Fabrication of Nano-filter Device for High Efficient Separation and Concentration of Biomolecules (고효율 바이오물질 분리 및 농축을 위한 나노필터소자제작)

  • Here, we develop a new nanofilter device for the rapid and efficient separation of nanoparticles and biomolecules, exploiting the use of AAO mebrane with ordered nanopores in the range from 20 nm to 200 nm. Briefly, the chip comprises of a series of the upper and lower PDMS channels containing embedded inlet and outlet ports, and $50{\mu}m$ width microfluidic channel, and AAO membrane to be made the filtering zone. After assembling these components, the acrylate plastic plates were used to fix the device on the top and bottom side. When introducing the samples into the inlet ports of the upper PDMS channel, we were able to separate and concentrate the nanoparticles and target molecules at the filtering zone, and to elute the solutions containing the unwanted materials toward the lower PDMS channels normal to the direction of AAO membrane. To demonstrate the usefulness of the device we apply it to the SERS detection of nucleic acid sequences associated with Dengue virus serotype 2. We report a limit of detection for Dengue sequences of 300 nM and show excellent enhancement of Raman signals from the filter zone of the nanofilter device.

  • The Fluorescence-Activated Cell Sorter (FACS) is a well-established instrument used for identifying, enumerating, classifying and sorting cells by their physical and optical characteristics. For a miniaturized FACS device, a disposable plastic microchip has been developed which has a hydrodynamic focusing chamber using soft lithography. As the characteristics of the spatially confined sample stream have an effect on sample throughput, detection efficiency, and the accuracy of cell sorting, systematic fluid dynamic studies are required. Flow visualization is conducted with a laser scanning confocal microscopy (LSCM), and three-dimensional flow structure of the focused sample stream is reconstructed from 2D slices acquired at $1\mutextrm{m}$ intervals in depth. It was observed that the flow structure in the focusing chamber is skewed by unsymmetrical velocity profile arising from trapezoidal cross section of the microchannel. For a quantitative analysis of a microscopic flow structure, Confocal Micro-PIV system has been developed to evaluate the accelerated flow field in the focusing chamber. This study proposes a method which defines the depth of the measurement volume using a detection pinhole. The trajectories of red blood cells (RBCs) and their interactions with surrounding flow field in the squeezed sample stream are evaluated to find optimal shape of the focusing chamber and fluid manipulation scheme for stable cell transporting, efficient detection, and sorting

  • The aim of this experiment is to compare the healing process of extraction sockets after immediate implant placement with those using autogenous bone grafts and guided tissue regeneration with Gore-Tex. The first lower premolars and the second premolars of six experimental dogs were extracted and Stryker fin type implants were placed into the extraction sockets immediately after extraction. In the control group, any graft materials were'not used and the dead space around implants was left in itself and covered with only periosteum. In the experimental group A, implants were covered with Gore tex without any bone grafts, and in the experimental group B, the dead space around implants was filled with the bone chips gained from drilling procedure. Each experimental dogs were sacrificed at the 1st, 2nd, 3rd, 6th, and 8th week and the specimens were observed by gross examination, radiological examination, and light microscopic examination. The following results were obtained. 1. Well healed soft tissue and no mobility of the implants were observed in control and two experimental groups. 2. In the radiogical examination, radiopacity around implants had been increased gradually. 3. In the microscopic examination, there were good healing process and active new bone formation in both in the experimental groups, Especially the more amount of new bone formation occurred in the experimental group B using bone chips. 4. Bone chip grafts and guided tissue regeneration (GTR) using Gore-Tex may be one of the successful methods in the immediate implantation.

GENE EXPRESSION AFTER THE APPLICATION OF THE FLUID-INDUCED SHEAR STRESS ON THE GINGIVAL FIBROBLAST (유체에 의해 유발된 전단력이 치은 섬유아세포 유전자 발현 변화에 미치는 영향에 관한 연구)

  • The oral cavity is humid environment mainly due to the continuous salivary flow. The reaction of oral mucosa to fluid flow is important for homeostasis and pathogenesis. The objective of this study is the screening the change of gene expression after the application of fluid induced shear stress (FISS) on the gingival fibroblast using cDNA microarray assay. The immortalized human gingival fibroblasts were grown and FISS was applied using a cone viscometer at a rotational velocity of 40 rpm, respectively for periods of 2 and 4 hours. The synthesis of cDNA was done from the extracted total RNA and cDNA microarray assay was done subsequently. The genes that showed over 1.6 in the Cy3/Cy5 or the Cy5/Cy3 value were regarded as genes influenced significantly by the FISS application ion (/M/>0.7). The " RUNX-1" was increased its expression in 2 hours group and " RUN and SH3 domain containing 1" was increased its expression in 4 hours group. The "CC020415", "cyclin L1", "interferon regulatory factor1", "early growth response 1", "immediate early response 2", and "immediate early response 3" genes were increased their expression in 2 and 4 hours after FISS application. In conclusion, we could find many genes that were probably related to the FISS application. Interestingly, most of them were placed in similar molecular pathways and these findings improve the reliability of chip data and usefulness in overall screening. From this experiment, we could find many items for further study and it will make improvement in the understanding of intracellular events in response to FISS.

Nonhermetic Plastic Packaged Optical Modules of Passive Optical Fiber Alignment Method (수동 광섬유정렬을 이용한 Nohermetic 플라스틱 패키지 광모듈)

  • In this paper, We proposed a efficient OSA(Optical Sub-Assembly) packaging method in use 1.31/1.49um bi-directional hybrid-integrated PLC chip for low-cost OSA in optical access network system applications as GE-PON in FTTH. Fabricated OSA with passive optical fiber alignment and nonhermetic plastic package method and measured optical coupling efficiency and electric-optical characteristics. Its performance is feasible to satisfy the GE-PON ONU specifications with the results as less than 0.5dB coupling losses within 40um alignment of z-axis and less than -24dBm sensitivity. It also has good temperature characteristics to sustain optical output power more than 1.5mW and 10dB extinction ratio, less than 0.3dB tracking error.

Effects of Coir Substrate Application and Substrate Volume on the Growth and Yields of Strawberry in a Hydroponically Cultured System (딸기 수경재배에 코이어 배지 적용과 근권부 배지 용량이 생육 및 수확량에 미치는 영향)

  • This study was conducted to examine an alternative cultivating method that uses coir substrates in a hydroponically cultured system. Three treatment conditions were applied with one-layer substrate (10 cm height) with a coir chip and dust ratio of 5:5 (Treatment A), two-layer coir substrate (20 cm height) with a coir chip and dust ratio of 5:5 (Treatment B), one-layer coir substrate (15 cm height) with a coir chip and dust ratio of 7:3 (Treatment C). The control condition was a plastic container filled with a coir chip and dust ratio of 5:5. Various criteria were measured and compared between the treatments and the control. The yield of strawberry was smaller in the control than in the treatments. No significant difference in growth characteristic was found in the height treatments of the coir substrates. The net photosynthetic rate of the treatments was 14.68-15.76 µmol CO2·m-2·s-1. This does not show a statistically significant difference. The root activity was better in treatment B and C than in treatment A and the control. The length and width of leaves were measured as 4.04-4.13 cm and 3.26-3.34 cm. These results are not statistically significant. The leaf length and width ratio was 1.27 in the control and 1.24 in the treatments. The findings show that no statistically significant benefit was found when utilizing coir substrates with different height treatments in the hydroponic culture system. However, the harvested fruit per plant weights 72.38 g in treatment A and 48.69 g in treatment C. The number of harvested fruit was least in treatment C in which a coir chip and dust ratio of 7:3 was applied. Therefore, further research is needed to examine how the chip and dust ratio in coir substrate affects growth characteristics.

  • Wear mechanisms may be briefly classified by mechanical, chemical and thermal wear. A plane strain finite element method is used with a new material stress and temperature fields to simulate orthogonal machining with continuous chip formation. Deformation of the workpiece material is healed as elastic-viscoplastic with isotropic strain hardening and the numerical solution accounts for coupling between plastic deformation and the temperature field, including treatment of temperature-dependent material properties. Effect of the uncertainty in the constitutive model on the distributions of strait stress and temperature around the shear zone are presented, and the model is validated by comparing average values of the predicted stress, strain, and temperature at the shear zone with experimental results.

  • [ $Moir{\acute{e}}$ ] interferometry is an optical method, providing whole field contour maps of in-plane displacements with high resolution. The demand for enhanced sensitivity in displacement measurements leads to the technique of microscopic $moir{\acute{e}}$ interferometry. The method is an extension of the $moir{\acute{e}}$ interferometry, and employs an optical microscope for the required spatial resolution. In this paper, the sensitivity of $moir{\acute{e}}$ interferometry is enhanced by an order of magnitude using an immersion interferometry and the optical/digital fringe multiplication(O/DFM) method. In fringe patterns, the contour interval represents the displacement of 52 nm per fringe order. In order to estimate the reliability and the applicability of the optical system implemented, the measurements of rigid body displacements of grating mold and the coefficient of thermal expansion(CTE) for an aluminium block are performed. The system developed is applied to the measurement of thermal deformation in a flip chip plastic ball grid array package.

A Study on the Grinding Characteristics of the Quartz (Quartz의 연삭 특성에 관한 연구(I))

  • This investigation reports the grinding characteristics of quartz. Grinding experiments were performed at various grinding conditions including wheel mesh, table speed and depth of cut. The grinding forces and specific grinding energies were measured to compare the grindability of quartz with those of structural ceramics such as A1$_2$O$_3$, SiC, Si$_3$N$_4$ and ZrO$_2$. Surface roughness was also measured with tracer and the ground surfaces were observed with SEM. The chip formation energy of quartz was about 6J/㎣, which is quite smaller than those of structural ceramics. Although plastic flows are occured in Si$_3$N$_4$ and ZrO$_2$, micro/macro cracks are occured in ground surface of quartz like in A1$_2$O$_3$ and SiC.

  • Thermo-mechanical and flexural behavior of a wire-bond plastic ball grid array (WB-PBGA) are characterized by high sensitive $moir{\acute{e}}$ interferometry. $Moir{\acute{e}}$ fringe patterns are recorded and analyzed at several various bending loads and temperature steps. At the temperature higher that $100^{\circ}C$, the inelastic deformation in solder balls became more dominant. As a result the bending of the molding compound decreased while temperature increased. The strain results show that the solder ball located at the edge of the chip has largest shear strain by the thermal load while the maximum average shear strain by the bending moment occurs in the end solder. The results also show that $moir{\acute{e}}$ interferometry is a powerful and effective tool in experimental studies of electronic packaging.
