 나타내었다. 또한, 고지방 식이로 혈소판 활성화를 유도한 랫드에서 CTE를 경구 투여 하였을 때, 간독성 없이 콜라겐으로 유도한 혈소판 응집반응을 thromboxane A2 생성을 억제함으로써 항혈소판 효과를 보였다. 이는 in vivo와 in vitro에서 같은 결과를 제시하고 있다. 결론적으로, CTE는 항혈소판 작용 및 심혈관계 질환 예방을 위한 천연물 유래의 안전하고, 새로운 물질임을 제시하는 바이다.
