Differential Expression Patterns of Gangliosides in the Liver and Heart of NIH-miniature Pigs (NIH-미니돼지의 간과 심장에서 갱글리오시드의 서로 다른 발현 패턴)

  • Gangliosides are a major component of the plasma membrane of mammalian cells, which are directly involved in a variety of immunological events, including cell-to cell or cell-to-protein interactions. In this study, we investigated whether gangliosides, sialic acid-containing glycosphingolipids, are related to rejection during the xenotransplantation of NIH-miniature pig livers and hearts to humans. Both high performance thin-layer chromatography and immunohistochemistry analyses revealed that the expression of gangliosides in the liver tissue of NIH-miniature pigs was higher than that in the heart. Gangliosides GD3, GD1a, GD1b, GT1b and GQ1b were observed in both the liver and heart, whereas GQ1b was detected only in the liver, indicating that the ganglioside expression profiles are tissue specific. Moreover, other ganglio-series gangliosides, including GM3, were not detected in the livers and hearts of NIH-miniature pigs. Taken together, these results suggest that gangliosides may play important roles in immune responses in clinical xenotransplants of pig livers and hearts.

  • 수소화된 비정질 실리콘 박막을 이용한 반도체는 현재 태양전지, 트랜지스터, 매트릭스 배열 및 이미지 센서 등의 분야에서 이용되고 있다. 자세히 이야기 하면, 여러 가지의 광전효과 물질에 대한 특성이 있으며, 가시광선영역에 대하여 > $10^5cm^{-1}$이상의 매우 높은 광흡수계수와 낮은 온도를 갖는 증착공정 등이 있다. 박막의 밴드갭은 약 1.6~1.8eV로서 태양전지의 흡수층과 passivation층으로 적절하다. 여러 가지 종류의 태양전지 중 비정질 실리콘 박막/결정질 실리콘 기판의 구조로 이루어진 이종접합 태양전지는 저온에서 공정이 가능한 대표적인 것으로서 HIT(Heterojunction with Intrinsic Thin layer)구조로 산요사에 의해 제안된 것이다. 이것은 결정질 실리콘 기판과 도핑된 비정질 실리콘 박막사이에 얇은 진성층 비정질실리콘 박막을 삽입함으로서, 캐리어 전송을 좋게하여 실리콘 기판 표면의 passivation효과를 증대시키는 결과를 가지고 온다. 실험실 규모에서는 약 20%이상의 효율을 보이고 있으며, 모듈에서는 19.5%의 높은 효율을 보이고 있어 실리콘 기판을 이용한 고효율 태양전지로서 각광을 받고 있다. 이러한 이종접합 태양전지의 대부분은 단결정 실리콘을 사용하고 있는데, 점차적으로 다결정 실리콘 기판으로 추세가 바뀌고 있어, 여기에 맞는 표면 passivation 공정 및 분석이 필요하다. 본 발표에서는 다결정 실리콘 기판위에 진성층 비정질 실리콘 박막을 유도결합 플라즈마 화학기상 증착법(ICP-CVD)을 이용하여 제조하여 passivation 효과를 분석한다. 일반적으로 ICP는 CCP(coupled charged plasma)에 비해 약 100배 이상 높은 플라즈마 밀도를 가지고 있으며, 이온 충돌같은 표면으로 작용하는 것들이 기존 방식에 비해서 작다라는 장점이 있다. 먼저, 유리기판을 사용하여 ICP-CVD 챔버내에 이송 한 후 플라즈마 파워, 온도 및 가스비(SiH4/H2)에 따른 진성층 비정질 실리콘 박막을 증착 한 후, 밴드갭, 전도도 및 결합구조 등에 대한 결과를 분석한 후, 최적의 값을 가지고 250um의 두께를 갖는 다결정 실리콘을 기판위에 증착을 한다. 두께(1~20nm)에 따라 표면의 passivation이 되는 정도를 QSSPCD(Quasi steady state Photoconductive Decay)법에 의하여 소수캐리어의 이동거리, 재결합율 및 수명 등에 대한 측정 및 분석을 통하여 다결정 실리콘 기판의 passivation effect를 확인한다. 제시된 데이터를 바탕으로 향후 다결정 HIT셀 제조를 통해 태양전지 효율에 대한 특성을 비교하고자 한다.

The Effects of Astragali Radix Pharmacopuncture at $CV_{12}$ on Osteoporosis of Senescence Accelerated Mice (SAM) P6 (중완(中脘)($CV_{12}$) 황기약침이 노화촉진 생쥐의 골다공증 개선에 미치는 영향)

  • Objectives : The purpose of this study was to examine the effects of the Astragali radix Pharmacopuncture on improvement of senile osteoporosis (Type 2) in SAM P6. Methods : At 10 weeks after birth, Astragali radix Pharmacopunctures were given 100mL, 200mL/kg/day, i.p. 3times a week for 4 weeks at $CV_{12}$ in SAM P6 mice. We measured complete blood cells (CBC) such as RBC, HGB, Hct, PLT, MPB and MCHC. And we analyzed the plasma concentrations of blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : The results were as follows. 1. At $CV_{12}$ in SAM P6, treatments of Astragali radix Pharmacopuncture were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales, femurand tibia in P6 mice. 2. At $CV_{12}$ in SAM P6, treatment of Astragali radix Pharmacopuncture increased in RBC, HB, HCT and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased in the treatment on groups of the Astragali radix Pharmacopuncture from that of the control group, but blood urea nitrogen was no significant. Conclusions : These results are suggested that at $CV_{12}$ the Astragali radix Pharmacopuncture help on improvement of osteoporosis in SAMs.

Diffuse Nodular Interstitial Infiltrations with Bilateral Hilar Lymphadenopathy (양측 폐문 림프절 종대를 동반한 간질성 결절성 폐침윤)

  • Lymphocytic interstitial pneumonia(LIP) is an uncommon condition in which the alveolar septa and extra-alveolar interstitial space are markedly expanded by small lymphocytes, plasma cells and histiocytes. Chest radiographs generally show nonspecific patterns with the most common pattern showing bibasilar reticular or reticulonodular infiltrates. Hilar or mediastinal lymphadenopathy and pleural effusions are usually absent. We encountered a 42-year-old female patient who was admitted to hospital because of exertional dyspnea and palpitation. The chest X-ray showed an enlarged bilateral hilar shadow and diffusely increased bronchovascular markings in both lung fields. The chest CT showed diffuse nodular infiltrations with mild septal thickening and combined patchy ground glass opacity in both lungs, and conglomerated mediastinal and bilateral hilar lymphadenopathy. A diagnosis of LIP was made from the tissue pathology taken by a thoracoscopic lung biopsy. The patient showed clinical and radiographic improvement after 3 months of treatment with prednisolone. We report a case of LIP presenting as diffuse nodular interstitial infiltrations with multiple mediastinal and bilateral hilar lymphadenopathy.

A Case of IgG4-Related Disease Presenting as Massive Pleural Effusion and Thrombophlebitis

  • Immunoglobulin (Ig) G4-related disease is a recently recognized systemic fibroinflammatory condition characterized by a lymphoplasmacytic infiltrate rich in IgG4-positive plasma cells with elevated circulating levels of IgG4. The disease can either be localized to one or two organs, or present as diffuse multi-organ disease. Furthermore, lesions in different organs can present simultaneously or metachronously. In the pulmonary manefestations, lesions associated with IgG4-related disease have been described in the lung parenchyma, airways and pleura, as well as the mediastinum. We report a case of IgG4-related disease presenting as massive pleural effusion and thrombophlebitis.

Effects of Antioxidant and Blood Flow Improvement of Grape Leaf Extract and Resveratrol from Vitis romaneti (남항(Vitis romaneti) 유래 포도잎 추출물과 Resveratrol의 항산화 및 혈행개선 효과)

  • The purpose of this study is to investigate the effects of 80% ethanol grape leaf extract (VGLE) and resveratrol (VGLR) from Vitis romaneti on antioxidant of red blood cells (RBC) of rat and efficacy of blood flow improvement in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. RBC oxidative hemolysis and plasma lipid peroxidation induced by the aqueous peroxyl radical generator 2,2'-azobis(2-amidinopropane) dihydrochloride were significantly suppressed by VGLE or VGLR in a dose-dependent manner. The $FeCl_3$ treatment seriously damaged the carotid artery: the walls of the artery and blood flow. However, VGLE or VGLR administration has ameliorated the blood flow and suppressed thrombus in blood vessels. These results suggest that VGLE or VGLR ameliorates the oxidative stress of RBC and thrombosis against blood vessel damage.

$^{99m}Tc-HMPAO$ Labelled WBC Scan in Experimental Abscess by Labelling Autologous Leukocytes With In-House-Synthesized HMPAO (국내 합성한 HMPAO 이용 $^{99m}Tc$ 표지 백혈구를 이용한 실험적 농양 스캔)

  • With HMPAO we have synthesized in our laboratory, we labelled $^{99m}Tc$ to canine leukocytes. Experimental abscess made by subcutaneous injection with Staphylococcus aureus was imaged with these $^{99m}Tc$ labelled leukocytes. Labelling efficiency of HMPAO with $^{99m}Tc$ was $66.2%{\pm}14.6%$ (N=9). Labelling efficiency of leukocytes with $^{99m}Tc-HMPAO$ was $54%{\pm}7.7%$ (N=7). Cell bound radioactivity in $^{99m}Tc-HMPAO$ labelled leukocytes was around 80% when these cells were incubated in plasma in vitro at $37^{\circ}C$ for 5 hours. In vivo cell bound activity was over 80% at 24 hours after injection. One day and four days after inoculation, uptake at the inflammatory focus was found with $^{99m}Tc$ labelled leukocytes. Uptake showed up in 4 hour image, and the uptake at the lesion was most prominent in 24 hour image. These findings show that in-house-synthesized HMPAO could be used for labelling leukocytes with $^{99m}Tc$, and that s$^{99m}Tc-HMPAO-labelled$ leukocytes were so stable and viable that inflammatory focus could be visualized with these $^{99m}Tc-labelled$ leukocytes.

Pathophysiology of olive flounder Paralichthys olivaceus suffering from emaciation (여윔증 넙치, Paralichthys olivaceus의 증상에 대한 병태생리학적 고찰)

  • This study was aimed to investigate the pathophysiological changes of olive flounder, Paralichthys olivaceus suffering from emaciation. A plasma osmolality was higher in the emaciated and control flounders than that of normal teleost, suggesting osmoregulatory failure in both of them. Also, the control in the same stock with emaciated flounder seem to be classified into a primary degree of emaciation. According to microscopic observations, the inflammatory responses were observed in the submucosal layer of anterior intestine, although the some of mucosal intestinal epithelium still remained. It was suggested that the pathological changes of the anterior part give rise to malabsorption of nutrients through the mucosa. In the posterior intestine and rectum, the mucosal epithelium were almostly sloughed off and severe inflammatory responses were observed in the submucosa. Immunoreaction for NKCC was not detected in the mucosal epithelial cells in intestine because of sloughing of epithelium. These changes would lead to functional disorder in the intestine, such as malabsorption of nutrients and osmoregulatory failure. Also important is to investigate the recovery phase.

Fermentation of Citrus unshiu Marc. and Functional Characteristics of the Fermented Products (감귤의 발효와 발효산물의 기능적 특성)

  • Functional characteristics of citrus products fermented with lactic acid bacterium and yeast were investigated. Flavonoid composition of fermented citrus extracts increased significantly compared to control, leading to increases of naringenin and hesperetin concentrations. All citrus extracts showed anti-apoptotic effects in HepG2 cells regardless of fermentation, with citrus-fermented products showing greater anti-apoptotic effect and intracellular Reactive Oxygen Species content reduction compared to native citrus extracts. Male Sprague-Dawley rats were orally dosed with native or fermented citrus extracts. Singnificantly higher body weight reductions were observed in higher fermented citrus-dosed (100 mg/kg body weight) group compared to the other groups. Plasma total cholesterol level was slightly, but not significantly, reduced. Fatty liver formation induced by high-fat diet was significantly suppressed in rats administered with fermented citrus extracts. Results suggest fermented citrus extracts have potent anti-apoptotic and anti-oxidative activities in vitro, and inhibitory activity against fatty liver formation by high-fat diet in vivo.

Evaluation for Biocompatibility of Gentamicin-collagen Sponge on the Experimental Animal Wound Model (실험동물 창상 모델에서 겐타마이신 함유 콜라겐 스폰지의 생체적합성 평가)

  • The objective of this study was to compare the biocompatibility for local irritation and bioavailability of $Gentacol^{TM}$ and $Collatamp^{TM}$, after single intramuscular loading in rats. Sixty-six male Sprague-Dawley rats were divided into 4 groups; (1) any test materials were not applied into the quadriceps muscles (control group, N = 6), (2) Gentamicin was injected into the quadriceps muscles (Gentamicin group, N = 6), (3) Collatamp was applied into the quadriceps muscles (Collatamp group, N = 27), and (4) Gentacol was applied into the quadriceps muscles (Gentacol group, N = 27). The concentration of gentamicin in muscles was gradually decreased with time after loaded in the both $Gentacol^{TM}$ and $Collatamp^{TM}$ loaded regions. No detectable gentamicin was observed in the plasma of $Gentacol^{TM}$ and $Collatamp^{TM}$ loaded rats. Histologically, moderate to severe inflammatory cell infiltrations including PMN, lymphoid cells and macrophages were observed with slight to moderate edematous changes of muscle fibers after intramuscular injection of gentamicin. However, these histopathological changes of gentamicin injection induced local irritations were dramatically decreases after intramuscular loading of $Collatamp^{TM}$ and $Gentacol^{TM}$. These results suggest $Gentacol^{TM}$ may show favorable local bioavailability and induce only slight local irritations as comparable as $Collatamp^{TM}$ without systemic exposures in the condition of this experiment.
